1Hepatic echinococcosis:Clinical and therapeutic aspects显示文摘Echinococcosis or hydatid disease(HD)is a zoonosis caused by the larval stages of taeniid cestodes belonging to the genus Echinococcus.Hepatic echinococcosis is a life-threatening disease,mainly differentiated into alveolar and cystic forms,associated with Echinoccus multilocularis(E.multilocularis)and Echinococcus granulosus(E.granulosus)infection,respectively.Cystic echinococcosis(CE)has a worldwide distribution,while hepatic alveolar echinococcosis(AE)is endemic in the Northern hemisphere,including North America and several Asian and European countries,like France,Germany and Austria.E.granulosus young cysts are spherical,unilocular vesicles,consisting of an internal germinal layer and an outer acellular layer.Cyst expan-sion is associated with a host immune reaction and the subsequent development of a fibrous layer,called the pericyst;old cysts typically present internal septations and daughter cysts.E.multilocularis has a tumorlike,infiltrative behavior,which is responsible for tissue destruction and finally for liver failure.The liver is the main site of HD involvement,for both alveolar and cystic hydatidosis.HD is usually asymptomatic for a long period of time,because cyst growth is commonly slow;the most frequent symptoms are fatigue and abdominal pain.Patients may also present jaundice,hepatomegaly or anaphylaxis,due to cyst leakage or rupture.HD diagnosis is usually accomplished with the combined use of ultrasonography and immunodiagnosis;furthermore,the improvement of surgical techniques,the introduction of minimally invasive treatments[such as puncture,aspiration,injection,re-aspiration(PAIR)]and more effective drugs(such as benzoimidazoles)have deeply changed life expectancy and quality of life of patients with HD.The aim of this article is to provide an up-todate review of biological,diagnostic,clinical and therapeutic aspects of hepatic echinococcosis.Giuseppe Nunnari Marilia R Pinzone Salvatore Gruttadauria Benedetto M Celesia Giordano Madeddu Giulia Malaguarnera Piero Pavone Alessandro Cappellani Bruno Cacopardo 2012World Journal of Gastroenterology2012,18,13:72
2Long-term outcome and prognostic factors of patients with hilar cholangiocarcinoma显示文摘AIM: To evaluate the long-term outcome and prognostic factors of patients with hilar cholangiocarinoma. METHODS: Ninety-six consecutive patients underwent treatment for malignant hilar bile duct tumors during 1995–2005. Of the 96 patients, 20 were initially treated with surgery (n = 2 R0 / n = 18 R1). In non-operated patients, data analysis was performed retrospectively. RESULTS: Among the 96 patients, 76 were treated with endoscopic transpapillary (ERC, n = 45) and/or percutaneous transhepatic biliary drainage (PTBD, n = 31). The mean survival time of these 76 patients undergoing palliative endoscopic and/or percutaneous drainage was 359 ± 296 d. The mean survival time of patients with initial bilirubin levels > 10 mg/dL was significantly lower (P < 0.001) than patients with bilirubin levels < 10 mg/dL. The mean survival time of patients with Bismuth stage Ⅱ (n = 8), Ⅲ (n = 28) and Ⅳ (n = 40) was 496 ± 300 d, 441 ± 385 d and 274 ± 218 d, respectively. Thus, patients with advanced Bismuth stage showed a reduced mean survival time, but the difference was not significant. The type of biliary drainage had no significant beneficial effect on the mean survival time (ERC vs PTBD, P = 0.806). CONCLUSION: Initial bilirubin level is a significant prognostic factor for survival of patients. In contrast, age, tumor stage according to the Bismuth-Corlette classification, and types of intervention are not significant prognostic parameters for survival. 3TRAIL-induced apoptosis of hepatocellular carcinoma cells is augmented by targeted therapies显示文摘AIM:To analyze the effect of chemotherapeutic drugs and specific kinase inhibitors,in combination with the death receptor ligand tumor necrosis factor-related apoptosis inducing ligand(TRAIL),on overcoming TRAIL resistance in hepatocellular carcinoma(HCC)and to study the efficacy of agonistic TRAIL antibodies,as well as the commitment of antiapoptotic BCL-2 proteins, in TRAIL-induced apoptosis. METHODS:Surface expression of TRAIL receptors (TRAIL-R1-4)and expression levels of the antiapoptotic BCL-2 proteins MCL-1 and BCL-xL were analyzed by flow cytometry and Western blotting,respectively. Knock-down of MCL-1 and BCL-xL was performed by transfecting specific small interfering RNAs.HCC cellswere treated with kinase inhibitors and chemotherapeutic drugs.Apoptosis induction and cell viability were analyzed via flow cytometry and 3-(4,5-Dimethyl-thiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. RESULTS:TRAIL-R1 and-R2 were profoundly expressed on the HCC cell lines Huh7 and Hep-G2. However,treatment of Huh7 and Hep-G2 with TRAIL and agonistic antibodies only induced minor apoptosis rates.Apoptosis resistance towards TRAIL could be considerably reduced by adding the chemotherapeutic drugs 5-fluorouracil and doxorubicin as well as the kinase inhibitors LY294002[inhibition of phosphoinositol- 3-kinase(PI3K)],AG1478(epidermal growth factor receptor kinase),PD98059(MEK1),rapamycin(mam- malian target of rapamycin)and the multi-kinase inhibitor Sorafenib.Furthermore,the antiapoptotic BCL-2 proteins MCL-1 and BCL-xL play a major role in TRAIL resistance:knock-down by RNA interference increased TRAIL-induced apoptosis of HCC cells.Additionally, knock-down of MCL-1 and BCL-xL led to a significant sensitization of HCC cells towards inhibition of both c-Jun N-terminal kinase and PI3K.CONCLUSION:Our data identify the blockage of survival kinases,combination with chemotherapeutic drugs and targeting of antiapoptotic BCL-2 proteins as promising ways to overcome TRAIL resistance in HCC.Bruno Christian Koehler Toni Urbanik Binje Vick Regina Johanna Boger Steffen Heeger Peter R Galle Marcus Schuchmann Henning Schulze-Bergkamen 2009World Journal of Gastroenterology2009,15,47:9
4A longitudinal study of SARS-CoV-2-infected patients reveals a high correlation between neutralizing antibodies and COVID-19 severity显示文摘Understanding the immune responses elicited by SARS-CoV-2 infection is critical in terms of protection against reinfection and,thus,for public health policy and vaccine development for COVID-19.In this study,using either live SARS-CoV-2 particles or retroviruses pseudotyped with the SARS-CoV-2 S viral surface protein(Spike),we studied the neutralizing antibody(nAb)response in serum samples from a cohort of 140 SARS-CoV-2 qPCR-confirmed infections,including patients with mild symptoms and also more severe forms,including those that required intensive care.We show that nAb titers correlated strongly with disease severity and with anti-spike IgG levels.Indeed,patients from intensive care units exhibited high nAb titers;conversely,patients with milder disease symptoms had heterogeneous nAb titers,and asymptomatic or exclusive outpatient-care patients had no or low nAbs.We found that nAb activity in SARS-CoV-2-infected patients displayed a relatively rapid decline after recovery compared to individuals infected with other coronaviruses.Moreover,we found an absence of cross-neutralization between endemic coronaviruses and SARS-CoV-2,indicating that previous infection by human coronaviruses may not generate protective nAbs against SARS-CoV-2.Finally,we found that the D614G mutation in the spike protein,which has recently been identified as the current major variant in Europe,does not allow neutralization escape.Altogether,our results contribute to our understanding of the immune correlates of SARS-CoV-2-induced disease,and rapid evaluation of the role of the humoral response in the pathogenesis of SARS-CoV-2 is warranted.Vincent Legros Solène Denolly Manon Vogrig Bertrand Boson Eglantine Siret Josselin Rigaill Sylvie Pillet Florence Grattard Sylvie Gonzalo Paul Verhoeven Omran Allatif Philippe Berthelot Carole Pélissier Guillaume Thiery Elisabeth Botelho-Nevers Guillaume Millet Jérôme Morel Stéphane Paul Thierry Walzer François-Loïc Cosset Thomas Bourlet Bruno Pozzetto 2021Cellular & Molecular Immunology2021,18,2:3

8The contribution of microorganisms and metazoans to mineral nutrition in bromeliads显示文摘Aims One critical challenge for plants is to maintain an adequate nutrient supply under fluctuating environmental conditions.This is particularly true for epiphytic species that have limited or no access to the pedosphere and often live in harsh climates.Bromeliads have evolved key innovations such as epiphytism,water-absorbing leaf trichomes,tank habit and Crassulacean acid metabolism(CAM)photosynthesis that enable them to survive under various environmental conditions.Bromeliads encompass diverse ecological types that live on different substrates(they can be terrestrial,epilithic or epiphytic)and vary in their ability to retain water(they can be tank-forming or tankless)and photosynthetic pathway(i.e.C3 or CAM).In this review,we outline the nutritional modes and specializations that enable bromeliads to thrive in a wide range of nutrient-poor(mostly nitrogen-depleted)environments.Important FindingsBromeliads have evolved a great diversity of morphologies and functional adaptations leading to the existence of numerous nutritional modes.Focusing on species that have absorptive foliar trichomes,we review evidence that bromeliads have evolved multi-faceted nutritional strategies to respond to fluctuations in the supply of natural nitrogen(N).These plants have developed mutualistic associations with many different and functionally diverse terrestrial and aquatic microorganisms and metazoans that contribute substantially to their mineral nutrition and,thus,their fitness and survival.Bacterial and fungal microbiota-assisted N provisioning,protocarnivory,digestive mutualisms and myrmecotrophic pathways are the main strategies used by bromeliads to acquire nitrogen.The combination of different nutritional pathways in bromeliads represents an important adaptation enabling them to exploit nutrient-poor habitats.Nonetheless,as has been shown for several other vascular plants,multiple partners are involved in nutrient acquisition indicating that there have been convergent adaptations to nutrient scarcity.Finally,we point out some gaps in the current knowledge of bromeliad nutrition that offer fascinating research opportunities.Céline Leroy Jean-François Carrias Régis Céréghino Bruno Corbara 2016Journal of Plant Ecology2016,9,3:2

12Competition between fusion-evaporation and multifragmentation in central collisions in ^(58)Ni+^(48)Ca at 25A MeV显示文摘The experimental data concerning the58Ni+48Ca reaction at Elab(Ni)=25A MeV,collected by using the CHIMERA 4π device,have been analyzed in order to investigate the competition among different reaction mechanisms for central collisions in the Fermi energy domain.As a main criterion for centrality selection we have chosen the flow angle(flow) method,making an event-by-event analysis that considers the shape of events,as it is determined by the eigenvectors of the experimental kinetic-energy tensor.For the selected central events(flow >60°) some global variables,good to characterize the pattern of central collisions have been constructed.The main features of the reaction products were explored by using different constraints on some of the relevant observables,like mass and velocity distributions and their correlations.Much emphasis was devoted,for central collisions,to the competition between fusion-evaporation processes with subsequent identification of a heavy residue and a possible multifragmentation mechanism of a well defined(if any) transient nuclear system.Dynamical evolution of the system and pre-equilibrium emission were taken into account by simulating the reactions in the framework of transport theories.Different approaches have been envisaged(dynamical stochastic BNV calculations + sequential SIMON code,QMD,CoMD,etc.).Preliminary comparison of the experimental data with BNV calculations shows reasonable agreement with the assumption of sequential multifragmentation emission in the mass region of IMFs close to the heavy residues.Possible deviations from sequential processes were found for those IMFs in the region of masses intermediate between the mass of heavy residues and the mass of light IMFs.Further simulations are in progress.The experimental analysis will be enriched also by information obtained inspecting the IMF-IMF correlation function,in order to elucidated the nature of space-time decay property of the emitting source associated with events having the largest IMF multiplicity.FRANCALANZA L ABBONDANNO U AMORINI F BARLINI S BINI M BOUGAULT R BRUNO M CARDELLA G CASINI G AGOSTINO M D' De FILIPPO V De SANCTIS J GERACI E GIUSSANI A GRAMEGNA F GUIOT B KRAVCHUK V La GUIDARA E LANZALONE G Le NEINDRE N MAIOLINO C MARINI P MORELLI L OLMI A PAGANO A PAPA M PIANTELLI S PIRRONE S POLITI G POGGI G PORTO F RUSSOTTO P RIZZO F VANNINI G VANNUCCI L 2013Nuclear Science and Techniques2013,24,5:2
