| 1 | Promotion of the articular cartilage proteoglycan degradation by T-2 toxin and selenium protective effect显示文摘Objective: To identify the relationship between T-2 toxin and Kashin-Beck disease (KBD),the effects of T-2 toxin on aggrecan metabolism in human chondrocytes and cartilage were investigated in vitro. Methods: Chondrocytes were isolated from human articular cartilage and cultured in vitro. Hyaluronic acid (HA),soluble CD44 (sCD44),IL-1β and TNF-α levels in super-natants were measured by enzyme-linked immunosorbent assay (ELISA). CD44 content in chondrocyte membrane was deter-mined by flow cytometry (FCM). CD44,hyaluronic acid synthetase-2 (HAS-2) and aggrecanases mRNA levels in chondrocytes were determined using reverse transcription polymerase chain reaction (RT-PCR). Immunocytochemical method was used to investigate expressions of BC-13,3-B-3(-) and 2-B-6 epitopes in the cartilage reconstructed in vitro. Results: T-2 toxin inhibited CD44,HAS-2,and aggrecan mRNA expressions,but promoted aggrecanase-2 mRNA expression. Meanwhile,CD44 expression was found to be the lowest in the chondrocytes cultured with T-2 toxin and the highest in control plus selenium group. In addition,ELISA results indicated that there were higher sCD44,IL-1β and TNF-α levels in T-2 toxin group. Similarly,higher HA levels were also observed in T-2 toxin group using radioimmunoprecipitation assay (RIPA). Furthermore,using monoclonal antibodies BC-13,3-B-3 and 2-B-6,strong positive immunostaining was found in the reconstructed cartilage cultured with T-2 toxin,whereas no positive staining or very weak staining was observed in the cartilage cultured without T-2 toxin. Selenium could partly inhibit the effects of T-2 toxin above. Conclusion: T-2 toxin could inhibit aggrecan synthesis,promote aggrecanases and pro-inflammatory cytokines production,and consequently induce aggrecan degradation in chondrocytes. This paper aims to examine the mechanisms contributing to coal burst occurrence, with an emphasis on the energy release concept. In this study, a numerical modelling study has been conducted to evaluate the roles and contributions of difference energy components. | 5 | Nicotine-induced upregulation of antioxidant protein Prx 1 in oral squamous cell carcinoma显示文摘Nicotine is a source of exogenous oxidative stress, which is associated with the pathogenesis of numerous diseases including oral squamous cell carcinoma (OSCC), whereas an antioxidant protein, peroxiredoxin 1 (Prx 1), plays an important role in the modulation of this condition. This study was to investigate the association between Prx 1 and tobacco-induced oxidative stress. The expression of Prx 1 and GST in OSCC Tca8113 cells, which were pre-treated with nicotine, was determined. In the present study, MTT assay, reactive oxygen species (ROS) assay, RT-PCR and Western blot analyses, respectively, were conducted to assess cell viability, ROS level, and expression level of Prx 1 and GST in nicotine-treated Tca8113 cells. Nuclear factor kappa B (NF- B) expression was detected by immuno-fluorescence. Our results showed the growth of Tca8113 cells was increased in a dose-dependent manner when cells were treated with nicotine at concentrations from 0.1 to 10 mol/L, but the proliferation of the cells decreased at 100 mol/L. ROS levels increased in all groups treated with nicotine at concentrations of 0.1, 1, 10, or 100 mol/L for 24h. Prx 1 and GST mRNA and protein expression were up-regulated in cells treated with nicotine for the same time at different concentrations or at the same concentration for different times (P<0.05). NF-B was translocated from cytoplasm to nucleus, the expression of NF- B was increased in nucleus. | 6 | Impaired granulocyte-macrophage colony-stimulating factor bioactivity accelerates surgical recurrence in ileal crohn's disease显示文摘AIM To examine the relationship between elevated granulocyte-macrophage colony-stimulating factor(GMCSF) auto-antibodies(Ab) level and time to surgical recurrence after initial surgery for Crohn's disease(CD). METHODS We reviewed 412 charts from a clinical database at tertiary academic hospital. Patients included in the study had ileal or ileocolonic CD and surgical resection of small bowel or ileocecal region for management of disease. Serum samples were analyzed for serological assays including GM-CSF cytokine, GM-CSF Ab, ASCA Ig G and Ig A, and genetic markers including SNPs rs2066843, rs2066844, rs2066845, rs2076756 and rs2066847 in NOD2, rs2241880 in ATG16 L1, and rs13361189 in IRGM. Cox proportional-hazards models were used to assess the predictors of surgical recurrence.RESULTS Ninety six percent of patients underwent initial ileocecal resection(ICR) or ileal resection(IR) and subsequently 40% of patients required a second ICR/IR for CD. GMCSF Ab level was elevated at a median of 3.81 mcg/mL. Factors predicting faster time to a second surgery included elevated GM-CSF Ab [hazard ratio(HR) 3.52, 95%CI: 1.45-8.53, P = 0.005] and elevated GM-CSF cytokine(HR = 2.48, 95%CI: 1.31-4.70, P = 0.005). Factors predicting longer duration between first and second surgery included use of Immunomodulators(HR = 0.49, 95%CI: 0.31-0.77, P = 0.002), the interaction effect of low GM-CSF Ab levels and smoking(HR = 0.60, 95%CI: 0.45-0.81, P = 0.001) and the interaction effect of low GM-CSF cytokine levels and ATG16 L1(HR = 0.65, 95%CI: 0.49-0.88, P = 0.006).CONCLUSION GM-CSF bioavailability plays a critical role in maintaining intestinal homeostasis. | 9 | Bioreduction of vanadium(V) in groundwater by autohydrogentrophic bacteria:Mechanisms and microorganisms显示文摘As one of the transition metals, vanadium(V)(V(V)) in trace amounts represents an essential element for normal cell growth, but becomes toxic when its concentration is above 1 mg/L. V(V) can alter cellular differentiation, gene expression, and other biochemical and metabolic phenomena. A feasible method to detoxify V(V) is to reduce it to V(IV), which precipitates and can be readily removed from the water. The bioreduction of V(V) in a contaminated groundwater was investigated using autohydrogentrophic bacteria and hydrogen gas as the electron donor. Compared with the previous organic donors,H2 shows the advantages as an ideal electron donor, including nontoxicity and less production of excess biomass. V(V) was 95.5% removed by biochemical reduction when autohydrogentrophic bacteria and hydrogen were both present, and the reduced V(IV)precipitated, leading to total-V removal. Reduction kinetics could be described by a first-order model and were sensitive to p H and temperature, with the optimum ranges of p H 7.5–8.0 and 35–40°C, respectively. Phylogenetic analysis by clone library showed that the dominant species in the experiments with V(V) bioreduction belonged to theβ-Proteobacteria. Previously known V(V)-reducing species were absent, suggesting that V(V)reduction was carried out by novel species. | 11 | A semi-quantitative coal burst risk classification system显示文摘Safety is the highest priority in the mining industry as underground mining in particular poses high safety risks to its workers. In underground coal mines, coal bursts are one of the most catastrophic hazards, which involves sudden and violent dynamic coal mass failure with rapid ejection of the broken material into the mine workings. Despite decades of research, the contributing mechanisms of coal bursts are still not completely understood. Hence, it remains challenging to forecast coal bursts and quantify their likelihood of occurrence. However, a range of geological and geotechnical factors are associated with coal bursts and can increase the coal burst proneness. This paper introduces a semi-quantitative coal burst risk classification system, namely, BurstRisk. Based on back-analysis of case histories from Australia, China and the United States, BurstRisk classifies the coal burst risk into three categories:low, medium and high risk. In addition, it allows mining engineers to modify the weighting of the selected factors based on specific conditions. The risk classification charts introduced are for both longwall retreat and development sections of long-wall mining operations. | 12 | Modeling Climate Change Impacts on the US Agricultural Exports显示文摘Climate change is expected to have substantial effects on agricultural productivity worldwide. However, these impacts will differ across commodities, locations and time periods. As a result, landowners will see changes in relative returns that are likely to induce modifications in production practices and land allocation. In addition, regional variations in impacts can alter relative competitiveness across countries and lead to adjustments in international trade patterns. Thus in climate change impact studies it is likely useful to account for worldwide productivity effects. In this study, we investigate the implications of considering rest of world climate impacts on projections of the US agricultural exports. We chose to focus on the US because it is one of the largest agricultural exporters. To conduct our analyses, we consider four alternative climate scenarios, both with and without rest of world climate change impacts. Our results show that considering/ignoring rest of world climate impacts causes signifi cant changes in the US production and exports projections.
