| 5 | Effect of Rare Earth Sc Addition on Microstructure and Mechanical Properties of an Al-Cu-Mg-Ag Alloy显示文摘Al-5.3Cu-0.8Mg-0.6Ag alloys containing 0, 0.1, 0.3 and 0.5 (mass. %) Sc were prepared by ingot metallurgy and thermomechanical treatment. The effect of Sc addition on the precipitation and microstructure of the alloys has been investigated using mechanical testing, optical microscope, scanning electron microscopy (SEM) and transmission electron microscopy (TEM). It has been shown that trace Sc element refines the grains of the casting alloys and the average grain size decreases from 85 μm to 30 μm. Increasing Sc content from 0.1 to 0.3% accelerates the aging hardening process, increases the maximum hardness and the tensile strength of the extruded alloys from room temperature to 300 ℃. | 7 | Effect of trace tungsten on the microstructure of TiAl alloys containing Nb and B显示文摘New TiAl alloys, containing 45 at.% Al, 7 at.% Nb, x at.% W, and 0.15 at.% B (x = 0, 0.2, 0.4, and 0.7) were prepared by arc melting and drop casting consequently. Using optical microscopy, scanning electron microscopy (SEM), and electron superprobe technologies, the effects of tungsten on the microstructural evolution of the TiAl alloys, including the colony size and lamellar spacing, were analyzed. It was found that cellular structures and dendrites were formed in the as-cast TiAl alloys, and heavy metals, such as niobium and tungsten, tend to segregate strongly at the interface of the cellu- lar structures and dendrites. | 8 | Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. | 9 | Novel SFTSV Phylogeny Reveals New Reassortment Events and Migration Routes显示文摘Severe fever with thrombocytopenia syndrome virus(SFTSV),the causative agent of a febrile human disease,was first identified from central and eastern provinces in China,and later in Japan and South Korea.Hubei Province is one of the major SFTS epidemic areas in the central part of China.This study reported the isolation of 11 new SFTSV strains from patients in Hubei Province collected in 2017.Extensive phylogenetic analyses were conducted based on the complete coding sequences of SFTSV segments including the new strains.It was suggested that five different SFTSV genotypes were circulating in Hubei,and 15 reassortment patterns and migration pathways correlated with each genotype were identified,which was more than previously recognized.Hubei Province was more involved in the evolutionary events of SFTSV than that previously thought in which the evolutionary events of SFTSV were reported to be independent from those in other epidemic regions.Further divergence of SFTSV strains was suggested by pairwise comparison of SFTSV sequences from each genotype and sequence identity normalized to representative strain in genotype C1.Subsequently,amino acid variations specific for genotype(s),strain(s),or cluster(s)were inspected,which may be related to differential biological activity of SFTSV strains/genotypes.In conclusion,we analyzed the current status of SFTSV phylogeny in Hubei Province and discussed the possible events correlated to SFTSV evolution.It provided an in-depth insight into SFTSV evolution,raising concerns for the use of proper SFTSV strains in future studies. | Xiaoli Wu Mingyue Li Yanfang Zhang Boyun Liang Junming Shi Yaohui Fang Zhengyuan Su Mengmeng Li Wenjing Zhang Ling Xu Jun Wang Qiaoli Wu Shuang Tang Hualin Wang Tao Zhang Cheng Peng Xin Zheng Fei Deng Shu Shen | 2021 | Virologica Sinica2021,36,2: | 2 |
