3Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently,the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however,the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown.In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo,accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment.The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death,indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together,our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis,which will hopefully provide a prospective compound for bladder cancer treatment.Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao 2021Acta Pharmaceutica Sinica B2021,11,12:23

5新疆准噶尔盆地北缘夺勒布勒津剖面的地层学及环境意义显示文摘新疆准噶尔盆地北缘夺勒布勒津台地西侧的索索泉组与上覆哈拉玛盖组为连续堆积地层,在该地测制的地层剖面,完善了索索泉组,并可与铁尔斯哈巴合剖面连接,组成时间跨度为25~13.5Ma的基本上连续的夺勒布勒津-铁尔斯哈巴合综合剖面.索索泉组厚约150m,是以风成沉积为主体的地层.该组堆积始于地磁极性时C6Cr顶,止于C5Cr,地质时间约为24~17Ma,跨越了最晚渐新世至早中新世晚期近7Ma的时间.夺-铁剖面岩性变化明显、含多层哺乳动物化石且古地磁记录较完整,是目前研究中国北方该时段中陆相哺乳动物年代和气候环境变化的理想剖面.剖面也为了解现代亚洲风系和风成沉积的形成过程、亚洲中纬度内陆地区的干旱化过程以及这个过程与全球环境变化、青藏高原隆升、副特提斯海的退缩等因素的关系,提供了新的认识和证据.叶捷 吴文裕 倪喜军 ShunDong BI 孙继敏 Jin MENG 2012中国科学:地球科学2012,42,10:12
6Atmospheric Deposition-Carried Pb, Zn, and Cd from a Zinc Smelter and Their Effect on Soil Microorganisms显示文摘Dust emissions from smelters, as a major contributor to heavy metal contamination in soils, could severely influence soil quality. Downwind surface soils within 1.5 km of a zinc smelter, which was active for 10 years but ceased in 2000, in Magu Town, Guizhou Province, China were selected to examine Pb, Zn, and Cd concentrations and their fractionation along a distance gradient from a zinc smelter, and to study the possible e?ects of Pb, Zn, and Cd accumulation on soil microorganisms by comparing with a reference soil located at a downwind distance of 10 km from the zinc smelter. Soils within 1.5 km of the zinc smelter accumulated high levels of heavy metals Zn (508 mg kg-1), Pb (95.6 mg kg-1), and Cd (5.98 mg kg-1) with low ratios of Zn/Cd (59.1-115) and Pb/Cd (12.4-23.4). Composite pollution indices (CPIs) of surface soils (2.52-15.2) were 3 to 13 times higher than the reference soils. In metal accumulated soils, exchangeable plus carbonate-bound fractions accounted for more than 10% of the total Zn, Pb, and Cd. The saturation degree of metals (SDM) in soils within 1.5 km of the smelter (averaging 1.25) was six times higher than that of the reference soils (0.209). A smaller soil microbial biomass was found more frequently in metal accumulated soils (85.1-438 μg C g-1) than in reference soils (497 μg C g-1), and a negative correlation (P < 0.01) of soil microbial biomass carbon to organic carbon ratio (Cmic/Corg) with SDM was observed. Microbial consumption of carbon sources was more rapid in contaminated soils than in reference soils, and a shift in the substrate utilization pattern was apparent and was negatively correlated with SDM (R = -0.773, P < 0.01). 11Pore Size Distribution of a Tight Sandstone Reservoir and its Effect on Micro Pore-throat Structure: A Case Study of the Chang 7 Member of the Xin’anbian Block, Ordos Basin, China显示文摘Pore distribution and micro pore-throat structure characteristics are significant for tight oil reservoir evaluation, but their relationship remains unclear. This paper selects the tight sandstone reservoir of the Chang 7 member of the Xin’anbian Block in the Ordos Basin as the research object and analyzes the pore size distribution and micro pore-throat structure using field emission scanning electron microscopy(FE-SEM), high-pressure mercury injection(HPMI), highpressure mercury injection, and nuclear magnetic resonance(NMR) analyses. The study finds that:(1) Based on the pore size distribution, the tight sandstone reservoir is characterized by three main patterns with different peak amplitudes. The former peak corresponds to the nanopore scale, and the latter peak corresponds to the micropore scale. Then, the tight sandstone reservoir is categorized into three types: type 1 reservoir contains more nanopores with a nanopore-to-micropore volume ratio of 82:18;type 2 reservoir has a nanopore-to-micropore volume ratio of 47:53;and type 3 reservoir contains more micropores with a nanopore-to-micropore volume ratio of 35:65.(2) Affected by the pore size distribution, the throat radius distributions of different reservoir types are notably offset. The type 1 reservoir throat radius distribution curve is weakly unimodal, with a relatively dispersed distribution and peak ranging from 0.01 μm to 0.025 μm. The type 2 reservoir’s throat radius distribution curve is single-peaked with a wide distribution range and peak from 0.1 μm to 0.25 μm. The type 3 reservoir’s throat radius distribution curve is single-peaked with a relatively narrow distribution and peak from 0.1 μm to 0.25 μm. With increasing micropore volume, pore-throat structure characteristics gradually improve.(3) The correlation between micropore permeability and porosity exceeds that of nanopores, indicating that the development of micropores notably influences the seepage capacity. In the type 1 reservoir, only the mean radius and effective porosity have suitable correlations with the nanopore and micropore porosities. The pore-throat structure parameters of the type 2 and 3 reservoirs have reasonable correlations with the nanopore and micropore porosities, indicating that the development of these types of reservoirs is affected by the pore size distribution. This study is of great significance for evaluating lacustrine tight sandstone reservoirs in China. 12Magnetic resonance imaging for chronic pain:diagnosis,manipulation,and biomarkers显示文摘Pain is a multidimensional subjective experience with biological,psychological,and social factors.Whereas acute pain can be a warning signal for the body to avoid excessive injury,long-term and ongoing pain may be developed as chronic pain.There are more than 100 million people in China living with chronic pain,which has raised a huge socioeconomic burden.Studying the mechanisms of pain and developing effective analgesia approaches are important for basic and clinical research.Recently,with the development of brain imaging and data analytical approaches,the neural mechanisms of chronic pain have been widely studied.In the first part of this review,we briefly introduced the magnetic resonance imaging and conventional analytical approaches for brain imaging data.Then,we reviewed brain alterations caused by several chronic pain disorders,including localized and widespread primary pain,primary headaches and orofacial pain,musculoskeletal pain,and neuropathic pain,and present meta-analytical results to show brain regions associated with the pathophysiology of chronic pain.Next,we reviewed brain changes induced by pain interventions,such as pharmacotherapy,neuromodulation,and acupuncture.Lastly,we reviewed emerging studies that combined advanced machine learning and neuroimaging techniques to identify diagnostic,prognostic,and predictive biomarkers in chronic pain patients.Yiheng Tu Jin Cao Yanzhi Bi Li Hu 2021Science China(Life Sciences)2021,64,6:5
13Effect of nitrogen limitation on antioxidant qualities is highly associated with genotypes of lettuce(Lactuca sativa L.)显示文摘Antioxidants are highly beneficial to human health, and their accumulation in lettuce, one of the most popular leafy vegetables, depends on both genetic and environmental factors. Nitrogen(N) availability plays an essential role in regulating antioxidant accumulation, but the influence of genotype × N interactions on the antioxidant qualities of lettuce is poorly understood. Therefore, the present study investigated the variation of growth and antioxidant qualities of 20 lettuce(Lactuca sativa L.) genotypes(10 green lettuce genotypes and 10 red lettuce genotypes) under limited N(low N, LN) conditions and standard N(high N, HN) conditions. For all 20 genotypes, LN conditions reduced shoot(i.e., leaf) growth, but increased plant concentrations of vitamin C,glutathione, and phenolic compounds, with the exception of carotenoids, compared with HN conditions. Because of reduced biomass under LN conditions, not all lettuce genotypes exhibited increased antioxidant yields or total antioxidant capacity yield. The variation in antioxidant quality was primarily genetically determined. Generally, the green lettuce genotypes exhibited more pronounced increases in antioxidant yields and total antioxidant capacity yield than the red lettuce genotypes under LN conditions. These results suggest that even though LN conditions generally tend to improve the antioxidant qualities of lettuce, the extent of this effect is highly dependent on genotype. 15Rapid and sensitive determination of fatty acids in edible oil by liquid chromatography-electrospray ionization tandem mass spectrometry显示文摘A sensitive and robust on-line LC/MS method was developed for quantitative determination of linoleic acid,docosahexaenoic acid and docosanoic acid from edible oil samples.The oil samples were dissolved in chloroform-isopropyl alcohol(20:80,v:v)solution and the three fatty acids were separated by HPLC with a C4 column using 10 mmol/L ammonium acetate-isopropyl alcohol-acetonitrile(20:40:40,v:v:v)mobile phase in isocratic elution.Electrospray ionization mass spectrometry with the selected ion recording monitoring was used to detect and quantify the fatty acid.The calibration curves were linear in the range of 10.00–5000 pg/mL for linoleic acid and docosanoic acid,and 1.000–500.0 pg/mL for docosahexaenoic acid.The limit of detection was 2.0 pg/mL for linoleic acid,3.0 pg/mL for docosanoic acid,and 0.20 pg/mL for docosahexaenoic acid.The results showed that the method described in this paper could be utilized for rapid determination of three fatty acids at picogram levels in edible oils.BI Xin JIN YiBao LI ShangFu GAO Dan JIANG YuYang LIU HongXia 2014Science China Chemistry2014,57,3:4

18Interconnected carbon nanocapsules with high N/S co-doping as stable and high-capacity potassium-ion battery anode显示文摘Carbonaceous materials have drawn much attention in potassium-ion batteries (PIBs) due to their low price and superior physicochemical properties. However, the application of carbonaceous materials in PIB anodes is hindered by sluggish kinetics and large volume expansion. Herein, N/S co-doped carbon nanocapsule (NSCN) is constructed for superior K+ storage. The NSCN possesses 3D nanocapsule framework with abundant meso/macropores, which guarantees structural robustness and accelerates ions/electrons transportation. The high-level N/S co-doping in carbon matrix not only generates ample defects and active sites for K+ adsorption, but also expands interlayer distance for facile K+ intercalation/deintercalation. As a result, the NSCN electrode delivers a high reversible capacity (408 mAh g^(−1) at 0.05 A g^(−1)), outstanding rate capability (149 mAh g^(−1) at 5 A g^(−1)) and favorable cycle stability (150m Ah g^(−1) at 2 A g^(−1) after 2000 cycles). Ex situ TEM, Raman and XPS measurements demonstrate the excellent stability and reversibility of NSCN electrode during potassiation/depotassiation process. 19Famitinib versus placebo in the treatment of refractory metastatic colorectal cancer:a multicenter,randomized,double-blinded,placebo-controlled,phaseⅡclinical trial显示文摘Background: Metastatic colorectal cancer(mCRC) patients with progressive disease after all available standard therapies need new medication for further treatment. Famitinib is a small-molecule multikinase inhibitor, with promising anticancer activities. This multicenter, randomized, double-blinded, placebo-controlled, phase II clinical trial was designed to evaluate the safety and efficacy of famitinib in mCRC.Methods: Famitinib or placebo was administered orally once daily. The primary endpoint was progression-free survival(PFS). Secondary endpoints included objective response rate(ORR), disease control rate(DCR), overall survival(OS), quality-of-life(QoL), and safety.Results: Between July 18,2012 and Jan 22,2014, a total of 167 patients were screened, and 154 patients were randomized in a 2:1 ratio to receive either famitinib(n = 99) or placebo(n = 55). The median PFS was 2.8 and 1.5 months in the famitinib and placebo groups(hazard ratio = 0.60,95% confidence interval = 0.41-0.86, P = 0.004). The DCR was 59.8% and 31.4%(P = 0.002) and the ORR was 2.2% and 0.0%(P = 0.540) in the famitinib and placebo groups,respectively. The most frequent grade 3-4 adverse events were hypertension(11.1%), hand-foot syndrome(10.1%),thrombocytopenia(10.1%) and neutropenia(9.1%). Serious adverse events occurred in 11(11.1%) patients in the famitinib group and 5(9.1%) in the placebo group(P = 0.788). 20LIN28 coordinately promotes nucleolar/ribosomal functions and represses the 2C-like transcriptional program in pluripotent stem cells显示文摘LIN28 is an RNA binding protein with important roles in early embryo development,stem cell differentiation/re-programming,tumorigenesis and metabolism.Previous studies have focused mainly on its role in the cytosol where it interacts with Let-7 microRNA precursors or mRNAs,and few have addressed LIN28's role within the nucleus.Here,we show that LIN28 displays dynamic temporal and spatial expression during murine embryo development.Maternal LIN28 expression drops upon exit from the 2-cell stage,and zygotic LIN28 protein is induced at the forming nucleolus during 4-cell to blas-tocyst stage development,to become dominantly expressed in the cytosol after implantation.In cultured pluripotent stem cells(PSCs),loss of LIN28 led to nucleolar stress and activation of a 2-cell/4-cell-like transcriptional program characterized by the expression of endogenous retrovirus genes.Mechanistically,LIN28 binds to small nucleolar RNAs and rRNA to maintain nucleolar integrity,and its loss leads to nucleolar phase separation defects,ribosomal stress and activation of P53 which in turn binds to and activates 2C transcription factor Dux.LIN28 also resides in a complex containing the nucleolar factor Nucleolin(NCL)and the transcrip-tional repressor TRIM28,and LIN28 loss leads to reduced occupancy of the NCL/TRIM28 complex on the Dux and rDNA loci,and thus de-repressed Dux and reduced rRNA expression.Lin28 knockout cells with nucleolar stress are more likely to assume a slowly cycling,translationally inert and anabolically inactive state,which is a part of previously unappreciated 2C-like transcriptional program.These findings elucidate novel roles for nucleolar LIN28 in PSCs,and a new mechanism linking 2C program and nucleolar functions in PSCs and early embryo development.Zhen Sun Hua Yu Jing Zhao Tianyu Tan Hongru Pan Yuqing Zhu Lang Chen Cheng Zhang Li Zhang Anhua Lei Yuyan Xu Xianju Bi Xin Huang Bo Gao Longfei Wang Cristina Correia Ming Chen Qiming Sun Yu Feng Li Shen Hao Wu Jianlong Wang Xiaohua Shen George Q.Daley Hu Li Jin Zhang 2022Protein & Cell2022,13,7:4
