1Genome-wide Dissection of Co-selected UV-B Responsive Pathways in the UV-B Adaptation of Qingke显示文摘Qingke (Tibetan hulless barley) has long been cultivated and exposed to long-term and strong UV-B radiation on the Tibetan Plateau, which renders it an ideal species for elucidating novel UV-B responsive mechanisms in plants. Here we report a comprehensive metabolite profiling and metabolite-based genome-wide association study (mGWAS) using 196 diverse qingke and barley accessions. Our results demonstrated both constitutive and induced accumulation, and common genetic regulation, of metabolites from different branches of the phenylpropanoid pathway that are involved in UV-B protection. A total of 90 significant mGWAS loci for these metabolites were identified in barley-qingke differentiation regions, and a number of high-level metabolite trait alleles were found to be significantly enriched in qingke, suggesting co-selection of various phenylpropanoids. Upon dissecting the entire phenylpropanoid pathway, we identified some key determinants controlling natural variation of phenylpropanoid content, including three novel proteins, a flavone C-pentosyltransferase, a tyramine hydroxycinnamoyl acyltransferase, and a MYB transcription factor. 2Residents’ perspectives and responses to environmental degradation in the upper Dadu River, eastern Tibetan Plateau显示文摘Environmental degeneration in the Tibetan Plateau attracts worldwide attention, whereas case studies on how the residents understand and respond to environmental degeneration are scarce. Using a Participatory Rural Appraisal method, this paper investigates how the people in different regions in the upper Dadu River understand and respond to environmental degeneration, based on comparative field surveys in three villages, in which Danzamu village is chosen from villages in the valley region, Kerma village from mountainside region, Rico village from the mountain and plateau region. The results show that: (1) although awakened to environmental degeneration, the residents in different regions have different responses. As agricultural labors have been transferred to the secondary and tertiary industries, population pressure in Danzamu and Kerma villages is mitigated. Residents in Danzamu village actively respond to natural disasters and forest degradation, as their livelihoods never rely on forests and rangelands again. Whereas the residents in Kerma village negatively respond to natural disasters, forest and meadow degradation and the ruin of wildlife resources, as their livelihoods still rely on stockbreeding. Labors in Rico village are hard to transfer to the secondary and tertiary industries, so they have to raise more livestock to make a living. Active measures are just taken to avoid livestock loss, not to avoid forest and meadow degradation and the ruin of wildlife resources. So the most fragile region is the mountain and plateau region and mountainside region, not the valley region. (2) Livelihood strategy is the key factor affecting the residents to respond to population pressure and environmental degeneration. So the framework of sustainable livelihood strategy should be used to explain and intervene in issues of population pressure and environmental degradation in ecotones. (3) Transferring agricultural labors to the secondary and tertiary industries were favorable to improving people’s livelihood. It is necessary to reduce the education fees to speed up the pace of labors transferring in the mountainside region. 3Effect of fluoride on the corrosion behavior of nanostructured Ti-24Nb-4Zr-8Sn alloy in acidulated artificial saliva显示文摘The surface of titanium dental implants is highly susceptible to aggressive fluoride ions in the oral environment. Nanotechnology has proven an effective approach to improve the stability and corrosion resistance of titanium by applying a passive film. In this study, we investigated the effects of fluoride on the corrosion behavior of nanostructured(NS) Ti-24 Nb-4 Zr-8 Sn(Ti2448) alloy in acidulated artificial saliva(AAS)at 37 ℃, and then conducted comparisons with its coarse grained(CG) counterpart. Electrochemical techniques, such as potentiodynamic polarization and electrochemical impedance spectroscopy(EIS), as well as surface analysis including X-ray photoelectron spectroscopy(XPS) with argon ion sputtering, and scanning electronic microscopy(SEM) were employed to evaluate the effects of fluoride on sensitivity to pitting and the tolerance of Ti2448 to fluoride in AAS solution. The results demonstrate that corrosion current density increased with F-concentration. In all respects, the NS Ti2448 alloy presented corrosion resistance superior to that of its coarse grained(CG) counterpart at low F-concentrations(<0.1%).Furthermore, a high content of F-(1%) was shown to promote the active dissolution of both alloys by increasing the rate of corrosion. Following immersion in the fluoridated AAS solution for 60 days, a tissuefriendly compound, Ca_3(PO_4)_2, was detected on the surface of the NS when F-= 0.01% and Na_2 TiF_6 was identified as the main component in the corrosion products of the CG as well as NS Ti2448 alloys when F-= 1%. High concentrations of F-produced pitting corrosion on the CG alloy, whereas NS Ti2448 alloy presented general corrosion in the form of lamellar separation under the same conditions. Its applications have been extended from engineering education and technology demonstration into various other fields, such as communication, remote sensing, navigation and scientific experiments just to name a few. 5Electrochemical behavior of open-cellular structured Ti-6Al-4V alloy fabricated by electron beam melting in simulated physiological fluid:the significance of pore characteristics显示文摘The cellular structured titanium alloys have attracted significant attention for implants because of their lower Young’s modulus,which is comparable to human bone and has the capability of providing space for bone tissue in-growth.However,there is a gap in the knowledge in regard to the relationship between the pore characteristics and the electrochemical performance of open-cellular structured titanium alloys.In this study,we elucidate the influence of pore characteristics on the electrochemical performance of open-cellular structured Ti-6Al-4V alloys produced by electron beam melting(EBM).Intriguingly,the passive film formed on cellular structured Ti-6Al-4V alloy with a larger pore size was more stable and protective,and the corrosion performance was superior compared to the samples with a smaller pore size in phosphate buffered saline(PBS),mainly because of relatively smaller exposed surface area and unlimited flow of electrolyte.However,in acidic PBS containing fluoride ions,the pore characteristics did not play an important role in the corrosion resistance.It was considered that the protective film breaks down such that the corrosion performance of cellular structured alloys was comparable to each other in this harsh environment.Xin Gai Rui Liu Yun Bai Shujun Li Yang Yang Shenru Wang Jianguo Zhang Wentao Hou Yulin Hao Xing Zhang Rui Yang R.D.K.Misra 2022Journal of Materials Science & Technology2022,,2:1

7MCA-TFP Model:A Short-Term Traffic Flow Prediction Model Based on Multi-characteristic Analysis显示文摘With the urbanization,urban transportation has become a key factor restricting the development of a city.In a big city,it is important to improve the efficiency of urban transportation.The key to realize short-term traffic flow prediction is to learn its complex spatial correlation,temporal correlation and randomness of traffic flow.In this paper,the convolution neural network(CNN)is proposed to deal with spatial correlation among different regions,considering that the large urban areas leads to a relatively deep Network layer.First three gated recurrent unit(GRU)were used to deal with recent time dependence,daily period dependence and weekly period dependence.Considering that each historical period data to forecast the influence degree of the time period is different,three attention mechanism was taken into GRU.Second,a twolayer full connection network was applied to deal with the randomness of short-term flow combined with additional information such as weather data.Besides,the prediction model was established by combining these three modules.Furthermore,in order to verify the influence of spatial correlation on prediction model,an urban functional area identification model was introduced to identify different functional regions.Finally,the proposed model was validated based on the history of New York City taxi order data and reptiles for weather data.The experimental results show that the prediction precision of our model is obviously superior to the mainstream of the existing prediction methods.Xiujuan Xu Lu Xu Yulin Bai Zhenzhen Xu Xiaowei Zhao Yu Liu 2020国际计算机前沿大会会议论文集2020,,2:0
8The First Crested Duck Genome Reveals Clues to Genetic Compensation and Crest Cushion Formation显示文摘The Chinese crested(CC)duck is a unique indigenous waterfowl breed,which has a crest cushion that affects its survival rate.Therefore,the CC duck is an ideal model to investigate the genetic compensation response to maintain genetic stability.In the present study,we first generated a chromosome-level genome of CC ducks.Comparative genomics revealed that genes related to tissue repair,immune function,and tumors were under strong positive selection,indicating that these adaptive changes might enhance cancer resistance and immune response to maintain the genetic stability of CC ducks.We also assembled a Chinese spot-billed(Csp-b)duck genome,and detected the structural variations(SVs)in the genome assemblies of three ducks(i.e.,CC duck,Csp-b duck,and Peking duck).Functional analysis revealed that several SVs were related to the immune system of CC ducks,further strongly suggesting that genetic compensation in the anti-tumor and immune systems supports the survival of CC ducks.Moreover,we confirmed that the CC duck originated from the mallard ducks.Finally,we revealed the physiological and genetic basis of crest traits and identified a causative mutation in TAS2R40 that leads to crest formation.Overall,the findings of this study provide new insights into the role of genetic compensation in adaptive evolution.Guobin Chang Xiaoya Yuan Qixin Guo Hao Bai Xiaofang Cao Meng Liu Zhixiu Wang Bichun Li Shasha Wang Yong Jiang Zhiquan Wang Yang Zhang Qi Xu Qianqian Song Rui Pan Lingling Qiu Tiantian Gu Xinsheng Wu Yulin Bi Zhengfeng Cao Yu Zhang Yang Chen Hong Li Jianfeng Liu Wangcheng Dai Guohong Chen 2023Genomics, Proteomics & Bioinformatics2023,21,3:0

10Research on Influence of Guilin Topography on Haze显示文摘This paper analyzed space-time distribution features of haze at different topographical representative stations using haze days observation data of 50 years in different meteorological observatories of Guilin to study the topography influence on haze. Different topographical representative stations were classified into four types of mountain,hill,river and city. Mountain stations were classified into two types,surrounded by mountains and faced with mountains on two sides. Under parallel comparison method,change features of wind direction,wind speed,temperature and haze days at different meteorological observatories were analyzed to investigate the topography influence on haze with pollution increase factors excluded. The study found that the city station had the biggest haze occurrence rate,followed by the mountain station surrounded by the mountains,and the river station had the smallest haze occurrence rate. 11An ultra-fast and highly efficient multiple proteases digestion strategy using graphene-oxide-based immobilized protease reagents显示文摘Highly efficient and rapid proteolytic digestion of proteins into peptides is a crucial step in shotgun-based proteome-analysis strategy.Tandem digestion by two or more proteases is demonstrated to be helpful for increasing digestion efficiency and decreasing missed cleavages,which results in more peptides that are compatible with mass-spectrometry analysis.Compared to conventional solution digestion,immobilized protease digestion has the obvious advantages of short digestion time,no self-proteolysis,and reusability.We proposed a multiple-immobilized proteases-digestion strategy that combines the advantages of the two digestion strategies mentioned above.Graphene-oxide(GO)-based immobilized trypsin and endoproteinase Glu-C were prepared by covalently attaching them onto the GO surface.The prepared GO-trypsin and GO-Glu-C were successfully applied in standard protein digestion and multiple immobilized proteases digestion of total proteins of Thermoanaerobacter tengcongensis.Compared to 12-hour solution digestion using trypsin or Glu-C,14%and 7%improvement were obtained,respectively,in the sequence coverage of BSA by one-minute digestion using GO-trypsin and GO-Glu-C.Multiple immobilized-proteases digestion of the total proteins of Thermoanaerobacter tengcongensis showed 24.3%and 48.7%enhancement in the numbers of identified proteins than was obtained using GO-trypsin or GO-Glu-C alone.The ultra-fast and highly efficient digestion can be contributed to the high loading capacity of protease on GO,which leads to fewer missed cleavages and more complete digestion.As a result,improved protein identification and sequence coverage can be expected.BAI HaiHong PAN YiTing REN XiaoJun HAO FeiRan DENG ShanShan FAN Chao YAN Hui SHEN BingQuan MA Lin TIAN Fang PENG Bo DENG YuLin QIN WeiJie QIAN XiaoHong 2014Science China Chemistry2014,57,5:0

13Proteomic and phosphoproteomic analysis reveal threonine deficiency increases hepatic lipid deposition in Pekin ducks via reducing STAT phosphorylation显示文摘Dietary threonine(Thr)deficiency enhances triglyceride(TG)deposition in the liver of Pekin ducks,which injures hepatic function and impairs growth performance.However,the underlying molecular mechanisms remain unclear.In the present study,we investigated the effects of dietary Thr deficiency on the expressions of proteins and phosphoproteins in liver of Pekin ducks,to identify the underlying molecular changes.A total of 300 one-day-old ducklings were divided into 3 groups with 10 replicates of 10 birds.All ducks were fed corn-wheat-peanut meal diets containing 0.46%,0.71%,and 0.96%Thr,respectively,from 1 to 21 days of age.Growth performance,serum parameters,hepatic TG content,and expression of genes involved in lipid metabolism of Pekin ducks were determined.A Thr deficiency group(Thr-D,0.46%Thr)and a Thr sufficiency group(Thr-S,0.71%Thr)were selected for subsequent proteomic and phosphoproteomic analysis.The results showed that Thr-D reduced the growth performance(P<0.001),and increased the plasma concentrations of cholesterol,high-density lipoprotein cholesterol,low-density lipoprotein cholesterol,and hepatic TG(P<0.05).Thr-D increased gene expression related to fatty acid and TG synthesis(P<0.05).A total of 176 proteins and 259 phosphosites(containing 198 phosphoproteins)were observed to be differentially expressed as a result of Thr-D.The upregulated proteins were enriched in the pathway related to amino acid metabolism,peroxisome.The down-regulated proteins were enriched in linolenic and arachidonic acid metabolism,and the Janus kinase-signal transducer and activator of transcription(JAK-STAT)signaling pathway.The upregulated phos-phoproteins were enriched in the pathways related to fatty acid biosynthesis,fructose and mannose metabolism,and glycolysis/gluconeogenesis.Thr-D reduced the phosphorylation of STAT1 at S729 and STAT3 at S728,and expression of STAT5B.In contrast,Thr-D increased non-receptor tyrosine-protein kinase(TYK2)expression and STAT1 phosphorylation at S649.Taken together,dietary Thr-D increased hepatic TG accumulation by upregulating the expression of genes and proteins,and phosphoproteins related to fatty acid and triglyceride synthesis.Furthermore,these processes might be regulated by the JAK-STAT signaling pathway,especially the phosphorylation of STAT1 and STAT3.Yong Jiang Zhong Zhuang Wenqia Jia Zhiguo Wen Ming Xie Hao Bai Yulin Bi Zhixiu Wang Guobin Chang Shuisheng Hou Guohong Chen 2023Animal Nutrition2023,,2:0
14Ex utero intrapartum therapy in infants with congenital diaphragmatic hernia:a propensity score matching analysis显示文摘Objective Previous studies have shown that ex utero intrapartum therapy(EXIT)is safe and feasible for newborns with congenital diaphragmatic hernia(CDH).This study reports our experience with EXIT in fetuses with CDH in an attempt to explore the efficacy of EXIT on the survival rate of this population.Methods A retrospective analysis of the clinical data of 116 children with CDH was conducted.The children were assigned to EXIT and non-EXIT groups.Propensity score matching(PSM)toward clinical data was performed,and the clinical characteristics and outcomes were compared.Taking survival at discharge as the main outcome,logistic regression analysis was carried out to explore the efficacy of EXIT on survival.Results During the study period,30 of 116 children received EXIT.After PSM,the survival rates of the EXIT group and the non-EXIT group were 82.76%(24/29)and 48.28%(14/29),respectively(p=0.006).EXIT(OR=0.083,95%CI=0.013to 0.525,p=0.008),liver herniation(OR=16.955,95%CI=2.342 to 122.767,p=0.005),and gestational age at diagnosis(OR=0.662,95%CI=0.497 to 0.881,p=0.005)were independent mortality-related risk factors of all children with CDH.Ninety-nine of 116 children underwent surgery.After PSM,the postoperative survival rates of the EXIT group and non-EXIT group were 84.6%(22/26)and 76.9%(20/26),respectively(p=0.754).Liver herniation(OR=10.451,95%CI=1.641 to 66.544,p=0.013)and gestational age at diagnosis(OR=0.736,95%CI=0.577 to 0.938,p=0.013)were independent mortality-related risk factors of children after surgery.Conclusion EXIT can be performed safely for selected prenatally diagnosed CDH neonates with potentially better survival and does not cause more maternal complications compared with traditional cesarean section.Yunlong Zhao Ying Wang Chao Liu Yulin Jiang Yandong Wei Hua Meng Shan Jian Xiting Zhu Lijian Pei Xiaochen Bai Feng Feng Yan Lv Xiya Zhou Qingwei Qi Jingna Li Lishuang Ma 2022World Journal of Pediatric Surgery2022,5,4:0

17Population structure and genome‑wide evolutionary signatures reveal putative climate‑driven habitat change and local adaptation in the large yellow croaker显示文摘The large yellow croaker(Larimichthys crocea)is one of the most economically valuable marine fsh in China and is a notable species in ecological studies owing to a serious collapse of wild germplasm in the past few decades.The stock division and species distribution,which have important implications for ecological protection,germplasm recovery,and fshery resource management,have been debated since the 1960s.However,it is still uncertain even how many stocks exist in this species.To address this,we evaluated the fne-scale genetic structure of large yellow croaker populations distributed along the eastern and southern Chinese coastline based on 7.64 million SNP markers.Compared with the widely accepted stock boundaries proposed in the 1960s,our results revealed that a climate-driven habitat change probably occurred between the Naozhou(Nanhai)Stock and the Ming-Yuedong(Mindong)Stock.The boundary between these two stocks might have shifted northwards from the Pearl River Estuary to the northern area of the Taiwan Strait,accompanied by highly asymmetric introgression.In addition,we found divergent landscapes of natural selection between the stocks inhabiting northern and southern areas.The northern population exhibited highly agminated signatures of strong natural selection in genes related to developmental processes,whereas moderate and interspersed selective signatures were detected in many immune-related genes in the southern populations.These fndings establish the stock status and genome-wide evolutionary landscapes of large yellow croaker,providing a basis for conservation,fsheries management and further evolutionary biology studies.Baohua Chen Yulin Bai Jiaying Wang Qiaozhen Ke Zhixiong Zhou Tao Zhou Ying Pan Renxie Wu Xiongfei Wu Weiqiang Zheng Peng Xu 2023Marine Life Science & Technology2023,5,2:0
