1Role of the renin angiotensin system in diabetic nephropathy显示文摘Diabetic nephropathy has been the cause of lot of mor bidity and mortality in the diabetic population.The renin angiotensin system(RAS) is considered to be involved in most of the pathological processes that result in diabetic nephropathy.This system has var ious subsystems which contribute to the disease pat ho logy.One of these involves angiotensin Ⅱ(Ang Ⅱ) which shows in c reased activity during diabetic nephropathy.This causes hypertrophy of various renal cells and has a pres sor effect on arteriolar smooth muscle resulting in increas ed vascular pressure.Ang Ⅱ also induces inflamm ation,apoptosis,cell growth,migration and differentiation.Monocyte chemoattractant protein-1 production responsible for renal f ibrosis is also regulated by RAS.Polymorphism of angiotensin converting enzym e(ACE) and Angiotensinogen has been shown to have effects on RAS.Available treatment modalities have proven effective in controlling the progression of nephropathy.Various drugs(based on antagonism of RAS) are currently in the market and others are still under trial.Amongst the approved drugs,ACE inhibitors and ang iot ensin receptor blockers(ARBs) are widely used in clinical practice.ARBs are shown to be superior to ACE inhibitors in terms of reducing proteinuria but the combined role of ARBs with ACE inhibitors in diabetic nephropathy is under debate.Tanuj Chawla Deepika Sharma Archana Singh 2010World Journal of Diabetes2010,1,5:34

3Changing role of histopathology in the diagnosis and management of hepatocellular carcinoma显示文摘Hepatocellular carcinoma (HCC) is one of the most common and fatal cancer in the world. HCC frequently presents with advanced disease, has a high recurrence rate and limited treatment options, which leads to very poor prognosis. This warrants urgent improvement in the diagnosis and treatment. Liver biopsy plays very important role in the diagnosis and prognosis of HCC, but with technical advancements and progression in the field of imaging, clinical guidelines have restricted the role of biopsy to very limited situations. Biopsy also has its own problems of needle tract seeding of tumor, small risk of complications, technical and sampling errors along with interpretative errors. Despite this, tissue analysis is often required because imaging is not always specific, limited expertise and lack of advanced imaging in many centers and limitations of imaging in the diagnosis of small, mixed and other variant forms of HCC. In addition, biopsy confirmation is often required for clinical trials of new drugs and targeted therapies. Tissue biomarkers along with certain morphological features, phenotypes and immune-phenotypes that serve as important prognostic and outcome predictors and as decisive factors for therapy decisions, add to the continuing role of histopathology. Advancements in cancer biology and development of molecular classification of HCC with clinic pathological correlation, lead to discovery of HCC phenotypic surrogates of prognostic and therapeutically significant molecular signatures. Thus tissue characteristics and morphology based correlates of molecular subtypes provide invaluable information for management and prognosis. 6Relationship of Periodontal Status and Dental Caries Status with Oral Health Knowledge, Attitude and Behavior among Professional Students in India显示文摘Aim To find the relationship of periodontal status and dental caries status with oral health knowledge, attitude, behavior, among professional students in India. Methodology In a cross sectional study, a total of 825 students (males: 577, females: 248) from six professions were surveyed using a self administered structured questionnaire including 41 multiple choice questions and the WHO Oral Health Assessment Form (1997). The data was analyzed using the SPSS version 13.0 to perform the Student's t-test, ANOVA test, Scheffe's test and Chi-square test, linear regression analysis. Results The mean percentage scores of the students for knowledge were 53.25 ± 15.05; for attitude 74.97 ±20.48; and for behavior 59.09 ± 18.77. The percentage of students with calculus score was found to be significantly high (43.8%). The percentage of professional students with DMFT>4 was 14.1% and the percentage of students with decayed teeth was 46.2%. The regression analysis showed that the oral health behavior of the students was dependent on the attitude (P<0.001), but showed no significant linear relation with the knowledge. Also, that the mean DMFT score was dependent on the oral health behavior (P<0.05), but showed no significant relationship with the knowledge and attitude of the students. The periodontal status was independent on the knowledge, but showed a significant relationship with attitude and behavior of the students. 7Pharmacological cyclin dependent kinase inhibitors: Implications for colorectal cancer显示文摘Colorectal cancer accounts for a significant proportion of cancer deaths worldwide. The need to develop more chemotherapeutic agents to combat this disease is critical. Cyclin dependent kinases(CDKs), along with its binding partner cyclins, serve to control the growth of cells through the cell cycle. A new class of drugs, termed CDK inhibitors, has been studied in preclinical and now clinical trials. These inhibitors are believed to act as an anti-cancer drug by blocking CDKs to block the uncontrolled cellular proliferation that is hallmark of cancers like colorectal cancer. 11Screening of Antioxidant Potential of Selected Barks of Indian Medicinal Plants by Multiple in vitro Assays显示文摘Objective To evaluate the antioxidant potential in herbal extract barks of five therapeutically important medicinal plants native to India, i.e. Crataeva nurvala Buch.-Ham., Buchanania lanzan Spreng., Aegle marmelos Corr., Dalbergia sissoo Roxb. ex DC., and Cedrela toona Roxb. Methods Standardized aqueous alcoholic extracts from the selected barks having different target radicals, such as superoxide radical, nitric oxide, ABTS radical, and peroxidative decomposition of phospholipids, were prepared and screened by multiple in vitro assays. These extracts were also tested for total phenolic and tannin content and correlated with antioxidant capacity. Results Total phenolic and tannin contents were found to be the highest in C. nurvala (195 GAE mg/g and 218.3 mg/g CE). SOD mimetic activity was found to be the highest in Crataeva nurvula, although all barks showed activity more than 100 units/mg extract. Lipid peroxidation inhibitory potential was found to be the highest in Crataeva nurvala (83.4% inhibition of MDA formation/10 μg extract), and also showed a comparatively high NO quenching capacity (45.5% per 10 μg extract). The highest NO quenching potential was found in Aegle marmelos (47.3% per 10 μg extract). Cedrela toona showed the lowest LPO inhibitory potential and NO quenching capacity (50.5% and 30.5%, respectively). Buchanania lanzan, a medicinal plant extensively used for inflammatory disorders and Dalbergia sissoo also showed 72.5% and 69.1% LPO inhibitory potential/10 μg extract. Trolox equivalent antioxidant capacity ranged from 0.24 to 0.39 mmol/L TEAC/mg extract, indicating that all the barks tested had ABTS+· radical quenching capacity. 12Leukemic stromal hematopoietic microenvironment negatively regulates the normal hematopoiesis in mouse model of leukemia显示文摘Background and Objective: Leukemic microenvironment has a major role in the progression of leukemia. Leukemic cells can induce reversible changes in microenvironmental components, especially the stromal function which results in improved growth conditions for maintaining the malignant leukemic cells. This study aimed to investigate the survival advantage of leukemic cells over normal hematopoietic cells in stromal microenvironment in long term. Methods: The mice were injected intraperitoneally with N-N’ ethylnitrosourea (ENU) to induce leukemia; the mice received injection of normal saline were used as control. At 180 days after ENU induction, the mice were killed and the bone marrows were cultured for 19 days. Colony-forming assays were used to analyze the formation of various cell colonies. The expression of Sca-1, CD146, VEGFR2, CD95, pStat3, pStat5, and Bcl-xL in marrow cells were detected by flow cytometry. Results: Long-term leukemic bone marrow culture showed abnormal elongated stromal fibroblasts with almost absence of normal hematopoietic cells. Adherent cell colonies were increased, but CFU-F and other hematopoietic cell colonies were significantly decreased in leukemia group (P< 0.001). Primitive progenitor-specific Sca-1 receptor expression was decreased with subsequent increased expression of CD146 and VEGFR-2 in leukemic bone marrow cells. Decreased Fas antigen expression with increased intracellular pStat3, pStat5 and Bcl-xL proteins were observed in leukemic bone marrow cells. Conclusions: Stromal microenvironment shows altered morphology and decreased maturation in leukemia. Effective progenitor cells are decreased in leukemia with increased leukemia-specific cell population. 13葡萄糖调节蛋白前体GRP78和肿瘤排斥抗原GP96为仓鼠附睾头部精子特有显示文摘哺乳动物睾丸中的精子经过“附睾成熟”期,由静止状态转变成为运动状态。该过程中精子从附睾头部向附睾尾部移动,同时精子发生了一系列的形态、生理和生化改变,如蛋白组成和蛋白修饰的改变可能会影响精子获能的潜能。本实验使用基质辅助激光解吸/电离串联质谱(MALDI-MS/MS)法分析仓鼠睾丸头部和尾部精子的蛋白组学,成功发现了113个蛋白质点。对113个蛋白质点进一步对照比较发现30个蛋白质点(对应20个蛋白)的密度发生了显著改变,其中附睾尾部精子5个蛋白的密度增加,11个蛋白密度减少;此外,葡萄糖调节蛋白前体GRP78和肿瘤排斥抗原GP96为仓鼠附睾头部精子特有,而纤维蛋白原样蛋白1为附睾尾部精子所特有。几个蛋白密度增加可能与附睾成熟过程中精子代谢和ATP产生相关。一些蛋白如ERp57,GRP78,GP96,Hsp60,Hsp70和二氢硫辛酰胺S-乙酰转移酶的密度改变通过免疫印迹法得到验证。本研究首次报道了仓鼠精子的蛋白质组学研究,全面展示了仓鼠精子附睾成熟过程中的蛋白结构改变。Duvvuri Butchi Kameshwari Satish Bhande Curam Sreenivasacharlu Sundaram Venkatesh Kota Archana B. 17SYNTHESIS, CHARACTERIZATION AND ION TRANSPORT PROPERTIES OF HOT-PRESSED SOLID POLYMER ELECTROLYTES (1-x) PEO:x KI显示文摘Synthesis and ion transport properties of hot-pressed solid polymer electrolytes (SPEs), (1-x) PEO: x KI, where x is the content of KI in wt%, are reported. A hot-press technique has been used for the formation of the polymeric membranes in place of the usual solution cast method. The composition (80PEO:20KI) was identified as the highest conducting polymer electrolyte on the basis of compositional dependent conductivity studies of PEO:KI films. A conductivity enhancement of more than two orders of magnitude from that of the pure PEO was achieved. 18Transport of chemical signals in systemic acquired resistance显示文摘Systemic acquired resistance(SAR) is a form of broad-spectrum resistance induced in response to local infections that protects uninfected parts against subsequent secondary infections by related or unrelated pathogens. SAR signaling requires two parallel branches,one regulated by salicylic acid(SA),and the other by azelaic acid(AzA) and glycerol-3-phosphate(G3P). Az A and G3P function downstream of the free radicals nitric oxide(NO) and reactive oxygen species(ROS). During SAR,SA,Az A and G3P accumulate in the infected leaves,but only a small portion of these is transported to distal uninfected leaves. SA is preferentially transported via the apoplast,whereas phloem loading of Az A and G3P occurs via the symplast. The symplastic transport of Az A and G3P is regulated by gating of the plasmodesmata(PD).
