| 3 | Surgical stress and cytoskeletal changes in lens epithelial cells following manual and femtosecond laser-assisted capsulotomy显示文摘AIM:To study the effect of mechanical stress on the cytoskeleton in lens epithelial cells following conventional phacoemulsification surgery(CPS)and femtosecond laserassisted cataract surgery(FLACS).METHODS:The cytoskeleton of the epithelial cells of the anterior lens capsules(ALC)removed by CPS and FLACS was examined by immunohistochemistry.Expression of the intermediate filament,glial fibrillary acidic protein(GFAP),and glutamine synthetase(GS)immunoreactivity were detected.In order to map the actin network of cells,fluorescently labeled phalloidin was used.The samples were examined using confocal laser scanning microscopy.RESULTS:GFAP expression was visible in a larger number of the epithelial cells after CPS compared to FLACS.In CPS sample’s epithelial cells,GFAP immunoreactivity indicated robust morphological change.Regarding the actin filaments,the presence of tubular elements connecting epithelial cells,regular actin pattern and marked cortical network after CPS were found.Following FLACS,the actin cytoskeleton of the epithelial cells remained densely structured,and the tubular elements were undetectable,however,the above-mentioned regular actin pattern and the marked cortical network were visible.CONCLUSION:The conventional removal of the ALC induces more robust changes of the cytoskeleton of the lens epithelial cells. | ANDrea Krisztina Sükosd Krisztina Szabadfi Edina Szabó-Meleg Beáta Gáspár Pavel Stodulka Gyorgy SétálóJr Róbert Gábriel Miklós Nyitrai Zsolt Biró Hajnalka Abrahám | 2020 | International Journal of Ophthalmology(English edition)2020,13,6: | 2 |

| 11 | An inter-laboratory study of DNA-based identity,parentage and species testing in animal forensic genetics显示文摘The probative value of animal forensic genetic evidence relies on laboratory accuracy and reliability.Inter-laboratory comparisons allow laboratories to evaluate their performance on specific tests and analyses and to continue to monitor their output.The International Society for Animal Genetics(ISAG)administered animal forensic comparison tests(AFCTs)in 2016 and 2018 to assess the limitations and capabilities of laboratories offering forensic identification,parentage and species determination services.The AFCTs revealed that analyses of low DNA template concentrations(≤300 pg/μL)constitute a significant challenge that has prevented many laboratories from reporting correct identification and parentage results.Moreover,a lack of familiarity with species testing protocols,interpretation guidelines and representative databases prevented over a quarter of the participating laboratories from submitting correct species determination results.Several laboratories showed improvement in their genotyping accuracy over time.However,the use of forensically validated standards,such as a standard forensic short tandem repeat(STR)kit,preferably with an allelic ladder,and stricter guidelines for STR typing,may have prevented some common issues from occurring,such as genotyping inaccuracies,missing data,elevated stutter products and loading errors.The AFCTs underscore the importance of conducting routine forensic comparison tests to allow laboratories to compare results from each other.Laboratories should keep improving their scientific and technical capabilities and continuously evaluate their personnel’s proficiency in critical techniques such as low copy number(LCN)analysis and species testing.Although this is the first time that the ISAG has conducted comparison tests for forensic testing,findings from these AFCTs may serve as the foundation for continuous improvements of the overall quality of animal forensic genetic testing. | Sreetharan Kanthaswamy Torsten Brendel Luis Cancela Denise A.Andrade de Oliveira Bertram Brenig Carmen Cons Julian A.Crespi Markéta Dajbychová Andreas Feldl Tomohito Itoh Vincenzo Landi Amparo Martinez Malgorzata Natonek-Wisniewska Robert F.Oldt Anna Radko Oscar Ramírez Clementina Rodellar Manuel Ruiz-Girón David Schikorski María Elena Turba Guillermo Giovambatista | 2022 | Forensic Sciences Research2022,7,4: | 0 |
| 12 | The Loss of Heterozygosity of FHIT Gene in Sporadic Breast Cancer显示文摘function.FHIT is a potential tumor suppressor gene.Although the precise FHIT molecular mechanism of action is not well understood,evidences suggest that Fhit protein reduced levels are involved in mammary carcinogenesis.The aim of this study was to investigate if FHIT LOH could influence on sporadic breast cancer(BC)biological behavior,through its association with prognostic factors for sporadic BC.Tumor tissue and peripheral blood samples were analyzed using the microsatellite marker D3S1300.The findings were associated with clinicopathological parameters including overall survival.LOH was detected in 31.1%(52/167)of the informative BC’cases.Considering clinical and pathological characteristics we have found no significant association with FHIT LOH status.The mean follow-up time was 80 months.After the Cox regression analysis two parameters remained associated with BC’s risk of death:TNM stage III and IV-HR=3.74(95%CI,1.16-12.1)P=0.027 and disease relapse HR=3.14(CI 95%1.26-7.80)P=0.014.This study shows that FHIT LOH by itself is not a prognostic factor for sporadic BC.Further researches are required to elucidate the functional role of FHIT LOH concerning to BC. | Lisiane Silveira Zavalhia Andrea Pires Souto Damin Grasiela Agnes Aline Weber Taís Frederes Kramer Alcalde Laura Marinho Dorneles Guilherme Watte Adriana Vial Roehe | 2021 | Journal of Oncology Research2021,3,2: | 0 |
