1Safety,tolerability,and pharmacokinetics of VV116,an oral nucleoside analog against SARS-CoV-2,in Chinese healthy subjects显示文摘VV116(JT001)is an oral drug candidate of nucleoside analog against SARS-CoV-2.The purpose of the three phase I studies was to evaluate the safety,tolerability,and pharmacokinetics of single and multiple ascending oral doses of VV116 in healthy subjects,as well as the effect of food on the pharmacokinetics and safety of VV116.Three studies were launched sequentially:Study 1(single ascending-dose study,SAD),Study 2(multiple ascending-dose study,MAD),and Study 3(food-effect study,FE).A total of 86 healthy subjects were enrolled in the studies.VV116 tablets or placebo were administered per protocol requirements.Blood samples were collected at the scheduled time points for pharmacokinetic analysis.116-N1,the metabolite of VV116,was detected in plasma and calculated for the PK parameters.In SAD,AUC and Cmax increased in an approximately dose-proportional manner in the dose range of 25–800 mg.T_(1/2)was within 4.80–6.95 h.In MAD,the accumulation ratio for Cmax and AUC indicated a slight accumulation upon repeated dosing of VV116.In FE,the standard meal had no effect on Cmax and AUC of VV116.No serious adverse event occurred in the studies,and no subject withdrew from the studies due to adverse events.Thus,VV116 exhibited satisfactory safety and tolerability in healthy subjects,which supports the continued investigation of VV116 in patients with COVID-19.Hong-jie Qian Yu Wang Meng-qi Zhang Yuan-chao Xie Qing-qing Wu Li-yu Liang Ye Cao Hua-qing Duan Guang-hui Tian Juan Ma Zhuo-bing Zhang Ning Li Jing-ying Jia Jing Zhang Haji Akber Aisa Jing-shan Shen Chen Yu Hua-liang Jiang Wen-hong Zhang Zhen Wang Gang-yi Liu 2022Acta Pharmacologica Sinica2022,43,12:13
2Potato Responds to Salt Stress by Increased Activity of Antioxidant Enzymes显示文摘To understand the response of potato to salt stress, antioxidant enzyme activities and ion content were analyzed for a sensitive and a tolerant cultivar. Nodal cuttings of the tolerant cultivar, Kennebec, and the sensitive cultivar, Concord, were exposed to media without or with 30, 60, 90 or 120 mmol/L NaCl for 4 weeks. On exposure to NaCl, the length and fresh and dry weight of both shoots and roots of Concord showed greater decrease than those of Kennebec. The decrease in shoot growth was more severe than that of the root for both cultivars. The K+ content of shoots and roots of both cultivars was reduced in a dose-dependent manner by exposure to NaCl; the Na+ content increased. Activities of ascorbate peroxidase, catalase and glutathione reductase were increased in NaCl-exposed shoots of Kennebec; the corresponding activities in NaCl-exposed shoots of Concord were decreased. Roots of both cultivars showed similar changes in the activities of these enzymes on exposure to NaCl. These studies established that enzyme activities in Concord shoots are inversely related to the NaCl concentration, whereas those in Kennebec do not show a dose dependency, which is also the case for the roots of both cultivars. 3Effect of cytokine gene polymorphism on histological activity index, viral load and response to treatment in patients with chronic hepatitis C genotype 3显示文摘AIM: To investigate the association between cytokine gene polymorphism and disease status in chronic hepatitis C genotype 3 by liver biopsy, ALT, HCV RNA levels and response to treatment.METHODS: Patients with chronic hepatitis C genotype 3 were analyzed for single nucleotide polymorphisms of interleukin (IL)-10, IL-1 beta, interferon-gamma (IFN-γ),tumor necrosis factor-alpha (TNF-α) and transforming growth factor-beta (TGF-β) by polymerase chain reaction using sequence-specific oligonucleotide primers. Liver biopsies were assessed by modified histological activity index (HAI) scoring system using a scale of 0-18 for grading the necro-inflammatory activity and 0-6 for staging the fibrosis. HCV RNA levels were determined by bDNA assay. The patients were treated with interferon alpha and ribavirin for 6 mo. Sustained virological response was assessed 6 mo after the completion of the treatment.RESULTS: Out of the 40 patients analyzed, 26 were males. Mean age was 40.5±12.5 years (range 18-65 years). The frequencies of different dimorphic polymorphisms based on single nucleotide substitution were as follows: IL-10-1082 G/A 85%, A/A 12.5%, G/G 2.5%; IL-10-819 A/C 87.5%, C/C 10%, A/A 2.5%;IL-10-592 C/A 72.5%, C/C 27.5%; IL-1 C 90%, U 10%;IFN-874 T/A 50%, T/T 27.5%, A/A 22.5%; TNF-308A/G 95%, G/G 5%; TGF-10 T/C 52.5%, C/C 35%, T/T 12.5%. The mean grades of necro-inflammatoryactivity of different genotypes of IL-10 at promoter site -1082were A/A = 3.6, A/G = 5.0, and G/G = 10.0 and the difference was significant (P = 0.029). The difference in the stage of disease at a scale of 0-6 was A/A 0.8, A/G 2.3, and G/G 4.0 (P = 0.079). The difference in the HAI seemed to be related to the presence of allele -1082G.For IL-10 -819 genotypes, mean scores of fibrosis were A/A = 6.0, A/C = 2.2, and C/C = 1.0 (P = 0.020)though the inflammatory activity was not much different.No significant differences in HAI were noted among polymorphisms of other cytokines. Moreover, ALT and HCV RNA levels were not significantly different among different cytokine polymorphisms. There was a significant correlation of HAI and HCV RNA levels with the duration of disease. TGFβ -10 genotype CC patients had a better end of treatment response than those with other genotypes (P = 0.020). Sustained virological response to the treatment was not influenced by the cytokine polymorphism. No effect of other factors like viral load,degree of fibrosis, gender, steatosis, was observed on sustained virological response in this population infected with genotype 3.CONCLUSION: There is no significant correlation between cytokine polymorphisms and HAI except for the polymorphisms of anti-inflammatory cytokine IL-10,which may influence hepatic inflammatory activity and fibrosis in patients with chronic hepatitis C genotype 3.Sustained virological response in this genotype does not seem to be influenced by cytokine gene polymorphisms.Zaigham Abbas Tariq Moatter Akber Hussainy Wasim Jafri 2005World Journal of Gastroenterology2005,11,42:7

5Alkaloids of Nitraria sibirica Pall. decrease hypertension and albuminuria in angiotensin II-salt hypertension显示文摘In traditional Chinese medicine, Nitraria sibirica Pall.(Nitrariaceae) is used to treat hypertension. This study determined the effects of the total alkaloids of the leaves of Nitraria sibirica(NSTA) on blood pressure and albuminuria in mice treated with angiotensin II and a high-salt diet(ANG/HS). Adult mice were divided into three groups: control; infused with angiotensin II and fed a diet containing 4% NaCl(ANG/HS; and ANG/HS plus injection of NSTA(1 mg·kg-1·d-1, i.p.). After treatment of these regimens, daily water and food intake, kidney weight, blood pressure, urinary albumin excretion, renal concentrations of inflammatory markers, including soluble intercellular adhesion molecule-1(sICAM-1) and monocyte chemoattractant protein-1(MCP-1), and the expression of renal fibrosis markers were determined. Compared to the control group, the ANG/HS group had higher blood pressure and urinary albumin excretion. Treatment with NSTA in ANG/HS mice for three weeks significantly reduced blood pressure and urinary albumin excretion. ANG/HS treatment caused elevated levels of sICAM-1 and MCP-1, as well as increased fibrosis markers. Concurrent treatment with ANG/HS and NSTA attenuated the levels and expression of renal inflammatory and fibrosis markers. 7The mechanisms of melanogenesis inhibition by glabridin:molecular docking,PKA/MITF and MAPK/MITF pathways显示文摘Glabridin is the main ingredient of hydrophobic fraction in licorice extract and has been shown to have anti-melanogenesis activity in skins.However,the underlying mechanism(s)remain not completely understood.The aim of this study is thus to elucidate the possible mechanisms related to the melanogenesis suppression by glabridin in cultured B16 murine melanoma cells and in UVA radiation induced hyperpigmentation model of BALB/c mice as well.Molecular docking simulations revealed that between catalytic core residues and the compound.The treatment by glabridin significantly downregulated both transcriptional and/or protein expression of melanogenesis-related factors including melanocyte stimulating hormone receptor(MC1R),microphthalmia-associated transcription factor(MITF),tyrosinase(TYR),TYR-related protein-1(TRP-1)and TRP-2 in B16 cells.Both PKA/MITF and MAPK/MITF signaling pathways were found to be involved in the suppression of melanogenesis by glabridin in B16 cells.Also in vivo glabridin therapy significantly reduced hyperpigmentation,epidermal thickening,roughness and inflammation induced by frequent UVA exposure in mice skins,thus beneficial for skin healthcare.These data further look insights into the molecular mechanisms of melanogenesis suppression by glabridin,rationalizing the application of the natural compound for skin healthcare.Chunxing Pan Xiaoying Liu Yating Zheng Zejun Zhang Yongliang Li Biao Che Guangrong Liu Lanyue Zhang Changzhi Dong Haji Akber Aisa Zhiyun Du Zhengqiang Yuan 2023Food Science and Human Wellness2023,12,1:2
