1All roads lead to Rome-a review of the potential mechanisms by which exerkines exhibit neuroprotective effects in Alzheimer’s disease显示文摘Age-related neurodegenerative disorders such as Alzheimer’s disease(AD)have become a critical public health issue due to the significantly extended human lifespan,leading to considerable economic and social burdens.Traditional therapies for AD such as medicine and surgery remain ineffective,impractical,and expensive.Many studies have shown that a variety of bioactive substances released by physical exercise(called“exerkines”)help to maintain and improve the normal functions of the brain in terms of cognition,emotion,and psychomotor coordination.Increasing evidence suggests that exerkines may exert beneficial effects in AD as well.This review summarizes the neuroprotective effects of exerkines in AD,focusing on the underlying molecular mechanism and the dynamic expression of exerkines after physical exercise.The findings described in this review will help direct research into novel targets for the treatment of AD and develop customized exercise therapy for individuals of different ages,genders,and health conditions.Yi-Yao Liang Li-Dan Zhang Xi Luo Li-Li Wu Zhao-Wei Chen Guang-Hao Wei Kai-Qing Zhang Ze-An Du Ren-Zhi Li Kwok-Fai So Ang Li 2022Neural Regeneration Research2022,17,6:2
2Luteolin delays photoreceptor degeneration in a mouse model of retinitis pigmentosa显示文摘Luteolin is neuroprotective for retinal ganglion cells and retinal pigment epithelial cells after oxidative injury,whereby it can inhibit microglial neurotoxicity.Therefore,luteolin holds the potential to be useful for treatment of retinal diseases.The purpose of this study was to investigate whether luteolin exhibits neuroprotective effects on rod cells in rd10 mice,a slow photoreceptor-degenerative model of retinitis pigmentosa.Luteolin(100 mg/kg)intraperitoneally injected daily from postnatal day 14(P14)to P25 significantly enhanced the visual performance and retinal light responses of rd10 mice at P25.Moreover,it increased the survival of photoreceptors and improved retinal structure.Mechanistically,luteolin treatment attenuated increases in reactive oxygen species,photoreceptor apoptosis,and reactive gliosis;increased mRNA levels of anti-inflammatory cytokines while lowering that of pro-inflammatory and chemoattractant cytokines;and lowered the ratio of phospho-JNK/JNK.Application of the JNK inhibitor SP600125 exerted a similar protective effect to luteolin,suggesting that luteolin delays photoreceptor degeneration and functional deterioration in rd10 mice through regulation of retinal oxidation and inflammation by inhibiting the JNK pathway.Therefore,luteolin may be useful as a supplementary treatment for retinitis pigmentosa.This study was approved by the Qualified Ethics Committee of Jinan University,China(approval No.IACUC-20181217-02)on December 17,2018.Xiao-Bin Liu Feng Liu Yi-Yao Liang Gang Yin Hui-Jun Zhang Xue-Song Mi Zai-Jun Zhang Kwok-Fai So Ang Li Ying Xu 2021Neural Regeneration Research2021,16,10:2

20Retinal safety and toxicity study of artesunate in vitro and in vivo显示文摘Background:Artesunate(ART),a member of the artemisinin family,possesses multi-properties,including antiinflammation,anti-oxidation,and anti-tumor.ART was recently reported to show anti-neovascularization effect on the cornea,iris,and retina.Compared to the expensive anti-VEGF treatment,this versatile,economical treatment option is attractive in the ophthalmic field.The safety and toxicity profile of ART intravitreal application are in utmost need.Methods:In this study,immortalized microglial(IMG)cells were treated with ART to determine the safe concentrations without inducing overt inflammatory reactions.Reverse transcription-polymerase chain reaction analysis was used to detect the cytokine expressions in IMG cells in response to ART stimulation.Various doses of ART were intravitreally injected into the right eyes of C57BL/6 mice.Retinal function was tested by electroretinogram,and retinal ganglion cell(RGC)survival was evaluated by counting Brn3a stained cells in flat-mounted retinas at 7 days after ART injection.Results:ART below 5μM was safe for IMG cells in vitro.Both 2.5 and 5μM ART treatment increased IL-10 gene expression in IMG cells while not changing IL-1β,IL-6,TNF-α,and Arg-1.In the in vivo study,intravitreal injection of ART below 100μM did not cause deterioration in the retinal function and RGC survival of the mouse eyes,while 1 mM ART treatment significantly attenuated both the scotopic and photopic b-wave amplitudes and impaired RGC survival.In addition,treatment with ART of 25,50,and 100μM significantly decreased TNF-αgene expression while ART of 100μM significantly increased IL-10 in the mouse retina.Conclusions:Intravitreal injection of 100μM ART could downregulate TNF-αwhile upregulate IL-10 in the mouse retina without causing retinal functional deterioration and RGC loss.ART might be used as anti-inflammatory agent for retinal disorders.Bing-Wen Lu Yu-Xiang Liang Jin-Feng Liu Zhong-Qing Sun Kwok-Fai So Kin Chiu 2023Advances in Ophthalmology Practice and Research2023,3,2:0
