| 1 | Identification of an HLA-A~* 0201-restricted CD8^+ T-cell epitope SSp-1 of SARS-CoV spike protein显示文摘A novel coronavirus, severe acute respiratory syndrome (SA RS)-associated coronavirus (SARS-CoV), has been identified as the causal agent of SARS. Spike (S) protein is a major structural glycoprotein of the SARS virus and a potential target for SARS-specific cell-mediated immune responses. A pa nel of S protein-derived peptides was tested for their binding affinity to HLA -A *0201 molecules. Peptides with high affinity for HLA-A *0201 were then as se ssed for their capacity to elicit specific immune responses mediated by cytotoxi c T lymphocytes (CTLs) both in vivo, in HLA-A2.1/K b transgenic mice, a nd in vitro, from peripheral blood lymphocytes (PBLs) harvested from healthy HLA-A 2.1 + donors. SARS-CoV protein-derived peptide-1 (SSp-1 RLNEVAKNL), induced pepti de-specific CTLs both in vivo (transgenic mice) and in vitro (human PBL s), which specifically released interferon-gamma (IFN-gamma) upon stimulation with SSp-1-pulsed autologous dendritic cells (DCs) or T2 cells. SSp-1-specif ic CTLs also lysed major histocompatibility complex (MHC)-matched tumor cell lines engineered to express S proteins. HLA-A *0201-SSp-1 tetramer staining re vealed the presence of significant populations of SSp-1-specific CTLs in SSp- 1-induced CD8 + T cells. | 2 | Microstructure and Mechanical Properties of Pulse MIG Welded 6061/A356 Aluminum Alloy Dissimilar Butt Joints显示文摘The microstructure and mechanical properties of pulse metal inert-gas(MIG) welded dissimilar joints between 4 mm thick wrought 6061-T6 and cast A356-T6 aluminum alloy plates were investigated. The tensile strength of the joints reached 235 MPa, which is 83% of that of 6061 aluminum alloy, and then decreased with the increase of travel speed while keeping other welding parameters constant. The microstructure, composition and fractography of joints were examined by the optical microscopy(OM),scanning electron microscopy(SEM) and electron probe microanalysis(EPMA). Grain boundary liquation and segregation occurred in the partially melted zone(PMZ) on 6061 aluminum alloy side, and brittle Fe-rich phases were observed in partially melted zone on A356 aluminum alloy side. The minimum microhardness appeared in heat-affected zone(HAZ) near A356 aluminum alloy substrate. | 3 | Phosphoinositide 3-Kinase/Akt and Nuclear Factor-κB Are Involved in Staphylococcus Aureus-induced Apoptosis in U937 Cells显示文摘Objective To explore the mechanisms involved in Staphylococcus aureus(S.aureus) invading human monocytic U937 cells.Methods S.aureus were added to U937 cells at multiplicity of infections(MOI) of 20:1 for 0,15,30,60,and 90 minutes,respectively.Cell apoptosis was analyzed with Hoechst 33258 staining and Annexin V-fluorescein isothiocyanate(FITC)/propidium iodide(PI) flow cytometry analysis.Akt and nuclear factor-κB(NF-κB) activities were detected by Western blotting.Results Infection of U937 cells with S.aureus induced rapid cell death in a time-dependent manner,and the cells displayed characteristic features of apoptosis.S.aureus-induced apoptosis was associated with a prominent downregulation of activated(phosphorylated) Akt and NF-κB.The inhibition of phosphorylated Akt by LY294002 led to the inhibition of NF-κB in a dose-dependent manner.Inhibition of Akt with LY294002 caused further increase in apoptosis of U937 cells.Conclusions S.aureus can stimulate the apoptosis of U937 cells.S.aureus induces apoptosis of U937 cells by inhibiting Akt-regulated NF-κB. | Jia-he Wang Yi-jun Zhoux Yi-jun Zhou Li Tian Ping He | 2009 | Chinese Medical Sciences Journal2009,24,4: | 6 |
