1Decoding Cortical Glial Cell Development显示文摘Mouse cortical radial glial cells(RGCs)are primary neural stem cells that give rise to cortical oligodendrocytes,astrocytes,and olfactory bulb(OB)GABAergic interneurons in late embryogenesis.There are fundamental gaps in understanding how these diverse cell subtypes are generated.Here,by combining single-cell RNA-Seq with intersectional lineage analyses,we show that beginning at around E16.5,neocortical RGCs start to generate ASCL1^(+)EGFR^(+)apical multipotent intermediate progenitors(MIPCs),which then differentiate into basal MIPCs that express ASCL1,EGFR,OLIG2,and MKI67.These basal MIPCs undergo several rounds of divisions to generate most of the cortical oligodendrocytes and astrocytes and a subpopulation of OB interneurons.Finally,single-cell ATAC-Seq supported our model for the genetic logic underlying the specification and differentiation of cortical glial cells and OB interneurons.Taken together,this work reveals the process of cortical radial glial cell lineage progression and the developmental origins of cortical astrocytes and oligodendrocytes.Xiaosu Li Guoping Liu Lin Yang Zhenmeiyu Li Zhuangzhi Zhang Zhejun Xu Yuqun Cai Heng Du Zihao Su Ziwu Wang Yangyang Duan Haotian Chen Zicong Shang Yan You Qi Zhang Miao He Bin Chen Zhengang Yang 2021Neuroscience Bulletin2021,37,4:2
2Recent advances in wireless epicortical and intracortical neuronal recording systems显示文摘An implantable brain-computer interface(BCI)has proven to be effective in the field of sensory and motor function restoration and in the treatment of neurological disorders.Using a BCI recording system,we can transform current methods of human interaction with machines and the environment,especially to help those with cognitive and mobility disabilities regain mobility and reintegrate into society.However,most reported work has focused on a simple aspect of the whole system,such as electrodes,circuits,or data transmission,and only a very small percentage of systems are wireless.A miniature,lightweight,wireless,implantable microsystem is key to realizing long-term,real-time,and stable monitoring on freely moving animals or humans in their natural conditions.Here,we summarize typical wireless recording systems,from recording electrodes,processing chips and controllers,wireless data transmission,and power supply to the system-level package for either epicortical electrocorticogram(ECoG)or intracortical local field potentials(LFPs)/spike acquisition,developed in recent years.Finally,we conclude with our vision of challenges in next-generation wireless neuronal recording systems for chronic and safe applications.Bowen JI Zekai LIANG Xichen YUAN Honglai XU Minghao WANG Erwei YIN Zhejun GUO Longchun WANG Yuhao ZHOU Huicheng FENG Honglong CHANG Jingquan LIU 2022Science China(Information Sciences)2022,65,4:2

4A flexible neural implant with ultrathin substrate for low-invasive brain–computer interface applications显示文摘Implantable brain–computer interface(BCI)devices are an effective tool to decipher fundamental brain mechanisms and treat neural diseases.However,traditional neural implants with rigid or bulky cross-sections cause trauma and decrease the quality of the neuronal signal.Here,we propose a MEMS-fabricated flexible interface device for BCI applications.The microdevice with a thin film substrate can be readily reduced to submicron scale for low-invasive implantation.An elaborate silicon shuttle with an improved structure is designed to reliably implant the flexible device into brain tissue.The flexible substrate is temporarily bonded to the silicon shuttle by polyethylene glycol.On the flexible substrate,eight electrodes with different diameters are distributed evenly for local field potential and neural spike recording,both of which are modified by Pt-black to enhance the charge storage capacity and reduce the impedance.The mechanical and electrochemical characteristics of this interface were investigated in vitro.In vivo,the small cross-section of the device promises reduced trauma,and the neuronal signals can still be recorded one month after implantation,demonstrating the promise of this kind of flexible BCI device as a low-invasive tool for brain–computer communication.Zhejun Guo Fang Wang Longchun Wang Kejun Tu Chunpeng Jiang Ye Xi Wen Hong Qingda Xu Xiaolin Wang Bin Yang Bomin Sun Zude Lin Jingquan Liu 2022Microsystems & Nanoengineering2022,8,6:1

10Comparative pharmacophore modeling of human adenosine receptor A1 and A3 antagonists显示文摘Adenosine receptors are promising therapeutic targets in drug discovery. In this study, three-dimensional pharmacophore models of human adenosine receptor A1 and A3 antagonists were developed based on 26 and 23 diverse compounds, respectively. The best A1 pharmacophore model (A 1 _Hopy1) consists of four features: one hydrogen bond donor, one hydrophobic point and two ring aromatics, while the best A 3 pharmacophore model (A3 _Hopy1) also has four features: one hydrogen bond acceptor, one hydrophobic point and two ring aromatics. The correlation coefficients were 0.840 for A 1 test set with 146 diverse compounds and 0.827 for A3 test set with 238 diverse compounds. In the simulated virtual screening experiments, high enrichment factors of 6.51 and 6.90 were obtained for A 1 _Hopy1 and A3 _Hopy1 models, respectively. Moreover, two models also showed high subtype-selectivity in the simulated virtual screening experiments. 11Homeobox Gene Six3 is Required for the Differentiation of D2-Type Medium Spiny Neurons显示文摘Medium spiny neurons(MSNs)in the striatum,which can be divided into D1 and D2 MSNs,originate from the lateral ganglionic eminence(LGE).Previously,we reported that Six3 is a downstream target of Sp8/Sp9 in the transcriptional regulatory cascade of D2 MSN development and that conditionally knocking out Six3 leads to a severe loss of D2 MSNs.Here,we showed that Six3 mainly functions in D2 MSN precursor cells and gradually loses its function as D2 MSNs mature.Conditional deletion of Six3 had little effect on cell proliferation but blocked the differentiation of D2 MSN precursor cells.In addition,conditional overexpression of Six3 promoted the differentiation of precursor cells in the LGE.We measured an increase of apoptosis in the postnatal striatum of conditional Six3-knockout mice.This suggests that,in the absence of Six3,abnormally differentiated D2 MSNs are eliminated by programmed cell death.These results further identify Six3 as an important regulatory element during D2 MSN differentiation.Xiaolei Song Haotian Chen Zicong Shang Heng Du Zhenmeiyu Li Yan Wen Guoping Liu Dashi Qi Yan You Zhengang Yang Zhuangzhi Zhang Zhejun Xu 2021Neuroscience Bulletin2021,37,7:0
12BMP7 expression in mammalian cortical radial glial cells increases the length of the neurogenic period显示文摘The seat of human intelligence is the human cerebral cortex,which is responsible for our exceptional cognitive abilities.Identifying principles that lead to the development of the large-sized human cerebral cortex will shed light on what makes the human brain and species so special.The remarkable increase in the number of human cortical pyramidal neurons and the size of the human cerebral cortex is mainly because human cortical radial glial cells,primary neural stem cells in the cortex,generate cortical pyramidal neurons for more than 130 days,whereas the same process takes only about 7 days in mice.The molecular mechanisms underlying this difference are largely unknown.Here,we found that bone morphogenic protein 7(BMP7)is expressed by increasing the number of corti-cal radial glial cells during mammalian evolution(mouse,ferret,monkey,and human).BMP7 expression in cortical radial glial cells promotes neurogenesis,inhibits gliogenesis,and thereby increases the length of the neurogenic period,whereas Sonic Hedgehog(SHH)signaling promotes cortical gliogenesis.We demonstrate that BMP7 sign-aling and SHH signaling mutually inhibit each other through regulation of GLI3 repressor formation.We propose that BMP7 drives the evolutionary expansion of the mammalian cortex by increasing the length of the neurogenic period.Zhenmeiyu Li Guoping Liu Lin Yang Mengge Sun Zhuangzhi Zhang Zhejun Xu Yanjing Gao Xin Jiang Zihao Su Xiaosu Li Zhengang Yang 2024Protein & Cell2024,15,1:0
