2The history and advances in cancer immunotherapy:understanding the characteristics of tumor-infiltrating immune cells and their therapeutic implications显示文摘Immunotherapy has revolutionized cancer treatment and rejuvenated the field of tumor immunology.Several types of immunotherapy,including adoptive cell transfer(ACT)and immune checkpoint inhibitors(ICIs),have obtained durable clinical responses,but their efficacies vary,and only subsets of cancer patients can benefit from them.Immune infiltrates in the tumor microenvironment(TME)have been shown to play a key role in tumor development and will affect the clinical outcomes of cancer patients.Comprehensive profiling of tumor-infiltrating immune cells would shed light on the mechanisms of cancer–immune evasion,thus providing opportunities for the development of novel therapeutic strategies.However,the highly heterogeneous and dynamic nature of the TME impedes the precise dissection of intratumoral immune cells.With recent advances in single-cell technologies such as single-cell RNA sequencing(scRNA-seq)and mass cytometry,systematic interrogation of the TME is feasible and will provide insights into the functional diversities of tumor-infiltrating immune cells.In this review,we outline the recent progress in cancer immunotherapy,particularly by focusing on landmark studies and the recent single-cell characterization of tumor-associated immune cells,and we summarize the phenotypic diversities of intratumoral immune cells and their connections with cancer immunotherapy.We believe such a review could strengthen our understanding of the progress in cancer immunotherapy,facilitate the elucidation of immune cell modulation in tumor progression,and thus guide the development of novel immunotherapies for cancer treatment.Yuanyuan Zhang Zemin Zhang 2020Cellular & Molecular Immunology2020,17,8:61
3Visualization of the complex structure and stress field inside rock by means of 3D printing technology显示文摘Accurate characterization and visualization of the complex inner structure and stress distribution of rocks are of vital significance to solve a variety of underground engineering problems. In this paper, we incorporate several advanced technologies, such as CT scan, three-dimensional(3D) reconstruction, and 3D printing, to produce a physical model representing the natural coal rock that inherently contains complex fractures or joints. We employ 3D frozen stress and photoelastic technologies to characterize and visualize the stress distribution within the fractured rock under uniaxial compression. The 3D printed model presents the fracture structures identical to those of the natural prototype. The mechanical properties of the printed model,including uniaxial compression strength, elastic modulus,and Poisson's ratio, are testified to be similar to those of the prototype coal rock. The frozen stress and photoelastic tests show that the location of stress concentration and the stress gradient around the discontinuous fractures are in good agreement with the numerical predictions of the real coalsample. The proposed method appears to be capable of visually quantifying the influences of discontinuous,irregular fractures on the strength, deformation, and stress concentration of coal rock. 4Oxford Nanopore MinION Sequencing and Genome Assembly显示文摘The revolution of genome sequencing is continuing after the successful secondgeneration sequencing(SGS) technology. The third-generation sequencing(TGS) technology, led by Pacific Biosciences(Pac Bio), is progressing rapidly, moving from a technology once only capable of providing data for small genome analysis, or for performing targeted screening, to one that promises high quality de novo assembly and structural variation detection for human-sized genomes. In 2014, the Min ION, the first commercial sequencer using nanopore technology, was released by Oxford Nanopore Technologies(ONT). Min ION identifies DNA bases by measuring the changes in electrical conductivity generated as DNA strands pass through a biological pore. Its portability,affordability, and speed in data production makes it suitable for real-time applications, the release of the long read sequencer Min ION has thus generated much excitement and interest in the genomics community. While de novo genome assemblies can be cheaply produced from SGS data, assembly continuity is often relatively poor, due to the limited ability of short reads to handle long repeats. Assembly quality can be greatly improved by using TGS long reads, since repetitive regions can be easily expanded into using longer sequencing lengths, despite having higher error rates at the base level. 7Melatonin alleviates intervertebral disc degeneration by disrupting the IL-1β/NF-κB-NLRP3 inflammasome positive feedback loop显示文摘The inflammatory response is induced by the overexpression of inflammatory cytokines, mainly interleukin(IL)-1β, and is one of the main causes of intervertebral disc degeneration(IVDD). NLR pyrin domain containing 3(NLRP3) inflammasome activation is an important source of IL-1β. As an anti-inflammatory neuroendocrine hormone, melatonin plays various roles in different pathophysiological conditions. However, its roles in IVDD are still not well understood and require more examination. First, we demonstrated that melatonin delayed the progression of IVDD and relieved IVDD-related low back pain in a rat needle puncture IVDD model;moreover, NLRP3 inflammasome activation(NLRP3, p20, and IL-1β levels) was significantly upregulated in severely degenerated human discs and a rat IVDD model. Subsequently, an IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop was found in nucleus pulposus(NP) cells that were treated with IL-1β. In these cells, expression of NLRP3 and p20 was significantly increased, NF-κB signaling was involved in this regulation, and mitochondrial reactive oxygen species(mt ROS)production increased. Furthermore, we found that melatonin disrupted the IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop in vitro and in vivo. Melatonin treatment decreased NLRP3, p20, and IL-1β levels by inhibiting NF-κB signaling and downregulating mt ROS production. Finally, we showed that melatonin mediated the disruption of the positive feedback loop of IL-1β in vivo. In this study, we showed for the first time that IL-1β promotes its own expression by upregulating NLRP3 inflammasome activation. 8Modeling and performance analysis of energy selective electron (ESE) engine with heat leakage and transmission probability显示文摘A model of an energy selective electron (ESE) engine with linear heat leakage and Lorentzian transmission probability is established in this paper.The expressions for the main performance parameters of the ESE engine operating as a heat engine or a refrigerator are derived by using the theory of finite time thermodynamics.The optimum performances of the ESE engine are explored and the influences of the heat leakage,the central energy level of the resonance,and the width of the resonance on the performance of the ESE engine are analyzed by using detailed numerical examples.The optimal operation regions of power output and efficiency (or cooling load and coefficient of performance (COP)) are also discussed.Moreover,the performances of the ESE engine with Lorentzian transmission probability are compared with those with rectangular transmission probability.It is shown that the power output versus efficiency (or cooling load versus COP) characteristic curves with and without heat leakage are all closed loop-shaped ones.The efficiency (or COP) of the ESE engine decreases as the heat leakage increases.It is found that as the resonance width increases,the power output and efficiency (or cooling load and COP) increase to a maximum and then decrease due to the finite range of energies which contribute positively to the power generation or refrigeration in the electron system.Especially,when heat leakage is taken into account,the characteristic curves of maximum efficiency (or maximum COP) versus half resonance width are parabolic-like ones,which are quite different from the monotonic decreasing characteristic curves obtained in previous analyses without considering heat leakage.The results obtained in this paper can provide some theoretical guidelines for the design and operation of practical electron energy conversion devices such as solid-state thermionic refrigerators.DING ZeMin,CHEN LinGen & SUN FengRui College of Naval Architecture and Power,Naval University of Engineering,Wuhan 430033,China Received December 24,2010 accepted May 30,2011 published online September 9,2011 2011Science China(Physics,Mechanics & Astronomy)2011,54,11:14
9Weldability, microstructure and mechanical properties of laser-welded selective laser melted 304 stainless steel joints显示文摘Laser welding is a promising process for joining small components produced by selective laser melting (SLM) to fabricate the large-scale and complex-shaped parts. In the work, the morphology, microstructure, microhardness, tensile properties and corrosion resistance of the laser welded stress-relieved SLMed 304 stainless steel joints are investigated, as the different sections of stress-relieved SLMed 304 stainless steel are joined. Results show that the SLMed 304 stainless steel plates have a good laser weldability. The microstructure of laser-welded joints consists of the cellular dendrites in austenite matrix within columnar grains, exhibiting a coarser dendrite structure, lower microhardness (~220HV) and tensile properties (tensile strength of ~750 MPa, and area reduction of ~27.6%), but superior corrosion resistance to those of SLMed plates. The dendrite arm spacing of the joints varies from ~3.7μm in center zone, to ~5.0 (xm in fusion zone, to ~2.5 in epitaxial zone. The SLMed anisotropy shows a negligible effect on the microstructure and performance of the laser-welded joints. 10Generalized constructal optimization for the secondary cooling process of slab continuous casting based on entransy theory显示文摘Based on constructal theory and entransy theory,a generalized constructal optimization of a solidification heat transfer process of slab continuous casting for a specified total water flow rate in the secondary cooling zone was carried out.A complex function was taken as the optimization objective to perform the casting.The complex function was composed of the functions of the entransy dissipation and surface temperature gradient of the slab.The optimal water distribution at the sections of the secondary cooling zone were obtained.The effects of the total water flow rate in the secondary cooling zone,casting speed,superheat and water distribution on the generalized constructal optimizations of the secondary cooling process were analyzed.The results show that on comparing the optimization results obtained based on the optimal water distributions of the 8 sections in the secondary cooling zone with those based on the initial ones,the complex function and the functions of the entransy dissipation and surface temperature gradient after optimization decreased by 43.25%,5.90%and 80.60%,respectively.The quality and energy storage of the slab had obviously improved in this case.The complex function,composed of the functions of the entransy dissipation and surface temperature gradient of the slab,was a compromise between the internal and surface temperature gradients of the slab.Essentially,it is also the compromise between energy storage and quality of the slab.The'generalized constructal optimization'based on the minimum complex function can provide an optimal alternative scheme from the point of view of improving energy storage and quality for the parameter design and dynamic operation of the solidification heat transfer process of slab continuous casting.FENG HuiJun CHEN LinGen XIE ZhiHui DING ZeMin SUN FengRui 2014Science China(Technological Sciences)2014,57,4:14
11Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. 12Thermodynamic characteristics of a Brownian heat pump in a spatially periodic temperature field显示文摘This paper has studied the thermodynamic performance of a thermal Brownian heat pump,which consists of Brownian particles moving at a periodic sawtooth potential with external forces and contacting with the alternating hot and cold reservoirs along the space coordinate.The heat flows driven by both potential and kinetic energies are taken into account.The analytical expressions for the heating load,coefficient of performance(COP) and power input of the Brownian heat pump are derived and the performance characteristics are obtained by numerical calculations.It is shown that due to the heat flow via the change of kinetic energy of the particles,the Brownian heat pump is always irreversible and the COP can never attain the Carnot COP.The study has also investigated the influences of the operating parameters,i.e.the external force,barrier height of the potential,asymmetry of the sawtooth potential and temperature ratio of the heat reservoirs,on the performance of the Brownian heat pump.The effective regions of external force and barrier height of the potential in which the Brownian motor can operates as a heat pump are determined.The results show that the performance of the Brownian heat pump greatly depends on the parameters;if the parameters are properly chosen,the Brownian heat pump may be controlled to operate in the optimal regimes.DING ZeMin,CHEN LinGen & SUN FengRui Postgraduate School,Naval University of Engineering,Wuhan 430033,China 2010Science China(Physics,Mechanics & Astronomy)2010,53,5:12
13Fine mapping of locus S-b for F_1 pollen sterility in rice (Oryza sativa L.)显示文摘Hybrid sterility is the main barrier in util-izing the heterosis of subspecies in rice. A knowledge of the underlying molecular mechanism of the hybrid sterility will be useful for overcoming the barrier. In this research, the F1 pollen sterility locus, S-b, was mapped between SSR markers PSM8 and PSM202. To fine map the locus, one F2 mapping population of 3910 plants was developed using the near-isogenic lines of the locus. Ninety-seven recombinants be-tween two markers were selected. Moreover, a series of markers, including two SSR markers, two InDel markers and four CAPS markers, were developed on the region. Linkage analysis showed that marker W4 was co-segregated with locus S-b, while makers A8 and A14 were located on the two sides of the locus with a distance of 0.026 and 0.038 cM, respectively. The markers were then integrated with the se-quences of the clones of the region. Results showed that all the polymorphic markers were anchored on the three end-to-end jointed clones AC093089, AC079021 and AC134931. According to the physical information of the markers, locus S-b was finally de-limited to a region of 27 kb between A8 and A14. Seven ORFs were identified on the region based on the annotation results of RiceGAAS system. 14Removal of polycyclic aromatic hydrocarbons from aqueous solution by raw and modified plant residue materials as biosorbents显示文摘Removal of polycyclic aromatic hydrocarbons(PAHs), e.g., naphthalene, acenaphthene, phenanthrene and pyrene, from aqueous solution by raw and modified plant residues was investigated to develop low cost biosorbents for organic pollutant abatement. Bamboo wood, pine wood, pine needles and pine bark were selected as plant residues, and acid hydrolysis was used as an easily modification method. The raw and modified biosorbents were characterized by elemental analysis, Fourier transform infrared spectroscopy and scanning electron microscopy. The sorption isotherms of PAHs to raw biosorbents were apparently linear, and were dominated by a partitioning process. In comparison, the isotherms of the hydrolyzed biosorbents displayed nonlinearity, which was controlled by partitioning and the specific interaction mechanism. The sorption kinetic curves of PAHs to the raw and modified plant residues fit well with the pseudo second-order kinetics model. The sorption rates were faster for the raw biosorbents than the corresponding hydrolyzed biosorbents, which was attributed to the latter having more condensed domains(i.e., exposed aromatic core). By the consumption of the amorphous cellulose component under acid hydrolysis, the sorption capability of the hydrolyzed biosorbents was notably enhanced, i.e., 6–18 fold for phenanthrene, 6–8 fold for naphthalene and pyrene and 5–8 fold for acenaphthene. The sorption coefficients(Kd) were negatively correlated with the polarity index [(O+N)/C], and positively correlated with the aromaticity of the biosorbents. For a given biosorbent, a positive linear correlation between logKoc and logKow for different PAHs was observed. Interestingly, the linear plots of logKoc–logKow were parallel for different biosorbents. 15Optimal performance regions of an irreversible energy selective electron heat engine with double resonances显示文摘A theoretical model for irreversible double resonance ESE(energy selective electron) device with phonon induced bypass heat leakage which is operating as heat engine system is proposed. The thermodynamic performance is optimized and the impacts of heat leakage and structure parameters of the electron system on its performance are discussed in detail by using FTT(finite time thermodynamics). Moreover, performances of the ESE system with multiple optimization objective functions, including power output, thermal efficiency, ecological function and efficient power, are explored by numerical examples. New optimal performance regions and the selection plans of optimization objective functions of the ESE system are obtained. It reveals that the characteristic of power versus efficiency behave as loop-shaped curves in spite of the heat leakage which will always decrease the efficiency of the electron engine. By properly choosing the design parameters, the ESE engine can be designed to operate at optimal conditions according to different design purpose. 16Lipoprotein metabolism in nonalcoholic fatty liver disease显示文摘Nonalcoholic fatty liver disease (NAFLD),an escalating health problem worldwide,covers a spectrum of pathologies characterized by fatty accumulation in hepatocytes in early stages,with potential progression to liver inflammation,fibrosis,and failure.A close,yet poorly understood link exists between NAFLD and dyslipidemia,a constellation of abnormalities in plasma lipoproteins including triglyceride-rich very low density lipoproteins.Apolipoproteins are a group of primarily liver-derived proteins found in serum lipoproteins;they not only play an extracellular role in lipid transport between vital organs through circulation,but also play an important intracellular role in hepatic lipoprotein assembly and secretion.The liver functions as the central hub for lipoprotein metabolism,as it dictates lipoprotein production and to a significant extent modulates lipoprotein clearance.Lipoprotein metabolism is an integral component of hepatocellular lipid homeostasis and is implicated in the pathogenesis,potential diagnosis,and treatment of NAFLD.Zhenghui Gordon Jiang Simon C. 17Novel estimators of equivalent number of looks in polarimetric SAR imagery based on sub-matrices显示文摘The equivalent number of looks(ENL) is an important parameter in the multilook statistical model of polarimetric synthetic aperture radar(Pol-SAR). Recently, the maximum likelihood(ML) method was proposed and gave a good performance in the Gaussian model case by using the full covariance matrix instead of the intensity of Pol-SAR data, but it generated underestimates in the product model case. In this paper several novel ENL estimators are presented via certain cumulants of the log-determinant of the sub-matrices of the multilook polarimetric covariance matrix. The texture effect to the ENL estimates is eliminated, and the analytic estimators are derived. The estimators use the full covariance matrix and sub-matrices information, rather than the intensities of polarization channels. All the novel estimators are suitable for any texture model and thus provide more accurate results than many existing ones. Experiments using simulated data and real data are presented to evaluate the performance of different estimators. The results show that the second log-determinant moment(SLDM3)-based method is the best one among the novel estimators. At the same time this estimator has much less computational complexity. 19Simultaneous incorporation of PTH(1-34) and nano-hydroxyapatite into Chitosan/Alginate Hydrogels for efficient bone regeneration显示文摘Tissue regeneration based on the utilization of artificial soft materials is considered a promising treatment for bone-related diseases.Here,we report cranial bone regeneration promoted by hydrogels that contain parathyroid hormone(PTH)peptide PTH(1-34)and nano-hydroxyapatite(nHAP).A combination of the positively charged natural polymer chitosan(CS)and negatively charged sodium alginate led to the formation of hydrogels with porous structures,as shown by scanning electron microscopy.Rheological characterizations revealed that the mechanical properties of the hydrogels were almost maintained upon the addition of nHAP and PTH(1-34).In vitro experiments showed that the hydrogel containing nHAP and PTH(1-34)exhibited strong biocompatibility and facilitated osteogenic differentiation of rat bone marrow mesenchymal stem cells(rBMSCs)via the Notch signaling pathway,as shown by the upregulated expression of osteogenic-related proteins.We found that increasing the content of PTH(1-34)in the hydrogels resulted in enhanced osteogenic differentiation of BMSCs.Implantation of the complex hydrogel into a rat cranial defect model led to efficient bone regeneration compared to the rats treated with the hydrogel alone or with nHAP,indicating the simultaneous therapeutic effect of nHAP and PTH during the treatment process.Both the in vitro and in vivo results demonstrated that simultaneously incorporating nHAP and PTH into hydrogels shows promise for bone regeneration,suggesting a new strategy for tissue engineering and regeneration in the future.Zhiyuan Zou Le Wang Zhifei Zhou Qing Sun Delong Liu Yan Chen Hao Hu Yu Cai Sixiong Lin Zhengran Yu Bizhi Tan Wei Guo Zemin Ling Xuenong Zou 2021Bioactive Materials2021,6,6:7
20Performance optimization of thermionic refrigerators based on van der Waals heterostructures显示文摘In this paper, an irreversible thermionic refrigerator model based on van der Waals heterostructure with various irreversibilities is established by utilizing combination of non-equilibrium thermodynamics and finite time thermodynamics. The basic performance characteristics of the refrigerator are obtained. The effects of key factors, such as bias voltages, Schottky barrier heights and heat leakages, on the performance are studied. Results show that cooling rates and coefficients of performances(COPs) can attain the double maximum with proper modulation of barrier heights and bias voltages. Increasing cross-plane thermal resistance as well as decreasing electrode-reservoir thermal resistance and reservoir-reservoir thermal resistance can enhance the performance of the device. The optimal performance region is the interval between the maximum cooling rate point and the maximum COP point. By modulating the bias voltage, the working state of the device can fall into the optimal performance region.
