| 1 | Piper sarmentosum as an antioxidant on oxidative stress in human umbilical vein endothelial cells induced by hydrogen peroxide显示文摘Endothelial cell death due to increased reactive oxygen species(ROS) may contribute to the initial endothelial injury,which promotes atherosclerotic lesion formation.Piper sarmentosum(PS),a natural product,has been shown to have an antioxidant property,which is hypothesized to inhibit production of ROS and prevent cell injury.Thus,the present study was designed to determine the effects of PS on the hydrogen peroxide(H2O2)-induced oxidative cell damage in cultured human umbilical vein endothelial cells(HUVECs).In this experiment,HUVECs were obtained by collagenase perfusion of the large vein in the umbilical cord and cultured in medium M200 supplemented with low serum growth supplementation(LSGS).HUVECs were treated with various concentrations of H2O2(0-1000 μmol/L) and it was observed that 180 μmol/L H2O2 reduced cell viability by 50% as denoted by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay.Using the above concentration as the positive control,the H2O2-induced HUVECs were concomitantly treated with various concentrations(100,150,250 and 300 μg/ml) of three different extracts(aqueous,methanol and hexane) of PS.Malondialdehyde(MDA),superoxide dismutase(SOD),catalase(CAT) and glutathione peroxidase(GPX) levels showed a significant increase(P<0.05) in HUVECs compared to the negative control.However,PS extracts showed a protective effect on HUVECs from H2O2-induced cell apoptosis with a significant reduction in MDA,SOD,CAT and GPX levels(P<0.05).Furthermore,PS had exhibited ferric reducing antioxidant power with its high phenolic content.Hence,it was concluded that PS plays a beneficial role in reducing oxidative stress in H2O2-induced HUVECs. | Abdul Hamid HAFIZAH Zakaria ZAITON Amom ZULKHAIRI Adenan MOHD ILHAM Megat Mohd Nordin NOR ANITA Abdullah Mahdy ZALEHA | 2010 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2010,11,5: | 13 |

| 3 | Correlation of serum insulin like growth factor-I with retinopathy in Malaysian pregnant diabetics显示文摘AIM:To study the association of serum insulin-like growth factor-I(IGF-I) with diabetic retinopathy.· METHODS:Serum IGF-1 levels were measured in 25 pregnant diabetic patients and 25 pregnant non-diabetic patients who were matched for age,ethnicity,parity and period of gestation.Fundus examination was performed in both groups at 28,32 and 36 weeks of gestation.RESULTS:The serum IGF-I level was significantly elevated in pregnant diabetics compared to pregnant non-diabetics(366±199μg/L vs 184±89μg/L,(P=0.0001) at 24 weeks,535±251μg/L vs 356±89μg/L,(P=0.007) at 32 weeks and 404±166μg/L vs 264±113μg/L,(P=0.003) at 36 weeks of gestation).The pregnant diabetics with established diabetes had significantly higher IGF-1 level than gestational diabetes at 28,32 and 36 weeks of gestation.The serum IGF-I level in pregnant diabetics with retinopathy was significantly higher than in those without retinopathy at all periods of gestation.· CONCLUSION:Increased serum IGF-1 in pregnancy may increase the risks for retinopathy. | S Adzura M Muhaya M Normalina A M Zaleha W P Sharifa Ezat I Tajunisah | 2011 | International Journal of Ophthalmology(English edition)2011,4,1: | 4 |

| 17 | Epithelial Layer Estimation Using Curvatures and Textural Features for Dysplastic Tissue Detection显示文摘Boundary effect in digital pathology is a phenomenon where the tissue shapes of biopsy samples get distorted during the sampling process.The morphological pattern of an epithelial layer is greatly affected.Theoretically,the shape deformation model can normalise the distortions,but it needs a 2D image.Curvatures theory,on the other hand,is not yet tested on digital pathology images.Therefore,this work proposed a curvature detection to reduce the boundary effects and estimates the epithelial layer.The boundary effect on the tissue surfaces is normalised using the frequency of a curve deviates from being a straight line.The epithelial layer’s depth is estimated from the tissue edges and the connected nucleolus only.Then,the textural and spatial features along the estimated layer are used for dysplastic tissue detection.The proposed method achieved better performance compared to the whole tissue regions in terms of detecting dysplastic tissue.The result shows a leap of kappa points from fair to a substantial agreement with the expert’s ground truth classification.The improved results demonstrate that curvatures have been effective in reducing the boundary effects on the epithelial layer of tissue.Thus,quantifying and classifying the morphological patterns for dysplasia can be automated.The textural and spatial features on the detected epithelial layer can capture the changes in tissue. | Afzan Adam Abdul Hadi Abd Rahman Nor Samsiah Sani Zaid Abdi Alkareem Alyessari Nur Jumaadzan Zaleha Mamat Basela Hasan | 2021 | Computers, Materials & Continua2021,,4: | 0 |
