Agrobacterium-Mediated Transformation of Rice: Constraints and Possible Solutions显示文摘Genetic transformation of rice(Oryza sativa L.) by introducing beneficial traits is now a central research instrument in plant physiology and a practical tool for plant improvement. Many approaches are verified for stable introduction of foreign genes into the plant genome. The review examined the different constraints that limit the success of rice genetic transformation via Agrobacterium-mediated approach and suggested possible solutions. Explant identification, gene transfer technique and construct to tailor the integration, transgene expression without collateral to genetic damage and transformant selection are among the technical challenges affecting the rice transformation. Despite the contests, Agrobacteriummediated transformation system has been a better option for producing transgenic rice varieties because of its exact T-DNA processing and simple integration of low copy-number transgene. This information is necessary for improving the transformation system for recalcitrant rice varieties. | Sulaiman MOHAMMED Azman Abd SAMAD Zaidah RAHMAT | 2019 | Rice science2019,26,3: | 6

Tuberculosis:an experience from Mycobacterium smears and culture analysis显示文摘Objective:Simple tests like direct smear of the acid fast bacilli(AFB) and Mycobacterium culture could assist the diagnosis of tuberculosis. This study is aimed at reviewing the outcome of smears, culture results and contamination rate among specimens requested for AFB smear and Mycobacterium culture. Methods:Retrospective laboratory data analysis requesting for Mycobacterium culture from January 2005 till December 2006 was done in a tertiary teaching hospital of Universiti Sains Malaysia,Kubang Kerian,Kelantan,Malaysia. Results: Four hundred and sixty seven(36.6%) isolates grew from 1 277 specimens.Of these isolates,314 (67.2%) grew Mycobacterium tuberculosis,23(4.9%) grew Mycobacterium other than tuberculosis and 38 (8.1%) grew contaminants.Among the M.tuberculosis cultures,165(52.5%) had growth of more than 100 confluent colonies, whereas 39 cultures(12.4%) had growth of less than 19 colonies.Direct smear for AFB among smear positive cases showed presence of more than 50 bacilli/line in 231(49.5%) cases and smear negative cases accounted for 63(13.5%).Among smear positive cases,291(94.5%) cultures grew Mycobacterium species and another 17(5.5%) cultures grew contaminants. In smear negative cases,32(62.7%) cultures grew Mycobacterium species and 19(37.3%) cultures grew contaminants. Conclusion:The results from data analysis of the Mycobacterium cultures should be critically utilized in order to review the laboratory performance and to improve its services in the future. Some of the data is also useful to the administrators of the hospital in terms of estimating the risk of occupational hazard faced by the health care workers. | Zeehaida M Siti Asma H Siti Hawa H Zaidah AR Norbanee TH | 2009 | Asian Pacific Journal of Tropical Medicine2009,2,2: | 0
Extra pulmonary tuberculosis in a tertiary teaching hospital:A five years review显示文摘Objective:To investigate the prevalence, clinical and bacteriological features and outcome of extrapulmonary tuberculosis in patients attending a tertiary care hospital at Kota Bharu,Kelantan. Methods:All culture positive tuberculosis cases recorded in Hospital Universiti Sains Malaysia at Kota Bharu during five years period from 2003-2007 were included in the study.Mycobacterium detected in samples originating from sites other than lungs was considered as a case of extrapulmonary tuberculosis.Age,site of infection, bacteriological findings and outcomes were analyzed. Results:Out of 437 tuberculosis cases recorded from 2002-2006,59 cases had culture positive extrapulmonary tuberculosis.Their mean age was 37 years and 44%were between the ages of 20-40 years.About 63%were negative for acid-fast bacilli by Z-N stain.Of the 59,97%was Mycobacterium tuberculosisc(M.tuberculosis) while two(3%) were atypical Mycobacterium Runyon Group-Ⅳ.Only 3 (5%) were isoniazid resistant.Extrapulmonary sites involved were lymph nodes 19(31%),central nervous system 12(20%),pleural 11(19%) spine 8(14%).,renal 3(5.1%) and others 6(10.2%).In this series, 11(18%) were positive for HIV.There was no significant association between extrapulmonary tuberculosis and HIV status. The outcome of CNS tuberculosis was the poorest with a mortality rate of 25%.Conclusion: M.tuberculosis is the predominant organism in extrapulmonary tuberculosis and culture is the most effective method for its diagnosis. | Siti Suraiya Md Noor Zaidah Abdul Rahman Sarimah Abdullah Zakuan Zainy Deris Che Wan Aminuddin | 2009 | Asian Pacific Journal of Tropical Medicine2009,2,5: | 0
Resistant patterns of Pseudomonas aeruginosa in a Malaysian teaching hospital显示文摘Objective:Pseudomonas aeruginosa is an opportunistic pathogen and the leading cause of nosocomial infections. Currently a notable increase in the prevalence of multidrug-resistant P.aeruginosa worldwide has been reported in hospitalized patients and was associated with high morbidity and mortality. Methods:A retrospective laboratory based analysis regarding the spectrum and distribution of P.aeruginosa from a wide range of clinical samples in Hospital Universiti Sains Malaysia since January 2003 to December 2007 was done. Results: Altogether, there were 2 308 clinical isolates analyzed. The main sources of P.aeruginosa were from swab, respiratory, urine and blood specimens which accounted for 28.2%,21.8%,13.2%and 12.8% respectively. Results showed significant reduction in percentage of resistant towards three antibiotic namely ciprofloxacin, ceftazidime and imipenem. However the percentage of pan-resistant P.aeruginosa increased steadily over these years. Conclusion:This data is helpful to the clinician in guiding the choice of appropriate antibiotic to treat P.aeruginosa infection. At the same time,it warrants a more aggressive infection control activity to be implemented to control the spread of pan resistant strain in this centre. | Zaidah AR Siti SMN Zahiruddin WM Zeehaida M | 2009 | Asian Pacific Journal of Tropical Medicine2009,2,6: | 0
Integrated Evolving Spiking Neural Network and Feature Extraction Methods for Scoliosis Classification显示文摘Adolescent Idiopathic Scoliosis(AIS)is a deformity of the spine that affects teenagers. The current method for detecting AIS is based on radiographic images which may increase the risk of cancer growth due to radiation.Photogrammetry is another alternative used to identify AIS by distinguishing the curves of the spine from the surface of a human’s back.Currently, detecting the curve of the spine is manually performed, making it a time-consuming task.To overcome this issue,it is crucial to develop a better model that automatically detects the curve of the spine and classify the types of AIS.This research proposes a new integration of ESNN and Feature Extraction(FE)methods and explores the architecture of ESNN for the AIS classification model. This research identifies the optimal Feature Extraction(FE)methods to reduce computational complexity. The ability of ESNN to provide a fast result with a simplicity and performance capability makes this model suitable to be implemented in a clinical setting where a quick result is crucial.A comparison between the conventional classifier(Support Vector Machine(SVM),Multi-layer Perceptron(MLP)and Random Forest(RF))with the proposed AIS model also be performed on a dataset collected by an orthopedic expert from Hospital Universiti Kebangsaan Malaysia(HUKM).This dataset consists of various photogrammetry images of the human back with different types ofMalaysian AIS patients to solve the scoliosis problem. The process begins by pre-processing the images which includes resizing and converting the captured pictures to gray-scale images. This is then followed by feature extraction, normalization, and classification. The experimental results indicate that the integration of LBP and ESNN achieves higher accuracy compared to the performance of multiple baseline state-of-the-art Machine Learning for AIS classification. This demonstrates the capability of ESNN in classifying the types of AIS based on photogrammetry images. | Nurbaity Sabri Haza Nuzly Abdull Hamed Zaidah Ibrahim Kamalnizat Ibrahim Mohd Adham Isa | 2022 | Computers, Materials & Continua2022,,12: | 0
Prediction of C-glycosylated apigenin (vitexin) biosynthesis in Ficus deltoidea based on plant proteins identified by LC-MS/MS显示文摘BACKGROUND: Plant secondary metabolites act as defence molecules to protect plants from biotic and abiotic stresses. Inparticular, C-glycosylated flavonoids are more stable and reactive than their O-glycosylated counterparts. Therefore, vitexin(apigenin 8-C glucoside) present in Ficus deltoidea is well-known for its antioxidant, anti-inflammatory, and antidiabeticProperties. | Farah Izana Abdullah Lee Suan Chua Zaidah Rahmat | 2017 | Frontiers in Biology2017,12,6: | 0
