1Genome survey uncovers the secrets of sex and lifestyle in caterpillar fungus显示文摘The caterpillar fungus Ophiocordyceps sinensis (best known as Cordyceps sinensis) mummifies ghost moth larvae exclusively in Tibetan Plateau alpine ecosystems. Touted as 'Himalayan Viagra', the fungus is highly prized due to its medical benefits and dwindling supplies. Attempts to culture the sexual fruiting-body have failed and the huge market demand has led to severe devastation of local ecosystems and to the fungus heading towards extinction. By genome sequencing, we establish that unlike related insect pathogens O. sinensis contains two compatible mating-type genes in its genome and is self-fertile, i.e. homothallic. However, sexual processes are only initiated under native environmental conditions. O. sinensis resembles biotrophic plant pathogens in having a genome shaped by retrotransposon-driven expansions. The resulting changes in gene content suggest that O. sinensis has a biphasic pathogenic mechanism beginning with stealth pathogenesis in early host instars. O. sinensis is the first psychrophilic fungus sequenced and is adapted to extreme cold with putative antifreeze proteins and mechanisms for increasing lipid accumulation and fatty acid unsaturation. 2A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 3Grafting Onto Artemisia annua Improves Drought Tolerance in Chrysanthemum by Enhancing Photosynthetic Capacity显示文摘Drought stress drastically influences the yield and quality of chrysanthemums,and thus grafting has been widely used to improve tolerance to biotic and abiotic stresses.To explore the mechanisms underlying improvements in drought resistance afforded by grafting,we investigated the changes in growth,gas exchange,ribulose-1,5-bisphosphate carboxylase/oxygenase(Rubisco)expression and activity,sugar components,and the relative expression of photosynthesis-related genes of chrysanthemum(Chrysanthemum morifolium Ramat.‘Hangbaiju')plants grafted onto Artemisia annua rootstocks under drought stress by withholding water for 6 d.The results revealed that the growth declines in the grafted chrysanthemums were relatively lower than those of the non-grafted plants under drought stress,and net photosynthetic rate,stomatal conductance,water use efficiency,and transpiration rate in the non-grafted chrysanthemums were significantly decreased.Moreover,the intercellular CO_2 concentrations were significantly increased compared with the grafted plants at 5 and 6 d following drought stress.The grafted plants exhibited higher relative expression of the Cmrbc L,Cmrbc S,Cmpsa B,and Cmcab genes,as well as higher Rubisco activity and chlorophyll content under the drought treatment.Sugar accumulation also increased under drought stress,particularly in the non-grafted plants.This result suggested that non-grafted chrysanthemums were less able to resist dehydration,and repressed the genes encoding the expression of photosynthetic components.In conclusion,using A.annua rootstock could alleviate drought stress in chrysanthemums by improving gas exchange capacity and maintaining Cmrbc L,Cmrbc S,Cmcab,and Cmpsa B gene expression,thereby increasing Rubisco activity and improving photosynthetic performance.CHEN Yue SUN Xianzhi ZHENG Chengshu ZHANG Sheng YANG Jinghui 2018Horticultural Plant Journal2018,4,3:3
