3Mapping loci controlling fatty acid profiles, oil and protein content by genome-wide association study in Brassica napus显示文摘Optimizing the profile and quantity of fatty acids in rapeseed(Brassica napus L.) is critical for maximizing the value of edible oil and biodiesel. However, selection of these complex seed quality traits is difficult before haplotypes controlling their contents are identified. To efficiently identify genetic loci influencing these traits and underlying candidate genes and networks, we performed a genome-wide association study(GWAS) of eight seed quality traits(oil and protein content, palmitic, stearic, oleic, linoleic, eicosenoic and erucic acids content). The GWAS population comprised 370 diverse accessions, which were phenotyped in five environments and genotyped using 60K SNP arrays. The results indicated that oil and protein contents generally showed negative correlations, while fatty acid contents showed positive or negative correlations,with palmitic and erucic acid contents directly affecting oil content. Seven SNPs on five chromosomes were associated with both seed oil and protein content, and five genes orthologous to genes in Arabidopsis thaliana were predicted as candidates. From resequencing data, besides known haplotypes in Bna A.FAE1.a and Bna C.FAE1.a, three accessions harboring a new haplotype conferring moderate erucic acid content were identified. Interestingly, in a haplotype block, one haplotype was associated with high palmitic acid content and low oil content, while the others showed the reverse effects. This finding was consistent with a negative correlation between palmitic acid and oil contents, suggesting historical selection for high oil content. 5Genome assembly of the Brassicaceae diploid Orychophragmus violaceus reveals complex whole-genome duplication and evolution of dihydroxy fatty acid metabolism显示文摘Orychophragmus violaceus is a Brassicaceae species widely cultivated in China,particularly as a winter cover crop in northern China because of its low-temperature tolerance and low water demand.Recently,O.violaceus has also been cultivated as a potential industrial oilseed crop because of its abundant 24-carbon dihydroxy fatty acids(diOH-FAs),which contribute to superior high-temperature lubricant properties.In this study,we performed de novo assembly of the O.violaceus genome.Whole-genome synteny analysis of the genomes of its relatives demonstrated that O.violaceus is a diploid that has undergone an extrawhole-genome duplication(WGD)after the Brassicaceae-specific a-WGD event,with a basic chromosome number of x=12.Formation of diOH-FAs is hypothesized to have occurred after the WGD event.Based on the genome and the transcriptome data from multiple stages of seed development,we predicted that OvDGAT1-1 and OvDGAT1-2 are candidate genes for the regulation of diOH-FA storage in O.violaceus seeds.These results may greatly facilitate the development of heat-tolerant and eco-friendly plant-based lubricants using O.violaceus seed oil and improve our understanding of the genomic evolution of Brassicaceae.Fan Huang Peng Chen Xinyu Tang Ting Zhong Taihua Yang Chinedu Charles Nwafor Chao Yang Xianhong Ge Hong An Zaiyun Li Edgar B.Cahoon Chunyu Zhang 2023Plant Communications2023,4,2:0

7Effect of nerve growth factor on angiogenesis in a rat model of sciatic nerve regeneration显示文摘BACKGROUND:It remains to be determined whether nerve growth factor(NGF) can promote angiogenesis in regenerating peripheral nerves during repairing peripheral nerve injury. OBJECTIVE:To evaluate the effects of NGF on angiogenesis,and to analyze the influencing mechanisms of NGF,according to the expression patterns of CD34,von Willebrand factor(vWF), vascular endothelial cell growth factor(VEGF),and the NGF receptor TrkA in proliferating vascular endothelial cells from a rat model of sciatic nerve injury. DESIGN,TIME AND SETTING:Randomized,controlled study performed at the Research Institute of Field Surgery,Daping Hospital affiliated to the Third Military Medical University of Chinese PLA,between October 2003 and July 2005. MATERIALS:Forty-five healthy,adult,Wistar rats underwent sciatic nerve injury.The rats were randomly divided into four groups:NGF+chitosan(n=15),NGF+chitosan+anti-VEGF(n=10), chitosan(n=10),and physiological saline(n=10). METHODS:A 1-cm defected sciatic nerve was bridged with a silica gel conduit.NGF+chitosan group:100μL chitosan and 5μL NGF(20 mg/L) were injected into the silica gel conduit;NGF + chitosan+anti-VEGF group:an additional 5μL anti-VEGF monoclonal antibody(1 g/L) was injected into the silica gel conduit;chitosan group:100μL chitosan and 5μL physiological saline were injected into the silica gel conduit;physiological saline group:only 5μL physiological saline was injected into the silica gel conduit. MAIN OUTCOME MEASURES:CD34 and vWf were used to label blood capillaries and large-diameter blood vessels in the regenerating peripheral nerves,respectively.At day 14 following surgery,immunohistochemistry was used to detect and semi-quantitatively analyze expressions of CD34,vWf,VEGF,and TrkA in proliferating vascular endothelial cells in the regenerating sciatic nerve.A confocal laser microscope was used to determine co-expression. RESULTS:Expressions of TrkA,CD34,vWf,and VEGF in the NGF+chitosan group were significantly greater than the physiological saline and chitosan groups(P<0.05-0.01). Expressions of CD34 and VEGF in the NGF+chitosan+anti-VEGF group were completely inhibited,while expressions of vWf and TrkA gradually decreased,compared with the NGF + chitosan group(P<0.01).Confocal microscopy revealed strong co-expression of VEGF and CD34 in the regenerating sciatic nerve,and CD34 expression positively correlated with VEGF expression.In addition,VEGF expression was greater than CD34 expression,and coexpression of VEGF and vWf was also strong.
