1Establishment of a transgenic mouse model with liver-specific expression of secretory immunoglobulin D显示文摘Mutation of mevalonate kinase(MVK) is thought to account for most cases of hyperimmunoglobulinemia D syndrome(HIDS) with recurrent fever.However,its mechanism and the relationship between elevated serum immunoglobulin D(IgD) and the clinical features of HIDS are unclear.In this study,we generated by fusion PCR a vector to express high levels of chimeric secretory IgD(csIgD) specifically in the liver.We then generated seven founder lines of transgenic mice by co-microinjection,and verified them using genomic PCR and Southern blotting.We detected the expression of csIgD by reverse transcription PCR,quantitative PCR,western blotting,and enzyme-linked immunosorbent assays.We demonstrated that csIgD could be specifically and stably expressed in the liver.We used flow cytometry to show that overexpression of csIgD in the bone marrow and spleen cells had no effect on B cell development.Morphologic and anatomical observation of the transgenic mice revealed skin damage,hepatosplenomegaly,and nephromegaly in some transgenic mice;in these mice,pathological sections showed high levels of cell necrosis and protein-like sediments in the liver,spleen,and kidney.We demonstrated that the genomic insertion sites of the transgenes did not disrupt the MVK gene on mouse chromosome 5.This transgenic mouse will be useful to explore the pathogenesis of HIDS.WANG Ping WEI ZhiGuo YAN BoWen HUANG Tan GOU KeMian DAI YunPing ZHENG Min WANG MeiLi CHENG XueQian WANG XiFeng XU Chen SUN Yi 2012Science China(Life Sciences)2012,55,3:6
2Animal diversities and characteristics of environmental change revealed by skeletons unearthed at Zhongba Site of Chongqing City,China显示文摘Nearly 200000 animal skeletons are unearthed in the Unit T0202 from Zhongba Site of Zhongxian County.According to the analysis of 129165 specimens,these skeletons may be classified into 5 kinds, namely,Mammalia,Osteichthys,Aves,Amphibian and Reptilia,which belong to 13 orders,28 families and 42 genera.In this paper,based on archeological dating and AMS 14 C data,through statistically analyzing the unearthed skeletons and studying the change of the smallest individual numbers,these research results detected the following:1)In almost all the time of 2370―200 BC,in Zhongba region, some animals distributed widely,such as Muntiacus sp,Elaphodus cephalophus and Muntiacus sp. inhabiting in glade and grassland,Scrofa sp.and Canis familiaris raised by the ancient people,rodentia rabbit and Rattus rattus,which suggested that a fairly good ecological environment of forest and grassland was preserved at that period and the predecessors began to raise domestic animals from 1750 to 1000 BC,which has lasted until today.2)Rhinoceros only lived during 2000―1750 BC,1000― 700 BC and 700―500 BC,which indicates that the ecological environment of grassland and wetland might be better in these phases.3)Macaca sp.and Ursus arctos appeared only after 1750―1000 BC, which may show that the forest condition is better for animals to live during 1750―200 BC.4)The smallest individual numbers of Bublus sp.,bos sp.and otters emerged during about 2370―1750 BC, which perhaps infers that water area during the early period was wider than that of the late period.5) Since skeletons of rhinoceros are discovered in strata of Zhongba Site during 2000―1750 BC and 1000―500 BC,according to the climate and ecology environment which rhinoceros live in now,the average annual temperature and precipitation during 2000―1750 BC and 1000―500 BC are supposed to be probably higher than that of today.Although Zhu Kezhen considered that the first low temperature period in the past 5000 years of China was between 1100 BC and 850 BC,massive pollen of Morus, Ulmus,Fagus,Quercus,Castanea,etc.were found in Dajiuhu peat at that time,which should indicate that the climate was still moderate for living things to live at least in Dajiuhu basin and Zhongba Site.6) Because skeletons of Gervus albirostris were unearthed during 310―200 BC at Zhongba Site,based onthe climate and the ecological environment those animals live in now,the average annual temperature and the average annual precipitation in Zhongba area between 310 BC and 200 BC should be lower than those of today,which is confirmed by the TOC research of Zhongba Site strata.ZHU Cheng MA ChunMei OUYANG Jie LI ZhongXuan YIN Qian SUN ZhiBin HUANG YunPing R.K.FLAD LI Lan LI YuMei 2008Chinese Science Bulletin2008,53,S1:4
3On conformal measures for infinitely renormalizable quadratic polynomials显示文摘We study a conformal measure for an infinitely renormalizable quadratic polynomial. We prove that the conformal measure is ergodic if the polynomial is unbranched and has complex bounds. 4Carbon and nitrogen stable isotope analyses of mammal bone fossils from the Zhongba site in the Three Gorges Reservoir region of the Yangtze River,China显示文摘Based on AMS 14 C dating data,carbon and nitrogen isotope analyses were conducted on mammal bone collagen of deer,cattle and pigs from the Zhongba site in the Three Gorges Reservoir region of the Yangtze River.These analyses were conducted to reconstruct palaeodiets of mammals,palaeoecology,palaeoenviroment and previous human activities in the study area.Results show that the collagen loss of bone did not change the in vivo isotopic composition of carbon and nitrogen stable isotopes,and most of the bone fossils were well preserved.The bone collagen of samples from deer had a mean δ 13 C of-23.1‰ and a mean δ 15 N of 4.7‰,suggesting that deer subsisted in a closed habitat and fed on branches and leaves.The bone collagen of cattle had a mean δ 13 C of-19.6‰ and a mean δ 15 N of 5.2‰,which indicates that cattle subsisted in an open habitat and fed on grasses and stems.The δ 13 C values show that both deer and cattle fed on C 3 plants and lived in the same ecosystem,but the t-test results show that deer δ 13 C and δ 15 N values were both more negative than those of cattle,indicating that they inhabited different niches.The δ 13 C and δ 15 N values of cattle partially overlapped those of deer,suggesting some competition in diets between them.The t-tests show that the δ 13 C and δ 15 N values of pigs were more positive than those of cattle and deer,which signifies that pigs occupied a higher trophic level compared to cattle and deer.The wide range of pig δ 13 C values demonstrates that pig trading had been taking place from early Neolithic Age to late Bronze Age.There were no significant differences in deer δ 13 C and δ 15 N values among different archaeological periods,making it clear that climatic,ecological and environmental conditions were kept relatively stable from 2200 to 4200 a BP.This stability may have been responsible for the extensive and complete cultural layers at the Zhongba site.The minimum number of samples required to estimate the mean δ 13 C values of deer,pigs and cattle are 8,73 and 16,respectively,and for mean δ 15 N values of deer,pigs and cattle,the minimum numbers are 4,5 and 6,respectively.TIAN XiaoSi ZHU Cheng SUN ZhiBin SHUI Tao HUANG YunPing FLAD Rowan Kimon LI YuMei 2011Chinese Science Bulletin2011,56,2:2

6The effect of end-capping groups in A-D-A type non-fullerene acceptors on device performance of organic solar cells显示文摘A series of novel wide bandgap small molecules(IFT-ECA, IFT-M, IFT-TH and IFT-IC) based on the A-D-A structure with indenofluorene core, thiophene bridge, and different electron-deficient end-capping groups, were synthesized and used as non-fullerene acceptors in organic solar cells. The influences of end-capping groups on the device performance were studied.The four materials exhibited different physical and chemical properties due to the variation of end-capping groups, which further affect the exciton dissociation, charge transport, morphology of the bulk-heterojunction films and device performance. Among them, IFT-IC-based device delivered the best power conversion efficiency of 7.16% due to proper nano-scale phase separation morphology and high electron mobility, while the devices based on the other acceptors achieved lower device performance(4.14% for IFT-TH, <1% for IFT-ECA and IFT-M). 8Design, synthesis and photovoltaic properties of a series of new acceptor-pended conjugated polymers显示文摘A series of novel acceptor-pended conjugated polymers featuring a newly developed carbazole-derived unit are designed and synthesized. The relationships between chemical structure and optoelectronic properties of the polymers are systematically investigated.The control of UV-Vis absorption spectra and energy levels in resulting polymers are achieved by introducing suitable pended acceptor units. The photovoltaic properties of the resulting polymers are evaluated by blending the polymers with(6,6)-phenyl-C71-butyric acid methyl ester. The resulting solar cells exhibit moderate performances with high open-circuit voltage. 9Effect of behavior training on learning and memory of young rats with fetal growth restriction显示文摘Objective: To investigate the effect of behavior training on the learning and memory of young rats with fetal growth restriction (FGR). Methods: The model of FGR was established by passive smoking method to pregnant rats. The new-born rats were divided into FGR group and normal group, and then randomly subdivided into trained and untrained group respectively. Morris water maze behavior training was performed on postnatal months 2 and 4, then learning and memory abilities of young rats were measured by dark-avoidance testing and step-down testing. Results: In the dark-avoidance and step-down testing, the young rats’ performance of FGR group was worse than that of control group, and the trained group was better than the untrained group significantly. Conclusion: FGR young rats have descended learning and memory abilities. 10Effect of behavior training on learning,memory and the expression of NR2B,GluR1 in hippocampus of rats offspring with fetal growth restriction显示文摘Objective:To study effects of behavior training on learning,memory and the expression of NR2B,GluR1 in hippocampus of rat's offspring with fetal growth restriction(FGR).Methods:The rat model of FGR was established by passive smoking method.The rats offspring were divided into the FGR group and the control group,then randomly divided into the trained and untrained group,respectively. Morris water maze test was proceeded on postnatal month(PM2/4)as a behavior training method,then the learning-memory of rats was detected through dark-avoidance and step-down tests.The expressions of NR2B and GluR1 subunits in hippocampal CA1 and CA3 areas were detected by immunohistochemical method.Results:In the dark-avoidance and step-down tests,the performance record of rats with FGR was worse than that of control rats,and the behavior-trained rats was better than the untrained rats,when the FGR model and training factors were analyzed singly.The model factor and training factor had significant interaction(P<0.05).The expressions of NR2B and GluR1 subunits in hippocampal CA1 and CA3 areas of rats with FGR reduced.In contrast,the expressions of GluR1 and NR2B subunits in CA1 area of behavior-trained rats increased,when the FGR model and training factors were analyzed singly. 12Overlapped Rectangle Image Representation and Its Application to Exact Legendre Moments Computatio显示文摘Linear quadtree is a popular image representation method due to its convenient imaging procedure. However, the excessive emphasis on the symmetry of segmentation, i.e. dividing repeatedly a square into four equal sub-squares, makes linear quadtree not an optimal representation. In this paper, a no-loss image representation, referred to as Overlapped Rectangle Image Representation (ORIR), is presented to support fast image operations such as Legendre moments computation. The ORIR doesn’t importune the symmetry of segmentation, and it is capable of representing, by using an identical rectangle, the information of the pixels which are not even adjacent to each other in the sense of 4-neighbor and 8-neighbor. Hence, compared with the linear quadtree, the ORIR significantly reduces the number of rectangles required to represent an image. Based on the ORIR, an algorithm for exact Legendre moments computation is presented. 13Diets, eco-environments and seasonal variations recorded in the oxygen and carbon isotopic compositions of mammal tooth enamel from the Shunshanji site,Sihong County,Jiangsu Province,China显示文摘The carbon and oxygen isotopic compositions of the tooth enamel of mammals,including deer,wild pigs,buffaloes and domesticated pigs from the Shunshanji site,Sihong County,Jiangsu Province,China,were analyzed to reconstruct the mammals’ecoenvironments and diets,and to evaluate seasonal variations in the study area.Carbon isotopic compositions of buck samples revealed that the deer ate completely C3 plants and the environments they inhabited were relatively open and that wild pigs ate primarily C3 plants.Oxygen isotopic compositions indicated that the body sources of these two mammals were different,i.e.the deer and pigs lived in different niches within a relatively similar ecosystem.Modern domesticated pigs were isotopically more positive than the ancient wild pigs in carbon 13C values,suggesting the former ingested more C3 plants relative to the latter.Although the 18O data showed modern domesticated and ancient pigs had similar oxygen isotope compositions,their water sources were different.The carbon and oxygen isotopic patterns of premolar microsamples of ancient and modern buffaloes indicated that the plants ingested by the ancient buffalo varied with seasonal shifts,but plants ingested by the modern buffalo were relatively constant.The eco-environment of the modern buffalo was more open,warmer and drier than eco-environment of the ancient buffalo,which may be the result of the deforestation and other human activities.Ancient and modern seasonal changes were clearly recorded in the isotopic patterns and the seasonal variation amplitudes of the ancient and modern eco-environments were similar.TIAN XiaoSi ZHU Cheng SHUI Tao HUANG YunPing 2013Chinese Science Bulletin2013,58,31:0
14Impairments of spatial learning and memory in rat offspring with fetal growth restriction显示文摘Objective:Throughout the world,fetal growth restriction(FGR) is one of the most severe complications occurring during pregnancy. It is subsequently associated with neurologic abnormalities in chldren. Our aim was to investigate the spatial learning and memory ability of rat offspring born with FGR. Methods:A rat model of FGR was constructed using the method of passive smoking. Spatial learning and memory were studied in rat offspring born with FGR by assessing the animals’ performance using the Morris water maze task. Results: At 1-and 2-months of age,both female and male offspring rats showed impairment of performance,while at 4 months of age,only female rats showed impaired performance. The FGR offspring spent a longer time swimming and used inefficient strategies(P < 0.05,respectively). However,there were no significant maze performance FGR effects in the 4 month old male rats. In all groups of FGR offspring,irrespective of age or sex,the time spent in the platform quadrant by the rat was significantly less than that in the control group(P < 0.05). Conclusion: The Morris water maze performance decreased in rat offspring born with FGR. 15Two-Stage Optimal Scheduling of Community Integrated Energy System显示文摘From the perspective of a community energy operator,a two-stage optimal scheduling model of a community integrated energy system is proposed by integrating information on controllable loads.The day-ahead scheduling analyzes whether various controllable loads participate in the optimization and investigates the impact of their responses on the operating economy of the community integrated energy system(IES)before and after;the intra-day scheduling proposes a two-stage rolling optimization model based on the day-ahead scheduling scheme,taking into account the fluctuation of wind turbine output and load within a short period of time and according to the different response rates of heat and cooling power,and solves the adjusted output of each controllable device.The simulation results show that the optimal scheduling of controllable loads effectively reduces the comprehensive operating costs of community IES;the two-stage optimal scheduling model can meet the energy demand of customers while effectively and timely suppressing the random fluctuations on both sides of the source and load during the intra-day stage,realizing the economic and smooth operation of IES.Ming Li Rifucairen Fu Tuerhong Yaxiaer Yunping Zheng Abiao Huang Ronghui Liu Shunfu Lin 2024Energy Engineering2024,121,2:0
16Comparative transcriptome analysis revealed the influence of sucrose on flavor and taste quality of Coffea arabica and C.canephora varieties显示文摘Coffea arabica(Arabica)and C.canephora(Robusta)are the most popular coffee species in the world,accounting for 99%of overall consumption of coffee beans.Arabica generally possesses better coffee quality than Robusta partly due its higher sucrose content.The flavor and taste(FT)of coffee,as important aspects of coffee quality,are mainly affected by the content of caffeine,phenolic acid and terpenoid compounds,which use sucrose as an important precursor.However,how sucrose affects the coffee FT remains unclear.In this study,coffee beans at different developmental stages from Arabica#161(A161,high-sucrose variety)and Robusta#6(R6,low-sucrose variety)were sampled to perform transcriptomic analysis.Most differentially expressed genes(DEGs)between them were enriched in sucrose-related metabolisms and the FTrelated metabolism processes:caffeine biosynthesis,phenylpropanoid and flavonoid pathway,terpenoid metabolism.Thirty-four candidate DEGs probably contributed to the higher content of sucrose,anthocyanin and linalool in A161,and higher content of caffeine and carotenoid in R6.Generally,sucrose-related metabolisms were strongly associated with FT-related substance accumulation.The content of sucrose and its influence on the downstream secondary metabolism probably play important roles in the FT quality of coffee beans.Our results provide efficient targets for investigation regarding the influence of sucrose on FT quality of coffee beans.Lifang Huang Lisong Hu Yu Fan Xiaoyang Wang Yunping Dong Fupeng Li Yuzhou Long Lin Yan Jie Meng 2021Beverage Plant Research2021,1,1:0
