| 1 | Curative effect of warming kidney and fortifying spleen recipe on diarrhea-predominant irritable bowel syndrome显示文摘OBJECTIVE:To observe the curative effect of a recipe for warming the kidney and fortifying the spleen on diarrhea-predominant irritable bowel syndrome(IBS-D).METHODS:This multi-center,double-blind,randomized,and controlled trial included 240 patients that met the inclusion criterion and were then divided into two groups of 120.Patients in the treatment group(group A)took modified Sishen Wan orally for warming the kidney and fortifying the spleen and patients in the control group(group B)took a placebo,Chao Maiya,for 4 weeks.28 days after withdrawal,there was a 6-month follow-up to observe patient recurrence condition.The total effective rate,curative effect,and recurrence rate were evaluated after treatment.RESULTS:There was statistical difference(P<0.01)between the two groups in total effective rate(92.24%in the treatment group and 49.07%in the control group),in curative effect of TCM syndrome(90.52%and 47.22%,respectively),and in the recurrence rate(15.79%and 56.86%,respectively)within6 months after treatment.CONCLUSION:Modified Sishen Wan,for warming the kidney and fortifying the spleen,can effectively treat IBS-D and better control its recurrence. | Xiaolan Su Yanping Tang Jing Zhang Yuebin Dong Wei Wei Yibing Bai Yong Liu | 2013 | Journal of Traditional Chinese Medicine2013,33,5: | 22 |

| 3 | Growth and metal uptake of energy sugarcane (Saccharum spp.) in different metal mine tailings with soil amendments显示文摘A pot experiment was conducted to investigate the feasibility of growing energy sugarcane(Saccharum spp.) in three different metal mine tailings(Cu, Sn and Pb/Zn tailings) amended with uncontaminated soil at different mixing ratios. The results indicated that sugarcane was highly tolerant to tailing environments. Amendments of 20% soil to Sn tailings and 30% soil to Cu tailings could increase the biomass of cane-stem for use as the raw material for bioethanol production. Heavy metals were mostly retained in roots, which indicated that sugarcane was useful for the stabilization of the tailings. Bagasse and juice, as the most valuable parts to produce bioethanol, only accounted for 0.6%– 3% and 0.6%–7% of the total metal content. | 15 | A telomere-to-telomere reference genome provides genetic insight into the pentacyclic triterpenoid biosynthesis in Chaenomeles speciosa显示文摘Chaenomeles speciosa(2n=34),a medicinal and edible plant in the Rosaceae,is commonly used in traditional Chinese medicine.To date,the lack of genomic sequence and genetic studies has impeded efforts to improve its medicinal value.Herein,we report the use of an integrative approach involving PacBio HiFi(third-generation)sequencing and Hi-C scaffolding to assemble a high-quality telomere-to-telomere genome of C.speciosa.The genome comprised 650.4 Mb with a contig N50 of 35.5 Mb.Of these,632.3 Mb were anchored to 17 pseudo-chromosomes,in which 12,4,and 1 pseudo-chromosomes were represented by a single contig,two contigs,and four contigs,respectively.Eleven pseudo-chromosomes had telomere repeats at both ends,and four had telomere repeats at a single end.Repetitive sequences accounted for 49.5%of the genome,while a total of 45515 protein-coding genes have been annotated.The genome size of C.speciosa was relatively similar to that of Malus domestica.Expanded or contracted gene families were identified and investigated for their association with different plant metabolisms or biological processes.In particular,functional annotation characterized gene families that were associated with the biosynthetic pathway of oleanolic and ursolic acids,two abundant pentacyclic triterpenoids in the fruits of C.speciosa.Taken together,this telomere-to-telomere and chromosome-level genome of C.speciosa not only provides a valuable resource to enhance understanding of the biosynthesis of medicinal compounds in tissues,but also promotes understanding of the evolution of the Rosaceae. | Shaofang He Duanyang Weng Yipeng Zhang Qiusheng Kong Keyue Wang Naliang Jing Fengfeng Li Yuebin Ge Hui Xiong Lei Wu De-Yu Xie Shengqiu Feng Xiaqing Yu Xuekui Wang Shaohua Shu Zhinan Mei | 2023 | Horticulture Research2023,10,10: | 0 |
| 16 | Synthesis of haptens and production of antibodies to bisphenol A显示文摘Three immunizing haptens of bisphenol A(BPA), including two new haptens, were used to produce highly sensitive and specific polyclonal antibodies. The spacer arms of haptens for coupling to the protein carrier were located at different positions in BPA, and different length spacer arms were tested. Highly sensitive polyclonal antibodies were obtained and characterized using indirect competitive enzyme-linked immunosorbent assay(ic ELISA). Under optimized conditions, the half maximal inhibitory concentration(IC50) value of the best polyclonal antibody was 2.1 mg·L^(-1), based on coating heterogeneous antigens, and this optimal polyclonal antibody was highly sensitive toward BPA and displayed negligible crossreactivity with bisphenol B and bisphenol E. A sensitive ic ELISA method utilizing the polyclonal antibody was developed for the determination of BPA in milk. In spiked samples(5, 10 and 20 mg·L^(-1)), the recovery ranged from 80% to 102% with a coefficient of variation(CV) value below 15.8%. The limit of detection of ic ELISA was1.95 mg·L^(-1). | 18 | Laparoscopic management of benign splenic tumors in children显示文摘Background The splenic tumor is relatively rare in children.However,diagnosing and managing this disease remain controversial.This study aimed to evaluate the clinical characteristics and pathological features of benign splenic tumors and to explore the feasibility and safety of laparoscopic splenectomy in children.Methods The clinical data of all patients who were diagnosed with benign splenic tumors and admitted to our center between January 2014 and December 2020 were analyzed retrospectively.Factors that were documented included demographic criteria,clinical manifestations,preoperative examinations,surgical methods,histopathological characteristics,postoperative complications and outcomes.Results Totally,24 consecutive patients with a mean age of 10.21±2.59 years were operated laparoscopically.Nine patients presented with recurrent abdominal pain,vomiting or backache.The remaining 15 cases were asymptomatic and were identified incidentally.Laparoscopic total splenectomy(LTS)was performed in 12 cases,laparoscopic partial splenectomy(LPS)in 9 cases and deroofing of splenic cyst in 3 cases.No significant differences were observed in operation time,intraoperative bleeding and postoperative abdominal drainage between the LTS and LPS groups.Histopathology examination reported 12 cases of splenic epithelial cyst,5 cases of splenic lymphangioma or lymphatic malformation,3 cases of splenic hamartoma,3 cases of splenic littoral cell angioma and 1 case of splenic hemangioma.No postoperative complications and tumor recurrence were observed.Conclusions It is still difficult to precisely diagnose the splenic tumor preoperatively.Laparoscopic splenectomy is safe and feasible in treating the benign splenic tumors,and partial splenectomy is of great significance in preserving the immune function in children. | Chen Qingjiang Linyan Wang Xiaoli Chen Yuebin Zhang Lifeng Zhang Kun Zhu Qixing Xiong Zhigang Gao | 2022 | World Journal of Pediatric Surgery2022,5,4: | 0 |
| 19 | Zipper head mechanism of telomere synthesis by human telomerase显示文摘Telomerase,a multi-subunit ribonucleoprotein complex,is a unique reverse transcriptase that catalyzes the processive addition of a repeat sequence to extend the telomere end using a short fragment of its own RNA component as the template.Despite recent structural characterizations of human and Tetrahymena telomerase,it is still a mystery how telomerase repeatedly uses its RNA template to synthesize telomeric DNA.Here,we report the cryo-EM structure of human telomerase holoenzyme bound with telomeric DNA at resolutions of 3.5 Åand 3.9 Åfor the catalytic core and biogenesis module,respectively.The structure reveals that a leucine residue Leu980 in telomerase reverse transcriptase(TERT)catalytic subunit functions as a zipper head to limit the length of the short primer–template duplex in the active center.Moreover,our structural and computational analyses suggest that TERT and telomerase RNA(hTR)are organized to harbor a preformed active site that can accommodate short primer–template duplex substrates for catalysis.Furthermore,our findings unveil a double-fingers architecture in TERT that ensures nucleotide addition processivity of human telomerase.We propose that the zipper head Leu980 is a structural determinant for the sequence-based pausing signal of DNA synthesis that coincides with the RNA element-based physical template boundary.Functional analyses unveil that the non-glycine zipper head plays an essential role in both telomerase repeat addition processivity and telomere length homeostasis.In addition,we also demonstrate that this zipper head mechanism is conserved in all eukaryotic telomerases.Together,our study provides an integrated model for telomerase-mediated telomere synthesis. | Futang Wan Yongbo Ding Yuebin Zhang Zhenfang Wu Shaobai Li Lin Yang Xiangyu Yan Pengfei Lan Guohui Li Jian Wu Ming Lei | 2021 | Cell Research2021,31,12: | 0 |
| 20 | Transmission of Carbapenem Resistance Between Human and Animal NDM-Positive Escherichia coli Strains显示文摘Although carbapenem use is prohibited in animals in China,carbapenem-resistant Escherichia coli(CREC),especially New Delhi metallo-β-lactamase(NDM)-producing strains,are widely prevalent in foodproducing animals.At present,the impact of livestock-associated CREC strains on human populations at the national level is unknown.Here,we conduct a retrospective cross-sectional study to investigate the prevalence of CREC from clinical settings across 22 Chinese provinces or municipalities and analyze anthropogenic factors associated with their presence.We also ascertain the blaNDMand blaKPCabundance among pig and chicken farms and present a detailed genomic framework for CREC of animal and human origin.Overall,631/29799(2.1%)clinical Escherichia coli(E.coli)isolates were identified as CREC.Multivariable analysis revealed that being male,an age below 1,an age between 13 and 18,provinces with greater chicken production,and provinces with higher pig production were associated with higher odds of CREC infection.In general,73.8%(n=45/61)of pig farms and 62.2%(n=28/45)of chicken farms had a blaNDMabundance of 1×10^(-5)to 1×10^(-3)and 1×10^(-3)to 1×10^(-2),respectively.Among all the Chinese NDM-positive E.coli(n=463)available at the National Center for Biotechnology Information(NCBI),the genomic analysis revealed that blaNDM-5and Inc X3 were the predominant carbapenemase gene-plasmid combination,while a highly homogeneous relationship between NDM-positive isolates from humans and animals was demonstrated at the plasmid and core genome levels.All the findings suggest frequent CREC transmission between humans and animals,indicating that further discussions on the use of antibiotics in animals and humans are needed,both in China and across the globe. | Yingbo Shen Fupin Hu Yongqiang Wang Dandan Yin Lu Yang Yiqiang Chen Chunyan Xu Jiyun Li Junyao Jiang Xueyang Wang Yulin Fu Dongyan Shao Dejun Liu Tengfei Ma Chang Cai Zhangqi Shen Shaolin Wang Juan Li Rong Zhang Yuebin Ke Congming Wu Jianzhong Shen Timothy R.Walsh Yang Wang | 2022 | Engineering2022,8,8: | 0 |
