1Transgenic rhesus monkeys carrying the human MCPH1 gene copies show human-like neoteny of brain development显示文摘Brain size and cognitive skills are the most dramatically changed traits in humans during evolution and yet the genetic mechanisms underlying these human-specific changes remain elusive.Here,we successfully generated 11 transgenic rhesus monkeys(8 first-generation and 3 second-generation)carrying human copies of MCPH1,an important gene for brain development and brain evolution.Brain-image and tissue-section analyses indicated an altered pattern of neural-cell differentiation,resulting in a delayed neuronal maturation and neural-fiber myelination of the transgenic monkeys,similar to the known evolutionary change of developmental delay(neoteny)in humans.Further brain-transcriptome and tissue-section analyses of major developmental stages showed a marked human-like expression delay of neuron differentiation and synaptic-signaling genes,providing a molecular explanation for the observed brain-developmental delay of the transgenic monkeys.More importantly,the transgenic monkeys exhibited better short-term memory and shorter reaction time compared with the wild-type controls in the delayed-matching-to-sample task.The presented data represent the first attempt to experimentally interrogate the genetic basis of human brain origin using a transgenic monkey model and it values the use of non-human primates in understanding unique human traits.Lei Shi Xin Luo Jin Jiang Yongchang chen Cirong Liu Ting Hu Min Li Qiang Lin Yanjiao Li Jun Huang Hong Wang Yuyu Niu Yundi Shi Martin Styner Jianhong Wang Yi Lu Xuejin Sun Hualin Yu Weizhi Ji Bing Su 2019National Science Review2019,6,3:12
2Exosome-loaded extracellular matrix-mimic hydrogel with anti-inflammatory property Facilitates/promotes growth plate injury repair显示文摘Growth plate cartilage has limited self-repair ability,leading to poor bone bridge formation post-injury and ultimately limb growth defects in children.The current corrective surgeries are highly invasive,and outcomes can be unpredictable.Following growth plate injury,the direct loss of extracellular matrix(ECM)coupled with further ECM depletion due to the inhibitory effects of inflammation on the cartilage matrix protein greatly hinder chondrocyte regeneration.We designed an exosome(Exo)derived from bone marrow mesenchymal stem cells(BMSCs)loaded ECM-mimic hydrogel to promote cartilage repair by directly supplementing ECM and anti-inflammatory properties.Aldehyde-functionalized chondroitin sulfate(OCS)was introduced into gelatin methacryloyl(GM)to form GMOCS hydrogel.Our results uncovered that GMOCS hydrogel could significantly promote the synthesis of ECM due to the doping of OCS.In addition,the GMOCS-Exos hydrogel could further promote the anabolism of chondrocytes by inhibiting inflammation and ultimately promote growth plate injury repair through ECM remodeling.Pengfei Guan Can Liu Denghui Xie Shichao Mao Yuelun Ji Yongchang Lin Zheng Chen Qiyou Wang Lei Fan Yongjian Sun 2022Bioactive Materials2022,7,4:3

5Polarization of plane wave propagating inside elastic hexagonal system solids显示文摘Based on the reported physical parameters for hexagonal system solids,we have calculated the effects of anisotropy on polarization of plane P-wave propagation.Herein we report the results of calculations and the newly observed physical phenomena.It is found that,for a given propagation,if the polarization is parallel to the wave vector,so also to the Poynting vector.In such a case,the phase velocity is identical to the energy velocity;the quasi P-wave degenerates to a pure P-wave along the propagation.It is also noted that if the polarization is parallel to the Poynting vector but not to the wave vector,the propagating wave cannot be a pure P-wave.Furthermore,the polarization in a quasi P-wave may deviate from the wave vector for more than 45°,but the deviation from the Poynting vector is always less than 45°.The energy velocity of a quasi SV-wave can be larger than that of the quasi P-wave in some propagation directions,even though the phase velocity of a quasi SV-wave may never be larger than either the phase velocity or energy velocity of the quasi P-wave.Finally,in case of parameters ε=0 and δ*≠0,the polarization of a quasi P-wave has an observed symmetry at a 45°phase angle.The anisotropy of a hexagonal system solid determines if a pure P-wave can be created and what the propagation direction is for a plane wave propagating inside such a hexagonal system solid.FA Lin ZHAO MeiShan LIU YongChang WANG Lei WANG YanQiang SUN JiGang 2014Science China(Physics,Mechanics & Astronomy)2014,57,2:2

13Low frequency noise reduction using stiff light composite panels显示文摘The experiment presented in this paper is to investigate and analyze the noise reduction at low frequency using stiff light composite panels. Since these composite panels are made of lightweight and stiff materials, this actuation strategy will enable the creation of composite panels for duct noise control without using traditional heavy structural mass. The results suggest that the mass-spring resonance absorption in the case of a comparatively stiff thick panel with a thin flexible plate is more efficient with minimum weight, when subjected to low-frequency (<500 Hz). 15Promoting Proton Migration Kinetics by Ni^(2+)Regulating Enables Improved Aqueous Zn-MnO_(2) Batteries显示文摘The energy storage behaviors of MnO_(2) for aqueous Zn-MnO_(2) batteries mainly depend on the Zn^(2+)/H^(+)intercalation but are limited by poor ion/electron migration dynamics and stability.Herein,a strategy is proposed that promoting proton migration kinetics ameliorates H^(+)storage activity by introducing Ni^(2+)intoγ-MnO_(2)(Ni-MnO_(2)).Ni^(2+)can lower the diffusion barrier of H^(+)and selectively induce the ion intercalation,thereby alleviating the electrostatic interaction with the lattice.Moreover,Ni^(2+)enables the adjacent[MnO6]octahedrons to have better electron conductivity.The Ni-MnO_(2) exhibits superior rate performance(nearly four times specific capacity compared with MnO_(2))and ultra-long-cycle stability(100%of capacity retention after 11000 cycles at 3.0 A g^(-1)).The calculation indicates that the Ni-MnO_(2) allows H^(+)migrate rapidly along the one-dimensional tunnel due to reduction of the activation energy caused by Ni^(2+)regulating,thus achieving excellent reaction kinetics.This work brings great potential for the development of high-performance aqueous Zn-MnO_(2) batteries.Jie Ji Jia Yao Yongchang Xu Houzhao Wan Bao Zhang Lin Lv Jingying Li Nengze Wang Zhaohan Zheng Jun Zhang Guokun Ma Li Tao Hanbin Wang Yi Wang Hao Wang 2023Energy & Environmental Materials2023,6,2:0

17Polypeptides inhibit HIV-1 replication by interfering viral Vpu-mediated tetherin degradation显示文摘Background:HIV-1 Vpu acts by counteracting the tethering function of tetherin and resulting in the release of HIV-1 virion.Disrupting Vpu-tetherin interactions may provide a promising new target for antiretroviral therapy.Methods:Polypeptides that covered the amino acid sequence on the interface of Vpu-tetherin complex were designed.Phenotypic susceptibilities and cellular toxicities to the polypeptides were measured.The mechanisms of the anti-HIV-1 polypeptides were determined by the Western blot analysis and laser confocal scanning.Seven 20-mer polypeptides from wild-type Vpu amino acid sequence were designed.Results:We report the design and identification of 3 novel anti-HIV-1 polypeptides that derived from Vpu se-quence which can efficiently inhibit HIV-1 infection.A pilot mechanism study showed that the active polypeptide could counteract Vpu-mediated tetherin downregulation.Laser confocal image scanning study showed that the polypeptides bound on the cell surface with a receptor specific binding manner,which may target tetherin that expressed on cell surface.Conclusion:Our work provided first evidence that counteracting Vpu-mediated tetherin downregulation could be a target for novel anti-HIV-1 drug design.Future works to provide direct evidence of inhibitors interact with teth-erin at atomic resolution and the development of small molecules inhibitors targeting Vpu-tetherin interactions may open a new avenue for novel antiretroviral therapy.Shuai Chang Lifeng Cai Yongchang Yang Binlian Sun Jingyun Li Jie Liu Lin Li 2023Infectious Medicine2023,2,3:0
