2Small molecule inhibitors reveal allosteric regulation of USP14 via steric blockade显示文摘The ubiquitin system is important for drug discovery,and the discovery of selective small-molecule inhibitors of deubiquitinating enzymes (DUBs) remains an active yet extremely challenging task. With a few exceptions,previously developed inhibitors have been found to bind the evolutionarily conserved catalytic centers of DUBs,resulting in poor selectivity. The small molecule IU1 was the first-ever specific inhibitor identified and exhibited surprisingly excellent selectivity for USP14 over other DUBs. However,the molecular mechanism for this selectivity was elusive. Herein,we report the high-resolution co-crystal structures of the catalytic domain of USP14 bound to IU1 and three IU1 derivatives. All the structures of these complexes indicate that IU1 and its analogs bind to a previously unknown steric binding site in USP14,thus blocking the access of the C-terminus of ubiquitin to the active site of USP14 and abrogating USP14 activity. Importantly,this steric site in USP14 is very unique,as suggested by structural alignments of USP14 with several known DUB X-ray structures. These results,in conjunction with biochemical characterization,Indicate a coherent steric blockade mechanism for U5P14 inhibition by compounds of the IU series. 3Design Schemes and Comparison Research of the End-effector of Large Space Manipulator显示文摘The end-effector of the large space manipulator is employed to assist the manipulator in handling and manipulating large payloads on orbit.Currently,there are few researches about the end-effector,and the existing end-effectors have some disadvantages,such as poor misalignment tolerance capability and complex mechanical components.According to the end positioning errors and the residual vibration characters of the large space manipulators,two basic performance requirements of the end-effector which include the capabilities of misalignment tolerance and soft capture are proposed.And the end-effector should accommodate the following misalignments of the mechanical interface.The translation misalignments in axial and radial directions and the angular misalignments in roll,pitch and yaw are ±100 mm,100 mm,±10°,±15°,±15°,respectively.Seven end-effector schemes are presented and the capabilities of misalignment tolerance and soft capture are analyzed elementarily.The three fingers-three petals end-effector and the steel cable-snared end-effector are the most feasible schemes among the seven schemes,and they are designed in detail.The capabilities of misalignment tolerance and soft capture are validated and evaluated,through the experiment on the micro-gravity simulating device and the dynamic analysis in ADAMS software.The results show that the misalignment tolerance capabilities of these two schemes could satisfy the requirement.And the translation misalignment tolerances in axial and radial directions and the angular misalignment tolerances in roll,pitch and yaw of the steel cable-snared end-effector are 30mm,15mm,6°,3° and 3° larger than those of the three fingers-three petals end-effector,respectively.And the contact force of the steel cable-snared end-effector is smaller and smoother than that of the three fingers-three petals end-effector.The end-effector schemes and research methods are beneficial to the developments of the large space manipulator end-effctor and the space docking mechanism.FENG Fei LIU Yiwei LIU Hong CAI Hegao 2012Chinese Journal of Mechanical Engineering2012,25,4:13
4Dihydroartemisinin regulates the immune system by promotion of CD8^+T lymphocytes and suppression of B cell responses显示文摘Artemisia annua is an anti-fever herbal medicine first described in traditional Chinese medicine 1,000 years ago.Artemisinin,the extract of A.annua,and its derivatives(dihydroartemisinin(DHA),artemether,and artesunate)have been used for the treatment of malaria with substantial efficacy.Recently,DHA has also been tested for the treatment of lupus erythematosus,indicating that it may function to balance the immune response in immunocompromised individuals.In the present study,the regulatory effect of artemisinin on the murine immune system was systematically investigated in mice infected with two different protozoan parasites(Toxoplasma gondii and Plasmodium berghei).Our results revealed that the mouse spleen index significantly increased(spleen enlargement)in the healthy mice after DHA administration primarily due to the generation of an extra number of lymphocytes and CD8^+T lymphocytes in both the spleen and circulation.DHA could increase the proportion of T helper cells and CD8^+T cells,as well as decrease the number of splenic and circulatory B cells.Further,DHA could reduce the production of proinflammatory cytokines.Our study revealed that apart from their anti-parasitic activity,artemisinin and its derivatives can also actively modulate the immune system to directly benefit the host.Ting Zhang Yiwei Zhang Ning Jiang Xu Zhao Xiaoyu Sang Na Yang Ying Feng Ran Chen Qijun Chen 2020Science China(Life Sciences)2020,63,5:12

6Stochastic processes in renewable power systems: From frequency domain to time domain显示文摘With the increasing penetration of renewable energy resources(RESs), the uncertainties of volatile renewable generations significantly affect the power system operation. Such uncertainties are usually modeled as stochastic variables obeying specific distributions by neglecting the temporal correlations. Conventional approaches to hedge the negative effects caused by such uncertainties are thus hard to pursue a trade-off between computation efficiency and optimality. As an alternative, the theory of stochastic process can naturally model temporal correlation in closed forms. Attracted by this feature, our research group has been conducting thorough researches in the past decade to introduce stochastic processes within renewable power systems. This paper summarizes our works from the perspective of both the frequency domain and the time domain, provides the tools for the analysis and control of power systems under a unified framework of stochastic processes, and discusses the underlying reasons that stochastic process-based approaches can perform better than conventional approaches on both computational efficiency and optimality. 8Serum macrophage migration inhibitory factor as a potential biomarker to evaluate therapeutic response in patients with allergic asthma:an exploratory study显示文摘Background:Previous studies have shown that macrophage migration inhibitory factor(MIF)is involved in the pathogenesis of asthma.This study aimed to investigate whether serum MIF reflects a therapeutic response in allergic asthma.Methods:We enrolled 30 asthmatic patients with mild-to-moderate exacerbations and 20 healthy controls,analyzing the parameter levels of serum MIF,serum total immunoglobulin E(tIgE),peripheral blood eosinophil percentage(EOS%),and fractional exhaled nitric oxide(FeNO).Lung function indices were used to identify disease severity and therapeutic response.Results:Our study showed that all measured parameters in patients were at higher levels than those of controls.After one week of treatment,most parameter levels decreased significantly except for serum tIgE.Furthermore,we found that serum MIF positively correlated with EOS%as well as FeNO,but negatively correlated with lung function indices.Receiver operator characteristic(ROC)curve analysis indicated that among the parameters,serum MIF exhibited a higher capacity to evaluate therapeutic response.The area under the curve(AUC)of MIF was 0.931,with a sensitivity of 0.967 and a specificity of 0.800.Conclusions:Our results suggested that serum MIF may serve as a potential biomarker for evaluating therapeutic response in allergic asthma with mild-to-moderate exacerbations.Huiyuan ZHU Shaochun YAN Jingshuo WU Zhong ZHANG Xiaolin LI Zheng LIU Xing MA Lina ZHOU Lin ZHANG Mingming FENG Yiwei GENG Aixin ZHANG Sabina JANCIAUSKIENE Aiguo XU 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,6:3
9A seismic gap on the Anninghe fault in western Sichuan, China显示文摘Through integrated analyses of time-varying patterns of regional seismicity, occurrence background of strong and large historical earthquakes along active faults, and temporal-spatial distribution of accu- rately relocated hypocenters of modern small earthquakes, this paper analyzes and discusses the im- plication of a 30-year-lasting seismic quiescence in the region along and surrounding the Anninghe and Zemuhe faults in western Sichuan, China. It suggests that the seismic quiescence for ML≥4.0 events has been lasting in the studied region since January, 1977, along with the formation and evaluation of a seismic gap of the second kind, the Anninghe seismic gap. The Anninghe seismic gap has the background of a seismic gap of the first kind along the Anninghe fault, and has resulted from evident fault-locking and strain-accumulating along the fault during the last 30 years. Now, two fault sections either without or with less small earthquakes exist along the Anninghe fault within the An- ninghe seismic gap. They indicate two linked and locked fault-sections, the northern Mianning section and the Mianning-Xichang section with lengths of 65 km and 75 km and elapsed time from the latest large earthquakes of 527 and 471 years, respectively. Along the Anninghe fault, characteristics of both the background of the first kind seismic gap and the seismicity patterns of the second seismic gap, as well as the hypocenter depth distribution of modern small earthquakes are comparable, respectively, to those appearing before the M=8.1 Hoh Xil earthquake of 2001 and to those emerging in the 20 years before the M=7.1 Loma Prieta, California, earthquake of 1989, suggesting that the Anninghe seismic gap is tending to become mature, and hence its mid- to long-term potential of large earthquakes should be noticeable. 10Preparation, microstructure and degradation performance of biomedical magnesium alloy fine wires显示文摘With the development of new biodegradable Mg alloy implant devices, the potential applications of biomedical Mg alloy fine wires are realized and explored gradually. In this study, we prepared three kinds of Mg alloy fine wires containing 4 wt% RE(Gd/Y/Nd) and 0.4 wt% Zn with the diameter less than 0.4 μm through casting, hot extruding and multi-pass cold drawing combined with intermediated annealing process. Their microstructures, mechanical and degradation properties were investigated. In comparison with the corresponding as-extruded alloy, the final fine wire has significantly refined grain with an average size of 3–4 μm, and meanwhile shows higher yield strength but lower ductility at room temperature. The degradation tests results and surface morphologies observations indicate that Mg–4Gd–0.4Zn and Mg–4Nd–0.4Zn fine wires have similar good corrosion resistance and the uniform corrosion behavior in SBF solution. 13Crystal structure of histidine-containing phosphocarrier protein from Thermoanaerobacter tengcongensis MB4 and the implications for thermostability显示文摘Protein thermostability is an inherent characteristic of proteins from thermophilic microorganisms,and therefore enables these organisms to survive at extreme temperatures.Although it is well-known that thermostable proteins are critical for the growth of thermophilic organisms,the structural basis of protein thermostability is not yet fully understood.The histidine-containing phosphocarrier (HPr) protein,a phosphate shuttle protein in the phosphoenolpyruvate-dependent sugar transport system (PTS) of bacterial species,is an ideal model for investigating protein thermostability with respect to its small size and deficiency in disulphide bonds or cofactors.In this study,the HPr protein from Thermoanaerobacter tengcongensis (TtHPr) is cloned and purified.Crystal structure with good quality has been determined at 2.3 resolution,which provides a firm foundation for exploring the thermostable mechanism.However,it shows that the crystal structure is conserved and no clue can be obtained from this single structure.Furthermore,detailed comparison of sequence and structure with the homologs from mesoor thermophilic bacteria shows no obvious rule for thermostability,but the extra salt-bridge existing only in thermophilic bacteria might be a better explanation for thermostability of HPr.Thus,mutations are performed to interrupt the salt-bridge in HPrs in thermophilic bacteria.Using site-directed mutations and the circular dichroism method,thermostability is evaluated,and the mutational variations are shown to have a faster denaturing rate than for wild-type viruses,indicating that mutations cause instability in the HPrs.Understanding the higher-temperature resistance of thermophilic and hyperthermophilic proteins is essential to studies on protein folding and stability,and is critical in engineering efficient enzymes that can work at a high temperature.FENG ChunYan GAO Feng LIU YiWei WANG GuiHua PENG Hao MA YanHe YAN JingHua GAO George Fu 2011Science China(Life Sciences)2011,54,6:2

15The effect of enzymes on release of trace elements in feedstuffs based on in vitro digestion model for monogastric livestock显示文摘Background: This experiment was conducted to study the effect of different feed enzymes(phytase,xylanase,β-glucanase) on release rate of trace elements(Fe,Cu,Mn and Zn) in 6 commonly used feedstuffs(corn,wheat,barley,soybean meal,wheat bran,wheat middlings) by using an in vitro model,simulating the digestive processes in stomach for 2 h and then in small intestine for 6 h at 39 °C.Results: Phytase raised(P < 0.05) the release rate of Cu and Zn in corn,Cu,Zn and Mn in wheat,Cu in barley,Cu,Zn and Mn in soybean meal,Zn,Fe in wheat bran and Zn,Fe,Mn in wheat middlings.The release rate of various trace elements in feedstuffs was increased after xylanase addition.Compared with the control group,the release rate of soluble Cu in corn,wheat,barley and soybean meal,soluble Zn in corn,wheat and wheat middlings and soluble of Mn in corn,wheat,barley and wheat bran increased(P < 0.05) after xylanase treatment.After the treatment of β-glucanase,the release rate of soluble Cu in corn,wheat and wheat bran,soluble Fe in barley,soybean meal and wheat bran and soluble Mn in corn and wheat bran all increased(P < 0.05) compared with the control group.In each feedstuff,after corresponding enzyme treatment,the contents of phytic acid,xylan and β-glucan were significantly lower than those of the control group(P < 0.05).Conclusions: Results showed that bound trace elements in feedstuffs can be released by feed enzymes.It may be necessary to take the trace elements in feedstuffs into account in the actual feed preparation including feed enzymes.Xiaonan Yu Jianan Han Haiyun Li Yiwei Zhang Jie Feng 2019Journal of Animal Science and Biotechnology2019,10,1:2

17The S190R mutation in the hemagglutinin protein of pandemic H1N1 2009 influenza virus increased its pathogenicity in mice显示文摘Human influenza viruses preferentially bind to sialic acid-α2,6-galactose (SAα2,6Gal) receptors, which are predominant in human upper respiratory epithelia, whereas avian influenza viruses preferentially bind to SAα2,3Gal receptors. However, variants with amino acid substitutions around the receptor-binding sites of the hemagglutinin (HA) protein can be selected after several passages of human influenza viruses from patients’ respiratory samples in the allantoic cavities of embryonated chicken eggs. In this study, we detected an egg-adapted HA S190R mutation in the pandemic H1N1 virus 2009 (pdmH1N1), and evaluated the effects of this mutation on receptor binding affinity and pathogenicity in mice. Our results revealed that residue 190 is located within the pocket structure of the receptor binding site. The single mutation to arginine at position 190 slightly increased the binding affinity of the virus to the avian receptor and decreased its binding to the long human α2,6-linked sialic acid receptor. Our study demonstrated that the S190R mutation resulted in earlier death and higher weight loss in mice compared with the wild-type virus. 18Ultra-robust stretchable electrode for e-skin:In situ assembly using a nanofiber scaffold and liquid metal to mimic water-to-net interaction显示文摘The development of stretchable electronics could enhance novel interface structures to solve the stretchability-conductivity dilemma,which remains a major challenge.Herein,we report a nano-liquid metal(LM)-based highly robust stretchable electrode(NHSE)with a self-adaptable interface that mimics water-tonet interaction.Based on the in situ assembly of electrospun elastic nanofiber scaffolds and electrosprayed LM nanoparticles,the NHSE exhibits an extremely low sheet resistance of 52 mΩsq^(-1).It is not only insensitive to a large degree of mechanical stretching(i.e.,350%electrical resistance change upon 570%elongation)but also immune to cyclic deformation(i.e.,5%electrical resistance increases after 330000 stretching cycles with 100%elongation).These key properties are far superior to those of the state-of-the-art reports.Its robustness and stability are verified under diverse circumstances,including long-term exposure to air(420 days),cyclic submersion(30000 times),and resilience against mechanical damages.The combination of conductivity,stretchability,and durability makes the NHSE a promising conductor/electrode solution for flexible/stretchable electronics for applications such as wearable on-body physiological signal detection,human-machine interaction,and heating e-skin.Jinwei Cao Fei Liang Huayang Li Xin Li Youjun Fan Chao Hu Jing Yu Jin Xu Yiming Yin Fali Li Dan Xu Hanfang Feng Huali Yang Yiwei Liu Xiaodong Chen Guang Zhu Run-Wei Li 2022InfoMat2022,4,4:1

20High-quality Cymbidium mannii genome and multifaceted regulation of crassulacean acid metabolism in epiphytes显示文摘Epiphytes with crassulacean acid metabolism(CAM)photosynthesis are widespread among vascular plants,and repeated evolution of CAM photosynthesis is a key innovation for micro-ecosystem adaptation.However,we lack a complete understanding of the molecular regulation of CAM photosynthesis in epiphytes.Here,we report a high-quality chromosome-level genome assembly of a CAM epiphyte,Cymbidium mannii(Orchidaceae).The 2.88-Gb orchid genome with a contig N50 of 22.7 Mb and 27192 annotated genes was organized into 20 pseudochromosomes,82.8%of which consisted of repetitive elements.Recent expansions of long terminal repeat retrotransposon families have made a major contribution to the evolution of genome size in Cymbidium orchids.We reveal a holistic scenario of molecular regulation of metabolic physiology using high-resolution transcriptomics,proteomics,and metabolomics data collected across a CAM diel cycle.Patterns of rhythmically oscillating metabolites,especially CAM-related products,reveal circadian rhythmicity in metabolite accumulation in epiphytes.Genomewide analysis of transcript and protein level regulation revealed phase shifts during the multifaceted regulation of circadian metabolism.Notably,we observed diurnal expression of several core CAM genes(especially bCA and PPC)that may be involved in temporal fixation of carbon sources.Our study provides a valuable resource for investigating post-transcription and translation scenarios in C.mannii,an Orchidaceae model for understanding the evolution of innovative traits in epiphytes.Weishu Fan Zheng-Shan He Mengqing Zhe Jing-Qiu Feng Le Zhang Yiwei Huang Fang Liu Jia-Lin Huang Ji-Dong Ya Shi-Bao Zhang Jun-Bo Yang Andan Zhu De-Zhu Li 2023Plant Communications2023,4,5:1
