| 2 | Mechanism of allosteric activation of SIRT6 revealed by the action of rationally designed activators显示文摘The recent discovery of activator compounds binding to an allosteric site on the NAD+-dependent protein lysine deacetylase,sirtuin 6(SIRT6)has attracted interest and presents a pharmaceutical target for aging-related and cancer diseases.However,the mechanism underlying allosteric activation of SIRT6 by the activator MDL-801 remains largely elusive because no major conformational changes are observed upon activator binding.By combining molecular dynamics simulations with biochemical and kinetic analyses of wild-type SIRT6 and its variant M136 A,we show that conformational rotation of 2-methyl-4-fluoro-5-bromo substituent on the right phenyl ring(R-ring)of MDL-801,which uncovers previously unseen hydrophobic interactions,contributes to increased activating deacetylation activity of SIRT6.This hypothesis is further supported by the two newly synthesized MDL-801 derivatives through the removal of the 5-Br atom on the R-ring(MDL-801-D1)or the restraint of the rotation of the R-ring(MDL-801-D2).We further propose that the 5-Br atom serves as an allosteric driver that controls the ligand allosteric efficacy.Our study highlights the effect of allosteric enzyme catalytic activity by activator binding and provides a rational approach for enhancing deacetylation activity. | Shaoyong Lu Yingyi Chen Jiacheng Wei Mingzhu Zhao Duan Ni Xinheng He Jian Zhang | 2021 | Acta Pharmaceutica Sinica B2021,11,5: | 2 |

| 4 | Study on Effects of Organic Nutrient Solution on Amino Acid Contents in Litchi (Litchi chinensis Sonn.) Fruit显示文摘[Objectives] This study was conducted to investigate the effect of organic nutrient solution on litchi quality. [Methods] Different concentrations of organic nutrient solutions were sprayed on litchi leaves to study their effects on amino acid contents in the fruit of litchi cultivar 'Qinzhouhongli'. [Results] The results showed that the organic nutrient solution could significantly promote the growth of litchi fruit at various development stages and improve fruit quality by spraying the organic nutrient solution for 3 times with a dilution concentration of 500-1 500 times during fruit development(at an interval of 15 d) of litchi. It could increase the total essential amino acid, total semi-essential amino acid, lysine, phenylalanine, threonine, methionine, valine, leucine, histidine, arginine, aspartic acid, glycine, serine, proline, alanine and tyrosine contents. Spraying the organic nutrient solution with a dilution concentration of 1 000 times achieved the best effect of promoting the growth and improving quality of litchi fruit. | 5 | PHF6 functions as a tumor suppressor by recruiting methyltransferase SUV39H1 to nucleolar region and offers a novel therapeutic target for PHF6-muntant leukemia显示文摘Mutations in the plant homeodomain-like finger protein 6(PHF6)gene are strongly associated with acute myeloid(AML)and T-cell acute lymphoblastic leukemia(T-ALL).In this study,we demonstrated that PHF6 can bind to H3K9me3 and H3K27me1 on the nucleolar chromatin and recruit histone methyltransferase SUV39H1 to the rDNA locus.The deletion of PHF6 caused a decrease in the recruitment of SUV39H1 to rDNA gene loci,resulting in a reduction in the level of H3K9me3 and the promotion of rDNA transcription.The knockdown of either SUV39H1 or PHF6 significantly attenuated the effects of increase in H3K9me3 and suppressed the transcription of rDNA induced by the overexpression of the other interacting partner,thereby establishing an interdependent relationship between PHF6 and SUV39H1 in their control of rRNA transcription.The PHF6 clinical mutants significantly impaired the ability to bind and recruit SUV39H1 to the rDNA loci,resulting in an increase in rDNA transcription activity,the proliferation of in vitro leukemia cells,and the growth of in vivo mouse xenografts.Importantly,significantly elevated levels of pre-rRNA were observed in clinical AML patients who possessed a mutated version of PHF6.The specific rDNA transcription inhibitor CX5461 significantly reduced the resistance of U937 AML cells deficient in PHF6 to cytarabine,the drug that is most commonly used to treat AML.Collectively,we revealed a novel molecular mechanism by which PHF6 recruits methyltransferase SUV39H1 to the nucleolar region in leukemia and provided a potential therapeutic target for PHF6-mutant leukemia. | Hsiang-i Tsai Yanping Wu Rui Huang Dandan Su Yingyi Wu Xiaoyan Liu Linglu Wang Zhanxue Xu Yuxin Pang Chong Sun Chao He Fan Shu Haitao Zhu Dongqing Wang Fang Cheng Laiqiang Huang Hongbo Chen | 2022 | Acta Pharmaceutica Sinica B2022,12,4: | 1 |

| 7 | A novel SIRT6 activator ameliorates neuroinflammation and ischemic brain injury via EZH2/FOXC1 axis显示文摘Ischemic stroke is the second leading cause of death worldwide with limited medications and neuroinflammation was recognized as a critical player in the progression of stroke,but how to control the overactive neuroinflammation is still a long-standing challenge.Here,we designed a novel SIRT6 activator MDL-811 which remarkably inhibited inflammatory response in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages and primary mouse microglia,which were abolished by silencing SIRT6.RNA-seq screening identified the forkhead box C1 (Foxc1) is a key gene evoked by MDL-811stimulation and is required for the anti-inflammatory effects of MDL-811.We found MDL-811-activated SIRT6 directly interacted with enhancer of zeste homolog 2 (EZH2) and promoted deacetylation of EZH2 which could bind to the promoter of Foxc1 and upregulate its expression to modulate inflammation.Moreover,our data demonstrated that MDL-811 not only ameliorated sickness behaviors in neuroinflammatory mice induced by LPS,but also markedly reduced the brain injury in ischemic stroke mice in addition to promoting long-term functional recovery.Importantly,MDL-811 also exhibited strong anti-inflammatory effects in human monocytes isolated from ischemic stroke patients,underlying an interesting translational perspective.Taken together,MDL-811 could be an alternative therapeutic candidate for ischemic stroke and other brain disorders associated with neuroinflammation. | Tailin He Jialin Shang Chenglong Gao Xin Guan Yingyi Chen Liwen Zhu Luyong Zhang Cunjin Zhang Jian Zhang Tao Pang | 2021 | Acta Pharmaceutica Sinica B2021,11,3: | 1 |
