| 1 | Discovery of mitochondrial chimeric-gene associated with cytoplasmic male sterility of HL-rice显示文摘The mitochondrial genome libraries of HL-type sterile line(A) and maintainer line(B) have been constructed. Mitochondrial gene, atp6, was used to screen libraries, due to the different Southern and Northern blot results between sterile and maintainer line. Sequencing analysis of positive clones proved that there were two copies of atp6 gene in sterile line and only one in maintainer line. One copy of atpt6 in sterile line was same to that in maintainer line; the other showed different flanking sequence from the 49th nucleotide downstream of the termination codon of atp6 gene. A new chimeric gene, orfH79, was found in the region. There may be some relationship between RNA editing and cytoplasmic male sterility (CMS), a kind of phenomenon that is attributed to mitochondrial genome mutations. The research materials used are the gametophytic male sterility line (A), maintainer line (B) and F1 hybrid (F1) of HL-type CMS rice. cDNAs and DNAs of atp6 and coxII have been obtained from A, B and F1 by PCR and RT-PCR. Comparing sequences of cDNAs and DNAs, 18 and 15 editing sites were found respectively in the transcripts of atp6 and coxll. A, B and F1 shared the same editing sites. | 11 | Positron emission tomography detector performance with different crystal pitches显示文摘Aims:In this work,we evaluated the performance of three pitches positron emission tomography(PET)detectors for building a high-resolution small-animal PET imaging experimental platform.Methods:The evaluation of PET detectors includes twodimensional crystal identification,energy resolution,and crystal look-up table.The nuclear instrumentation module and PXI data acquisition(DAQ)system were both used to obtain flood images.The energy spectrum of the Na-22 source was measured.PET detectors of 2.0 and 1.6 mm pitches used in our present experiment were composed of lutetium yttrium oxyorthosilicate(LYSO)crystals directly coupled to position-sensitive photomultiplier tubes(PSPMTs).The 1.3-mm pitch detector comprises a LYSO crystal,a tapered light-guide,and a PSPMT.The crystal sizes of the detectors were a 10×10 array of 2.0×2.0×10 mm^(3) crystals,a 10×10 array of 1.6×1.6×10 mm^(3) crystals,and an 18×18 array of 1.3×1.3×10 mm^(3) crystals,respectively.Due to the lower sampling frequency of the DAQ system,a spline interpolation algorithm was introduced in the signal waveform to reconstruct the peak values.Results:The experimental results indicated that all three PET detector crystals could be clearly identified.A detailed analysis demonstrated that the key characteristic full width at half maximum(FWHM)values of the crystals profile could reach 0.46±0.03,0.32±0.03,and 0.21±0.02 mm,while the crystal average energy resolutions were 16.8%±1.76%,20.1%±2.24%,and 31.5%±3.76%,which correspond to pitches of 2.0-,1.6-,and 1.3-mm,respectively.Conclusions:The PET detector modules of LYSO crystal arrays could be potentially used in the small-animal PET imaging applications and are promising to achieve submillimeter PET images. | LinGe Tong YaMing Fan Ming Dong Liang Zhao ZhongXing Li YingGuo Li TianQuan Wang YongZhi Yin XiMeng Chen | 2017 | Radiation Detection Technology and Methods2017,1,2: | 1 |
| 12 | Overexpression of OsKTN80a, a katanin P80ortholog, caused the repressed cell elongation and stalled cell division mediated by microtubule apparatus defects in primary root in Oryza sativa显示文摘Katanin, a microtubule‐severing enzyme, consists of two subunits: the catalytic subunit P60, and the regulatory subunit P80. In several species, P80 functions in meiotic spindle organization, the flagella biogenesis, the neuronal development, and the male gamete production. However,the P80 function in higher plants remains elusive. In this study, we found that there are three katanin P80 orthologs(OsKTN80a, OsKTN80b, and OsKTN80c) in Oryza sativa L.Overexpression of OsKTN80a caused the retarded root growth of rice seedlings. Further investigation indicates that the retained root growth was caused by the repressed cell elongation in the elongation zone and the stalled cytokinesis in the division zone in the root tip. The in vivo examination suggests that OsKTN80a acts as a microtubule stabilizer.
