3Association between homologous recombination deficiency and outcomes with platinum and platinum-free chemotherapy in patients with triple-negative breast cancer显示文摘Objective:The choice of chemotherapeutic regimen for triple-negative breast cancer(TNBC)remains controversial.Homologous recombination deficiency(HRD)has attracted increasing attention in informing chemotherapy treatment.This study was aimed at investigating the feasibility of HRD as a clinically actionable biomarker for platinum-containing and platinum-free therapy.Methods:Chinese patients with TNBC who received chemotherapy between May 1,2008 and March 31,2020 were retrospectively analyzed with a customized 3D-HRD panel.HRD positivity was defined by an HRD score≥30 or deleterious BRCA1/2 mutation.A total of 386 chemotherapy-treated patients with TNBC were screened from a surgical cohort(NCT01150513)and a metastatic cohort,and 189 patients with available clinical and tumor sequencing data were included.Results:In the entire cohort,49.2%(93/189)of patients were identified as HRD positive(40 with deleterious BRCA1/2 mutations and 53 with BRCA1/2 intact with an HRD score of≥30).In the first-line metastatic setting,platinum therapy was associated with longer median progression-free survival(mPFS)than platinum-free therapy[9.1 vs.3.0 months;hazard ratio(HR),0.43;95%confidence interval 0.22–0.84;P=0.01].Among HRD-positive patients,the mPFS was significantly longer in those treated with platinum rather than platinum-free therapy(13.6 vs.2.0 months;HR,0.11;P=0.001).Among patients administered a platinum-free regimen,HRD-negative patients showed a PFS significantly superior to that of HRD-positive patients(P=0.02;treatment-biomarker P-interaction=0.001).Similar results were observed in the BRCA1/2-intact subset.In the adjuvant setting,HRD-positive patients tended to benefit more from platinum chemotherapy than from platinum-free chemotherapy(P=0.05,P-interaction=0.02).Conclusions:HRD characterization may guide decision-making regarding the use of platinum treatment in patients with TNBC in both adjuvant and metastatic settings.Yimeng Chen Xue Wang Feng Du Jian Yue Yiran Si Xiaochen Zhao Lina Cui Bei Zhang Ting Bei Binghe Xu Peng Yuan 2023Cancer Biology & Medicine2023,20,2:1
4Tracking the in vivo spatio-temporal patterns of neovascularization via NIR-II fluorescence imaging显示文摘The in vivo spatio-temporal patterns of neovascularization are still poorly understood because it is limited to multi-scale techniques from the cellular level to living animal level.Owing to deep tissue-penetration and zero autofluorescence background,the second near-infrared(NIR-II,1,000–1,700 nm)fluorescence imaging recently shows promise in breaking through this dilemma by dynamically tracking the pathophysiological process of neovascularization in vivo.Here,NIR-II fluorescence imaging was recruited for monitoring blood vessels in order to visualize the vascular injury and quantitively assess neovascularization in mouse models of acute skeleton muscle contusion and hindlimb ischemia.The temporal analysis of real-time NIR-II fluorescence intensity demonstrated that the blood flow perfusion of ischemia area was able to rapidly restore to 96%of pre-ischemic state within one week.Moreover,the spatial analysis revealed that the lower and outer quadrants of ischemia area in the mouse model of hindlimb ischemia always had relatively high blood flow perfusion compared with other quadrants during three weeks post-ischemia,and even exceeded pre-ischemic quantity at 21 days post-ischemia.In conclusion,this in vivo imaging technique has significant potential utility for studying the spatio-temporal patterns of neovascularization in vivo.Mo Chen Sijia Feng Yimeng Yang Yunxia Li Jian Zhang Shiyi Chen Jun Chen 2020Nano Research2020,13,11:1

7Highly Efficient Multifunctional Luminescent Radicals显示文摘Stable organic luminescent radicals are a special class of compounds integrating optical,electrical,and magnetic properties.Luminescent radicals not only have potential applications in the field of the organic light-emitting diodes(OLEDs)but can also be applied in the fields of fluorescence sensing,bioimaging,and so forth.Nevertheless,due to the adverse effects of solvent polarity on the luminescent performance of radicals,no feasible approaches have been found in the literature toward fluorescent sensing.In this work,we report two luminescent radicals,2αPyID-TTM and 2δPyID-TTM,whose emissions show high efficiency and less dependence on solvent polarity.Both radicals show remarkable protonation–deprotonation properties.Besides,2αPyID-TTM exhibits significant fluorescence quenching and colorimetric response toward Fe^(3+)in aqueous solution.This suggests the possibility of a fluorometric/colorimetric dual-channel probe for Fe^(3+).Moreover,an optimized OLED using 2δPyID-TTM as an emissive dopant shows pure red emission and a maximum external quantum efficiency(EQE)of 10.6%.These results show promise for luminescent radicals as fluorescent probes and electroluminescent emitters.Yihan Zhao Alim Abdurahman Yimeng Zhang Ping Zheng Ming Zhang Feng Li 2022CCS Chemistry2022,4,2:0
8A Universal Hybrid Precoding Scheme for Massive MIMO Communications显示文摘Based on an analog radio frequency(RF)network,hybrid precoding(HPC)for massive MIMO can achieve very high spectral efficiencies with moderate hardware cost and power consumption.Despite the extensive research efforts in recent years,the practioners are still looking for HPCs that are efficient and easy-to-implement.In this paper,we present a new method termed as the universal hybrid precoding(UHP),which is nearly optimal,computationally efficient,and applicable to various types of RF network(thus,the name universal):the components of the network can be phase shifters(with finite or infinite resolutions),switches,or their combinations;the topology of the network can be fully-connected or partiallyconnected.Besides the standard UHP,we also propose a simplified version termed as sUHP to trade a negligible performance loss for much reduced computational complexity.The analysis shows that the computational complexity of the proposed UHP/sUHP is one to two orders of magnitude lower than the state-of-theart methods.Simulation results verify the(near-)optimality of the proposed UHP scheme for various forms of the analog networks.Yimeng Feng Yi Jiang Mahesh K.Varanasi 2022China Communications2022,19,11:0

10Emerging role of ubiquitination/deubiquitination modification of PD-1/PD-L1 in cancer immunotherapy显示文摘As members of the immune checkpoint family, PD-1 and its ligand PD-L1 play critical roles in maintaining the balance between autoimmunity and tolerance. The interaction of PD-1/PD-L1 is also involved in tumor evasion inside the tumor microenvironment, caused by reduced T cell activation, proliferation, cytotoxic secretion, and survival. Previous research has shown that the expression level of PD-1/PD-L1 may be regulated by ubiquitin-mediated proteasome degradation, which is an important mode of post-translational modification (PTM). PD-1/PD-L1 ubiquitin modification research in tumor immunotherapy is the subject of the present review, which aims to assess the most recent developments in this area. We offer a short explanation of PD-1/PD-L1 as well as some basic background information on the UPS system and discuss many routes that target E3s and DUBs, respectively, in the regulation of PD-1/PD-L1 in tumor immunotherapy. In addition, we offer numerous innovative prospective research areas for the future, as well as novel immunotherapy concepts and ideas. 11Nitrogen ion irradiation effect on enhancing photocatalytic performance of CdTe/ZnO heterostructures显示文摘To deal with the increasingly deteriorating environment problems, more and more harsh requirements are put forward for photocatalysis application. Building semiconductor heterostructures has been proven to be an efficient way to enhance photocatalytic performance. A kind of CdTe/ZnO heterostructures were synthesized by a hydrothermal and successive ionic layer absorption and reaction (SILAR) method and achieved obviously efficient photocatalytic performance. Moreover, after the N ion irradiation treatment, the photocatalytic activity was further enhanced, which can be ascribed to the introduction of oxygen vacancy defects. 12Dimensional crossover tuned by pressure in layered magnetic NiPS_(3)显示文摘The physical properties of most 2D materials are highly dependent on the nature of their interlayer interaction.In-depth studies of the interlayer interaction are beneficial to the understanding of the physical properties of 2D materials and permit the development of related devices.Layered magnetic NiPS_(3)has unique magnetic and electronic properties.The electronic band structure and corresponding magnetic state of NiPS_(3)are expected to be sensitive to the interlayer interaction,which can be tuned by external pressure.Here,we report an insulator-metal transition accompanied by the collapse of magnetic order during the 2D-3D structural crossover induced by hydrostatic pressure.A two-stage phase transition from a monoclinic(C2/m)to a trigonal(P31m)lattice is identified via ab initio simulations and confirmed via high-pressure X-ray diffraction and Raman scattering;this transition corresponds to a layer-by-layer slip mechanism along the a-axis.Temperature-dependent resistance measurements and room temperature infrared spectroscopy under different pressures demonstrate that the insulator-metal transition and the collapse of the magnetic order occur at~20 GPa,which is confirmed by low-temperature Raman scattering measurements and theoretical calculations.These results establish a strong correlation between the structural change,electric transport,and magnetic phase transition and expand our understanding of layered magnetic materials.Moreover,the structural transition caused by the interlayer displacement has significance for designing similar devices at ambient pressure.Xiaoli Ma Yimeng Wang Yunyu Yin Binbin Yue Jianhong Dai Jinguang Cheng Jianting Ji Feng Jin Fang Hong Jian-Tao Wang Qingming Zhang Xiaohui Yu 2021Science China(Physics,Mechanics & Astronomy)2021,64,9:0

14Test of magnetic susceptibility and grain-size age models of loess显示文摘Ages of the stratigraphic boundary MIS1/2 and MIS3/4 of the Yuanbu loess section in Linxia are used as the basis of the nodal control age. The age of MIS1/2 and MIS3/4 are obtained from the latest international research result—the climatic events recorded in the stalagmite in the Hulu Cave in Nanjing, that MIS1/2 is 11.5 kaB.P. and MIS3/4 is 59.8 kaB.P.. The ages of the two climatic events contain three nodal age control models (Model 1: 0 kaB.P.—59.8 kaB.P.; Model 2: 0 kaB.P.—11.5 kaB.P. and 11.5 kaB.P.—59.8 kaB.P.; Model 3: 11.5 kaB.P.—59.8 kaB.P.), which are used as the nodal control age separately. The deposition times of various stratigraphic horizons are calculated by using the magnetic susceptibility age model and grain-size age model, and then compared with each other. In addition, the AMS14C age, OSL age and the ages of YD and H events are compared with the ages of the corresponding horizons calculated by the three models of nodal control ages. From the analyses of lithologic characters and climatic stages it has been found that both the magnetic susceptibility age model and the grain-size age model have some defects. Because the accurate control ages are selected as the nodal points of the glacial period or interglacial period, the stratigraphic deposition times determined by the high resolution of magnetic susceptibility age model and grain-size age model approximate to the actual ages. 16Inhibition of aryl hydrocarbon receptor signaling promotes the terminal differentiation of human erythroblasts显示文摘The aryl hydrocarbon receptor(AHR)plays an important role during mammalian embryo development.Inhibition of AHR signaling promotes the development of hematopoietic stem/progenitor cells.AHR also regulates the functional maturation of blood cells,such as T cells and megakaryocytes.However,little is known about the role of AHR modulation during the development of erythroid cells.In this study,we used the AHR antagonist StemRegenin 1(SR1)and the AHR agonist 2,3,7,8-tetrachlorodibenzo-p-dioxin during different stages of human erythropoiesis to elucidate the function of AHR.We found that antagonizing AHR signaling improved the production of human embryonic stem cell derived erythrocytes and enhanced erythroid terminal differentiation.RNA sequencing showed that SR1 treatment of proerythroblasts upregulated the expression of erythrocyte differentiation-related genes and downregulated actin organization-associated genes.We found that SR1 accelerated F-actin remodeling in terminally differentiated erythrocytes,favoring their maturation of the cytoskeleton and enucleation.We demonstrated that the effects of AHR inhibition on erythroid maturation were associated with F-actin remodeling.Our findings help uncover the mechanism for AHRmediated human erythroid cell differentiation.We also provide a new approach toward the large-scale production of functionally mature human pluripotent stem cell-derived erythrocytes for use in translational applications.Yijin Chen Yong Dong Xulin Lu Wanjing Li Yimeng Zhang Bin Mao Xu Pan Xiaohong Li Ya Zhou Quanming An Fangxin Xie Shihui Wang Yuan Xue Xinping Cai Mowen Lai Qiongxiu Zhou Yan Yan Ruohan Fu Hong Wang Tatsutoshi Nakahata Xiuli An Lihong Shi Yonggang Zhang Feng Ma 2022Journal of Molecular Cell Biology2022,14,2:0
17Application of a ddRT-PCR to quantify seasonal influenza virus for viral isolation显示文摘Viral isolation in cell cultures has been regarded for decades as the“gold standard”for the laboratory diagnosis of influenza viral infections.Not all viral strains could be isolated from clinical samples.This study aimed to quantify the viral load in the samples before isolation to save working time and improve working efficiency.Four hundred samples from patients with influenza-like cases were confirmed pdmH1N1 positive(200 cases)and B Victoria(BV)positive(200 cases)by whole-genome sequencing and analyzed by ddPCR for viral load in samples before isolation,and isolation results were verified by hemagglutination(HA)assay and hemagglutination-inhibition(HI)tests.Probit regression analysis was used to calculate the isolation viral load limit with a 95%probability level by SPSS 19.0 software.The results showed that the isolation limit of viral load was 4.9×10^(4)(95%CI:2.5×10^(4)–9.0×10^(4))copies/mL for pdmH1N1 and 1.9×10^(4)(95%CI:7.8×103–3.6×10^(4))copies/mL for BV.The isolation rate of clinical samples is positively correlated with the viral load in clinical samples,which can be used for viral culture,providing important guidance for daily work.Yimeng Liu Jiachen Zhao Xiaomin Peng Guilan Lu Weixian Shi Zhaomin Feng Hui Xu Shujuan Cui Yang Pan Daitao Zhang Peng Yang Quanyi Wang 2022Biosafety and Health2022,4,5:0
