1Identification of the novel recessive gene pi55(t) conferring resistance to Magnaporthe oryzae显示文摘The elite rice cultivar Yuejingsimiao 2 (YJ2) is characterized by a high level of grain quality and yield, and resistance against Magnaporthe oryzae. YJ2 showed 100% resistance to four fungal populations collected from Guangdong, Sichuan, Liaoning, and Heilongjiang Provinces, which is a higher frequency than that shown by the well-known resistance (R) gene donor cultivars such as Sanhuangzhan 2 and 28zhan. Segregation analysis for resistance with F 2 and F 4 populations indicated the resistance of YJ2 was controlled by multiple genes that are dominant or recessive. The putative R genes of YJ2 were roughly tagged by SSR markers, located on chromosomes 2, 6, 8, and 12, in a bulked-segregant analysis using genome-wide selected SSR markers with F 4 lines that segregated into 3 resistant (R):1 susceptible (S) or 1R:3S. The recessive R gene on chromosome 8 was further mapped to an interval 1.9 cM/152 kb in length by linkage analysis with genomic position-ready markers in the mapping population derived from an F 4 line that segregated into 1R:3S. Given that no major R gene was mapped to this interval, the novel R gene was designated as pi55(t). 2Estimating biophysical parameters of rice with remote sensing data using support vector machines显示文摘Hyperspectral reflectance (350-2500 nm) measurements were made over two experimental rice fields containing two cultivars treated with three levels of nitrogen application.Four different transformations of the reflectance data were analyzed for their capability to predict rice biophysical parameters,comprising leaf area index (LAI;m-2 green leaf area m-2 soil) and green leaf chlorophyll density (GLCD;mg chlorophyll m 2 soil),using stepwise multiple regression (SMR) models and support vector machines (SVMs).Four transformations of the rice canopy data were made,comprising reflectances (R),first-order derivative reflectances (D1),second-order derivative reflectances (D2),and logarithm transformation of reflectances (LOG).The polynomial kernel (POLY) of the SVM using R was the best model to predict rice LAI,with a root mean square error (RMSE) of 1.0496 LAI units.The analysis of variance kernel of SVM using LOG was the best model to predict rice GLCD,with an RMSE of 523.0741 mg m-2.The SVM approach was not only superior to SMR models for predicting the rice biophysical parameters,but also provided a useful exploratory and predictive tool for analyzing different transformations of reflectance data.YANG XiaoHua HUANG JingFeng WU YaoPing WANG JianWen WANG Pei WANG XiaoMing Alfredo R. 3Modeling the radiation balance of different urban underlying surfaces显示文摘An urban net all-wave radiation parameterization scheme is evaluated using annual datasets for 2010 recorded at a Beijing urban observation site.The statistical relationship between observed data and simulation data of net radiation has a correlation coefficient of 0.98 and model efficiency of 0.93.Therefore,it can be used to simulate the radiation balance of Beijing.This study analyzes the variation in the radiation balance for different underlying surfaces.To simulate radiation balance differences,we set four pure land-cover types(forest,grass,roads,and buildings).Keeping all other conditions inputted unchanged,we model the radiation balance by changing the land-cover type.The results show that the effects of different underlying surfaces on radiation differ,and that there is much upward long-wave radiation,accounting for 84.3% of the total radiation energy falling incident on the land surface.The annual averages of net radiation for the four land-cover types are in the range of 38.2-53.4 W/m2.The net radiation of the grass surface is minimal while that of the roads surface is maximal.Additionally,with urbanization the net radiation values of common types of land-cover change,such as conversion from forest to roads,grass to roads,and grass to buildings,all have increasing trends,indicating that net radiation usually increases with urban sprawl.CUI YaoPing LIU JiYuan HU YunFeng WANG JunBang KUANG WenHui 2012Chinese Science Bulletin2012,57,9:9
42.5-Dimensional Parylene C micropore array with a large area and a high porosity for high-throughput particle and cell separation显示文摘Large-area micropore arrays with a high porosity are in high demand because of their promising potential in liquid biopsy with a large volume of clinical sample.However,a micropore array with a large area and a high porosity faces a serious mechanical strength challenge.The filtration membrane may undergo large deformation at a high filtration throughput,which will decrease its size separation accuracy.In this work,a keyhole-free Parylene molding process has been developed to prepare a large(>20 mm×20 mm)filtration membrane containing a 2.5-dimensional(2.5D)micropore array with an ultra-high porosity(up to 91.37% with designed pore diameter/space of 100μm/4μm).The notation 2.5D indicates that the large area and the relatively small thickness(approximately 10μm)of the fabricated membranes represent 2D properties,while the large thickness-to-width ratio(10μm/<4μm)of the spaces between the adjacent pores corresponds to a local 3D feature.The large area and high porosity of the micropore array achieved filtration with a throughput up to 180 mL/min(PBS solution)simply driven by gravity.Meanwhile,the high mechanical strength,benefiting from the 2.5D structure of the micropore array,ensured a negligible pore size variation during the high-throughput filtration,thereby enabling high size resolution separation,which was proven by single-layer and multi-layer filtrations for particle separation.Furthermore,as a preliminary demonstration,the prepared 2.5-dimensional Parylene C micropore array was implemented as an efficient filter for rare cancer cell separation from a large volume,approximately 10 cells in 10 mL PBS and undiluted urine,with high recovery rates of 87±13% and 56±13%,respectively.Yaoping Liu Han Xu Wangzhi Dai Haichao Li Wei Wang 2018Microsystems & Nanoengineering2018,4,1:2

9Hydrogen production by draTGB hupL double mutant of Rhodospirillum rubrum under different light conditions显示文摘To increase H2 yield of Rhodospirillum rubrum in two-stage hydrogen production process, two deletion mutants were constructed. One is single mutant designated R. rubrum UR801 that deleted hupL gene encoding the large subunit of uptake hy- drogenase, and the other is a double mutant desig- nated R. rubrum UR805 lacked both draTGB encod- ing regulators for the activity of nitrogenase and hupL. Comparing H2 yields of two mutants with R. rubrum UR2 (wild type) and UR472 (ΔdraTGB) under differ- ent light conditions, the results showed that the H2 yield of R. rubrum UR801 under continuous light is the highest (5700 mL of H2 per liter culture), and it is 1.56, 2.24 and 2.32-fold that of R. rubrum UR2, UR472 and UR805, respectively. However, the total H2 yield of R. rubrum UR805 in two-stage hydrogen production process is the highest (4303 mL/L), and it is 1.35, 1.21 and 1.04-fold that of R. rubrum UR2, UR801 and UR472, respectively. 11A micropore array-based solid lift-off method for highly efficient and controllable cell alignment and spreading显示文摘Interpretation of cell–cell and cell-microenvironment interactions is critical for both advancing knowledge of basic biology and promoting applications of regenerative medicine.Cell patterning has been widely investigated in previous studies.However,the reported methods cannot simultaneously realize precise control of cell alignment and adhesion/spreading with a high efficiency at a high throughput.Here,a novel solid lift-off method with a micropore array as a shadow mask was proposed.Efficient and precise control of cell alignment and adhesion/spreading are simultaneously achieved via an ingeniously designed shadow mask,which contains large micropores(capture pores)in central areas and small micropores(spreading pores)in surrounding areas contributing to capture/alignment and adhesion/spreading control,respectively.The solid lift-off functions as follows:(1)protein micropattern generates through both the capture and spreading pores,(2)cell capture/alignment control is realized through the capture pores,and(3)cell adhesion/spreading is controlled through previously generated protein micropatterns after lift-off of the shadow mask.High-throughput(2.4–3.2×10^(4) cells/cm 2)cell alignments were achieved with high efficiencies(86.2±3.2%,56.7±9.4%and 51.1±4.0%for single-cell,double-cell,and triple-cell alignments,respectively).Precise control of cell spreading and applications for regulating cell skeletons and cell–cell junctions were investigated and verified using murine skeletal muscle myoblasts.To the best of our knowledge,this is the first report to demonstrate highly efficient and controllable multicell alignment and adhesion/spreading simultaneously via a simple solid lift-off operation.This study successfully fills a gap in literatures and promotes the effective and reproducible application of cell patterning in the fields of both basic mechanism studies and applied medicine.Tingting Hun Yaoping Liu Yechang Guo Yan Sun Yubo Fan Wei Wang 2020Microsystems & Nanoengineering2020,6,1:1
12A dual-use probe for nano-metric photoelectric characterization using a confined light field generated by photonic crystals in the cantilever显示文摘We propose a plasmonic atomic force microscopy (AFM) probe, which takes a part of the laser beam for monitoring cantilever deflection as the excitation light source. Photonic crystal cavities are integrated near the cantilever’s free end where the laser spot locates. The transmitted light excites surface plasmon polaritons on the metal-coated tip and induces a confined hot-spot at the tip apex. Numerical simulations demonstrate that the plasmonic probe can couple a tilted, linearly polarized beam efficiently and yield a remarkable local electromagnetic enhancement with the intensity being around 21 times stronger than that of the original probe. For demonstration, we employ the plasmonic probe in electrostatic force microscopy and scanning Kelvin probe microscopy to study the impact of local light field on the photoelectric characteristics of SiO_(2) and Au nanoparticles. Compared with the original probe, obvious differences are observed in the electrostatic force gradients on SiO_(2) nanoparticles and in the surface potentials of Au nanoparticles. 13Enhanced YOLOv5 network-based object detection(BALFilter Reader)promotes PERFECT filter-enabled liquid biopsy of lung cancer from bronchoalveolar lavage fluid(BALF)显示文摘Liquid biopsy of cancers,detecting tumor-related information from liquid samples,has attracted wide attentions as an emerging technology.Our previously reported large-area PERFECT(Precise-Efficient-Robust-Flexible-Easy-ControllableThin)filter has demonstrated competitive sensitivity in recovering rare tumor cells from clinical samples.However,it is time-consuming and easily biased to manually inspect rare target cells among numerous background cells distributed in a large area(Φ≥13 mm).This puts forward an urgent demand for rapid and bias-free inspection.Hereby,this paper implemented deep learning-based object detection for the inspection of rare tumor cells from large-field images of PERFECT filters with hematoxylin-eosin(HE)-stained cells recovered from bronchoalveolar lavage fluid(BALF).CenterNet,EfficientDet,and YOLOv5 were trained and validated with 240 and 60 image blocks containing tumor and/or background cells,respectively.YOLOv5 was selected as the basic network given the highest mAP@0.5 of 92.1%,compared to those of CenterNet and EfficientDet at 85.2%and 91.6%,respectively.Then,tricks including CIoU loss,image flip,mosaic,HSV augmentation and TTA were applied to enhance the performance of the YOLOv5 network,improving mAP@0.5 to 96.2%.This enhanced YOLOv5 network-based object detection,named as BALFilter Reader,was tested and cross-validated on 24 clinical cases.The overall diagnosis performance(~2 min)with sensitivity@66.7%±16.7%,specificity@100.0%±0.0%and accuracy@75.0%±12.5%was superior to that from two experienced pathologists(10–30 min)with sensitivity@61.1%,specificity@16.7%and accuracy@50.0%,with the histopathological result as the gold standard.The AUC of the BALFilter Reader is 0.84±0.08.Moreover,a customized Web was developed for a user-friendly interface and the promotion of wide applications.The current results revealed that the developed BALFilter Reader is a rapid,bias-free and easily accessible AI-enabled tool to promote the transplantation of the BALFilter technique.This work can easily expand to other cytopathological diagnoses and improve the application value of micro/nanotechnology-based liquid biopsy in the era of intelligent pathology.Zheng Liu Jixin Zhang Ningyu Wang Yun’ai Feng Fei Tang Tingyu Li Liping Lv Haichao Li Wei Wang Yaoping Liu 2023Microsystems & Nanoengineering2023,9,5:0
14Radiofrequency ablation of hepatocellular carcinoma: Current status,challenges, and prospects显示文摘Local ablation technologies, such as radiofrequency ablation (RFA), microwave ablation (MWA) andcryoablation, have become a standard treatment option for hepatocellular carcinoma (HCC) less than5 cm in size, particularly in individuals who are not candidates for hepatectomy. Except for equivalentprognosis and efficiency, RFA has various advantages over surgical excision, including a lower rate ofcomplications, a cheaper cost, more normal tissue preservation, and a shorter hospital stay. However, therate of tumor recurrence and/or distant metastasis after RFA therapy is still high. RFA has been widelyemployed in multiple cancers, large cancer, and lesion identified at “high-risk” sites in recent years, withthe advancement of ablation types and operating techniques, particularly the combined use of manytechnologies. 16Interannual variability and climatic sensitivity of global wildfire activity显示文摘Understanding historical wildfire variations and their environmental driving mechanisms is key to predicting and mitigating wildfires. However, current knowledge of climatic responses and regional contributions to the interannual variability (IAV) of global burned area remains limited. Using recent satellite-derived wildfire products and simulations from version v1.0 of the land component of the U.S. Department of Energy's Energy Exascale Earth System Model (E3SM land model [ELM] v1) driven by three different climate forcings, we investigated the burned area IAV and its climatic sensitivity globally and across nine biomes from 1997 to 2018. We found that 1) the ELM simulations generally agreed with the satellite observations in terms of the burned area IAV magnitudes, regional contributions, and covariations with climate factors, confirming the robustness of the ELM to the usage of different climate forcing sources;2) tropical savannas, tropical forests, and semi-arid grasslands near deserts were primary contributors to the global burned area IAV, collectively accounting for 71.7%–99.7% of the global wildfire IAV estimated by both the satellite observations and ELM simulations;3) precipitation was a major fire suppressing factor and dominated the global and regional burned area IAVs, and temperature and shortwave solar radiation were mostly positively related with burned area IAVs;and 4) noticeable local discrepancies between the ELM and remote-sensing results occurred in semi-arid grasslands, croplands, boreal forests, and wetlands, likely caused by uncertainties in the current ELM fire scheme and the imperfectly derived satellite observations.
