1Status and development of high-power laser facilities at the NLHPLP显示文摘In this paper, we review the status of the multifunctional experimental platform at the National Laboratory of High Power Laser and Physics(NLHPLP). The platform, including the SG-II laser facility, SG-II 9th beam, SG-II upgrade(SG-II UP) facility, and SG-II 5 PW facility, is operational and available for interested scientists studying inertial confinement fusion(ICF) and a broad range of high-energy-density physics. These facilities can provide important experimental capabilities by combining different pulse widths of nanosecond, picosecond, and femtosecond scales. In addition, the SG-II UP facility, consisting of a single petawatt system and an eight-beam nanosecond system, is introduced including several laser technologies that have been developed to ensure the performance of the facility. 2Embryonic endothelial evolution towards first hematopoietic stem cells revealed by single-cell transcriptomic and functional analyses显示文摘Hematopoietic stem cells(HSCs)in adults are believed to be born from hemogenic endothelial cells(HECs)in mid-gestational embryos.Due to the rare and transient nature,the HSC-competent HECs have never been stringently identified and accurately captured,let alone their genuine vascular precursors.Here,we first used high-precision single-cell transcriptomics to unbiasedly examine the relevant EC populations at continuous developmental stages with intervals of 0.5 days from embryonic day(E)9.5 to E11.0.As a consequence,we transcriptomically identified two molecularly different arterial EC populations and putative HSCprimed HECs,whose number peaked at El 0.0 and sharply decreased thereafter,in the dorsal aorta of the aorta-gonadmesonephros(AGM)region.Combining computational prediction and in vivo functional validation,we precisely captured HSCcompetent HECs by the newly constructed Neurl3-EGFP reporter mouse model,and realized the enrichment further by a combination of surface markers(Procr+Kit+CD44+,PK44).Surprisingly,the endothelial-hematopoietic dual potential was rarely but reliably witnessed in the cultures of single HECs.Noteworthy,primitive vascular ECs from E8.0 experienced two-step fate choices to become HSC-primed HECs,namely an Initial arterial fate choice followed by a hemogenic fate conversion.This finding resolves several previously observed contradictions.Taken together,comprehensive understanding of endothelial evolutions and molecular programs underlying HSC-primed HEC specification in vivo will facilitate future investigations directing HSC production in vitro.Siyuan Hou Zongcheng Li Xiaona Zheng Yun Gao Ji Dong Yanli Ni Xiaobo Wang Yunqiao Li Xiaochen Ding Zhilin Chang Shuaili Li Yuqiong Hu Xiaoying Fan Yu Hou Lu Wen Bing Liu Fuchou Tang Yu Lan 2020Cell Research2020,30,5:8
3The Bethesda System for Reporting Thyroid Cytopathology(TBSRTC):A report of 2,781 cases in a Chinese population显示文摘Objective:To evaluate the diagnostic utility of The Bethesda System for Reporting Thyroid Cytology(TBSRTC)at Peking University Cancer Hospital,the incidence of noninvasive follicular thyroid neoplasms with papillary-like nuclear features(NIFTP),and the impact of reclassification on cytopathological outcomes.Methods:We performed a retrospective study of thyroid fine-needle aspiration(FNA)cases between April 2014 and March 2019.The FNA results were classified according to TBSRTC.Post-surgery histological findings were followed up.Results:A total of 2,781 thyroid FNAs were performed.The incidences of the 6 diagnostic categories(DCs I-VI)were 14.8%,17.1%,15.8%,2.3%,11.6%and 38.5%,respectively.A total of 1,122 patients(40.3%)had corresponding histological results.NIFTP accounted for 0.4%of papillary thyroid carcinoma(PTC)cases,and there was no significant difference in the risk of malignancy(ROM)for each TBSRTC DC between'NIFTP=carcinoma(Ca)'and'NIFTP≠Ca'.When'NIFTP=Ca',the sensitivity,specificity,positive predictive value(PPV),negative predictive value(NPV),and accuracy of TBSRTC were 98.0%,84.0%,99.4%,58.3%,and97.5%,respectively.When'NIFTP≠Ca',the sensitivity,specificity,PPV,NPV and accuracy of the TBSRTC were 98.1%,81.5%,99.3%,61.1%,and 97.5%,respectively.Conclusions:TBSRTC is effective in the preoperative diagnosis of thyroid nodules in Peking University Cancer Hospital.The impact of the reclassification of NIFTP on cytopathological outcomes is limited because of its low incidence,and the revised ROMs are not suitable for Asian patients.Yanli Zhu Yuntao Song Guohui Xu Zhihui Fan Wenhao Ren 2020Chinese Journal of Cancer Research2020,32,2:4
4Development of transplanting manipulator for hydroponic leafy vegetables显示文摘The production of hydroponic leafy vegetable plug-seedlings uses coco-peat as culture substrate in South China.Coco-peat has lowered density than peat-moss,and the friction between substrate block and pickup tool is small.So,it is hard to pick up in mechanism transplantation.In order to increase the friction,the existing transplanting manipulator had relatively complex structures.To simplify the structure of transplanting manipulator and improve the stability of picking up substrate block,four stainless steel fingers with rectangular cross-section were used in this research.A vertical driving was used to realize the coupling effect that could insert and shrink at the same time,by applying different combination of constraints to the steel fingers.This could increase friction between the steel fingers and the substrate block,and then enhance the stability of the substrate block.Different combinations of constraints were applied to the rectangular stainless steel fingers(3 mm×0.8 mm).The working videos of steel fingers were taken by high-speed photography.High-speed motioned analysis software was used to acquire and analyze traces of steel fingers movements.When the length which top end of the steel fingers moved outward(M)is equal to 1.5 mm,the length which guiding part widened(N)is equal to 1 mm,the shrinking distance of steel fingers is 4.2 mm.In this research,16-day hydroponic leafy vegetable plug-seedlings were used for performance,which cultivated with coco-peat substrate with the moisture in the substrate at 81%.The transplanting manipulator was attached to a Denso robotic arm to conduct transplanting performance test.When the shrinking distance of steel fingers increased from 0 mm to 3.2 mm and the inserting angle decreased from 80°to 77°,the lifting force of substrate block increased by 118%from 1.45 N to 3.16 N.However,excessive shrinkage stirred the substrate block,which would reduce the friction between the substrate block and pickup parts and lowered the lifting force of pickup part in the substrate block.The experimental results also demonstrated that when the shrinking distance of the steel fingers reached 3.2 mm and the root distribution rate reached 46%,the success rate of transplantation was 80%.When the leafy vegetable plug-seedlings root distribution rate reached 92%,the success rate of transplantation was 96.67%.The degree of root distribution rate was positively correlated with the transplantation success rate.Therefore,in order to ensure an acceptable success rate of transplantation,the root distribution rate of leafy vegetable plug-seedlings should be at least 90%.This study provides a technical reference for developing simplified transplanting manipulator that can be used to transplant the hydroponic leafy vegetable plug-seedlings with coco-peat as the culture substrate.Bo Li Song Gu Qi Chu Yanli Yang Zhongjian Xie Kaijun Fan Xiaogeng Liu 2019International Journal of Agricultural and Biological Engineering2019,12,6:4

6A Novel Strategy to Fabricate Core-Sheath Structure Piezoelectric Yarns for Wearable Energy Harvesters显示文摘Wearable and portable electronic devices based on textile structure have been widely used owing to their wearability and comfortableness.However,yarn fineness and the comfort of the fabric cannot satisfy the requirements of smart wearable devices.This work presents a novel strategy to prepare highly integrated PVDF/conductive nylon core-sheath structure piezoelectric yarns for wearable which is fabricated by combining electrospinning strategy with 2D braiding technology.The fineness of single yarns as well as strength are both improved significantly compared to previous works.The piezoelectric outputs of the yarn are still stable after 800 s fatigue test at a frequency of 4 Hz,and the cycle stability can maintain more than 3200 cycles.Furthermore,the piezoelectric yarns are further woven into piezoelectric plain fabric.According to the electrical performance,the length of the piezoelectric yarn and the thickness of the piezoelectric layer would both affect the output electrical performance.The yarn of the 10 cm in length and 600μm in fineness can produce an output voltage of 120 mV.Meanwhile,Both the piezoelectric yarn and the fabric could generate piezoelectric output signals through human movement,such as bending,walking.Therefore,the electrical and mechanical performance of the piezoelectric yarns prepared in our work could be improved significantly,and the comfortableness and durability performance of the piezoelectric fabric can satisfy most wearing requirements,which would provide some help in the field of piezoelectric wearable devices based on yarns and fabrics.Lili Xue Wei Fan Yang Yu Kai Dong Chengkun Liu Yanli Sun Cong Zhang Weichun Chen Ruixin Lei Kai Rong Qi Wang 2021Advanced Fiber Materials2021,3,4:3

8Molecular self-assembly behavior of mono[6-O-6-(4-carboxyl-phenyl)]-β-CD in solution and solid state显示文摘A novel modified cyclodextrin, mono[6-O-6-(4- carboxyl-phenyl)]-b-CD (1), has been synthesized by the reaction of mono[6-(p-toluenesulfonyl)]- b-CD with 4-hydro- xybenzoate, and its molecular self-assembly behavior in both solution and solid state was studied by means of crystallography, NMR spectroscopy and microcalorimetry. The results indicate that the bezoic acid groups are successively penetrated intermolecularly into the adjacent b-CD cavities to form helical columnar supramolecules in the solid state. As compared with crystal, the similar self-assembly behavior of 1 in aqueous solution has also been confirmed by the 1H ROESY spectroscopy. 10On-chip multiplexed single-cell patterning and controllable intracellular delivery显示文摘Conventional electroporation approaches show limitations in the delivery of macromolecules in vitro and in vivo.These limitations include low efficiency,noticeable cell damage and nonuniform delivery of cells.Here,we present a simple 3D electroporation platform that enables massively parallel single-cell manipulation and the intracellular delivery of macromolecules and small molecules.A pyramid pit micropore array chip was fabricated based on a silicon wet-etching method.A controllable vacuum system was adopted to trap a single cell on each micropore.Using this chip,safe single-cell electroporation was performed at low voltage.Cargoes of various sizes ranging from oligonucleotides(molecular beacons,22 bp)to plasmid DNA(CRISPR-Cas9 expression vectors,>9 kb)were delivered into targeted cells with a significantly higher transfection efficiency than that of multiple benchmark methods(e.g.,commercial electroporation devices and Lipofectamine).The delivered dose of the chemotherapeutic drug could be controlled by adjusting the applied voltage.By using CRISPR-Cas9 transfection with this system,the p62 gene and CXCR7 gene were knocked out in tumor cells,which effectively inhibited their cellular activity.Overall,this vacuum-assisted micropore array platform provides a simple,efficient,high-throughput intracellular delivery method that may facilitate on-chip cell manipulation,intracellular investigation and cancer therapy.Zaizai Dong Yanli Jiao Bingteng Xie Yongcun Hao Pan Wang Yuanyuan Liu Junfeng Shi Chandani Chitrakar Stephen Black Yu-Chieh Wang LJames Lee Mo Li Yubo Fan Lingqian Chang 2020Microsystems & Nanoengineering2020,6,1:2

17Visualized and cascade-enhanced gene silencing by smart DNAzyme-graphene nanocomplex显示文摘BCL-2 gene as well as its products is recognized as a promising target for the molecular targeted therapy of tumors.However,due to certain defense measures of tumor cells,the therapeutic effect based on the gene silencing of BCL-2 is greatly reduced.Here we fabricate a smart response nucleic acid therapeutic that could silence the gene effectively through a dual-targeted and cascade-enhanced strategy.In brief,nano-graphene oxide(GO),working as a nano-carrier,is loaded with a well-designed DNAzyme,which can target and silence the BCL-2 mRNA.Furthermore,upon binding with the BCL-2 mRNA,the enzymatic activity of the DNAzyme can be initiated,cutting a substrate oligonucleotide to produce an anti-nucleolin aptamer AS1411.Nucleolin,a nucleolar phosphoprotein,is known as a stabilizer of BCL-2 mRNA.Via binding and inactivating the nucleolin,AS1411 can destabilize BCL-2 mRNA.By this means of simultaneously targeting mRNA and its stabilizer in an integrated system,effective silencing of the BCL-2 gene of tumor cells is achieved at both the cellular and in vivo levels.After being dosed with this nucleic acid therapeutic and without any chemotherapeutics,apoptosis of tumor cells at the cellular level and apparent shrinkage of tumors in vivo are observed.By labeling a molecular beacon on the substrate of DNAzyme,visualization of the enzymatic activity as well as the tumor in vivo can be also achieved.Our work presents a pure bio-therapeutic strategy that has positive implications for enhancing tumor treatment and avoiding side effects of chemotherapeutics.Lingjie Ren Xiaoxia Chen Chang Feng Lei Ding Xiaomin Liu Tianshu Chen Fan Zhang Yanli Li Zhongliang Ma Bo Tian Xiaoli Zhu 2020Nano Research2020,13,8:1

19Clinicopathological Significance of E-cadherin and PCNA Expression in Hunman Non-small Cell Lung Cancer显示文摘OBJECTIVE This study was designed to assess E-cadherin (E-cad)and proliferating cell nuclear antigen(PCNA)expression as well as their clinicopathological significance in hunman non- small cell lung cancers(NSCLCs).Possible molecular mechanisms of differentiation and metastasis of NSCLCs are discussed. METHODS Immunohistochemical and immunofluorescence double staining were performed to examine the expression of E-cad and PCNA in 68 primary NSCLCs cases. RESULTS The E-cad expression in squamous cell carcinomas and adenocarcinomas showed no significant difference.E-cad expression had a positive correlation with the histological- differentiated grade.A significant difference of E-cad expression was found between metastatic and non-metastatic groups.PCNA expression in squamous cell carcinomas and adenocarcinomas showed no significant difference.The PCNA expression had a reverse correlation with the histological-differentiated grade.A significant difference of PCNA expression was found between metastatic and non-metastatic groups.The E-cad and PCNA expression presented a reverse correlation. 20Embryonic lineage tracing with Procr-CreER marks balanced hematopoietic stem cell fate during entire mouse lifespan显示文摘The functional heterogeneity of hematopoietic stem cells(HSCs) has been comprehensively investigated by single-cell transplantation assay.However,the heterogeneity regarding their physiological contribution remains an open question,especially for those with life-long hematopoietic fate of rigorous selfrenewing and balanced differentiation capacities.In this study,we revealed that Procr expression was detected principally in phenotypical vascular endothelium co-expressing DII4 and CD44 in the midgestation mouse embryos,and could enrich all the HSCs of the embryonic day 11.5(E11.5) aortagonad-mesonephros(AGM) region.We then used a temporally restricted genetic tracing strategy to irreversibly label the Procr-exp res sing cells at E9.5.Interestingly,most labeled mature HSCs in multiple sites(such as AGM) around E11.5 were functionally categorized as lymphomyeloid-balanced HSCs assessed by direct transplantation.Furthermore,the labeled cells contributed to an average of 7.8% of immunophenotypically defined HSCs in E14.5 fetal liver(FL) and 6.9% of leukocytes in peripheral blood(PB) during one-year follow-up.Surprisingly,in aged mice of 24 months,the embryonically tagged cells displayed constant contribution to leukocytes with no bias to myeloid or lymphoid lineages.Altogether,we demonstrated,for the first time,the existence of a subtype of physiologically long-lived balanced HSCs as hypothesized,whose precise embryonic origin and molecular identity await further characterization.Xiaona Zheng Guangyu Zhang Yandong Gong Xiaowei Ning Zhijie Bai Jian He Fan Zhou Yanli Ni Yu Lan Bing Liu 2019Journal of Genetics and Genomics2019,46,10:1
