| 2 | Gastrodin inhibits neuroinflammation in rotenone-induced Parkinson's disease model rats显示文摘The present study showed that the latency of rats moving on a vertical grid was significantly prolonged,and the number of rats sliding down from the declined plane was increased remarkably,in rotenone-induced Parkinson's disease model rats compared with control rats.The moving latency recovered to normal levels,but the number of slides was significantly increased at 28 days after model establishment.The slope test is a meaningful approach to evaluate the symptoms of Parkinson's disease model rats treated with rotenone.In addition,loss of substantia nigral dopaminergic neurons in model rats was observed at 1 day after the model was established,and continued gradually at 14 and 28 days.The expression of tyrosine hydroxylase-positive cells was significantly increased in gastrodin-treated rats at 14 days.Significant numbers of activated microglia cells were observed in model rats at 14 and 28 days;treatment of rats with Madopar at 28 days suppressed microglial activation.Treatment of rats with gastrodin or Madopar at 28 days significantly reduced interleukin-1β expression.The loss of substantia nigral dopaminergic neurons paralleled the microglial activation in Parkinson's disease model rats treated with rotenone.The inflammatory factors tumor necrosis factor-α and interleukin-1β are involved in the substantia nigral damage.Gastrodin could protect dopaminergic neurons via inhibition of interleukin-1β expression and neuroinflammation in the substantia nigra. | Chun Li Xin Chen Nan Zhang Yangwen Song Yang Mu | 2012 | Neural Regeneration Research2012,7,5: | 15 |
| 3 | Integrated simulation of the dualistic water cycle and its associated processes in the Haihe River Basin显示文摘Water resource shortages, water environmental deterioration, and water ecological degradation are becoming increasingly prominent in the Haihe River Basin under the effects of global climate change and intense human activity. Finding ways to solve these problems reasonably is an urgent task in basin water resources management. The water cycle describes the formation and transformation of every water drop, while it interacts with associated water ecology and water environment processes. Therefore, to fundamentally solve the above problems, the three processes must be combined and the evolutionary trends of the ecology and water environment driven by the water cycle must be determined. To comprehensively diagnose and solve the problems of water resources, ecology and water environment in the basin, an integrated simulation platform of the dualistic water cycle and its associated processes in the Haihe River Basin was established by fully analyzing the 'natural-social' dualistic characteristics of the evolution of the basin water cycle in this study. Accordingly, it is a supporting tool to evaluate the future trends of water resources, ecology and the water environment by setting scenarios based on the predictions of a climate mode and water control conditions. The results show that with climate change, the water consumption, groundwater overexploitation, and the amount of water flowing to the sea could develop soundly and satisfy the requirements of national economic growth in the future planning year 2030 through the implementation of water consumption control management and the South-North Water Diversion Project. Meanwhile, associated water environmental deterioration can be alleviated, and a growing trend for production capacities of the natural ecosystem and agro-ecosystem is seen. | 4 | Divalent metal transporter 1 expression and iron deposition in the substantia nigra of a rat model of Parkinson's disease显示文摘Extensive iron deposition has been observed in the midbrain substantia nigra(SN) of Parkinson's disease(PD) patients,but the mechanisms of iron deposition in the SN remain poorly understood.The present study investigated the relationship between dopaminergic neuronal damage,iron content changes,and divalent metal transporter 1(DMT1) in the midbrain SN of PD rats to explore the relationship between time of iron deposition and DMT1 expression.Frozen midbrain SN sections from model rats were stained with Perls' iron.Results showed massive loss of tyrosine hydroxylase(TH)-positive cells in the SN and increased DMT1 expression in model group rats.No obvious iron deposition was observed in the SN during early stages after damage,but significant iron deposition was detected at 8 weeks post-injury.Results demonstrate that the loss of TH-positive cells in the SN appeared simultaneously with increased DMT1 expression.Extensive iron deposition occurred at 8 weeks post injury,which could be regarded as an early time window of iron deposition. | Yangwen Song Xin Chen Chun Li Nan Zhang | 2010 | Neural Regeneration Research2010,5,22: | 8 |
| 5 | Parallel relationship between microglial activation and substantia nigra damage in a rotenone-induced Parkinson's disease rat model显示文摘BACKGROUND:Inflammatory injury induced by microglial activation plays an important role in the occurrence and development of Parkinson's disease (PD). However, few studies have examined the relationship between microglia and substantia nigra damage or dopaminergic neuron loss in animals with rotenone-induced PD. OBJECTIVE: To explore the relationship between activated microglia and loss of the substantia nigra, and the changes in concentration and dose of rotenone in the brain of rats with rotenone-induced PD. DESIGN, TIME AND SETTING: The neuropathological experiment was performed at the School of Traditional Chinese Medicine, Capital Medical University, China, from July 2007 to July 2008. MATERIALS: Rotenone was purchased from Sigma, USA. METHODS: The Parkinson's model was induced by injection of a rotenone oily-emulsion (2 mg/kg daily) subcutaneously into the back of 58 male adult Wistar rats for 3-6 weeks. Another three rats served as normal controls. MAIN OUTCOME MEASURES: Neurobehavioral changes were observed and recorded following rotenone treatment. Tyrosine hydroxylase and complement receptor OX42 were separately analyzed by immunohistochemical staining within 4 weeks following stopping rotenone treatment. Rotenone content was measured using high performance liquid chromatography in the cerebellum of rats that scored 2, 4-6. RESULTS: Rotenone induced a loss of dopaminergic neurons in the substantia nigra as well as microglial activation, with increased behavior scores. Dopaminergic loss was still ongoing even when rotenone was stopped. Dopaminergic neuronal degeneration in the substantia nigra was ini- tially 6%, but was 85% at 2 weeks after scoring, and degeneration depended on activated microglia. Rotenone was detected in the cerebellum at concentrations between 78.9 μg/L and 309.6 μg/L. CONCLUSION: Nigrostriatal dopaminergic degeneration paralleled the microglial activation. | 6 | Necessity and feasibility for an ET-based modern water resources management strategy: A case study of soil water resources in the Yellow River Basin显示文摘The necessity and feasibility of an ET-based modern water resources management was analyzed to improve assessment of critical water resources scarcity in the region/basin. This analysis was based on the whole water cycle process and its analysis object is evapotranspiration (ET), a main consumption component in the water resources dynamic transformation process. A case study was undertaken by selecting soil water resources in the Yellow River Basin and employing the WEP-L distributed hydrological model with physics mechanisms. This paper discusses the amount and consumption efficiency of soil-water resources according to completely simulated results of water cycle elements throughout the basin. Results indicate that it is important for the ET-based modern water resources management strategy to alleviate water resources scarcity because it may not only avoid unused water wasting but also improve water use efficiency. | 9 | Alternation in Lipid Composition of Wheat Leaves Induced by Phosphate Deficiency Is Related to Both Lipid Biosynthesis and Phosphatidylglycerol Degradation显示文摘In this study, the causes of the changes in lipid composition induced by different phosphatenutrient levels were investigated. Wheat plants were grown in phosphate-deficient and phosphate-suffcient conditions, respectively, and lipid compositions in the leaves of 9-day-old and 16-day-old plants wereanalyzed. We found that phosphate deficiency induced a dramatic change at the lipid levels in photosyntheticmembranes of wheat leaves and the extent of changes in lipid composition depended on the leaf ages.Phosphate deficiency induced a gradual decrease in PG and MGDG and a concomitant increase in DGDGand SQDG from the first leaf to the second and the third leaf on 16-day-old plants. In addition, as comparedto leaves grown under phosphate sufficient solution, PG content in the first leaf of 16-day-old plants wassignificantly lower than that of 9-day-old leaf with 2.5 mol% versus 5.5 mol% when these plants were grownunder phosphate deficient condition. From these results, it is suggested that the alternation in lipidcomposition in wheat leaves induced by phosphate deficiency is related to both lipid biosynthesis and PGdegradation. | 10 | A cryptic inhibitor of cytokinin phosphorelay controls rice grain size显示文摘Plant hormone cytokinin signals through histidine-aspartic acid(H-D)phosphorelay to regulate plant growth and development.While it is well known that the phosphorelay involves histidine kinases,histidine phosphotransfer proteins(HPs),and response regulators(RRs),how this process is regulated by external components remains unknown.Here we demonstrate that protein phosphatase with kelch-like domains(PPKL1),known as a signaling component of steroid hormone brassinosteroid,is actually a cryptic inhibitor of cytokinin phosphorelay in rice(Oryza sativa).Mutation at a specific amino acid D364 of PPKL1 activates cytokinin response and thus enlarges grain size in a semi-dominant mutant named s48.Overexpression of PPKL1 containing D364,either with the deletion of the phosphatase domain or not,rescues the s48 mutant phenotype.PPKL1 interacts with OsAHP2,one of authentic HPs,and D364 resides in a region resembling the receiver domain of RRs.Accordingly,PPKL1 can utilize D364 to suppress OsAHP2-to-RR phosphorelay,whereas mutation of D364 abolishes the effect.This function of PPKL1 is independent of the phosphatase domain that is required for brassinosteroid signaling.Importantly,editing of the D364-residential region produces a diversity of semi-dominant mutations associated with variously increased grain sizes.Further screening of the edited plants enables the identification of two genotypes that confer significantly improved grain yield.Collectively,our study uncovers a noncanonical cytokinin signaling suppressor and provides a robust tool for seed rational design. | Dapu Liu He Zhao Yunhua Xiao Guoxia Zhang Shouyun Cao Wenchao Yin Yangwen Qian Yanhai Yin Jinsong Zhang Shouyi Chen Chengcai Chu Hongning Tong | 2022 | Molecular Plant2022,15,2: | 3 |
| 11 | Study on consumption efficiency of soil water resources in the Yellow River Basin based on regional ET structure显示文摘Based on the regional water resources character, the concept of soil water resources is first redefined, and then associated with their transfer relationship in the hydrological cycle, Evapotranspiration (ET)-based consumption structure and consumption efficiency of soil water resources are analyzed. According to ET 's function in productivity, the consumption efficiency of soil water resources is di- vided into three classes: high efficient consumption from vegetation transpiration, low efficient con- sumption from soil evaporation among plants with high vegetation coverage and inefficient consump- tion from soil evaporation among plants with low vegetation coverage and bare soil evaporation. The high efficient and low efficient consumption were further classified as productive consumption. The ineffi- cient consumption is considered non-productive consumption because it is significant in the whole hydrological cycle process. Finally, according to these categories, and employing a WEP-L dis- tributed hydrological model, this paper analyzes the consumption efficiency of soil water resources in the Yel- low River Basin. The results show that there are 2078.89×108 m3 soil water resources in the whole basin. From the viewpoint of consumption structure, the soil water resources are comprised of 381.89×108 m3 transpiration consumption from vegetation and 1697.09×108 m3 evaporation consumption from soil among plants and bare soil. From the viewpoint of consumption efficiency, soil water re- sources are composed of 920.11×108 m3 efficient consumption and 1158.86×108 m3 of inefficient con- sumption. High efficient consumption accounts for 41.5 percent of the total efficient consumption of the whole basin, low efficient for 58.5 percent. Furthermore, consumption efficiency varies by region. Compared with ET from different land use conditions, the whole basin appears to follow the trend of having the greatest proportion of consumption as inefficient consumption, followed by low efficient consumption, and then the least proportion as high efficient consumption. The amount of inefficient consumption in some regions with vegetation is less than in other regions without vegetation. The amount of inefficient consumption in grasslands is much greater than in forestlands. However, the proportion of low efficient consumption is the greatest in crop fields. The amount of high efficient con- sumption in grasslands and forelands is similar to the corresponding low efficient consumption. However, the low efficient consumption in grasslands is larger than in the forelands.
