2A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 5Sedum alfredii H:A new Zn hyperaccumulating plant first found in China显示文摘Field survey and greenhouse experiments were carried out to identify and characterize zinc (Zn) uptake and accumulation by a new Zn hyperaccumulating plant species (Sedum alfredii Hance) native to China. Zinc concentration in the shoots of Sedum alfredii Hance grown on an ancient mined area ranged from 4134 to 5000 mg/kg, with a mean of 4515 mg/kg. It suggests that Sedum alfredii could not only grow on heavily Pb/Zn contaminated soils, but also could accumulate extraordinarily high concentration of Zn. Under nutrient solution culture conditions, Sedum alfredii Hance grew healthy at Zn supplying levels from 0.006 to 240 mg · L-1. Zinc concentration in the shoots increased with external Zn levels increasing. The Zn concentration and accumulation in the shoots reached the highest at Zn supply level of SO mg/L, with 19.67 g/kg and 19.83 nig/plant, respectively. All the results showed that Sedum alfredii Hance is a new Zn hyperaccumulating plant. 6Effects of dietary protein/energy ratio on growth performance,carcass trait,meat quality,and plasma metabolites in pigs of different genotypes显示文摘Background:The protein/energy ratio is important for the production performance and utilization of available feed resources by animals.Increased protein consumption by mammals leads to elevated feed costs and increased nitrogen release into the environment.This study aimed to evaluate the effects of dietary protein/energy ratio on the growth performance,carcass traits,meat quality,and plasma metabolites of pigs of different genotypes.Methods:Bama mini-pigs and Landrace pigs were randomly assigned to two dietary treatment groups(Chinese conventional diet with low protein/energy ratio or National Research Council diet with high protein/energy ratio;n = 24 per treatment) in a 2x2 factorial arrangement.Blood and muscle samples were collected at the end of the nursery,growing,and finishing phases.Results:We observed significant interactions(P< 0.05) between breed and diet for total fat percentage,intramuscular fat(IMF) content,protein content in biceps femoris(BF) muscle,and plasma urea nitrogen(UN) concentration in the nursery phase;for average daily gain(ADG),average daily feed intake(ADFI),dry matter,IMF content in psoas major(PM) muscle,and plasma total protein and albumin concentrations in the growing phase;and for drip loss and plasma UN concentration in the finishing phase.Breed influenced(P < 0.05) growth performance,carcass traits,and meat quality,but not plasma metabolites.Throughout the trial,Landrace pigs showed significantly higher(P < 0.05) ADG,ADFI,dressing percentage,lean mass rate,and loin-eye area than did Bama mini-pigs,but significantly lower(P< 0.05)feed/gain ratio,fat percentage,backfat thickness,and IMF content.Dietary protein/energy ratio influenced the pH value,chemical composition of BF and PM muscles,and plasma activities of glutamic-pyruvic transaminase and gamma-glutamyl transpeptidase,and plasma concentration of UN.Conclusions:Compared with Landrace pigs,Bama mini-pigs showed slower growth and lower carcass performance,but had better meat quality.Moreover,unlike Landrace pigs,the dietary protein/energy ratio did not affect the growth performance of Bama mini-pigs.These results suggest that,in swine production,low dietary protein/energy ratio may be useful for reducing feed costs and minimizing the adverse effects of ammonia release into the environment.Yingying Liu Xiangfeng Kong Guoli Jiang Bi'e Tan Jinping Deng Xiaojian Yang Fengna Li Xia Xiong Yulong Yin 2015Journal of Animal Science and Biotechnology2015,6,4:37
7Phylogenetic reconstruction of Chirita and allies (Gesneriaceae) with taxonomic treatments显示文摘Chirita D. Don, a large genus in the subfamily Cyrtandroideae of Gesneriaceae, has been the subject of much debate whether it is a natural group or not. In addition, the highly heterogeneous Chirita has also been very problematic with regard to delimitation and subdivision. Here we used the nrDNA internal transcribed spacer and cpDNA trnL-F for molecular phylogenetic analaysis, combined with morphological data. Our results suggest that Chirita is an artificial, polyphyletic genus. The most important character that defines Chirita, the dorso-ventrally oblique and bilamellar stigma, has evolved convergently in different clades of diandrous Cyrtandroideae. Chirita sensu stricto only includes the species of Chirita sect. Chirita, whereas Chirita sect. Microchirita is an independent clade located at the basal node of the phylogenetic tree. Chirita sect. Liebigia is closely related to Didymocarpus with an entire stigma unlike other species of Chirita. The species of Chirita sect. Gibbosaccus, Chiritopsis, Primulina, and Wentsaiboea form a monophyletic group that is sister to a strongly supported clade comprising four monotypic genera Paralagarosolen, Calcareoboea, Petrocodon, and Tengia. 8An autotrophic nitrogen removal process:Short-cut nitrification combined with ANAMMOX for treating diluted effluent from an UASB reactor fed by landfill leachate显示文摘A combined process consisting of a short-cut nitrification (SN) reactor and an anaerobic ammonium oxidation upflow anaerobic sludge bed (ANAMMOX) reactor was developed to treat the diluted effluent from an upflow anaerobic sludge bed (UASB) reactor treating high ammonium municipal landfill leachate.The SN process was performed in an aerated upflow sludge bed (AUSB) reactor (working volume 3.05 L),treating about 50% of the diluted raw wastewater.The ammonium removal efficiency and the ratio of NO 2 N to NOx-N in the effluent were both higher than 80%,at a maximum nitrogen loading rate of 1.47 kg/(m 3 ·day).The ANAMMOX process was performed in an UASB reactor (working volume 8.5 L),using the mix of SN reactor effluent and diluted raw wastewater at a ratio of 1:1.The ammonium and nitrite removal efficiency reached over 93% and 95%,respectively,after 70-day continuous operation,at a maximum total nitrogen loading rate of 0.91 kg/(m 3 ·day),suggesting a successful operation of the combined process.The average nitrogen loading rate of the combined system was 0.56 kg/(m 3 ·day),with an average total inorganic nitrogen removal efficiency 87%.The nitrogen in the effluent was mostly nitrate.The results provided important evidence for the possibility of applying SN-ANAMMOX after UASB reactor to treat municipal landfill leachate.Jie Liu Jian'e Zuo Tang Yang Shuquan Zhu Sulin Kuang Kaijun Wang 2010Journal of Environmental Sciences2010,22,5:26

12Chimeric antigen receptor-engineered T-cell therapy for liver cancer显示文摘Background: Chimeric antigen receptor-engineered T-cell(CAR-T) therapy is a newly developed immunotherapy used in the treatment of cancers. Because CAR-T therapy has shown great success in treating CD19-positive hematological malignancies, its application has been explored in the treatment of solid tumors, such as liver cancer. In this review, we discuss the immune characteristics of liver cancer, the obstacles encountered during the application of CAR-T therapy, and preclinical and clinical progress in the use of CAR-T therapy in patients with liver cancer.Data sources: The data on CAR-T therapy related to liver cancers were collected by searching Pub Med and the Web of Science databases prior to December 2017 with the keywords 'chimeric antigen receptor','CAR-T', 'liver cancer', 'hepatocellular carcinoma', and 'solid tumor'. Additional articles were identified by manual search of references found in the primary articles. The data for clinical trials were collected by searching Clinical Trials.gov.Results: The liver has a tolerogenic nature in the intrahepatic milieu and its tumor microenvironment significantly affects tumor progression. The obstacles that reduce the efficacy of CAR-T therapy in solid tumors include a lack of specific tumor antigens, limited trafficking and penetration of CAR-T cells to tumor sites, and an immunosuppressive tumor microenvironment. To overcome these obstacles, several strategies have emerged. In addition, several strategies have been developed to manage the side effects of CAR-T, including enhancing the selectivity of CARs and controlling CAR-T activity. To date, no clinical trials of CAR-T therapy against HCC have been completed. However, preclinical studies in vitro and in vivo have shown potent antitumor efficacy. Glypican-3, mucin-1, epithelial cell adhesion molecule, carcinoembryonic antigen, and other targets are currently being studied.Conclusions: The application of CAR-T therapy for liver cancer is just beginning to be explored and more research is needed. 13Bone morphogenetic protein-2 gene controls tooth root development in coordination with formation of the periodontium显示文摘Formation of the periodontium begins following onset of tooth-root formation in a coordinated manner after birth. Dental follicle progenitor cells are thought to form the cementum, alveolar bone and Sharpey's fibers of the periodontal ligament (PDL). However, little is known about the regulatory morphogens that control differentiation and function of these progenitor cells, as well as the progenitor cells involved in crown and root formation. We investigated the role of bone morphogenetic protein-2 (Bmp2) in these processes by the conditional removal of the Bmp2 gene using the Sp7-Cre-EGFP mouse model. Sp7-Cre-EGFP first becomes active at E18 in the first molar, with robust Cre activity at postnatal day 0 (P0), followed by Cre activity in the second molar, which occurs after P0. There is robust Cre activity in the periodontium and third molars by 2 weeks of age. When the Bmp2 gene is removed from Sp7+ (Osterix+) cells, major defects are noted in root, cellular cementum and periodontium formation. First, there are major cell autonomous defects in root-odontoblast terminal differentiation. Second, there are major alterations in formation of the PDLs and cellular cementum, correlated with decreased nuclear factor IC (Nfic), periostin and α-SMA+ cells. Third, there is a failure to produce vascular endothelial growth factor A (VEGF-A) in the periodontium and the pulp leading to decreased formation of the microvascular and associated candidate stem cells in the Bmp2-cKOSp7-Cre-EGFP . Fourth, ameloblast function and enamel formation are indirectly altered in the Bmp2-cKOSp7-Cre-EGFP . 14Atmospheric particulate pollution of Lanzhou using magnetic measurements显示文摘To differentiate between natural and anthropogenic particulate sources in the atmosphere in Lanzhou City, samples were collected in different sites. The dust flux was calculated and magnetic measurements were conducted. Results show a distinct pattern of variation of dust flux within a year and it agrees with the shifts of atmospheric circulation regime. The magnetic parameters indicate that natural sources are the major components of atmospheric particulate during late spring and early summer, while anthropogenic sources contribute much more during winter months. 15Intravenous formulation of Panax notoginseng root extract:human pharmacokinetics of ginsenosides and potential for perpetrating drug interactions显示文摘XueShuanTong,a lyophilized extract of Panax notoginseng roots(Sanqi)for intravenous administration,is extensively used as add-on therapy in the treatment of ischemic heart and cerebrovascular diseases and comprises therapeutically active ginsenosides.Potential for XueShuanTong-drug interactions was determined;the investigation focused on cytochrome P450(CYP)3A induction and organic anion-transporting polypeptide(OATP)1 B inhibition.Ginsenosides considerably bioavailable for drug interactions were identifed by dosing XueShuanTong in human subjects and their interaction-related pharmacokinetics were determined.The CYP3A induction potential was determined by repeatedly dosing XueShuanTong for 15 days in human subjects and by treating cryopreserved human hepatocytes with circulating ginsenosides;midazolam served as a probe substrate.Joint inhibition of OATP1B by XueShuanTong ginsenosides was assessed in vitro,and the data were processed using the Chou-Talalay method,Samples were analyzed by liquid chromatography/mass spectrometry.Ginsenosides Rb1,Rd,and Rg1 and notoginsenoside R were the major circulating XueShuanTong compounds;their interaction-related pharmacokinetics comprised compound dose-dependent levels of systemic exposure and,for ginsenosides Rb,and Rd,long terminal hal-lives(32-57 and 58-307 h,respectively)and low unbound fractions in plasma(0.8%-2.9%and 0.4%-3.0%,respectively).Dosing XueShuanTong did not induce CYP3A.Based on the pharmacokinetics and inhibitory potency of the ginsenosides,XueShuanTong was predicted to have high potential for OATP1B3-mediated drug interactions(attributed chiefly to ginsenoside Rb,)suggesting the need for further model-based determination of the interaction potential for XueShuanTong and,if necessary,a clinical drug interaction study.Increased awareness of ginsenosides'pharmacokinetics and XueShuanTong-drug interaction potential will help ensure the safe use of XueShuanTong and coadministered synthetic drugs.Salisa Pintusophon Wei Niu Xiao-na Duan Olajide E Olaleye Yu-hong Huang Feng-qing Wang Yan-fen Li Jun-ling Yang Chuan Li 2019Acta Pharmacologica Sinica2019,40,10:13

17Birth defects in pregestational diabetes:Defect range,glycemic threshold and pathogenesis显示文摘Currently,60 million women of reproductive age(18-44 years old) worldwide,and approximately 3 million American women have diabetes mellitus,and it has been estimated that this number will double by 2030.Pregestational diabetes mellitus(PGD) is a significant public health problem that increases the risk for structural birth defects affecting both maternal and neonatal pregnancy outcome.The most common types of human structural birth defects associated with PGD are congenital heart defects and central nervous system defects.However,diabetes can induce birth defects in any other fetal organ.In general,the rate of birth defects increases linearly with the degree of maternal hyperglycemia,which is the major factor that mediates teratogenicity of PGD.Stringent prenatal care and glycemic control are effective means to reduce birth defects in PGD pregnancies,but cannot reduce the incidence of birth defects to the rate of that is seen in the nondiabetic population.Studies in animal models have revealed that PGD induces oxidative stress,which activates cellular stress signalling leading to dysregulation of gene expression and excess apoptosis in the target organs,including the neural tube and embryonic heart.Activation of the apoptosis signalregulating kinase 1(ASK1)-forkhead transcription factor 3a(Fox O3a)-caspase 8 pathway causes apoptosis in the developing neural tube leading to neural tube defects(NTDs).ASK1 activates the c-Jun-N-Terminal kinase 1/2(JNK1/2),which leads to activation of the unfolded protein response and endoplasmic reticulum(ER) stress.Deletion of the ASK1 gene,the JNK1 gene,or the JNK2 gene,or inhibition of ER stress by 4-Phenylbutyric acid abrogates diabetes-induced apoptosis and reduces the formation of NTDs.Antioxidants,such as thioredoxin,which inhibits the ASK1-Fox O3a-caspase 8 pathway or ER stress inhibitors,may prevent PGD-induced birth defects.Rinat Gabbay-Benziv E Albert Reece Fang Wang Peixin Yang 2015World Journal of Diabetes2015,6,3:10

19Effect of prophylactic clip placement following endoscopic mucosal resection of large colorectal lesions on delayed polypectomy bleeding: A meta-analysis显示文摘BACKGROUND The role of prophylactic clipping for the prevention of delayed polypectomy bleeding(DPB) remains unclear and conclusions from prior meta-analyses are limited due to the inclusion of variety of resection techniques and polyp sizes.AIM To conduct a meta-analysis on the effect of clipping on DPB following endoscopic mucosal resection(EMR) of colorectal lesions ≥ 20 mm.METHODS We performed a search of PubMed and the Cochrane library for studies comparing the effect of clipping vs no clipping on DPB following endoscopic resection. The Cochran Q test and I^2 were used to test for heterogeneity. Pooling was conducted using a random-effects model.RESULTS Thirteen studies with a total of 7794 polyps were identified, of which data was available on 1701 cases of EMR of lesions ≥ 20 mm. Prophylactic clipping was associated with a lower rate of DPB(1.4%) when compared to no clipping(5.2%)(pooled OR: 0.24, 95%CI: 0.12-0.50, P < 0.001) following EMR of lesions ≥ 20 mm.There was no significant heterogeneity among the studies(I^2 = 0%, P = 0.67).CONLUSION Prophylactic clipping may reduce DPB following EMR of large colorectal lesions.Future trials are needed to further identify risk factors and stratify high risk cases in order to implement a cost-effective preventive strategy.Fares Ayoub Donevan R Westerveld Justin J Forde Christopher E Forsmark Peter V Draganov Dennis Yang 2019World Journal of Gastroenterology2019,25,18:9
