1Neutrophil-mimicking therapeutic nanoparticles for targeted chemotherapy of pancreatic carcinoma显示文摘Due to the critical correlation between inflammation and carcinogenesis, a therapeutic candidate with anti-inflammatory activity may find application in cancer therapy. Here, we report the therapeutic efficacy of celastrol as a promising candidate compound for treatment of pancreatic carcinoma via na?ve neutrophil membrane-coated poly(ethylene glycol) methyl ether-block-poly(lactic-co-glycolic acid)(PEG-PLGA) nanoparticles. Neutrophil membranecoated nanoparticles(NNPs) are well demonstrated to overcome the blood pancreas barrier to achieve pancreas-specific drug delivery in vivo. Using tumor-bearing mice xenograft model, NNPs showed selective accumulations at the tumor site following systemic administration as compared to nanoparticles without neutrophil membrane coating. In both orthotopic and ectopic tumor models,celastrol-loaded NNPs demonstrated greatly enhanced tumor inhibition which significantly prolonged the survival of tumor bearing mice and minimizing liver metastases. 2Effect of doped strontium on catalytic properties of La_(1-x)SrxMnO_(3) for rhodamine B degradation显示文摘A series of La_(1-x)Sr_(x)MnO_(3) samples were prepared by sol-gel method and used to degrade rhodamine B(RhB) in water.All samples were characterized by X-ray diffraction(XRD),X-ray photoelectron spectroscopy(XPS),N_(2) adsorption-desorption,temperature-programmed reduction of H_(2)(H_(2)-TPR) and temperature-programmed desorption of O_(2)(O_(2)-TPD).The results show that the degradation of RhB is highly dependent on the initial pH value of solution.Sr doping enhances the degradation ability of LaMnO_(3) for RhB in the time range of 0-40 min under a strong acidic environment,but all samples exhibit similar degradation rate from 40 to 60 min.In La_(0.7)Sr_(0.3)MnO_(3)-RhB reaction system,there are two different degradation pathways,including N-de-ethylation,chromophore cleavage,ring-opening and mineralization.La_(1-x)Sr_(x)MnO_(3)(x ≤0.3) has the perovskite structure of La-Mn oxides,while La_(0.6)Sr_(0.4)MnO_(3) exhibits a Sr_(0.4)MnLa_(0.6)O_(2).98 perovskite phase.Sr doping leads to distortion of rhombohedral crystal structure and increases the relative content of Mn^(4+).The perovskite structure is stable in strong acidic environment during RhB degradation,but the relative content of Mn^(4+)and Mn^(3+) on the material surface changes.Sr doped LaMnO_(3) achieves specific surface area of 58.8 m^(2)/g and total pore volume of 0.152 cm^(3)/g.Furthermore,Sr^(2+)doping improves redox properties of La-Mn oxides,and the presence of defects makes oxygen diffusion easier compared with the undoped samples.Jiaxiu Guo YueJing Ting Shen Hongdi Luo Juan Liang Shandong Yuan 2021Journal of Rare Earths2021,39,11:2
3Phosphoproteomics Reveals the AMPK Substrate Network in Response to DNA Damage and Histone Acetylation显示文摘AMP-activated protein kinase(AMPK)is a conserved energy sensor that plays roles in diverse biological processes via phosphorylating various substrates.Emerging studies have demonstrated the regulatory roles of AMPK in DNA repair,but the underlying mechanisms remain to be fully understood.Herein,using mass spectrometry-based proteomic technologies,we systematically investigate the regulatory network of AMPK in DNA damage response(DDR).Our system-wide phosphoproteome study uncovers a variety of newly-identified potential substrates involved in diverse biological processes,whereas our system-wide histone modification analysis reveals a link between AMPK and histone acetylation.Together with these findings,we discover that AMPK promotes apoptosis by phosphorylating apoptosis-stimulating of p53 protein 2(ASPP2)in an irradiation(IR)-dependent manner and regulates histone acetylation by phosphorylating histone deacetylase 9(HDAC9)in an IR-independent manner.Besides,we reveal that disrupting the histone acetylation by the bromodomain BRD4 inhibitor JQ-1 enhances the sensitivity of AMPKdeficient cells to IR.Therefore,our study has provided a resource to investigate the interplay between phosphorylation and histone acetylation underlying the regulatory network of AMPK,which could be beneficial to understand the exact role of AMPK in DDR.Yuejing Jiang Xiaoji Cong Shangwen Jiang Ying Dong Lei Zhao Yi Zang Minjia Tan Jia Li 2022Genomics, Proteomics & Bioinformatics2022,20,4:1
