1Ala-Gln improves varicocele-induced testicular injury by increasing HSP70 and antioxidant activity in male rats显示文摘This worked aimed to test the hypothesis that L-alanyl-L-glutamine(Ala-Gln)improves the varicocele-induced testicular injury,which causes male infertility.For this purpose,fifty adult male Wistar rats received the varicocele(VC)surgery at the left renal vein.Biomarkers were determined 2,4,and 8 weeks after VC(n=10/each detection).Four weeks after VC,rats received Ala-Gln(1.125 g/kg)treatment with and or saline for 1 week(n=10/each group).Rats in the sham group were also detected for biomarkers at 2,4,and 8 weeks(n=10/each detection).VC caused testicular injury detected by hematoxylin–eosin(H&E)staining,immunohistochemistry,and TUNEL assay.HSP70 mRNA was detected quantitative RT-PCR,SOD,and CAT by nitroblue tetrazolium(NBT)method and 8-OHDG by ELISA.The results showed that varicocele induced injury in the testes.The weight of the left testes was lower than that of the right testes in VC-bearing rats(p<0.01).The relative numbers of sustentacular and spermatogenic cells were decreased after VC(p<0.01).In sham-4 wk,VC-4wk,VC-5wk and Ala-Gln groups,the apoptosis index was 5.10±1.14,13.22±3.63,33.62±3.56 and 22.33±2.61,relative level of HSP70 mRNA 1.00±0.12,0.53±0.05,0.51±0.04 and 1.62±0.15 fold,SOD 16.4±0.23,13.4±0.17,10.01±1.06 and 19.53±2.26 U/mg protein,CAT 2.16±0.31,1.07±0.28,and 1.31±0.26 and 3.46±0.71 U/mg,8-OHDG 5.23±0.67,6.81±0.78,7.16±1.22 and 4.14±0.73 pg/μg DNA,respectively(p<0.01).Our results suggest that Ala-Gln prevented the VC-induced testicular injury.We have firstly reported that Ala-Gln protects against varicocele-induced testicular injuries by up-regulation of HSP70 and antioxidants,SOD and CAT,and down-regulation of oxidant 8-OHDG,resulting in reducing apoptosis in the testis.LEI YU ZHEN FAN HONGQIANG WANG WENXIAN LI TAO JING QIANG LI SHENQIAN LI PEITAO WANG XUJUN XUAN 2021BIOCELL2021,45,2:3
2Geological and geochemical prediction on deeply-seated ore-bodies in Haigou gold deposit of Jilin,China显示文摘Lode 28 is the largest gold-bearing quartz vein in Haigou gold deposit,and the lode itself contains more than 30t of gold. Geochemical study shows that 12 trace elements related gold mineralization can be divided into 5 metallogenic factors(element assemblages) .The high values of F1,F2 and F4 together indicate overlapped of multiple stages of gold mineralization,revealing high potential of gold mineralization at depth;the single high value of F4 represents the root of an existing ore body while high values of F3 and F4 show the intervals of barren sectors in the lode. Comprehensive geochemical studies show the high prospection potential below the 14th level and between lines 39-71. 4Dual-engineered cartilage-targeting extracellular vesicles derived from mesenchymal stem cells enhance osteoarthritis treatment via miR-223/NLRP3/pyroptosis axis:Toward a precision therapy显示文摘Osteoarthritis(OA)is the most common disabling joint disease with no effective disease modifying drugs.Extracellular vesicles released by several types of mesenchymal stem cells could promote cartilage repair and ameliorate OA pathology in animal models,representing a novel therapeutic strategy.In this study,we demonstrated that extracellular vesicles derived from human umbilical cord mesenchymal stem cells(hUC-EVs)could maintain chondrocyte homeostasis and alleviate OA,and further revealed a novel molecular mechanism of this therapeutic effect.miR-223,which could directly bind with the 3′UTR of NLRP3 mRNA,was found to be a key miRNA for hUC-EVs to exert beneficial effects on inflammation inhibiting and cartilage protecting.For enhancing the effect on mitigating osteoarthritis,exogenous miR-223 was loaded into hUC-EVs by electroporation,and a collagen II-targeting peptide(WYRGRL)was modified onto the surface of hUC-EVs by genetic engineering to achieve a more targeted and efficient RNA delivery to the cartilage.The dual-engineered EVs showed a maximal effect on inhibiting the NLRP3 inflammasome activation and chondrocyte pyroptosis,and offered excellent results for the treatment of OA.This study provides a novel theoretical basis and a promising therapeutic strategy for the application of engineered extracellular vesicles in OA treatment.Weixuan Liu Anqi Liu Xujun Li Ziyang Sun Zhenghua Sun Yaru Liu Gang Wang Dan Huang Hao Xiong Shiyang Yu Xintao Zhang Cunyi Fan 2023Bioactive Materials2023,,12:0
5Phase-transforming mechanical metamaterials with dynamically controllable shape-locking performance显示文摘Active mechanical metamaterials with customizable structures and deformations,active reversible deformation,dynamically controllable shape-locking performance and stretchability are highly suitable for applications in soft robotics and flexible electronics,yet it is challenging to integrate them due to their mutual conflicts.Here,we introduce a class of phase-transforming mechanical metamaterials(PMMs)that integrate the above properties.Periodically arranging basic actuating units according to the designed pattern configuration a nd positional relationship,PMMs can customize complex and diverse structures and deformations.Liquid-vapor phase transformation provides active reversible large deformation while a silicone matrix offers stretchability.The contained carbonyl iron powder endows PMMs with dynamically controllable shape-locking performance,thereby achieving magnetically assisted shape locking and energy storing in different working modes.We build a theoretical model and finite element simulation to guide the design process of PMMs,so as to develop a variety of PMMs with different functions suitable for different applications,such as a programmed PMM,reconfigurable antenna,soft lens,soft mechanical memory,biomimetic hand,biomimetic flytrap and self-contained soft gripper.PMMs are applicable to achieve various 2D deformations and 2D-to-3D deformations,and integrate multiple properties,including customizable structures and deformations,active reversible deformation,rapid reversible shape locking,adjustable energy storing and stretchability,which could open a new application avenue in soft robotics and flexible electronics.Yiding Zhong Wei Tang Huxiu Xu Kecheng Qin Dong Yan Xujun Fan Yang Qu Zhaoyang Li Zhongdong Jiao Huayong Yang Jun Zou 2023National Science Review2023,10,9:0
6Geological and geochemical indicators and prediction of deep-seated ore bodies in Jiudian gold deposit of Pingdu,Shandong Province显示文摘Based on the geology and geochemistry of gold-bearing quartz veins in the Jiudian gold deposit of Pingdu,Shandong Province,the geological and geochemical indicators of occurrence of deep-seated ore bodies in the Jiudian gold deposit have been summarized.Extent occurrence of wide,intensive,varied alteration zones around the gold-bearing quartz veins and continual presence of diorite porphyrite and lamprophyry indicate the probable occurrence of deep-seated ore bodies at depth of gold lodes.As,Sb and Hg belong to the head halo elements, and their anomalies could reveal the probable occurrence of the deep-seated ore bodies below.LIANG Bensheng LI Xujun FAN Wenliang WEN Zhimin JIANG Shenguang 2012Global Geology2012,15,4:0
