1Intestinal antigen-presenting cells in mucosal immune homeostasis:Crosstalk between dendritic cells,macrophages and B-cells显示文摘The intestinal immune system maintains a delicate balance between immunogenicity against invading pathogens and tolerance of the commensal microbiota.Inflammatory bowel disease(IBD)involves a breakdown in tolerance towards the microbiota.Dendritic cells(DC),macrophages(MΦ)and B-cells are known as professional antigen-presenting cells(APC)due to their specialization in presenting processed antigen to T-cells,and in turn shaping types of T-cell responses generated.Intestinal DC are migratory cells,unique in their ability to generate primary T-cell responses in mesenteric lymph nodes or Peyer’s patches,whilst MΦand B-cells contribute to polarization and differentiation of secondary T-cell responses in the gut lamina propria.The antigen-sampling function of gut DC and MΦenables them to sample bacterial antigens from the gut lumen to determine types of T-cell responses generated.The primary function of intestinal B-cells involves their secretion of large amounts of immunoglobulin A,which in turn contributes to epithelial barrier function and limits immune responses towards to microbiota.Here,we review the role of all three types of APC in intestinal immunity,both in the steady state and in inflammation,and how these cells interact with one another,as well as with the intestinal microenvironment,to shape mucosal immune responses.We describe mechanisms of maintaining intestinal immune tolerance in the steady state but also inappropriate responses of APC to components of the gut microbiota that contribute to pathology in IBD.Elizabeth R Mann Xuhang Li 2014World Journal of Gastroenterology2014,20,29:19
2New serological biomarkers of inflammatory bowel disease显示文摘Serological biomarkers in inflammatory bowel disease (IBD) are a rapidly expanding list of non-invasive tests for objective assessments of disease activity, early diagnosis, prognosis evaluation and surveillance. This review summarizes both old and new biomarkers in IBD, but focuses on the development and character-ization of new serological biomarkers (identifi ed since 2007). These include fi ve new anti-glycan antibodies, anti-chitobioside IgA (ACCA), anti-laminaribioside IgG (ALCA), anti-manobioside IgG (AMCA), and antibod-ies against chemically synthesized (∑) two major oligomannose epitopes, Man α-1,3 Man α-1,2 Man (∑Man3) and Man α-1,3 Man α-1,2 Man α-1,2 Man (∑Man4). These new biomarkers serve as valuable complementary tools to existing biomarkers not only in differentiating Crohn's disease (CD), ulcerative colitis (UC), normal and other non-IBD gut diseases, but also in predicting disease involvement (ileum vs colon), IBD risk (as subclinical biomarkers), and disease course (risk of complication and surgery). Interestingly, the prevalence of the antiglycan antibodies, including anti-Saccharomyces cerevisiae antibodies (ASCA), ALCA and AMCA, was found to be associated with single nucleotide polymorphisms (SNPs) of IBD susceptible genes such as NOD2/CARD15, NOD1/CARD4, toll-likereceptors (TLR) 2 and 4, and β-defensin-1. Further-more, a gene dosage effect was observed: anti-glycan positivity became more frequent as the number of NOD2/CARD15 SNPS increased. Other new serum/ plasma IBD biomarkers reviewed include ubiquitination factor E4A (UBE4A), CXCL16 (a chemokine), resistin, and apolipoprotein A-IV. This review also discusses the most recent studies in IBD biomarker discovery by the application of new technologies such as proteomics, fourier transform near-infrared spectroscopy, and mul-tiplex enzyme-linked immunosorbent assay (ELISA)'s (with an emphasis on cytokine/chemokine profiling). 3Thermal Performance Analysis of PCM Capsules Packed-Bed System with Biomimetic Leaf Hierarchical Porous Structure显示文摘As an efficient natural selection nutrient transport system,biomimetic leaf hierarchical porous structure has unique advantages in material transportation and energy transfer.Biomimetic leaf hierarchical porous structure has been widely used in solar thermochemical reactions,photocatalysis,and energy storage.To improve the thermal efficiency and reduce the power consumption,the authors introduce the idea of bionic leaf hierarchical porous structure packed-bed latent heat thermal energy storage(LHTES)system.Under the same porosity,the diameter of the PCM capsules is designed to change along the flow direction to optimize the thermal performance.The effects of velocity on temperature distribution,pressure drop,liquid fraction,and thermal storage capacity of the conventional uniform model and bionic leaf hierarchical porous model are analyzed.The results show that the bionic leaf hierarchical porous structure can thin the thickness of the thermocline,reduce the pressure drop,increase the heat transfer area,and improve the thermal response of the packed-bed compared with the conventional uniform model.The maximum increases of liquid fraction and completion rate are 36.6%and 20.3%with pressure drop reduction of 25 Pa,respectively.The maximum decrease of the above-melting point(MP)thermocline is 51.7%as well.These results provide suggestions to optimize the packed-bed LHTES system and improve its thermal performance under practical conditions.DONG Yan WANG Fuqiang YANG Luwei SHI Xuhang ZHANG Guoliang SHUAI Yong 2021Journal of Thermal Science2021,30,5:2

6scGET:Predicting Cell Fate Transition During Early Embryonic Development by Single-cell Graph Entropy显示文摘During early embryonic development,cell fate commitment represents a critical transition or“tipping point”of embryonic differentiation,at which there is a drastic and qualitative shift of the cell populations.In this study,we presented a computational approach,scGET,to explore the gene–gene associations based on single-cell RNA sequencing(scRNAseq)data for critical transition prediction.Specifically,by transforming the gene expression data to the local network entropy,the single-cell graph entropy(SGE)value quantitatively characterizes the stability and criticality of gene regulatory networks among cell populations and thus can be employed to detect the critical signal of cell fate or lineage commitment at the single-cell level.Being applied to five scRNA-seq datasets of embryonic differentiation,scGET accurately predicts all the impending cell fate transitions.After identifying the“dark genes”that are non-differentially expressed genes but sensitive to the SGE value,the underlying signaling mechanisms were revealed,suggesting that the synergy of dark genes and their downstream targets may play a key role in various cell development processes.The application in all five datasets demonstrates the effectiveness of scGET in analyzing scRNA-seq data from a network perspective and its potential to track the dynamics of cell differentiation.The source code of scGET is accessible at http://gffzz188fe103f8f1460asoocc5f9ubbcf6xwq.ffgz.tsg.suse.edu.cn/zhongjiayuna/scGET_Project.Jiayuan Zhong Chongyin Han Xuhang Zhang Pei Chen Rui Liu 2021Genomics, Proteomics & Bioinformatics2021,19,3:1
7Impact history and origin of lunar meteorite Northwest Africa 15528显示文摘No samples from the Moon’s far side have been returned to Earth.Thus,lunar meteorite study can deepen the understanding of the Moon’s far side(if we can identify which lunar meteorites came from the Moon’s far side).The Northwest Africa(NWA)15528 meteorite is a lunar feldspathic regolith breccia meteorite and we speculated it might originate from the Feldspathic Highlands Terrane(FHT)on the far side of the Moon.Here,we report detailed petrography,major and trace elements,and noble gas(He,Ne,and Ar)isotopes for the clasts and matrix,respectively.The results show that the NWA 15528 lunar meteorite contains diverse clasts,including anorthosite,granulite,basalt,and impact melt clasts.The coarse,wellcrystallized,uniform chemical composition minerals may come from intrusive plutonic rocks.Among the anorthosite clasts,the norite/olivine clasts originate from the deep lunar crust,whereas the other anorthosite clasts are from lunar highlands.The Sm concentrations in NWA 15528 were similar to those in the fourth group of Apollo 16 melt samples,demonstrating that NWA 15528 has a typical plagioclase highland meteorite composition.Compared with the Apollo sample data and remote sensing results,the chemical composition of NWA 15528 indicated strong affinities with the FHT area and ferroan anorthosite(FAN)material from the far side of the Moon.The noble gas isotopic composition of NWA 15528 is consistent with a binary mixture of solar wind and cosmogenic components;during stepwise pyrolytic extractions,we observed that the abundance of cosmogenic components decreased,whereas that of solar wind components increased with increasing temperature.The average cosmic-ray exposure(CRE)age of the matrix and granulite is 42±6 Ma,with a shielding depth in the same range of 10-20 g cm^(−2).The gas retention age of NWA 15528 is 2.14 Ga,and the antiquity age of NWA 15528 is(0.69-0.74)±0.2 Ga(considering 50%40Arm is 40Artrap)which indicates the different clasts of NWA 15528 are assembled after 0.69-0.74 Ga.Runchuan LIU Huaiyu HE Thomas SMITH Hanqing YE Huapei WANG Xuhang ZHANG Fei SU Ziheng LIU Wei GUO Peng YANG Chuantong ZHANG Dingshuai Xue Ting CAO Jiannan LI Xuelong JIANG Ye HE Chao LU 2023Science China Earth Sciences2023,66,6:0
8A novel fabrication technique for three-dimensional concave nanolens arrays显示文摘A novel facile technique is proposed for fabricating three-dimensional(3D)concave nanolens arrays on a silicon substrate.The technique leverages an inherent characteristic of the polymethyl methacrylate(PMMA)resist during inductively coupled plasma(ICP)etching.The tendency for plasma ions to accumulate at the edge of the PMMA resist helps create a local electric field that causes the ions to etch the sidewall of the PMMA resist.This process progressively increases the uncovered area,resulting in a graded etched depth or a concave structure in the substrate.In addition,using a given ICP etching recipe,the time required for a PMMA resist to be removed by sidewall etching is determined by its width.The use of PMMA resist of different widths enables one to achieve structures of varying etched depths and thus a 3D lens array.Optical characteristics of the fabricated nanolens were simulated using the FDTD(Finite-difference time-domain)method,and focal lengths ranging from 150 nm to 420 nm were obtained.This type of nanolens is very useful in ultraviolet optical devices and CMOS image sensors.Tianli Duan Kang Xu Zhihong Liu Chenjie Gu Jisheng Pan Diing Shenp Ang Rui Zhang Yao Wang Xuhang Ma 2020Journal of Materiomics2020,6,3:0
9Brain Functional Network Generation Using Distribution-Regularized Adversarial Graph Autoencoder with Transformer for Dementia Diagnosis显示文摘The topological connectivity information derived from the brain functional network can bring new insights for diagnosing and analyzing dementia disorders.The brain functional network is suitable to bridge the correlation between abnormal connectivities and dementia disorders.However,it is challenging to access considerable amounts of brain functional network data,which hinders the widespread application of data-driven models in dementia diagnosis.In this study,a novel distribution-regularized adversarial graph auto-Encoder(DAGAE)with transformer is proposed to generate new fake brain functional networks to augment the brain functional network dataset,improving the dementia diagnosis accuracy of data-driven models.Specifically,the label distribution is estimated to regularize the latent space learned by the graph encoder,which canmake the learning process stable and the learned representation robust.Also,the transformer generator is devised to map the node representations into node-to-node connections by exploring the long-term dependence of highly-correlated distant brain regions.The typical topological properties and discriminative features can be preserved entirely.Furthermore,the generated brain functional networks improve the prediction performance using different classifiers,which can be applied to analyze other cognitive diseases.Attempts on the Alzheimer’s Disease Neuroimaging Initiative(ADNI)dataset demonstrate that the proposed model can generate good brain functional networks.The classification results show adding generated data can achieve the best accuracy value of 85.33%,sensitivity value of 84.00%,specificity value of 86.67%.The proposed model also achieves superior performance compared with other related augmentedmodels.Overall,the proposedmodel effectively improves cognitive disease diagnosis by generating diverse brain functional networks.Qiankun Zuo Junhua Hu Yudong Zhang Junren Pan Changhong Jing Xuhang Chen Xiaobo Meng Jin Hong 2023Computer Modeling in Engineering & Sciences2023,137,12:0
10Domesticated HERV-W env contributes to the activation of the small conductance Ca^(2+)-activated K^(+)type 2 channels via decreased 5-HT4 receptor in recent-onset schizophrenia显示文摘The human endogenous retroviruses type W family envelope(HERV-W env)gene is located on chromosome 7q21-22.Our previous studies show that HERV-W env is elevated in schizophrenia and HERV-W env can increase cal-cium influx.Additionally,the 5-HTergie system and particularly 5-hydroxytryptamine(5-HT)receptors play a prominent role in the pathogenesis and treatment of schizophrenia.5-hydroxytryptamine receptor 4(5-HT4R)agonist can block calcium channels.However,the underlying relationship between HERV-W env and 5-HT4R in the etiology of schizophrenia has not been revealed.Here,we used enzyme-linked immunosorbent assay to detect the concentration of HERV-W env and 5-HT4R in the plasma of patients with schizophrenia and we found that there were decreased levels of 5-HT4R and a negative correlation between 5-HT4R and HERV-W env in schizophrenia.Overexpression of HERV-W env decreased the transcription and protein levels of 5-HT4R but increased small conductance Ca^(2+)-activated K^(+)type 2 channels(SK2)expression levels.Further studies revealed that HERV-w env could interact with 5-HT4R.Additionally,luciferase assay showed that an essential region(-364 to-176 from the transcription start site)in the SK2 promoter was required for HERV-W env-induced SK2 expression.Importantly,5-HT4R participated in the regulation of SK2 expression and promoter activity.Electrophysiological recordings suggested that HERV-Wenv could increase SK2 channel currents and the increase of SK2 currents was inhibited by 5-HT4R.In condusion,HERV-W env could activate SK2 channels via decreased 5-HT4R,which might exhibit a novel mechanism for HERV-Wenv to influence neuronal activity in schizophrenia.Xiulin Wua Qiujin Yan Lianzhong Liu Xing Xue Wei Yao Xuhang Li Wenshi Li Shuang Ding Yaru Xia Dongyan Zhang Fan Zhu 2023Virologica Sinica2023,38,1:0
