1Transplantation of Human Bone Marrow Mesenchymal Stem Cell Ameliorates the Autoimmune Pathogenesis in MRL/lpr Mice显示文摘Recent evidence indicates that mesenchymal stem cells (MSC) possess immunosuppressive properties both in vitro and in vivo. We previously demonstrated the functional abnormality of bone marrow derived MSC in patients with systemic lupus erythematosus (SLE). In this study, we aimed to investigate whether transplantation of human bone marrow derived MSC affects the autoimmune pathogenesis in MRL/lpr mice. We found that human MSC from healthy donors reduced the proliferation of T lymphocytes from MRL/lpr mice in a dose-dependent fashion. Two weeks after in vivo transfer of MSC, we detected significantly reduced serum levels of anti ds-DNA antibodies and 24 hour proteinuria in MRL/lpr mice as compared with control groups without MSC transplantation. Moreover, flow cytometric analysis revealed markedly reduced number of CD4+ T cells while increased Th1 subpopulation in MSC group and MSC + CTX group when compared with controls. Histopathological examination showed significantly reduced renal pathology in MSC-treated mice. Immunohistochemical studies further revealed reduced expression of TGF-β, FN, VEGF and the deposition of complement C3 in renal tissue after MSC and MSC + CTX treatment. 3Activated carbon adsorption of quinolone antibiotics in water:Performance,mechanism,and modeling显示文摘The extensive use of antibiotics has led to their presence in the aquatic environment, and introduces potential impacts on human and ecological health. The capability of powdered activated carbon(PAC) to remove six frequently used quinolone(QN) antibiotics during water treatment was evaluated to improve drinking water safety. The kinetics of QN adsorption by PAC was best described by a pseudo second-order equation, and the adsorption capacity was well described by the Freundlich isotherm equation. Isotherms measured at different p H showed that hydrophobic interaction, electrostatic interaction,and π–π dispersion force were the main mechanisms for adsorption of QNs by PAC. A p H-dependent isotherm model based on the Freundlich equation was developed to predict the adsorption capacity of QNs by PAC at different p H values. This model had excellent prediction capabilities under different laboratory scenarios. Small relative standard derivations(RSDs), i.e., 0.59%–0.92% for ciprofloxacin and 0.09%–3.89% for enrofloxacin,were observed for equilibrium concentrations above the 0.3 mg/L level. The RSDs increased to 11.9% for ciprofloxacin and 32.1% for enrofloxacin at μg/L equilibrium levels, which is still acceptable. 4Effect of particle size on the microstructure and thermal conductivity of Al/diamond composites prepared by spark plasma sintering显示文摘Spark plasma sintering (SPS) was used to fabricate Al/diamond composites. The influence of diamond particle size on the microstructure and thermal conductivity (TC) of composites was investigated by combining experimental results with model prediction. The results show that both composites with 40 μm particles and 70 μm particles exhibit high density and good TC, and the composite with 70 μm particles indicates an excellent TC of 325 W·m-1·K-1. Their TCs lay between the theoretical estimated bounds. In contrast, the composite with 100 μm particles demonstrates low density as well as poor TC due to its high porosity and weak interfacial bonding. Its TC is even considerably less than the lower bound of the predicted value. 5Survival of transplanted neurotrophin-3 expressing human neural stem cells and motor function in a rat model of spinal cord injury显示文摘<正>BACKGROUND:Many methods have been attempted to repair nerves following spinal cord injury, including peripheral nerve transplantation,Schwann cell transplantation,olfactory ensheathing cell transplantation,and embryonic neural tissue transplantation.However,there is a need for improved outcomes. OBJECTIVE:To investigate the repair feasibility for rat spinal cord injury using human neural stem cells(hNSCs) genetically modified by lentivirus to express neurotrophin-3. DESIGN,TIME AND SETTING:In vitro cell biological experiment and in vivo randomized,controlled, genetic engineering experiment were performed at the Third Military Medical University of Chinese PLA and First People's Hospital of Yibin,China from March 2006 to December 2007. MATERIALS:A total of 64 adult,female,Wistar rats were used for the in vivo study.Of them,48 rats were used to establish models of spinal cord hemisection,and were subsequently equally and randomly assigned to model,genetically modified hNSC,and normal hNSC groups.The remaining 16 rats served as normal controls. METHODS:hNSCs were in vitro genetically modified by lentivirus to secrete both green fluorescence protein and neurotrophin-3.Neurotrophin-3 expression was measured by Western blot. Genetically modified hNSC or normal hNSC suspension(5×10~5) was injected into the rat spinal cord following T_(10) spinal cord hemisection.A total of 5μL Dulbecco's-modified Eagle's medium was infused into the rat spinal cord in the model group.Transgene expression and survival of transplanted hNSCs were determined by immunohistochemistry.Motor function was evaluated using the Basso,Beattie,and Bresnahan(BBB) scale. MAIN OUTCOME MEASURES:The following parameters were measured:expression of neurotrophin-3 produced by genetically modified hNSCs,transgene expression and survival of hNSCs in rats,motor function in rats. RESULTS:hNSCs were successfully genetically modified by lentivirus to stably express neurotrophin-3.The transplanted hNSCs primarily gathered at,or around,the injection site two weeks following transplantation,and gradually migrated towards the surrounding tissue. Transplanted hNSCs were observed 7.0-8.0 mm away from the injection site.In addition,hNSCs were observed 10 weeks after transplantation.At week 4,BBB locomotor scores were significantly greater in the genetically modified hNSC and normal hNSC groups,compared with the model group (P<0.05),and scores were significantly greater in the genetically modified hNSC group compared with the normal hNSC group(P<0.05). 6Effect of powder mixing process on the microstructure and thermal conductivity of Al/diamond composites fabricated by spark plasma sintering显示文摘Two powder mixing processes, mechanical mixing (MM) and mechanical alloying (MA), were used to prepare mixed Al/diamond powders, which were subsequently consolidated using spark plasma sintering (SPS) to produce bulk Al/diamond composites. The effects of the powder mixing process on the morphologies of the mixed powders, the microstructure and the thermal conductivity of the composites were investigated. The results show that the powder mixing process can significantly affect the microstructure and the thermal conductivity of the composites. Agglomerations of the particles occurred in mixed powders using MM for 30 min, which led to high pore content and weak interfacial bonding in the composites and resulted in low relative density and low thermal conductivity for the composites. Mixed powders of homogeneous distribution of diamond particles could be obtained using MA for 10 min and MM for 2 h. The composite prepared through MA indicated a high relative density but low thermal conductivity due to its defects, such as damaged particles, Fe impurity, and local interfacial debonding, which were mainly introduced in the MA process. 7MicroRNA-663 induces immune dysregulation by inhibiting TGF-β1 production in bone marrow-derived mesenchymal stem cells in patients with systemic lupus erythematosus显示文摘Mesenchymal stem cells(MSCs)are critical for immune regulation.Although several microRNAs(miRNAs)have been shown to participate in autoimmune pathogenesis by affecting lymphocyte development and function,the roles of miRNAs in MSC dysfunction in autoimmune diseases remain unclear.Here,we show that patients with systemic lupus erythematosus(SLE)display a unique miRNA signature in bone marrow-derived MSCs(BMSCs)compared with normal controls,among which miR-663 is closely associated with SLE disease activity.MiR-663 inhibits the proliferation and migration of BMSCs and impairs BMSC-mediated downregulation of follicular T helper(Tfh)cells and upregulation of regulatory T(Treg)cells by targeting transforming growth factorβ1(TGF-β1).MiR-663 overexpression weakens the therapeutic effect of BMSCs,while miR-663 inhibition improves the remission of lupus disease in MRL/lpr mice.Thus,miR-663 is a key mediator of SLE BMSC regulation and may serve as a new therapeutic target for the treatment of lupus.Linyu Geng Xiaojun Tang Kangxing Zhou Dandan Wang Shiying Wang Genhong Yao Weiwei Chen Xiang Gao Wanjun Chen Songtao Shi Nan Shen Xuebing Feng Lingyun Sun 2019Cellular & Molecular Immunology2019,16,3:16
8Relationship between the -455G/A and -148C/T polymorphisms in the beta-fibrinogen gene and cerebral infarction in the Xinjiang Uygur and Han Chinese populations显示文摘We sought to investigate the correlation between the -455G/A and -148C/T polymorphisms of the β-fibrinogen gene and plasma fibrinogen levels in patients with cerebral infarction and in healthy subjects among the Xinjiang Uygur and Han Chinese populations, by using polymerase chain reaction-restriction enzyme digestion analysis. Results showed that there were no statistically significant differences in the distributions of the -455G/A genotype and allele frequency between the Uygurs and the Han. Plasma fibrinogen levels in cerebral infarction patients among the Uygurs and the Han were higher than those among healthy subjects. In particular, the frequencies of the -455G/A AA and -148C/T TT genotypes were significantly higher than in healthy subjects. Individuals carrying the A or T allele had a higher incidence of cerebral infarction compared with those carrying the G or C allele. Our experimental findings indicate that the -148C/T and -455G/A polymorphisms are associated with cerebral infarction in Xinjiang Uygur and Han Chinese subjects. 9Therapeutic potential of intravenous Xuebijing on transforming growth factor β_1 and procollagen type III peptide in patients with acute paraquat poisoning显示文摘OBJECTIVE:Paraquat(PQ) poisoning-induced pulmonary fibrosis causes asphyxiation and death.The therapeutic potential of intravenous Xuebijing therapy in PQ poisoning patients and its underlying immunomodulatory effects on transforming growth factor(TGF)-β1 and procollagen type III peptide(PIIIP) were investigated.METHODS:Thirty-six acute PQ poisoning patients were randomly assigned to conventional therapy(Group A) and intravenous Xuebijing administration plus conventional therapy(Group B).Twenty volunteers served as controls(Group C).Blood samples were collected upon admission(day 0) and at post-treatment days 5,10,and 14.TGF-β1 and PIIIP concentrations were determined by ELISA and analyzed for intra-and inter-group differences over time.One-month follow-up was conducted for determining the mortality rate.RESULTS:TGF-β1 and PIIIP levels were significantly higher in PQ poisoning patients and increased over time(Groups A and B vs C,P<0.01).However,the TGF-β1 and PIIIP levels were consistently significantly lower in Group B compared with those of Group A(P<0.01).The 1-month mortality rate was also lower in Group B compared with that of Group A(P<0.05).PQ poisoning patients showed remarkably high levels of TGF-β1 and PIIIP,which increased as PQ-induced pulmonary fibrosis progressed.CONCLUSION:Treatment with intravenous Xuebijing plus conventional therapy significantly lowered TGF-β1 and PIIIP levels,which indicates therapeutic efficacy in the treatment of PQ poisoning patients.Xuebing Zheng Xiaoli Sun Piyong Ma Zhongming Liu Nan Jiang 2012Journal of Traditional Chinese Medicine2012,32,4:11

12Lignocellulosic biomass as sustainable feedstock and materials for power generation and energy storage显示文摘Lignocellulosic biomass has attracted great interest in recent years for energy production due to its renewability and carbon-neutral nature.There are various ways to convert lignocellulose to gaseous,liquid and solid fuels via thermochemical,chemical or biological approaches.Typical biomass derived fuels include syngas,bio-gas,bio-oil,bioethanol and biochar,all of which could be used as fuels for furnace,engine,turbine or fuel cells.Direct biomass fuel cells mediated by various electron carriers provide a new direction of lignocellulose conversion.Various metal and non-metal based carriers have been screened for mediating the electron transfer from biomass to oxygen thus generating electricity.The power density of direct biomass fuel cells can be over 100 mW cm^(-2),which shows promise for practical applications.Lignocellulose and its isolated components,primarily cellulose and lignin,have also been paid considerable attention as sustainable carbonaceous materials for preparation of electrodes for supercapacitors,lithium-ion batteries and lithium-sulfur batteries.In this paper,we have provided a state-of-the-art review on the research progress of lignocellulosic biomass as feedstock and materials for power generation and energy storage focusing on the chemistry aspects of the processes.It was recommended that process integration should be performed to reduce the cost for thermochemical and biological conversion of lignocellulose to biofuels,while efforts should be made to increase efficiency and improve the properties for biomass fuelled fuel cells and biomass derived electrodes for energy storage.Fangqian Wang Denghao Ouyang Ziyuan Zhou Samuel JPage Dehua Liu Xuebing Zhao 2021Journal of Energy Chemistry2021,30,6:8
13Predicted interfacial thermal conductance and thermal conductivity of diamond/Al composites with various interfacial coatings显示文摘The interfacial thermal conductance (ITC) and thermal conductivity (TC) of diamond/Al composites with various coatings were theoretically studied and discussed. A series of predictions and numerical analyses were performed to investigate the effect of thickness, sound velocity, and other parameters of coating layers on the ITC and TC. It is found that both the ITC and TC decline with increasing coating thickness, especially for the coatings with relatively low thermal conductivity. Nevertheless, if the coating thickness is close to zero, or quite a small value, the ITC and TC are mainly determined by the constants of the coating material. Under this condition, coatings such as Ni, TiC, Mo 2 C, SiC, and Si can significantly improve the ITC and TC of diamond/Al composites. By contrast, coatings like Ag will exert the negative effect. 14Ketogenic diet alleviates colitis by reduction of colonic group 3 innate lymphoid cells through altering gut microbiome显示文摘Accumulating evidence suggests that ketogenic diets(KDs)mediate the rise of circulating ketone bodies and exert a potential antiinflammatory effect;however,the consequences of this unique diet on colitis remain unknown.We performed a series of systematic studies using a dextran sulfate sodium(DSS)animal model of inflammatory colitis.Animals were fed with a KD,low-carbohydrate diet(LCD),or normal diet(ND).Germ-free mice were utilized in validation experiments.Colon tissues were analyzed by transcriptome sequencing,RT2 profiler PCR array,histopathology,and immunofluorescence.Serum samples were analyzed by metabolic assay kit.Fecal samples were analyzed by 16S rRNA gene sequencing,liquid chromatography-mass spectrometry and gas chromatography-mass spectrometry.We observed that KD alleviated colitis by altering the gut microbiota and metabolites in a manner distinct from LCD.Quantitative diet experiments confirmed the unique impact of KD relative to LCD with a reproducible increase in Akkermansia,whereas the opposite was observed for Escherichia/Shigella.After colitis induction,the KD protected intestinal barrier function,and reduced the production of R0Ryt+CD3_group 3 innate lymphoid cells(ILC3s)and related inflammatory cytokines(IL-17a,IL-18,IL-22,Ccl4).Finally,fecal microbiota transplantation into germ-free mice revealed that the KD-mediated colitis inhibition and ILC3 regulation were dependent on the modification of gut microbiota.Taken together,our study presents a global view of microbiome-metabolomics changes that occur during KD colitis treatment,and identifies the regulation of gut microbiome and ILC3s as novel targets involving in IBD dietary therapy.Cheng Kong Xuebing Yan Yongqiang Liu Linsheng Huang Yefei Zhu Jide He Renyuan Gao Matthew F.Kalady Ajay Goel Huanlong Qin Yanlei Ma 2021Signal Transduction and Targeted Therapy2021,6,5:8
15Alterations in intestinal microbiota of colorectal cancer patients receiving radical surgery combined with adjuvant CapeOx therapy显示文摘An intricate relationship exists and interactions occur between gut microbiota and colorectal cancer(CRC).Radical surgery combined with adjuvant chemotherapy(AC) serves as the mainstream therapeutic scheme for most CRC patients.The current research was conducted to assess the effect of surgery or chemotherapy on gut microbiota.Forty-three CRC patients who received radical surgery and AC were enrolled.Fecal samples were collected preoperatively,postoperatively,and after the first to fifth cycles of postoperative chemotherapy.The microbial community of each sample was analyzed using high throughput 16S rRNA amplicon sequencing.Compared with preoperative samples,fecal samples collected postoperatively exhibited a significant decrease of obligate anaerobes,tumor-related bacteria,and butyric acid-producing bacteria.However,a significant increase of some conditional pathogens was observed.In addition,the AC regimen(CapeOx) was found to alter intestinal microbiota dramatically.In particular,several changes were observed after chemotherapy including an increase of pathogenic bacteria,the 'rebound effect' of chemotherapy-adapted bacteria,the shift of lactate-utilizing microbiota from Veillonella to Butyricimonas and Butyricicoccus,as well as the decrease of probiotics.Both radical surgery and CapeOx chemotherapy exert a non-negligible effect on the gut microbiota of CRC patients.Microbiota-based intervention may be beneficial for patients during postoperative clinical management.Cheng Kong Renyuan Gao Xuebing Yan Linsheng Huang Jide He Hao Li Jie You Huanlong Qin 2019Science China(Life Sciences)2019,62,9:7
16Effect of processing parameters on the microstructure and thermal conductivity of diamond/Ag composites fabricated by spark plasma sintering显示文摘Diamond/metal composites with 50 vol.% diamond have been produced by spark plasma sintering(SPS) using pure Ag as a matrix and diamond particles as reinforcement.Three kinds of powder mixing processes were used to prepare the mixture of diamond/Ag powders:dry mixing without milling medium,wet mixing and magnetic blending.Subsequently,they were all consolidated by SPS at various processing parameters to produce bulk diamond/Ag composites.Then samples were heat treated in order to obtain a higher thermal conductivity.The effect of processing parameters on the morphologies of the mixed powders,the microstructure and the thermal conductivity of the composites were investigated by comparing the experimental data.It reveals that particles were easy to agglomerate and the distribution of mixed powders was inhomogeneous by dry mixing method,and wet mixing method is too complex.The most favorable mixing process is magnetic blending by which the powders can be homogenously mixed and the composites prepared by optimized SPS processing parameters can obtain the highest relative density and the best thermal conductivity among the composites prepared by different processes.The magnetic blending diamond/Ag composites even have a 23% increase in thermal conductivity compared with pure silver sintered by SPS.GAO Wenjia,JIA Chengchang,JIA Xian,LIANG Xuebing,CHU Ke,ZHANG Luman,HUANG Hai,and LIU Meng School of Materials Science and Engineering,University of Science and Technology Beijing,Beijing 100083,China 2010Rare Metals2010,29,6:6

19Lipase-Catalyzed Synthesis of Sn-2 Palmitate: A Review显示文摘Human milk fat(HMF)is an important source of nutrients and energy for infants.Triacylglycerols(TAGs)account for about 98%of HMF and have a unique molecular structure.HMF is highly enriched in palmitic acid(PA)at the sn-2 position of the glycerol backbone(more than 70%)and in unsaturated fatty acids at the sn-1,3 position.The specific TAG structure in HMF plays a valuable function in infant growth.Sn-2 palmitate(mainly 1,3-dioleoyl-2-palmitoyl-glycerol)is one of the structured TAGs that is commonly supplemented into infant formula in order to enable it to present a similar structure to HMF.In this review,the development of the lipase-catalyzed synthesis of sn-2 palmitate over the last 25 years are summarized,with a focus on reaction schemes in a laboratory setting.Particular attention is also paid to the commercialized sn-1,3 regioselective lipases that are used in structured TAGs synthesis,to general methods of TAG analysis,and to successfully developed sn-2 palmitate products on the market.Prospects for the lipase-catalyzed synthesis of sn-2 palmitate are discussed.Wei Wei Cong Sun Xiaosan Wang Qingzhe Jin Xuebing Xu Casimir CAkoh Xingguo Wang 2020Engineering2020,6,4:5
20Fault detection of flywheel system based on clustering and principal component analysis显示文摘Considering the nonlinear, multifunctional properties of double-flywheel with closedloop control, a two-step method including clustering and principal component analysis is proposed to detect the two faults in the multifunctional flywheels. At the first step of the proposed algorithm,clustering is taken as feature recognition to check the instructions of ‘‘integrated power and attitude control' system, such as attitude control, energy storage or energy discharge. These commands will ask the flywheel system to work in different operation modes. Therefore, the relationship of parameters in different operations can define the cluster structure of training data. Ordering points to identify the clustering structure(OPTICS) can automatically identify these clusters by the reachability-plot. K-means algorithm can divide the training data into the corresponding operations according to the reachability-plot. Finally, the last step of proposed model is used to define the relationship of parameters in each operation through the principal component analysis(PCA) method.Compared with the PCA model, the proposed approach is capable of identifying the new clusters and learning the new behavior of incoming data.
