| 1 | Grasping institutional complexity in infrastructure megaprojects through the multi-level governance system: A case study of the Hong Kong-Zhuhai-Macao Bridge construction显示文摘This study analyzes the design and operation of multi-level governance system for the smooth delivery of infrastructure mega-projects with high institutional complexity caused by market transition. From an institutional perspective, this study scrutinizes the structure,elements, and dynamics of the governance system of infrastructure mega-projects and then proposes an integrative framework based on the inductive case study of the Hong Kong-Zhuhai-Macao Bridge mega-project. Multiple evidences of archives, field studies, and interviews related to the case project are triangulated to further analyze the institutional effects, specifically those with government logics and market structures, on the design and operation of the three-level governance system. Results reveal that the co-evolution between governments and markets in China has shaped the vertical levels of the mega-project governance system and has further affected their evolution and operation across various stages of project development. Based on the results of a field study, an in-depth interview, and archive data, this paper introduces the ecological compensation for Sousa chinensis(the Indo-Pacific humpbacked dolphin) during the construction of the Hong Kong-Zhuhai-Macao Bridge. It studies the concrete measures, decision-making processes,and organizational collaboration of the ecological compensation, using the method of a case study. | 8 | Enhanced a novel β-agarase production in recombinant Escherichia coli BL21(DE3) through induction mode optimization and glycerol feeding strategy显示文摘Agarases are hydrolytic enzymes that act on the hydrolysis of agar and have a broad range of applications in food,cosmetics and pharmaceutical industries. In this study, a glycerol feeding strategy based on induction mode optimization for high cell density and β-agarase production was established, which could effectively control acetate yield. First, exponential feeding strategy of glycerol with different overall specific growth rates(μ) was applied in the pre-induction phase. The results showed that the low μ(μ=0.2) was suggested to be the optimal for cell growth and β-agarase production. Second, the effects of induction temperature and the inducer concentration on cell growth and β-agarase production were investigated in the post-induction phase. When induced by isopropyl-β-d-thiogalactoside(IPTG), the strategy of 0.8 mmol/L IPTG induction at 20℃ was found to be optimal for β-agarase production. When cultivation was induced by continuous lactose feeding strategy of 1.0 g/(L·h), the β-agarase activity reached 112.5 U/mL, which represented the highest β-agarase production to date.Furthermore, the β-agarase was capable of degrading G. lemaneiformis powder directly to produce neoagarooligosaccharide, and the hydrolysates were neoagarotetraose(NA4) and neoagarohexaose(NA6). | 9 | Chiral effect on Aβ fibrillation from molecular-scale to nanoscale显示文摘β-Amyloid(Aβ)peptide fibrillation,one of the characteristic hallmarks of Alzheimer’s disease,is determined by many interfacial physical-chemical factors,e.g.,charge,hydrophobicity,etc.Despite extensive research,chiral effect in different-scales on the fibrillation process of Aβremains unclear.Herein,molecular-scale,sub-nanoscale,and nanoscale chiral-structures were constructed to investigate their chiral effect on the fibrillation of Aβ_(40) peptides.Chiral structures from molecular-scale to nanoscale were obtained from the different periods of the chemosynthesis process of chiral ZnS quantum-dots(QDs),confirmed by real-time monitoring of circular dichroism spectra.For molecular-scale,both L-penicillamine(L-P)and D-P ligands accelerated the fibrillation of Aβ_(40),and the speed-up effect of D-P was slightly stronger than L-P.For sub-nanoscale,both two chiral Zncomplexes(L-Zn and D-Zn)induced the agglomeration of Aβ_(40) without chirality discrimination.For nanoscale,both L-ZnS and DZnS QDs inhibited the fibrillation of Aβ_(40),and the inhibition effect of L-ZnS was notably better than that of D-ZnS.In-situ kinetics experiments of Aβ_(40) co-incubated with two chiral QDs demonstrated that L-ZnS completely prevents the misfolding of Aβ_(40) from unfolded toβ-sheet,while D-ZnS cannot achieve this.Further site-replacement experiments and simulation results revealed the underlying molecular mechanisms of the different inhibition efficiency of chiral ZnS QDs on Aβ_(40) fibrillation,which mainly attribute to the stereoselectivity interaction between the chiral ligands of ZnS QDs and electro-positive amino acid residues(R5,K16,and K28)of Aβ_(40).This work offers a microscopic insight of chiral effect on Aβfibrillation exerted by structures in different-scales,and provides a guidance in precise regulation of protein fibrillation via manipulating chiral structures in different-scales. | Guanbin Gao Guowei Zhu Liangchong Yu Zijun Zhang Ting Zhang Xinglin Liu Cheng Zhang Lin Zhou Taolei Sun | 2022 | Nano Research2022,15,7: | 0 |

Moreover, this paper proposes the theoretical concept, framework, and functions for the IMP complex ecosystem based on complex ecosystem theory.First, the coupling process between IMP complex ecosystem subsystems is analyzed through material flows, energy flows, information flows, and value streams.Second, a logistic model of the IMP complex ecosystem is proposed by analyzing the evolution conditions and motivations. Third, the evolution pattern of the IMP complex ecosystem is determined. Fourth, the positive evolution strategy of the IMP complex ecosystem based on dissipative structure theory and the influencing factors of the evolutionary process is introduced. Finally, the Hong Kong-Zhuhai-Macao Bridge and Sousa chinensis are used as the case study.
