2Identification of genes contributing to quantitative disease resistance in rice显示文摘Despite the importance of quantitative disease resistance during a plant’s life, little is known about the molecular basis of this type of host-pathogen interaction, because most of the genes underlying resistance quantitative trait loci (QTLs) are unknown. To identify genes contributing to resistance QTLs in rice, we analyzed the colocalization of a set of characterized rice defense-responsive genes and resistance QTLs against different pathogens. We also examined the expression patterns of these genes in response to pathogen infection in the parents of the mapping populations, based on the strategy of validation and functional analysis of the QTLs. The results suggest that defense-responsive genes are important resources of resistance QTLs in rice. OsWRKY45-1 is the gene contributing to a major resistance QTL.NRR,OsGH3-1,and OsGLP members on chromosome 8 contribute alone or collectively to different minor resistance QTLs. 3Temperature dependent Raman and photoluminescence of vertical WS2/MoS2 monolayer heterostructures显示文摘Heterostructures from two-dimensional transition-metal dichalcogenides MX_2 have emerged as a hot topic in recent years due to their various fascinating properties. Here, we investigated the temperature dependent Raman and photoluminescence(PL) spectra in vertical stacked WS_2/MoS_2 monolayer heterostructures. Our result shows that both E_(2g)~1 and A_(1g) modes of WS_2 and MoS_2 vary linearly with temperature increasing from 300 to 642 K. The PL measurement also reveals strong temperature dependencies of the PL intensity and peak position. The activation energy of the thermal quenching of the PL emission has been found to be equal to 69.6 meV. 4Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research.Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao 2021Cancer Biology & Medicine2021,18,1:5

6Mechanistic-empirical pavement design guide(MEPDG):a bird's-eye view显示文摘Past editions of the American Association of State Highway and Transportation Officials(AASHTO) Guide for Design of Pavement Structures have served well for several decades;nevertheless,many serious limitations exist for their continued use as the nation's primary pavement design procedures.Researchers are now incorporating the latest advances in pavement design into the new Mechanistic-Empirical Pavement Design Guide(MEPDG),developed under the National Cooperative Highway Research Program(NCHRP) 1-37A project and adopted and published by AASHTO.The MEPDG procedure offers several dramatic improvements over the current pavement design guide and presents a new paradigm in the way pavement design is performed.However,MEPDG is substantially more complex than the AASHTO Design Guide by considering the input parameters that influence pavement performance,including traffic,climate,pavement structure and material properties,and applying the principles of engineering mechanics to predict critical pavement responses.It requires significantly more input from the designer.Some of the required data are either not tracked previously or are stored in locations not familiar to designers,and many data sets need to be preprocessed for use in the MEPDG.As a result,tremendous research work has been conducted and still more challenges need to be tackled both in federal and state levels for the full implementation of MEPDG.This paper,for the first time,provides a comprehensive bird's eye view for the MEPDG procedure,including the evolvement of the design methodology,an overview of the design philosophy and its components,the research conducted during the development,improvement,and implementation phases,and the challenges remained and future developments directions.It is anticipated that the efforts in this paper aid in enhancing the mechanistic-empirical based pavement design for future continuous improvement to keep up with changes in trucking,materials,construction,design concepts,computers,and so on.Qiang LI Danny X. 9Coating performance on glutaraldehyde-modified wood显示文摘Scots pine(Pinus sylvestris L.) panels were modified with glutaraldehyde(GA) to various weight percent gains and subsequently coated with several commercial coatings. The drying rate and adhesion of the coatings on the modified wood were measured; the coated/modified woods were exposed outdoors to analyze how the wood modifications influence the coating deterioration. The results showed that GA modification caused an increase in the drying rate of the waterborne coatings, but had no influence on drying of tested solvent-borne coatings. GAmodification did not change the dry adhesion but reduced the wood strength in a pull-off test. Wet adhesion of waterborne coatings was improved, while that of the solvent-borne coatings tended to be somewhat reduced. 11Optimization of technical measures for improving high-temperature performance of asphalt-rubber mixture显示文摘Asphalt-rubber pavements often become damaged in high-temperature regions and appear rutted or wavy,and experience slippage.To improve the high-temperature performance of the asphalt-rubber mixture,technical measurements,such as,the optimal adjustment of gradation,technique of composite modification,and control of compaction were investigated.An optimal adjustment of aggregate gradation based on stone matrix asphalt improves the high-temperature stability of the asphaltrubber mixture significantly.Through composite modification,the effect of asphalt-rubber modification was enhanced,and the dynamic stability and relative deformation indices of the asphalt-rubber mixture were improved significantly.Furthermore,compaction parameters had a significant influence on the high-temperature stability of the asphalt-rubber mixture.The rolling times for compacting the asphalt-rubber mixture should be controlled to within 18-20 round-trips at a molding temperature at 180°C;if the rolling time is a 12 round-trip,the compaction temperature of the asphalt-rubber mixture should be controlled between 180 and 190°C.Chuan Xiao Tianqing Ling Yanjun Qiu 2013Journal of Modern Transportation2013,21,4:2
12Deciphering the contributions of spectral and structural data to wheat yield estimation from proximal sensing显示文摘Accurate, efficient, and timely yield estimation is critical for crop variety breeding and management optimization. However, the contributions of proximal sensing data characteristics(spectral, temporal, and spatial) to yield estimation have not been systematically evaluated. We collected long-term, hypertemporal, and large-volume light detection and ranging(Li DAR) and multispectral data to(i) identify the best machine learning method and prediction stage for wheat yield estimation,(ii) characterize the contribution of multisource data fusion and the dynamic importance of structural and spectral traits to yield estimation, and(iii) elucidate the contribution of time-series data fusion and 3 D spatial information to yield estimation. Wheat yield could be accurately(R^(2)= 0.891) and timely(approximately-two months before harvest) estimated from fused Li DAR and multispectral data. The artificial neural network model and the flowering stage were always the best method and prediction stage, respectively. Spectral traits(such as CIgreen) dominated yield estimation, especially in the early stage, whereas the contribution of structural traits(such as height) was more stable in the late stage. Fusing spectral and structural traits increased estimation accuracy at all growth stages. Better yield estimation was realized from traits derived from complete 3 D points than from canopy surface points and from integrated multi-stage(especially from jointing to heading and flowering stages) data than from single-stage data. 20Two new reinforcement techniques applicable in high steep slopes along the Chengdu-Lhasa Highway, Southwest China显示文摘Anti-slide pile and anchoring hole are two types of reinforcement techniques popularly used in slope reinforcement. However, some defects of these two techniques are found in slope engineering practices. Utilizing higher tensile strength of pre-stressed structures, the authors devise two new types of slope reinforcement techniques, i.e. pre-stressed anchoring beam and pre-stressed anti-slide pile, in an attempt to overcome those defects of the existing techniques. These two new techniques are characterized by gathering multiple functions, e.g. engineering investigation, reinforcement, drainage, monitoring and strength supplement, so as to achieve high reinforcement efficiency, convenient checking, and higher ratio of function to cost.
