| 2 | Improvement of wheat drought and salt tolerance by expression of a stress- inducible transcription factorGmDREB of soybean (Glycinemax)显示文摘Under stress conditions such as droughthigh-salinity and low-temperature, the transcription factorof DREB (dehydration responsive element binding proteins)improved efficiently stress resistance by regulating the ex-pression of its downstream genes with various environmentastress resistance in plants. GmDREB gene (GenBank Acces-sion No. AF514908) encoding a stress-inducible transcriptionfactor was cloned by screening a cDNA library of Glycinemax cv. Jinong 27 with yeast one-hybrid method. GmDREBgene was 910 bp in length and encoded 174 amino acids con-taining a conserved AP2/EREBP DNA-binding domain of 58amino acids. Two conserved functional amino acids, valineand glutamic acid, were located on the 14th and the 19thamino acid residues in the conserved structural domain. Analkaline amino acid region (KKR) related to a nuclear local-ization signal was at the N-terminal, while an acidic aminoacid region (DDD) related to trans-activation was at theC-terminal. Plant expression vectors were constructed andtransformed into wheat by bombardment. In total, 13 trans-genic plants with Ubi::GmDREB and 11 transgenic plantswith rd29A::GmDREB were identified from 103 regenerationplants by molecular analysis. The drought and salt tolerancesof T1 transgenic lines with Ubi::GmDREB orrd29A::GmDREB were demonstrated to be improved ascompared to wild type. | 3 | Influence of environment and substrate quality on the decomposition of wetland plant root in the Sanjiang Plain,Northeast China显示文摘The litterbag method was used to study the decomposition of wetland plant root in three wetlands along a water level gradient in the Sanjiang Plain,Northeast China.These wetlands are Calamagrostis angustifolia(C.aa),Carex meyeriana(C.ma)and Carex lasiocarpa (C.la).The objective of our study is to evaluate the influence of environment and substrate quality on decomposition rates in the three wetlands.Calico material was used as a standard substrate to evaluate environmental influences.Roots native to each wetland were used to evaluate decomposition dynamics and substrate quality influences.Calico mass loss was statistically different among the three wetlands in the upper soil profile(0–10 cm)and in the lower depth range(10–20 cm).Hydrology,temperature and pH all influence calico decomposition rates in different ways at different depths of the soil profiles.The decomposition rates of native roots declined differentially with the increase of depth in the soil profiles.The mass loss of native roots showed a statistical decrease among the three wetlands in the upper soil profile(0–10 cm)and in the lower depth range(10–20 cm)as C.ma wetland>C.aa wetland>C.la wetland. | 4 | Fuzzy Synthetic Evaluation of Water Quality of Naoli River Using Parameter Correlation Analysis显示文摘In order to improve the effectiveness of Fuzzy Synthetic Evaluation (FSE) models, a Parameter Correlation Analysis (PCA) was introduced into the FSE and a case study was carried out in the Naoli River in the Sanjiang Plain, Northeast China. The basic principle of the PCA is that the pairs of parameters which are highly correlated and linear with each other would contribute the same information to an assessment and one of them should be eliminated. The method of the PCA is that a correlation relationship among candidate parameters is examined before the FSE. If there is an apparent nonlinear or curvilinear relationship between two parameters, then both will be retained; if the correlation is significant (p<0.01), and the scatter plot suggests a linear relationship, then one of them will be deleted. However, which one will be deleted? For solving this problem, a sensitivity test was conducted and the higher sensitivity parameters remained. The results indicate that the original data should be preprocessed through the PCA for redundancy and variability. | 5 | The determination of 52 elements in marine geological samples by an inductively coupled plasma optical emission spectrometry and an inductively coupled plasma mass spectrometry with a high-pressure closed digestion method显示文摘An improved analytical method to determine the content of 52 major, minor and trace elements in marine geological samples, using a HF-HCl-HNO_3 acid system with a high-pressure closed digestion method(HPCD), is studied by an inductively coupled plasma optical emission spectrometry(ICP-OES) and an inductively coupled plasma mass spectrometry(ICP-MS). The operating parameters of the instruments are optimized, and the optimal analytical parameters are determined. The influences of optical spectrum and mass spectrum interferences, digestion methods and acid systems on the analytical results are investigated. The optimal spectral lines and isotopes are chosen, and internal standard element of rhodium is selected to compensate for matrix effects and analytical signals drifting. Compared with the methods of an electric heating plate digestion and a microwave digestion, a high-pressure closed digestion method is optimized with less acid, complete digestion,less damage for digestion process. The marine geological samples are dissolved completely by a HF-HCl-HNO_3 system, the relative error(RE) for the analytical results are all less than 6.0%. The method detection limits are 2–40μg/g by the ICP-OES, and 6–80 ng/g by ICP-MS. The methods are used to determine the marine sediment reference materials(GBW07309, GBW07311, GBW07313), rock reference materials(GBW07103, GBW07104,GBW07105), and cobalt-rich crust reference materials(GBW07337, GBW07338, GBW07339), the obtained analytical results are in agreement with the certified values, and both of the relative standard deviation(RSD) and the relative error(RE) are less than 6.0%. | 6 | Freeze-thaw Effects on Sorption/Desorption of Dissolved Organic Carbon in Wetland Soils显示文摘The effects of freeze-thaw cycles on sorption/desorption of dissolved organic carbon (DOC) in two wetland soils and one reclaimed wetland soil were investigated. DOC concentrations added were 0-600 mg/L. Laboratory incubations of sorption/desorption of DOC had been carried out at -15℃ for 10 h, and then at +5℃ for 13 h. Soil samples were refrozen and thawed subsequently for 5 cycles. Initial Mass model was used to describe sorption behavior of DOC. The results indicate that freeze-thaw cycles can significantly increase the sorption capacity of DOC and reduce the desorption capacity of DOC in the three soils. The freeze-thaw effects on desorption of DOC in soils increase with the increasing freeze-thaw cycles. The conversion of natural wetlands to soybean farmland can decrease the sorption capacity and increase the desorption capacity of DOC in soils. | 8 | Determination of Lignin in Marine Sediment Using Alkaline Cupric Oxide Oxidation-Solid Phase Extraction-on-Column Derivatization-Gas Chromatography显示文摘Lignin serves as one of the most important molecular fossils for tracing Terrestrial Organic Matters (TOMs) in marine environment. Extraction and derivatization of lignin oxidation products (LOPs) are crucial for accurate quantification of lignin in marine sediment. Here we report a modification of the conventional alkaline cupric oxide (CuO) oxidation method, the modification consisting in a solid phase extraction (SPE) and a novel on-column derivatization being employed for better efficiency and reproducibility. In spiking blanks, recoveries with SPE for the LOPs are between 77.84% and 99.57% with relative standard deviations (RSDs) ranging from 0.57% to 8.04% (n=3), while those with traditional liquid-liquid extraction (LLE) are from 44.52% to 86.16% with RSDs being from 0.53% to 13.14% (n=3). Moreover, the reproducibility is greatly improved with SPE, with less solvent consumption and shorter processing time. The average efficiency of on-column derivatization for LOPs is 100.8% ± 0.68%, which is significantly higher than those of in-vial or in-syringe derivatization, thus resulting in still less consumption of derivatizing reagents. Lignin in the surface sediments sampled from the south of Yangtze River estuary, China, was determined with the established method. Recoveries of 72.66% to 85.99% with standard deviation less than 0.01mg/10g dry weight are obtained except for p-hydroxyben-zaldehyde. The lignin content Σ8 (produced from 10 g dry sediment) in the research area is between 0.231 and 0.587 mg. | 12 | Transfer and Transformation of Soil Iron and Implications for Hydrogeomorpholocial Changes in Naoli River Catchment, Sanjiang Plain, Northeast China显示文摘Wetland soils are characterized by alternating redox process due to the fluctuation of waterlogged conditions. Iron is an important redox substance, and its transfer and transformation in the wetland ecosystem could be an effective indicator for the environment changes. In this paper, we selected the Naoli River catchment in the Sanjiang Plain, Northeast China as the study area to analyze the dynamics of transfer and transformation of soil iron, and the relationship between iron content change and environmental factors. The results show that the total and crystalline iron contents reach the peak in the depth of 60 cm in soil profile, while the amorphous iron content is higher in the topsoil. In the upper reaches, from the low to high landscape positions, the total and crystalline iron contents decrease from 62.98 g/kg to 41.61 g/kg, 22.82 g/kg to 10.53 g/kg respectively, while the amorphous iron content increases from 2.42 g/kg to 8.88 g/kg. Amorphous iron content has positive correlation with organic matter and soil water contents, while negative correlation with pH. Moreover, both the crystalline and amorphous iron contents present no correlation with total iron content, indicating that environmental factors play a more important role in the transfer and transformation of iron other than the content of the total iron. Different redoximorphic features were found along the soil profile due to the transfer and transformation of iron. | 14 | Spatial Variation of Dissolved Organic Carbon in Soils of Riparian Wetlands and Responses to Hydro-geomorphologic Changes in Sanjiang Plain, China显示文摘Spatial variation of dissolved organic carbon(DOC) in soils of riparian wetlands and responses to hydro-geomorphologic changes in the Sanjiang Plain were analyzed through in situ collecting soil samples in the Naoli River and the Bielahong River. The results showed that the average contents of DOC for soil layer of 0–100 cm were 730.6 mg/kg, 250.9 mg/kg, 423.0 mg/kg and 333.1 mg/kg respectively from riverbed to river terrace along the transverse directions of the Naoli watershed. The content of the soil DOC was the highest in the riverbed, lower in the high floodplain and much lower in the river terrace, and it was the lowest in the low floodplain. The difference in the content and vertical distribution of DOC between the riverbed and the three riparian wetlands was significant, while it was not significant among the low floodplain, the high floodplain and the river terrace. The variability of soil DOC was related to the hydrological connectivity between different landscape position of the riparian wetlands and the adjacent stream. Extremely significant correlations were observed between DOC and total organic carbon(TOC), total iron(TFe), ferrous iron(Fe(II)) whose correlation coefficients were 0.819, –0.544 and –0.709 in riparian wetlands of the Naoli River. With the increase of wetland destruction, soil p H increased and soil DOC content changed. The correlation coefficients between soil DOC and TOC, TFe, Fe(II) also changed into 0.759, –0.686 and –0.575 respectively in the Bielahong River. Under the impact of drainage ditches, the correlations between soil DOC and TFe, Fe(II) were not obvious, while the soil p H was weakly alkaline and was negatively correlated with soil DOC in the previous high floodplain. | 15 | Magnetic and Microwave-absorption Properties of Graphite-coated (Fe, Ni) Nanocapsules显示文摘The structure, magnetic and microwave-absorption properties of graphite-coated (Fe, Ni) alloy nanocapsules, synthesized by the arc-discharge method, have been studied. High-resolution transmission electron microscopy shows that the nanocapsules have a core/shell structure with (Fe, Ni) alloy as the core and graphite as the shell. All (Fe, Ni) alloy nanocapsules/paraffin composites show good microwave-absorption properties. The optimal reflection loss (RL) was found for (Fe70Ni30)/C nanocapsules/paraffin composites, being -47.84 dB at 14.6 GHz for an absorber thickness of 1.99 mm, while the RL values exceeding -10 dB were found in the 12.4- 17.4 GHz range, which almost covers the Ku band (12.4-18 GHz). | 16 | Mercury distribution and bioaccumulation up the soil-plant-grasshopper-spider food chain in Huludao City, China显示文摘The purpose of this study is to investigate total mercury (THg) distribution and its bioaccumulation up the soil-plant-grasshopperspider in the Huludao City, which is polluted seriously by chlor-alkali and zinc smelting industry in Northeast of China. Results indicated that average THg concentrations in soil, plant leaves, grasshopper Locusta migratoria manilensis and Acrida chinensis, and spider were 0.151, 0.119, 0.167 and 0.134 mg/kg, respectively. THg spatial distribution suggested that most of mercury came from the chlor-alkali plant and the two zinc smelteries. The highest mercury concentration was found in the wings among different grasshoppers’ organs. Although spiders are the predatory, THg concentrations in their bodies were not high, and only on the same level as in grasshoppers, which might be due to spiders’ special living habits. In the light of the mercury transportation at every stage of the soil-plant-grasshopper-spider food chain, the bioaccumulation factors were 0.03, 0.79–1.11 and 0.80–1.13 respectively. | 17 | Exosomal LINC00355 derived from cancer-associated fibroblasts promotes bladder cancer cell resistance to cisplatin by regulating miR-34b-5p/ABCB1 axis显示文摘Cisplatin resistance is a major challenge for bladder cancer(BC).Evidence indicates that exosome derived from cancer-associated fibroblasts(CAF-Exo)can promote chemotherapy resistance in various human tumors by delivering bioactive molecules.We have previously demonstrated that CAF-derived exosomal LINC00355 promotes BC cell proliferation and invasion.However,the underlying mechanisms are still unclear.In this study,we aimed to investigate the role and mechanisms of CAF-derived exosomal LINC00355 in BC cell resistance to cisplatin.Exosomes were isolated from normal fibroblasts(NFs)and BC tumor-derived CAFs,namely,NF-Exo and CAF-Exo.CAFs were transfected with si-Ctrl or si-LINC00355 and then different exosomes were isolated,namely,CAF^(si-Ctrl)-Exo and CAF^(si-LINC00355)-Exo.The human BC cell lines(T24 and 5367)were incubated with NF-Exo,CAF-Exo,CAF^(si-Ctrl)-Exo,and CAF^(si-LINC00355)-Exo in the presence of cisplatin.MTT proliferation assay and flow cytometric analysis showed that CAF-Exo promoted BC cell resistance to cisplatin and upregulated ABCB1 expression in BC cells by transferring LINC00355 to BC cells.Luciferase activity assay confirmed the interaction between miR-34b-5p and LINC00355 or ABCB1.qRT-PCR and western blot analysis further showed that LINC00355 sponged miR-34b-5p to upregulate ABCB1 expression.However,the promoting effects of CAF-Exo on BC cell resistance to cisplatin were abolished by miR-34b-5p overexpression and ABCB1 silencing.In conclusion,exosomal LINC00355 derived from CAFs promotes BC cell resistance to cisplatin by regulating the miR-34b-5p/ABCB1 axis. | Guangyue Luo Yangyang Zhang Zhonghui Wu Ligang Zhang Chaozhao Liang Xianguo Chen | 2021 | Acta Biochimica et Biophysica Sinica2021,53,5: | 6 |
| 18 | The geometric factor of high energy protons detector on FY-3 satellite显示文摘Geometric factor is the key parameter for inversion of particle spectrum in space particle detection. Traditional geometric factor is obtained through the method of numerical calculation with the actual structure of the detector as the input condition. The degree of accuracy for data inversion is reduced since traditional geometric factor fails to take into account the physical process of interaction between the particle and substance as well as the influence of factors such as the particle interference between different energy channels on the measurement result. Here we propose an improved geometrical factor calculation method, the concept of which is to conduct actual structural modelling of the detector in the GEANT4 program, consider the process of interaction between the particle and substance, obtain the response function of the detector to particles of different energy channels through the method of Monte Carlo simulation, calculate the influence of contaminated particle on the geometrical factor, and finally get the geometrical factors for different energy channels of the detector. The imrpoved geometrical factor obtained through the method has carried out inversion for the data of high energy protons detector on China's FY-3 satellite, the energy spectrum after which is more in line with the power law distribution recognized by space physics. | 20 | Impacts of Soil Fauna on Litter Decomposition at Different Succession Stages of Wetland in Sanjiang Plain,China显示文摘Litter decomposition is the key process in nutrient recycling and energy flow. The present study examined the impacts of soil fauna on decomposition rates and nutrient fluxes at three succession stages of wetland in the Sanjiang Plain, China using different mesh litterbags. The results show that in each succession stage of wetland, soil fauna can obviously increase litter decomposition rates. The average contribution of whole soil fauna to litter mass loss was 35.35%. The more complex the soil fauna group, the more significant the role of soil fauna. The average loss of three types of litter in the 4mm mesh litterbags was 0.3-4.1 times that in 0.058mm ones. The decomposition function of soil fauna to litter mass changed with the wetland succession. The average contribution of soil fauna to litter loss firstly decreased from 34.96% (Carex lasiocapa) to 32.94% (Carex meyeriana), then increased to 38.16% (Calamagrostics an- gustifolia). The contributions of soil fauna to litter decomposition rates vary according to the litter substrata, soil fauna communities and seasons. Significant effects were respectively found in August and July on C. angustifolia and C. lasiocapa, while in June and August on C. meyeriana. Total carbon (TC), total nitrogen (TN) and total phosphorus (TP) contents and the C/N and C/P ratios of decaying litter can be influenced by soil fauna. At different wetland succession stages, the effects of soil fauna on nutrient elements also differ greatly, which shows the significant difference of influencing element types and degrees. Soil fauna communities strongly influenced the TC and TP concentrations of C. meyeriana litter, and TP content of C. lasiocapa.
