| 1 | Geochemistry and petrogenesis of Jurassic high Sr/low Y granitoids in eastern China: Constrains on crustal thickness显示文摘The Jurassic high Sr/low Y granitoids in eastern China are characterized by high Sr/Y (27-166) and La/Yb (14-66) ratios, low abundance in Y (6-21μg/g) and Yb (0.5-2.0μg/g), comparable with those of adakites defined by Defant et al. Thus, they were recently considered as adakitic rocks by some researchers. Compared with the typical adakites in circum-Pacific margins, however, these high Sr/low Y granitoids have higher K2O (-3.5%) but lower A12O3 (-16.0%) as well as lower Mg#(-38) and δSrN (-1.23) values. Furthermore, they show relatively flat HREE patterns with Y/Yb values of -10 close to the chondritic value. These geochemical characteristics indicate a residue mineral assemblage of hornblende, garnet and plagioclase for these high Sr/low Y granitoids melt. Thus, they were generated by partial melting at 9-13 kbar (30-45 km in depth), similar to the Archaean high-Al TTG rather than the modern adakites. | 2 | Sorafenib inhibits growth and metastasis of hepatocellular carcinoma by blocking STAT3显示文摘AIM: To investigate the inhibitory role and the underlying mechanisms of sorafenib on signal transducer and activator of transcription 3 (STAT3) activity in hepatocellular carcinoma (HCC).METHODS: Human and rat HCC cell lines were treated with sorafenib. Proliferation and STAT3 dephosphorylation were assessed. Potential molecular mechanisms of STAT3 pathway inhibition by sorafenib were evaluated. In vivo antitumor action and STAT3 inhibition were investigated in an immunocompetent orthotopic rat HCC model.RESULTS: Sorafenib decreased STAT3 phosphorylationat the tyrosine and serine residues (Y705 and S727), but did not affect Janus kinase 2 (JAK2) and phosphatase shatterproof 2 (SHP2), which is associated with growth inhibition in HCC cells. Dephosphorylation of S727 was associated with attenuated extracellular signal-regulated kinase (ERK) phosphorylation, similar to the effects of a mitogen-activated protein kinase (MEK) inhibitor U0126, suggesting that sorafenib induced S727 dephosphorylation by inhibiting MEK/ERK signaling. Meanwhile, sorafenib could also inhibit Akt phosphorylation, and both the phosphatidylinositol-3-kinase (PI3K) inhibitor LY294002 and Akt knockdown resulted in Y705 dephosphorylation, indicating that Y705 dephosphorylation by sorafenib was mediated by inhibiting the PI3K/Akt pathway. | 5 | Diagnosis of bile duct hepatocellular carcinoma thrombus without obvious intrahepatic mass显示文摘AIM:To study the diagnosis of hepatocellular carcinoma(HCC)presenting as bile duct tumor thrombus with no detectable intrahepatic mass.METHODS:Six patients with pathologically proven bile duct HCC thrombi but no intrahepatic mass demonstrated on the preoperative imaging or palpated intrahepatic mass during operative exploration,were collected.Their clinical and imaging data were retrospectively analyzed.The major findings or signs on comprehensive imaging were correlated with the surgical and pathologic findings.RESULTS:Jaundice was the major clinical symptom of the patients.The elevated serum total bilirubin,direct bilirubin and alanine aminotransferase levels were in concordance with obstructive jaundice and the underlying liver disease.Of the 6 patients showing evidence of viral hepatitis,5 were positive for serum alpha fetoprotein and carbohydrate antigen 19-9,and 1 was positive for serum carcinoembryonic antigen.No patient was correctly diagnosed by ultrasound.The main features of patients on comprehensive imaging were filling defects with cup-shaped ends of the bile duct,with large filling defects presenting as casting moulds in the expanded bile duct,hypervascular intraluminal nodules,debris or blood clots in the bile duct.No obvious circular thickening of the bile duct walls was observed.CONCLUSION:Even with no detectable intrahepatic tumor,bile duct HCC thrombus should be considered in patients predisposed to HCC,and some imaging signs are indicative of its diagnosis. | Long XY Li YX Wu W Li L Cao J. | 2010 | World Journal of Gastroenterology2010，16，39： | 11 |
| 6 | Efficacies of β-L-D4A against Hepatitis B virus in 2.2.15 cells显示文摘AIM:To investigate the antiviral effect of beta-L-enantiomer of 2',3'-didehydro-2',3'-dideoxyadenosine (β-L-D4A) on 2.2.15 cells transfected with the hepatitis B virus (HBV) genome. METHODS:Lamivudine (3TC) as a positive control. Then, HBV DNA in treated 2.2.15 cells and the Hepatitis B surface antigen (HBsAg) in the culture supernatants were detected to determine the inhibitory effect of β-L-D4A. At the same time, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) was used to detect the survival ratio of 2.2.15 cells. RESULTS:β-L-D4A has a dose-dependent inhibitory effect on HBV DNA replication;this effect was apparent when the concentration was above 1 mol/L. When β-L-D4A was at the highest concentration, 100 mol/L, the HBsAg inhibition ratio was above 50%. The Therapeutic index (TI) of β-L-D4A was above 2.1. | 11 | Lipopolysaccharide decreasing albumin expression in rat hepatocytes显示文摘The relations of the severity of hypoalbu-minemia to morbidity and mortality of patients with critical illness illustrate the need for better understanding of molecular mechanism of hypoalbuminemia. This study was undertaken to investigate the response of albumin synthesis to lipopolysaccharide(LPS)in rat hepatocytes in vitro in early acute phase of sepsis. METHODS:Hepatocytes were cultured at an initial cell density of 1.5×106 cells/well in 3 ml culture medium. There were two groups of samples which received either normal saline or 1 μg/L LPS randomly. Albumin mRNA in hepatocytes was assessed by reverse transcription-poly-merase chain reaction(RT-PCR) and albumin level in the supernatant was measured by ELISA at 0,2,8,12,24 hours after exposure. Meanwhile, the albumin precursor was evaluated at the same time points by flow cytometry. RESULTS:The quantitative changes of mRNA, albumin precursor and its protein were analogous. All of them tended to decline at 12 hours post-treatment and did not decrease significantly until 24 hours after LPS exposure. Meanwhile, albumin mRNA decreased about 30% and the levels of albumin precursor and albumin reduced approximately 50%. CONCLUSIONS: LPS can inhibit albumin synthesis in rat hepatocytes by prevention of albumin transcription. Moreover, the response of hepatic albumin synthesis to LPS changes with the stage of sepsis process.
