A large time step Godunov scheme for free-surface shallow water equations显示文摘An algorithm for simulating free surface flows is presented using large time step based on the wave-propagation method proposed by LeVeque, and an exact Riemann solver is used.A multiple wave approximation approach was suggested for eliminating the discontinuities found in the rarefaction fans of dam-breaking flows. In addition,we use the random choice method to reduce non-physical oscillations.Applications demonstrate that the algorithm proposed in this paper can considerably increase the CFL number up to 25when modeling dam-break flows, while retaining satisfactory accuracy and efficiency. This suggests that our algorithm has the potential to be applied to modeling free surface flows.Renyi Xu Deyu Zhong Baosheng Wu Xudong Fu Runze Miao 2014Chinese Science Bulletin2014,59,21:5
Highly Dispersed Cobalt Nanoparticles Embedded in Nitrogen-Doped Graphitized Carbon for Fast and Durable Potassium Storage显示文摘Potassium-ion batteries(KIBs)have great potential for applications in large-scale energy storage devices. However, the larger radius of K+leads to sluggish kinetics and inferior cycling performance, severely restricting its practical applicability.Herein,we propose a rational strategy involving a Prussian blue analogue-derived graphitized carbon anode with fast and durable potassium storage capability, which is constructed by encapsulating cobalt nanoparticles in nitrogen-doped graphitized carbon(Co-NC).Both experimental and theoretical results show that N-doping effectively promotes the uniform dispersion of cobalt nanoparticles in the carbon matrix through Co-N bonds. Moreover, the cobalt nanoparticles and strong Co-N bonds synergistically form a threedimensional conductive network, increase the number of adsorption sites, and reduce the diffusion energy barrier, thereby facilitating the adsorption and the diffusion kinetics. These multiple effects lead to enhanced reversible capacities of 305 and 208.6 mAh g^−1 after 100 and 300 cycles at 0.05 and 0.1 A g^−1,respectively,demonstrating the applicability of the Co-NC anode for KIBs.Xiaodong Shi Zhenming Xu Cheng Han Runze Shi Xianwen Wu Bingan Lu Jiang Zhou Shuquan Liang 2021Nano-Micro Letters2021,13,2:4
The Variation of Stone Cell Content in 236 Germplasms of Sand Pear(Pyrus pyrifolia)and Identification of Related Candidate Genes显示文摘Stone cells have been described to substantially influence pear fruit quality,as lignin and cellulose are the main components of stone cells. However, there are limited studies on the relationship between the variation and molecular basis of stone cells, lignin and cellulose content among different pear varieties. Here,to reveal the variation of stone cell content within different cultivated species,we collected 236 germplasms of sand pear(Pyrus pyrifolia)at 50 days after flower blooming(DAFB),the key stage of stone cell formation. In our results,we measured the content of stone cells, lignin and cellulose and found that these contents ranged from2.82%to 29.00%,8.84%to 55.30%and 11.52%to 30.55%,respectively.Further analysis showed that the variation coefficient of stone cell, lignin and cellulose content was 39.10%,28.03%and 16.71%,respectively.Additionally,a significant correlation between stone cell, lignin and cellulose content were detected, and the correlation coefficient between the contents of stone cell and lignin(0.912)was higher than between the contents of stone cell and cellulose(0.796).Moreover,the average lignin content(29.73%)was higher than the average cellulose content(18.03%)in stone cells in pear fruits, indicating that lignin is the main component of stone cell in pears. Finally,on the basic of the transcriptome data,we identified 10 transcription factors belonging to bHLH,ERF,MYB,and NAC transcript families, which might be involved in lignin formation in stone cells.qRT-PCR experiments verified coincident trends between expression of candidate genes and stone cell content. This research laid foundation for future studies on genetic variation of stone cells in pear fruits and provided important gene resources for stone cell regulation.Jiaying Zhang Jiaming Li Cheng Xue Runze Wang Mingyue Zhang Kaijie Qi Jing Fan Hongju Hu Shaoling Zhang Jun Wu 2021Horticultural Plant Journal2021,7,2:3

Electrochemical deposited amorphous FeNi hydroxide electrode for oxygen evolution reaction显示文摘The electrodeposition approach is significant in electrode fabrication for practical application.Herein, the electrodeposited amorphous NiFe hydroxide species for oxygen evolution reaction (OER) in water splitting reaction is demonstrated by revealing the synergistic effect influenced by the support electrode of Fe and Ni foil and the contents of Fe and Ni in the electrolyte. All the electrodeposited samples have an amorphous structure and similar profiles of binding energy and chemical states for Fe and Ni as characterized by the spectroscopic techniques. While the support effect and Fe/Ni synergistic effect are indeed observed for the varied catalytic performances observed for the different electrodes;the Ni foil supported catalyst exhibits much higher performance than that of the Fe foil supported catalyst, and the different redox potentials of Ni species in the different Fe/Ni electrode resulting from the Fe–Ni synergism are observed in the cyclic voltammetry curve analysis. The surface roughness and the electrochemical surface area are also influenced by the support effect and the Fe/Ni ratio in the plating electrolyte. The optimal electrode shows a very low overpotential of~200 mV to reach 10 mA cm^(-2),and very high catalytic stability by the consecutive cyclic voltammetry measurements and 20 h stability test.Though it has the largest electrochemical surface area, the highest catalytic efficiency for these active sites is also indicated by the specific activity and turnover frequency polarization curves. The current work shows the effective experience for the electrodeposited Fe/Ni based catalysts in large-scale fabrication, which can be more practical for hydrogen generation in the alkaline water electrolysis.Zhengzhi Yin Runze He Yongcai Zhang Ligang Feng Xiang Wu Thomas Wågberg Guangzhi Hu 2022Journal of Energy Chemistry2022,31,6:2
The Solar Upper Transition Region Imager(SUTRI)Onboard the SATech-01 Satellite显示文摘The Solar Upper Transition Region Imager(SUTRI)onboard the Space Advanced Technology demonstration satellite(SATech-01),which was launched to a Sun-synchronous orbit at a height of~500 km in 2022 July, aims to test the on-orbit performance of our newly developed Sc/Si multi-layer reflecting mirror and the 2k×2k EUV CMOS imaging camera and to take full-disk solar images at the Ne VII 46.5 nm spectral line with a filter width of~3 nm.SUTRI employs a Ritchey-Chrétien optical system with an aperture of 18 cm.The on-orbit observations show that SUTRI images have a field of view of~416×416 and a moderate spatial resolution of~8″without an image stabilization system. The normal cadence of SUTRI images is 30 s and the solar observation time is about16 hr each day because the earth eclipse time accounts for about 1/3 of SATech-01's orbit period.Approximately15 GB data is acquired each day and made available online after processing.SUTRI images are valuable as the Ne VII 46.5 nm line is formed at a temperature regime of~0.5 MK in the solar atmosphere, which has rarely been sampled by existing solar imagers.SUTRI observations will establish connections between structures in the lower solar atmosphere and corona, and advance our understanding of various types of solar activity such as flares, filament eruptions, coronal jets and coronal mass ejections.Xianyong Bai Hui Tian Yuanyong Deng Zhanshan Wang Jianfeng Yang Xiaofeng Zhang Yonghe Zhang Runze Qi Nange Wang Yang Gao Jun Yu Chunling He Zhengxiang Shen Lun Shen Song Guo Zhenyong Hou Kaifan Ji Xingzi Bi Wei Duan Xiao Yang Jiaben Lin Ziyao Hu Qian Song Zihao Yang Yajie Chen Weidong Qiao Wei Ge Fu Li Lei Jin Jiawei He Xiaobo Chen Xiaocheng Zhu Junwang He Qi Shi Liu Liu Jinsong Li Dongxiao Xu Rui Liu Taijie Li Zhenggong Feng Yamin Wang Chengcheng Fan Shuo Liu Sifan Guo Zheng Sun Yuchuan Wu Haiyu Li Qi Yang Yuyang Ye Weichen Gu Jiali Wu Zhe Zhang Yue Yu Zeyi Ye Pengfeng Sheng Yifan Wang Wenbin Li Qiushi Huang Zhong Zhang 2023Research in Astronomy and Astrophysics2023,23,6:1
Serial deletion reveals structural basis and stability for the core enzyme activity of human glutaminase 1 isoforms:relevance to excitotoxic neurodegeneration显示文摘Background:Glutaminase 1 is a phosphate-activated metabolic enzyme that catalyzes the first step of glutaminolysis, which converts glutamine into glutamate.Glutamate is the major neurotransmitter of excitatory synapses, executing important physiological functions in the central nervous system. There are two isoforms of glutaminase 1,KGA and GAC,both of which are generated through alternative splicing from the same gene.KGA and GAC both transcribe 1–14 exons in the N-terminal,but each has its unique C-terminal in the coding sequence. We have previously identified that KGA and GAC are differentially regulated during inflammatory stimulation and HIV infection.Furthermore, glutaminase 1 has been linked to brain diseases such as amyotrophic lateral sclerosis,Alzheimer’s disease, and hepatic encephalopathy.Core enzyme structure of KGA and GAC has been published recently. However, how other coding sequences affect their functional enzyme activity remains unclear. Methods:We cloned and performed serial deletions of human full-length KGA and GAC from the N-terminal and the C-terminal at an interval of approximately 100 amino acids(AAs).Prokaryotic expressions of the mutant glutaminase 1 protein and a glutaminase enzyme activity assay were used to determine if KGA and GAC have similar efficiency and efficacy to convert glutamine into glutamate. Results:When 110 AAs or 218 AAs were deleted from the N-terminal or when the unique portions of KGA and GAC that are beyond the 550 AA were deleted from the C-terminal,KGA and GAC retained enzyme activity comparable to the full length proteins. In contrast, deletion of 310 AAs or more from N-terminal or deletion of 450 AAs or more from C-terminal resulted in complete loss of enzyme activity for KGA/GAC.Consistently,when both Nand C-terminal of the KGA and GAC were removed, creating a truncated protein that expressed the central 219 AA-550 AA,the protein retained enzyme activity.Furthermore, expression of the core 219 AA-550 AA coding sequence in cells increased extracellular glutamate concentrations to levels comparable to those of full-length KGA and GAC expressions, suggesting that the core enzyme activity of the protein lies within the central 219 AA-550 AA.Full-length KGA and GAC retained enzyme activities when kept at 4°C.In contrast,219 AA-550 AA truncated protein lost glutaminase activities more readily compared with full-length KGA and GAC,suggesting that the Nterminal and C-terminal coding regions are required for the stability KGA and GAC.Conclusions:Glutaminase isoforms KGA and GAC have similar efficacy to catalyze the conversion of glutamine to glutamate. The core enzyme activity of glutaminase 1 protein is within the central 219 AA-550 AA.The N-terminal and C-terminal coding regions of KGA and GAC help maintain the long-term activities of the enzymes.Yuju Li Justin Peer Runze Zhao Yinghua Xu Beiqing Wu Yi Wang Changhai Tian Yunlong Huang Jialin Zheng 2017Translational Neurodegeneration2017,6,1:1
Homotypic clustering of L1 and B1/Alu repeats compartmentalizes the 3D genome显示文摘Organization of the genome into euchromatin and heterochromatin appears to be evolutionarily conserved and relatively stable during lineage differentiation. In an effort to unravel the basic principle underlying genome folding, here we focus on the genome itself and report a fundamental role for L1(LINE1 or LINE-1)and B1/Alu retrotransposons, the most abundant subclasses of repetitive sequences,in chromatin compartmentalization. We find that homotypic clustering of L1 and B1/Alu demarcates the genome into grossly exclusive domains, and characterizes and predicts Hi-C compartments.Spatial segregation of L1-rich sequences in the nuclear and nucleolar peripheries and B1/Alu-rich sequences in the nuclear interior is conserved in mouse and human cells and occurs dynamically during the cell cycle. In addition,de novo establishment of L1 and B1 nuclear segregation is coincident with the formation of higher-order chromatin structures during early embryogenesis and appears to be critically regulated by L1 and B1 transcripts.Importantly, depletion of L1 transcripts in embryonic stem cells drastically weakens homotypic repeat contacts and compartmental strength, and disrupts the nuclear segregation of L1-or B1-rich chromosomal sequences at genome-wide and individual sites.Mechanistically, nuclear co-localization and liquid droplet formation of L1 repeat DNA and RNA with heterochromatin protein HP1αsuggest a phase-separation mechanism by which L1 promotes heterochromatin compartmentalization.Taken together,we propose a genetically encoded model in which L1 and B1/Alu repeats blueprint chromatin macrostructure. Our model explains the robustness of genome folding into a common conserved core,on which dynamic gene regulation is overlaid across cells.J.Yuyang Lu Lei Chang Tong Li Ting Wang Yafei Yin Ge Zhan Xue Han Ke Zhang Yibing Tao Michelle Percharde Liang Wang Qi Peng Pixi Yan Hui Zhang Xianju Bi Wen Shao Yantao Hong Zhongyang Wu Runze Ma Peizhe Wang Wenzhi Li Jing Zhang Zai Chang Yingping Hou Bing Zhu Miguel Ramalho-Santos Pilong Li Wei Xie Jie Na Yujie Sun Xiaohua Shen 2021Cell Research2021,31,6:1

Herbal formula BaWeiBaiDuSan alleviates polymicrobial sepsis-induced liver injury via increasing the gut microbiota Lactobacillus johnsonii and regulating macrophage anti-inflammatory activity in mice显示文摘Sepsis-induced liver injury(SILI)is an important cause of septicemia deaths.BaWeiBaiDuSan(BWBDS)was extracted from a formula of Panax ginseng C.A.Meyer,Lilium brownie F.E.Brown ex Miellez var.viridulum Baker,Polygonatum sibiricum Delar.ex Redoute,Lonicera japonica Thunb.,Hippophae rhamnoides Linn.,Amygdalus Communis Vas,Platycodon grandiflorus(Jacq.)A.DC.,and Cortex Phelloderdri.Herein,we investigated whether the BWBDS treatment could reverse SILI by the mechanism of modulating gut microbiota.BWBDS protected mice against SILI,which was associated with promoting macrophage anti-inflammatory activity and enhancing intestinal integrity.BWBDS selectively promoted the growth of Lactobacillus johnsonii(L.johnsonii)in cecal ligation and puncture treated mice.Fecal microbiota transplantation treatment indicated that gut bacteria correlated with sepsis and was required for BWBDS anti-sepsis effects.Notably,L.johnsonii significantly reduced SILI by promoting macrophage anti-inflammatory activity, increasing interleukin-10+M2 macrophage production and enhancing intestinal integrity.Furthermore, heat inactivation L.johnsonii(HI-L.johnsonii)treatment promoted macrophage anti-inflammatory activity and alleviated SILI.Our findings revealed BWBDS and gut microbiota L.johnsonii as novel prebiotic and probiotic that may be used to treat SILI.The potential underlying mechanism was at least in part, via L.johnsonii-dependent immune regulation and interleukin-10+M2 macrophage production.Xiaoqing Fan Chutian Mai Ling Zuo Jumin Huang Chun Xie Zebo Jiang Runze Li Xiaojun Yao Xingxing Fan Qibiao Wu Peiyu Yan Liang Liu Jianxin Chen Ying Xie Elaine Lai-Han Leung 2023Acta Pharmaceutica Sinica B2023,13,3:1
Constructing CdSe QDs modified porous g-C_(3)N_(4)heterostructures for visible light photocatalytic hydrogen production显示文摘Constructing heterostructures with narrow-band-gap semiconductors is a promising strategy to extend light absorption range of graphitic carbon nitride(g-C_(3)N_(4))and simultaneously promote charge separation for its photocatalytic activity improvement. However, its highly localized electronic states of g-C_(3)N_(4)hinder photo-carrier migration through bulk towards heterostructure interfaces, resulting in low charge carrier separation efficiency of solid bulk g-C_(3)N_(4)-based heterostructures.Herein, porous g-C_(3)N_(4)(PCN)material with greatly shortened migration distance of photo-carriers from bulk to surface was used as an effective substrate to host Cd Se quantum dots to construct type II heterostructure of Cd Se/PCN for photocatalytic hydrogen production. The homogeneous modification of the Cd Se quantum dots throughout the whole bulk of PCN together with proper band alignments between Cd Se and PCN enables the effective separation of photo-generated charge carriers in the heterostructure.Consequently, the Cd Se/PCN heterostructure photocatalyst gives the greatly enhanced photocatalytic hydrogen production activity of192.3μmol h^(-1),which is 4.4 and 8.1 times that of Cd Se and PCN,respectively. This work provides a feasible strategy to construct carbon nitride-based heterostructure photocatalysts for boosting visible light driven water splitting performance.Zheng Zhang Yuyang Kang Li-Chang Yin Ping Niu Chao Zhen Runze Chen Xiangdong Kang Fayu Wu Gang Liu 2021Journal of Materials Science & Technology2021,,36:0
Dynamic human retinal pigment epithelium(RPE)and choroid architecture based on single-cell transcriptomic landscape analysis显示文摘The retinal pigment epithelium(RPE)and choroid are located behind the human retina and have multiple functions in the human visual system.Knowledge of the RPE and choroid cells and their gene expression profiles are fundamental for understanding retinal disease mechanisms and therapeutic strategies. Here,we sequenced the RNA of about 0.3 million single cells from human RPE and choroids across two regions and seven ages, revealing regional and age differences within the human RPE and choroid.Cell–cell interactions highlight the broad connectivity networks between the RPE and different choroid cell types. Moreover, the transcription factors and their target genes change during aging. The coding of somatic variations increases during aging in the human RPE and choroid at the single-cell level. Moreover,we identified ELN as a candidate for improving RPE degeneration and choroidal structure during aging. The mapping of the molecular architecture of the human RPE and choroid improves our understanding of the human vision support system and offers potential insights into the intervention targets for retinal diseases.Lulin Huang Lin Ye Runze Li Shanshan Zhang Chao Qu Shujin Li Jie Li Mu Yang Biao Wu Ran Chen Guo Huang Bo Gong Zheng Li Hongjie Yang Man Yu Yi Shi Changguan Wang Wei Chen Zhenglin Yang 2023Genes & Diseases2023,10,6:0
Genomic selection of eight fruit traits in pear显示文摘Genomic selection (GS) has the potential to improve selection efficiency and shorten the breeding cycle in fruit tree breeding. In this study,we evaluated the effect of prediction methods, marker density and the training population (TP) size on pear GS for improving its performance and reducing cost. We evaluated GS under two scenarios:(1) five-fold cross-validation in an interspecific pear family;(2) independent validation. Based on the cross-validation scheme, the prediction accuracy (PA) of eight fruit traits varied between 0.33 (fruit core vertical diameter)and 0.65 (stone cell content). Except for single fruit weight, a slightly better prediction accuracy (PA) was observed for the five parametrical methods compared with the two non-parametrical methods. In our TP of 310 individuals, 2 000 single nucleotide polymorphism (SNP) markers were sufficient to make reasonably accurate predictions. PAs for different traits increased by 18.21%-46.98%when the TP size increased from 50to 100, but the increment was smaller (-4.13%-33.91%) when the TP size increased from 200 to 250. For independent validation, the PAs ranged from 0.11 to 0.45 using rrBLUP method. In summary, our results showed that the TP size and SNP numbers had a greater impact on the PA than prediction methods. Furthermore, relatedness among the training and validation sets, and the complexity of traits should be considered when designing a TP to predict the test panel.Manyi Sun Mingyue Zhang Satish Kumar Mengfan Qin Yueyuan Liu Runze Wang Kaijie Qi Shaoling Zhang Wenjing Chang Jiaming Li Jun Wu 2024Horticultural Plant Journal2024,10,2:0
Consecutive Methylation Catalyzed by TsrM,an Atypical Class B Radical SAM Methylase显示文摘TsrM is a cobalamin-dependent radical S-adenosylmethionine(SAM)methyltransferase belonging to the Class B radical SAM methylase(RSM)family. This enzyme catalyzes the C-2 methylation of L-tryptophan to produce 2-methyltrytophan(2-MeTrp),an intermediate involved in the biosynthesis of thiostrepton A.In this work,we report characterization of an unexpected activity of TsrM,which carries out an additional methylation reaction on the product 2-MeTrp.A series of isotopic labeling studies and assays with different Trp analogs revealed that TsrM is able to transfer a methyl group from SAM to the C4 of 2-MeTrp to produce 2,4-dimethyltryptophan.These results reveal the intriguing substrate specificity of TsrM,further expanding the reaction promiscuity of the radical SAM superfamily enzymes.Runze Wu Wei Ding Qi Zhang 2022Chinese Journal of Chemistry2022,40,14:0
Genome-wide identification of the MATE gene family and functional characterization of PbrMATE9 related to anthocyanin in pear显示文摘Plant multidrug and toxic compound extrusion(MATE) genes play an important role in the process of detoxification, plant morphogenesis, and anthocyanin accumulation. However, whether the MATE gene family functions in pear peel coloration is still unknown. To evaluate and identify the MATE gene family members which are involving in anthocyanin accumulation and coloration in pear. In this study, 85 MATE genes were identified in the reference pear genome of ‘Dangshansuli’ through genome-wide identification. Based on gene structure and phylogenetic tree analysis, the MATE family was divided into five subfamilies. RNA sequencing and quantitative real-time polymerase chain reaction(qRTPCR) indicated that the expression patterns of PbrMATEs were tissue-specific. 28.24%(24) of PbrMATE genes were expressed in the fruits, and44.71%(38) of PbrMATE genes were expressed in the leaves. Additionally, we found that the expression levels of PbrMATE9, PbrMATE26,PbrMATE50, and PbrMATE69 in debagged fruits with red peel were significantly higher than those in bagged fruits without red peel, according to our bagging/debagging treatment of ‘Mantianhong’. The expression pattern of PbrMATE9 was consistent with the variation trend in anthocyanin content, suggesting that it might play an important role in anthocyanin accumulation in response to light exposure. Subcellular localization showed that PbrMATE9 was a membrane protein. More strikingly, the transient overexpression of PbrMATE9 promoted anthocyanin accumulation in the peel of pear, and the expression of structural genes(PbrCHI, PbrANS, PbrDFR, and PbrUFGT) in the anthocyanin biosynthesis pathway also increased significantly. Through co-expression network analysis, the transcription factors were identified, such as WRKY, COL,GATA, and BBX, which might be involved in the regulation of PbrMATE9. The study has enriched the genetic resources and improved the understanding of the regulation network of anthocyanin accumulation in pear.Jiawen Sheng Xuening Chen Bobo Song Hainan Liu Jiaming Li Runze Wang Jun Wu 2023Horticultural Plant Journal2023,9,6:0

Self-confocal NIR-II fluorescence microscopy for multifunctional in vivo imaging显示文摘Fluorescence imaging in the second near-infrared window(NIR-II,900–1880 nm)with less scattering background in biological tissues has been combined with the confocal microscopic system for achieving deep in vivo imaging with high spatial resolution. However, the traditional NIR-IIfluorescence confocal microscope with separate excitation focus and detection pinhole makes it possess low confocal e±ciency,as well as di±cultly to adjust. Two types of upgraded NIR-IIfluorescence confocal microscopes, sharing the same pinhole by excitation and emission focus, leading to higher confocal e±ciency, are built in this work. One type is-ber-pinhole-based confocal microscope applicable to CW laser excitation. It is constructed forfluorescence intensity imaging with large depth, high stabilization and low cost, which could replace multiphotonfluorescence microscopy in some applications(e.g.,cerebrovascular and hepatocellular imaging). The other type is air-pinhole-based confocal microscope applicable to femtosecond(fs)laser excitation. It can be employed not only for NIR-IIfluorescence intensity imaging, but also for multi-channelfluorescence lifetime imaging to recognize different structures with similarfluorescence spectrum. Moreover,it can be facilely combined with multiphotonfluorescence microscopy.A single fs pulsed laser is utilized to achieve up-conversion(visible multiphotonfluorescence)and down-conversion(NIR-II one-photonfluorescence)excitation simultaneously, extending imaging spectral channels, and thus facilitates multi-structure and multi-functional observation.Jing Zhou Tianxiang Wu Runze Chen Liang Zhu Hequn Zhang Yifei Li Liying Chen Jun Qian 2024Journal of Innovative Optical Health Sciences2024,17,1:0
