| 2 | Oral bacteria colonize and compete with gut microbiota in gnotobiotic mice显示文摘The oral microbiota is associated with oral diseases and digestive systemic diseases.Nevertheless,the causal relationship between them has not been completely elucidated,and colonisation of the gut by oral bacteria is not clear due to the limitations of existing research models.The aim of this study was to develop a human oral microbiota-associated (HOMA) mouse model and to investigate the ecological invasion into the gut.By transplanting human saliva into germ-free (GF) mice,a HOMA mouse model was first constructed.16S rRNA gene sequencing was used to reveal the biogeography of oral bacteria along the cephalocaudal axis of the digestive tract.In the HOMA mice,84.78% of the detected genus-level taxa were specific to the donor.Principal component analysis (PCA) revealed that the donor oral microbiota clustered with those of the HOMA mice and were distinct from those of specific pathogen-free (SPF) mice.In HOMA mice,OTU counts decreased from the stomach and small intestine to the distal gut.The distal gut was dominated by Streptococcus,Veillonella,Haemophilus,Fusobacterium,Trichococcus and Actinomyces.HOMA mice and human microbiota-associated (HMA) mice along with the GF mice were then cohoused.Microbial communities of cohoused mice clustered together and were significantly separated from those of HOMA mice and HMA mice.The Source Tracker analysis and network analysis revealed more significant ecological invasion from oral bacteria in the small intestines,compared to the distal gut,of cohoused mice.In conclusion,a HOMA mouse model was successfully established.By overcoming the physical and microbial barrier,oral bacteria colonised the gut and profiled the gut microbiota,especially in the small intestine. | Bolei Li Yang Ge Lei Cheng Benhua Zeng Jinzhao Yu Xian Peng Jianhua Zhao Wenxia Li Biao Ren Mingyun Li Hong Wei Xuedong Zhou | 2019 | International Journal of Oral Science2019,11,3: | 7 |
| 3 | Fatigue crack propagation behavior of Ni-based superalloys after overloading at elevated temperatures显示文摘The fatigue crack propagation behavior of three superalloys subjected to a single overloading at elevated temperatures was investigated. The fatigue crack propagation rate FCPR versus stress intensity factor range data da/dN—ΔK were calculated using the two-point secant method. It was found that the crack growth rates of the investigated materials were retarded after overloading with an overload ratio R OL=1.6. The size of the plastic zone in the front of the crack tip and its relation to loading level were discussed. The overload retardation effects are attributed to crack closure. | 5 | The novel ER stress inducer Sec C triggers apoptosis by sulfating ER cysteine residues and degrading YAP via ER stress in pancreatic cancer cells显示文摘Pancreatic adenocarcinoma(PAAD) is one of the most lethal malignancies. Although gemcitabine(GEM) is a standard treatment for PAAD, resistance limits its application and therapy. Secoemestrin C(Sec C) is a natural compound from the endophytic fungus Emericella, and its anticancer activity has not been investigated since it was isolated. Our research is the first to indicate that Sec C is a broad-spectrum anticancer agent and could exhibit potently similar anticancer activity both in GEM-resistant and GEM-sensitive PAAD cells. Interestingly, Sec C exerted a rapid growth-inhibiting effect(80% death at 6 h), which might be beneficial for patients who need rapid tumor shrinkage before surgery. Liquid chromatography/mass spectrometry and N-acetyl-L-cysteine(NAC) reverse assays show that Sec C sulfates cysteines to disrupt disulfide-bonds formation in endoplasmic reticulum(ER) proteins to cause protein misfolding, leading to ER stress and disorder of lipid biosynthesis. Microarray data and subsequent assays show that ER stress-mediated ER-associated degradation(ERAD) ubiquitinates anddownregulates YAP to enhance ER stress via destruction complex(YAP-Axin-GSK-βTr CP), which also elucidates a unique degrading style for YAP. | 7 | EGFR TKIs impair lysosome-dependent degradation of SQSTM1 to compromise the effectiveness in lung cancer显示文摘Tyrosine kinase inhibitors for epidermal growth factor receptor(EGFR TKIs)greatly improved clinical outcomes of patients with non-small cell lung cancer(NSCLC).Unfortunately,primary and acquired resistance limits their clinical benefits.To overcome such resistance,new generations of EGFR TKIs have been developed by targeting newly identified mutations in EGFR.However,much less effort has been put into alternative strategies,such as targeting the intrinsic protective responses to EGFR TKIs.In this study,we found that EGFR TKIs,including gefitinib and AZD9291,impaired lysosome-dependent degradation of SQSTM1,thus compromising their anti-cancer efficiency.By accumulating in the lysosome lumen,gefitinib and AZD9291 attenuated lysosomal acidification and impaired autolysosomal degradation of SQSTM1 owing to their intrinsic alkalinity.As a result,SQSTM1 protein was stabilized in response to gefitinib and AZD9291 treatment and conferred EGFR TKI resistance.Depleting SQSTM1 significantly increased the sensitivity of NSCLC cells to gefitinib and AZD9291 both in vitro and in vivo.Furthermore,a chemically modified gefitinib analog lacking alkalinity displayed stronger inhibitory effects on NSCLC cells.Therefore,targeting accumulated SQSTM1 or chemically modified EGFR TKIs may represent new strategies to increase the effectiveness of EGFR targeted therapy. | Lixian Yang Shilong Ying Shiman Hu Xiangtong Zhao Muchun Li Miaoqin Chen Yiran Zhu Ping Song Liyuan Zhu Tingting Jiang Huimin An Neelum Aziz Yousafzai Wenxia Xu Zhiguo Zhang Xian Wang Lifeng Feng Hongchuan Jin | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 2 |
| 8 | An ultrasound-triggered cation chelation and reassembly route to one-dimensional Ni-rich cathode material enabling fast charging and stable cycling of Li-ion batteries显示文摘Ni-rich oxides,LiNixMnyCozO2(NMC),are among leading candidates for cathode materials in Li-ion batteries.However,they are mostly fabricated by coprecipitation approach under complex conditions,which usually produces large secondary particles composed of aggregated primary particles.Undesirable cation mixing and crack propagation upon cycling block ion and electron transport,result in fast capacity fading and poor rate capability.Herein,we present an ultrasound-triggered cation chelation and reassembly route for synthesizing one-dimensional precursor with homogeneous element distribution at the atomic level.This process is accomplished by the synergistic combination of ultrasound and surfactant,which can disperse reactants and remove hydration shells surrounding cations so as to accelerate chelating reaction,and then separate and assemble chelates into one dimensional structure.The whole synthesis time is only 20 min(8.9 min of ultrasonic working time)in an open vessel under natural ambient conditions.One-dimensional LiNi0.6Mn0.2Co0.2O2 has a high reversible capacity(184 mAh·g^−1 at 0.1 C)and long cycling stability(95.1%and 82.4%capacity retention for 100 and 1000 cycles,respectively).The short charging time of 76 s is realized at super high current rate of 20 C,which is very important to improve the competitiveness of electric vehicles relative to fuel vehicles.Our synthetic approach can provide a general strategy for the growth of mixed-metal-EDTA chelate precursors by changing the feeding ratio of Ni2+,Mn2+and Co2+cations in reaction for fabricating NMC cathode materials with other compositions. | Yongjian Lai Zhaojie Li Wenxia Zhao Xiaoning Cheng Shuo Xu Xiao Yu Yong Liu | 2020 | Nano Research2020,13,12: | 2 |
| 9 | Hyperactive PI3Kδ predisposes naive T cells to activation via aerobic glycolysis programs显示文摘Activated phosphoinositide 3-kinaseδsyndrome(APDS)is an autosomal-dominant combined immunodeficiency disorder resulting from pathogenic gain-of-function(GOF)mutations in the PIK3CD gene.Patients with APDS display abnormal T cell homeostasis.However,the mechanisms by which PIK3CD GOF contributes to this feature remain unknown.Here,with a cohort of children with PIK3CD GOF mutations from multiple regions of China and a corresponding CRISPR/Cas9 gene-edited mouse model,we reported that hyperactive PI3Kδdisrupted TNaive cell homeostasis in the periphery by intrinsically promoting the growth,proliferation,and activation of TNaive cells.Our results showed that PIK3CD GOF resulted in loss of the quiescence-associated gene expression profile in naive T cells and promoted naive T cells to overgrow,hyperproliferate and acquire an activated functional status.Naive PIK3CD GOF T cells exhibited an enhanced glycolytic capacity and reduced mitochondrial respiration in the resting or activated state.Blocking glycolysis abrogated the abnormal splenic T cell pool and reversed the overactivated phenotype induced by PIK3CD GOF in vivo and in vitro.These results suggest that enhanced aerobic glycolysis is required for PIK3CD GOF-induced overactivation of naive T cells and provide a potential therapeutic approach for targeting glycolysis to treat patients with APDS as well as other immune disorders. | Yanjun Jia Qiuyun Yang Yanping Wang Wenyan Li Xuemei Chen Tao Xu Zhirui Tian Minxuan Feng Liang Zhang Wenjing Tang Na Tian Lina Zhou Wenxia Song Xiaodong Zhao | 2021 | Cellular & Molecular Immunology2021,18,7: | 2 |

| 17 | Change in Precipitation over the Tibetan Plateau Projected by Weighted CMIP6 Models显示文摘Precipitation over the Tibetan Plateau(TP)is important to local and downstream ecosystems.Based on a weighting method considering model skill and independence,changes in the TP precipitation for near-term(2021-40),mid-term(2041-60)and long-term(2081-2100)under shared socio-economic pathways(SSP1-1.9,SSP1-2.6,SSP2-4.5,SSSP3-7.0,SSP5-8.5)are projected with 27 models from the latest Sixth Phase of the Couple Model Intercomparison Project.The annual mean precipitation is projected to increase by 7.4%-21.6%under five SSPs with a stronger change in the northern TP by the end of the 21st century relative to the present climatology.Changes in the TP precipitation at seasonal scales show a similar moistening trend to that of annual mean precipitation,except for the drying trend in winter precipitation along the southern edges of the TP.Weighting generally suggests a slightly stronger increase in TP precipitation with reduced model uncertainty compared to equally-weighted projections.The effect of weighting exhibits spatial and seasonal differences.Seasonally,weighting leads to a prevailing enhancement of increase in spring precipitation over the TP.Spatially,the influence of weighting is more remarkable over the northwestern TP regarding the annual,summer and autumn precipitation.Differences between weighted and original MMEs can give us more confidence in a stronger increase in precipitation over the TP,especially for the season of spring and the region of the northwestern TP,which requires additional attention in decision making. | Yin ZHAO Tianjun ZHOU Wenxia ZHANG Jian LI | 2022 | Advances in Atmospheric Sciences2022,39,7: | 1 |
