| 2 | Respiratory Virus Multiplex RT-PCR Assay Sensitivities and Influence Factors in Hospitalized Children with Lower Respiratory Tract Infections显示文摘Multiplex RT-PCR assays have been widely used tools for detection and differentiation of a panel of respiratory viral pathogens. In this study, we evaluated the Qiagen ResPlex II V2.0 kit and explored factors influencing its sensitivity. Nasopharyngeal swab (NPS) specimens were prospectively collected from pediatric inpatients with lower respiratory tract infections at the time of admission in the Shenzhen Children's Hospital from May 2009 to April 2010. Total nucleic acids were extracted using the EZ1 system (Qiagen, Germany) and 17 respiratory viruses and genotypes including influenza A virus (FluA), FluB, parainfluenza virus 1 (PIV1), PIV2, PIV3, PIV4, respiratory syncytial virus (RSV), human metapneumovirus (hMPV), rhinoviruses (RhV), enteroviruses (EnV), human bocaviruses (hBoV), adenoviruses (AdV), four coronaviruses (229E, OC43, NL63 and HKU1), and FluA 2009 pandemic H1N1(H1N1-p) were detected and identified by the ResPlex II kit. In parallel, 16 real-time TaqMan quantitative RT-PCR assays were used to quantitatively detect each virus except for RhV. Influenza and parainfluenza viral cultures were also performed. Among the total 438 NPS specimens collected during the study period, one or more viral pathogens were detected in 274 (62.6%) and 201(45.9%) specimens by monoplex TaqMan RT-PCR and multiplex ResPlex, respectively. When results from monoplex PCR or cell culture were used as the reference standard, the multiplex PCR possessed specificities of 92.9-100.0%. The sensitivity of multiplex PCR for PIV3, hMPV, PIV1 and BoV were 73.1%, 70%, 66.7% and 55.6%, respectively, while low sensitivities (11.1%-40.0%) were observed for FluA, EnV, OC43, RSV and H1N1. Among the seven viruses/genotypes detected with higher frequencies, multiplex PCR sensitivities were correlated significantly with viral loads determined by the TaqMan RT-PCR in FluA, H1N1-p and RSV (p=0.011-0.000). The Qiagen ResPlex II multiplex RT-PCR kit possesses excellent specificity for simultaneous detection of 17 viral pathogens in NPS specimens in pediatric inpatients at the time of admission. | 14 | The measurement of internal surface characteristics of fuel nozzle orifices using the synchrotron X-ray micro CT technology显示文摘The design of fuel nozzle orifices at micrometer scales is crucial for generating desired fuel spray patterns, and consequently optimizing fuel combustion and emission in internal combustion engines. Although there have been several recent advancements in the characterization of orifice internal geometries, quantitative studies on the orifice internal wall surface characteristics are still challeges due to the lack of effective measuring methods. A new method for quantifying the internal wall surface characteristics of fuel nozzle micro-orifices is presented in this study to achieve a better understanding and prediction of spray characteristics: Firstly, by using the synchrotron X-ray micro CT technology, a three-dimensional digital model of the fuel nozzle tip was constructed. Secondly, a data post-processing technique was then applied to unfold the orifice internal wall surface to a flat base plane. Finally, the conventional surface characteristic quantification techniques can be used to evaluate the wall surface characteristics. Two diesel nozzles with identical orifice geometry design but different hydraulic grinding time were measured using this method. One nozzle was hydro-ground for 2 s while the other was not. The internal wall surfaces of the two orifices were successfully unfolded to base planes and their surface characteristics were respectively analyzed. The surface fluctuation data were perfectly reproduced by a Gaussian distribution function. The standard deviations of the distribution demonstrate the fluctuation range and the distribution of the entire surface fluctuation profiles. | 17 | Pd/silicalite-1: An highly active catalyst for the oxidative removal of toluene显示文摘Catalytic combustion is thought as an efficient and economic pathway to remove volatile organic compounds, and its critical issue is the development of high-performance catalytic materials. In this work, we used the in situ synthesis method to prepare the silicalite-1(S-1)-supported Pd nanoparticles(NPs). It is found that the as-prepared catalysts displayed a hexagonal prism morphology and a surface area of 390-440 m^(2)/g. The sample(0.28Pd/S-1-H)derived after reduction at 500°C in 10 vol% H_(2)showed the best catalytic activity for toluene combustion(T50%= 180℃ and T90%= 189℃ at a space velocity of 40,000 m L/(g·hr), turnover frequency(TOFPd) at 160℃ = 3.46 × 10^(-3)sec^(-1), and specific reaction rate at 160℃ = 63.8μmol/(gPd·sec)), with the apparent activation energy(41 k J/mol) obtained over the bestperforming 0.28Pd/S-1-H sample being much lower than those(51-70 k J/mol) obtained over the other samples(0.28Pd/S-1-A derived from calcination at 500℃ in air, 0.26Pd/S-1-im derived from the impregnation route, and 0.27Pd/ZSM-5-H prepared after reduction at 500℃ in 10 vol% H_(2)). Furthermore, the 0.28Pd/S-1-H sample possessed good thermal stability and its partial deactivation due to CO_(2) or H_(2)O introduction was reversible, but SO_(2) addition resulted in an irreversible deactivation. The possible pathways of toluene oxidation over 0.28Pd/S-1-H was toluene → p-methylbenzoquinone → maleic anhydride, benzoic acid, benzaldehyde → carbon dioxide and water.
