1A mimotope of Pre-S_2 region of surface antigen of viral hepatitis B screened by phage display显示文摘To acquire the phage-displayed mimotopes which mimic the specificity of hepatitis B virus surface antigen (HBsAg), a random peptide library expressing linear peptide with 12 amino asids in length were used to screen with the serum from a hepatitis B virus infected patient in the recovery phase. After 3 rounds of biopanning, the positive phages were confirmed by competitive ELISA using HBsAg/P33. Two phagotopes were identified and one of them was confirmed as mimotope by competition experiment. Based on the mimotpe, a multiple antigenic peptide with four branches was synthesized by solid phase peptide synthesis. The antiginicity and specificity of the synthesized antigen was tested in BALB/c mice compared with the native epitope-based antigen. The results showed that the mimotope-based antigen could evoke higher titer of antibodies with the same specificity of the epitope-based antigen. 3Optimal Design and Force Control of a Nine-Cable-Driven Parallel Mechanism for Lunar Takeoff Simulation显示文摘Traditional simulation methods are unable to meet the requirements of lunar takeo simulations, such as high force output precision, low cost, and repeated use. Considering that cable-driven parallel mechanisms have the advantages of high payload to weight ratio, potentially large workspace, and high-speed motion, these mechanisms have the potential to be used for lunar takeo simulations. Thus, this paper presents a parallel mechanism driven by nine cables. The purpose of this study is to optimize the dimensions of the cable-driven parallel mechanism to meet dynamic workspace requirements under cable tension constraints. The dynamic workspace requirements are derived from the kinematical function requests of the lunar takeo simulation equipment. Experimental design and response surface methods are adopted for building the surrogate mathematical model linking the optimal variables and the optimization indices. A set of dimensional parameters are determined by analyzing the surrogate mathematical model. The volume of the dynamic workspace increased by 46% after optimization. Besides, a force control method is proposed for calculating output vector and sinusoidal forces. A force control loop is introduced into the traditional position control loop to adjust the cable force precisely, while controlling the cable length. The e ectiveness of the proposed control method is verified through experiments. A 5% vector output accuracy and 12 Hz undulation force output can be realized. 4Characterization and Proteomic Analysis of Novel Rice Lesion Mimic Mutant with Enhanced Disease Resistance显示文摘Lesion mmic mutants(LMMs)are plants that spontaneously form lesions without pathogeninfection or external stimulus and exhibit resistance to pathogens.Here,a rice LMM was created by ethylmethane sulfonate mutagenesis,named as hpil(hydrogen peroxide induced lesion).Diaminobenzidineand trypan blue staining showed that large amounts of H_(2)O_(2) were produced and cell death was occurredat and around the parts of lesion mimic in the rice leaves.The phenotype of hpil is controlled by a singlerecessive gene,localized at a 2 Mb interval on chromosome 2.The data suggested that hpil is a novelLMM with enhanced bacterial and fungal disease resistance,and multiple pathogenesis-related proteins(PRs)were up-regulated.The proteomes of leaves at three positions(different degrees of lesion mimicseverity)were characterized in hpil compared with its wild type plant.Differentially expressed proteinswere detected by two dimensional difference gel electrophoresis and 274 proteins were identified byMALDITOF/TOFTM.These proteins were related to metabolic process,cellular process and response tostimulus,with mostly down-regulated in hpil leaves.Many of these proteins were related to the Calvincycle,photosynthetic electron transport chain,glycolysis/gluconeogenesis and phosphonates pathways.Some resistance-related proteins including 14-3-3 proteins,OsPR10 and antioxidases such asperoxidase,superoxide dismutase and ascorbate peroxidase were up-regulated in leaves with lesionmimic.These results provide the foundation for cloning of the target gene and shed light on themechanism involved in autaimmunity of rice.Yang Yong Lin Qiujun Chen Xinyu Liang Weifang Fu Yuwen Xu Zhengjin Wu Yuanhua Wang Xuming Zhou Jie Yu Chulang Yan Chengqi Mei Qiong Chen Jianping 2021Rice science2021,28,5:2
5Effects of psycho-scene-drama on empathic ability of patients with chronic schizophrenia显示文摘Objective:The purpose of the current study was to determine the effects of psycho-scene drama on the empathic ability of patients with chronic schizophrenia.Methods:One hundred sixteen in-patients with chronic schizophrenia were randomly di-vided into study and control groups(n=58 cases each).The original anti-psychotic drugs were not changed in either group.In the study group,the therapy was combined with psychological scene drama with a treatment course of 3 months.Before and after treatment,the interpersonal reaction index scale(Chinese version[IRI-C]),negative syndrome scale(SANS),social function assessment scale for patients with mental diseases(SSPI),Rosenberg self-esteem scale(SES),the emotional-social loneliness inventory(ESLI),and the Texas social behavior inventory(TSBI)were utilized for measurements.Results:A comparison of the IRI-C total,PT,FS,and EC factor scores before and after treatment in the study group was significantly different(P<0.01);the PD factor score before and after treatment was not statistically significant(P>0.05).A comparison of the IRI-C total score and each factor score before and after treatment in the control group was not significantly different(P>0.05).A comparison of the IRI-C total,PT,and EC factor scores between the study and control groups after treatment was significantly different(P<0.05).A comparison of the FS factor score between the study and control groups before and after treatment was significantly different(P<0.01);the PD factor score was not significantly different(P>0.05).A comparison of the SANS total,SSPI total,SES total,and ESLI total scores between the two groups after treatment was significantly different(P<0.05).Conclusion:The psycho-scene drama can improve the empathic ability of patients with chronic schizophrenia,improve the negative symptoms and social function,increase the self-esteem and self-confidence,and reduce the loneliness in social interactions.Hongmei Tian Weifang Niu Ping Liu Na Wang Huijing Yu Zhanhong Zhao 2014Family Medicine and Community Health2014,2,4:2
6CC chemokine ligand 2 (CCL2)enhances TTX-sensitive sodium channel activity of primary afferent neurons in the complete Freud adjuvant-induced inflammatory pain model显示文摘CC chemokine ligand 2 (CCL2)has been implicated in pathological pain,but the mechanism underlying the pronociceptive effect of CCL2 is not fully understood.Voltage-gated sodium (Nav)channels are important determinants of the excitability of sensory neurons.Hence we tested the hypothesis that CCL2 contributes to inflammatory pain via modulating Nay channel activity of primary afferent neurons. Chronic inflammatory pain was induced in rats by intraplantar injection of the complete Freud adjuvant (CFA)to one of the hind paws.Control rats received intraplantar injection of equal volume of saline.A significant increase of CCL2 mRNA and CCL2 receptor (CCR2)protein expression was detected in the ipsilateral dorsal root ganglion (DRG)in CFA-treated rats.lntrapiantar injection of CCL2 protein in the control rats had minimal effect on the paw withdrawal threshold (PWT)in response to mechanical stimulation.However,in CFA-treated rats,intraplantar CCL2 led to an increase in pain responses.Patchclamp recording of acutely dissociated DRG neurons revealed that CCL2 had minimum effect on the excitability of sensory neurons from control rats.However,CCL2 directly depolarized a large proportion of small to medium-sized sensory neurons from CFA-treated rats.In addition,CCL2 was found to enhance whole-cell TTX-sensitive sodium currents without significantly affecting the TTX-resistant sodium currents and the potassium currents.These results are in agreement with previous reports concerning the involvement of CCL2-CCR2 signaling in inflammatory hyperalgesia and further indicate that enhanced TTX-sensitive channel activity may partly underlie the pronociceptive effects of CCL2.Ping Luo Jiayun Shao Yingfu Jiao Weifeng Yu Weifang Rong 2018Acta Biochimica et Biophysica Sinica2018,50,12:2

12SIRCM1,which encodes tomato Lutescent1,is required for chlorophyll synthesis and chloroplast development in fruits显示文摘In plants,chloroplasts are the sites at which photosynthesis occurs,and an increased abundance of chloroplasts increases the nutritional quality of plants and the resultant color of fruits.However,the molecular mechanisms underlying chlorophyll synthesis and chloroplast development in tomato fruits remain unknown.In this study,we isolated a chlorophyll-de fi cient mutant,reduced chlorophyll mutant 1(rcml),by ethylmethanesulfonate mutagenesis;this mutant produced yellowish fruits with altered chloroplast development.MutMap revealed that Solyc08g005010 is the causal gene underlying the rcm1 mutant phenotype.A single-nucleotide base substitution in the second exon of SIRCM1 results in premature termination of its translated protein.SIRCM1 encodes a chloroplast-targeted metalloendopeptidase that is orthologous to the BCM1 protein of Arabidopsis and the stay-green G protein of soybean(Glycine max L.Merr.).Notably,the yellowish phenotype of the lutescent1 mutant can be restored with the allele of SlRCM1 from wild-type tomato.In contrast,knockout of SlRCM1 by the CRISPR/Cas9 system in Alisa Craig yielded yellowish fruits at the mature green stage,as was the case for lutescent1.Amino acid sequence alignment and functional complementation assays showed that SlRCM1 is indeed Lutescent1.These fi ndings provide new insights into the regulation of chloroplast development in tomato fruits.Genzhong Liu Huiyang Yu Lei Yuan Changxing Li Jie Ye Weifang Chen Ying Wang Pingfei Ge Junhong Zhang Zhibiao Ye Yuyang Zhang 2021Horticulture Research2021,8,1:1
13Distal axis sulfide mineralization on the ultraslow-spreading Southwest Indian Ridge:an LA-ICP-MS study of pyrite from the East Longjing-2 hydrothermal field显示文摘The newly discovered East Longjing-2 hydrothermal field(ELHF-2)is located on the Dragon Horn oceanic core complex of the ultraslow-spreading Southwest Indian Ridge,approximately 12 km from the ridge axis.This study measured the chemical compositions of pyrite from ELHF-2 using a laser ablation inductively coupled plasma mass spectrometry(LA-ICP-MS)to investigate the genesis of the field.Three generations of pyrite were classified,and found that:Py1 and Py2,rich in V,Mn,U,and Se,occur in altered basalt debris and the silica alteration matrix,respectively.Py3 was mainly intergrown with chalcopyrite in quartz veins and had higher Cu,In,Ag,Sb,and Au contents than Py1 and Py2.Some elements,such as Au,Se,and Pb,are likely presented as direct substitution with Fe^(2+)in pyrite,while Cu,Zn,Co,Ni,and Ag probably occur both as direct substitution with Fe and as distributed micro-to nanoparticle-sized sulfides.Meanwhile,the occurrence of V,Mn,and U is likely presented as oxide inclusions.Trace element geochemistry suggested that the pyrite was formed under hightemperature conditions,and the ore forming elements were likely derived from ultramafic rocks.In addition,Py1and Py2 were formed under higher water/rock ratio and higher temperature conditions,with more seawater involvement compared with Py3.The formation of ELHF-2 was probably driven by exothermic serpentinization reactions with an additional magmatic heat.This study shows that high-temperature hydrothermal circulation driven by magmatic activity can be developed on distal rift flank areas of magma-starved ultraslow-spreading ridges.Shili Liao Chuanwei Zhu Jianping Zhou Weiyong Liu Junyu Yu Jin Liang Weifang Yang Wei Li Jia Liu Chunhui Tao 2021Acta Oceanologica Sinica2021,40,5:1
14NATURAL HISTORY OBSERVATION FOR ESOPHAGEAL AND CARDIA PRECURSORS BY REPETITIVE ENDOSCOPIC SCREENING OF 301 SUBJECTS IN SHEXIAN显示文摘OBJECTIVE To investigate the natural history of fast developing esopha- geal and cardia precursors. METHODS Repetitive endoscopic screenings were performed among 40-69-year-olds in the high-incidence areas for esophageal cancer in Shexian. RESULTS The initial diagnosis and the lag-time for 7 subsequently identified severe dysplasia(SD)subjects were as follows:in one subject 13 months after a baseline diagnosis of normal epithelium,in another subject 7 months after a baseline diagnosis of base cell hyperplasia(BCH),in four subjects 3,4,4,and 10.5 months after baseline diagnosis of mild dysplasia (mD),and in one subject 12.5 months after a baseline diagnosis of moder- ate dysplasia(MD).The initial diagnosis and the lag-time for 6 subsequently identified carcinomas in situ or intramucosal carcinoma cases were:in one case 48 months after a baseline diagnosis of mD,in 2 cases 4 and 13 months after baseline diagnoses of MD,and in the other 3 cases 3.5,9,and 17.5 months after baseline diagnoses of SD.The initial diagnosis and lag- time for 3 subsequently identified invasive cancer cases were:in one case 50 months after a baseline diagnosis of MD,in 2 cases 14 and 19 months after baseline diagnoses of SD.In addition,during a 4-year-fol ow-up of 18 subjects after endoscopic mucosa resection,9 of them were found to have developed precursors again at other sites,and also additional findings were obtained for 11 of the 16 dysplasia cases by repetitive biopsy in less than 2 months after the initial endoscopy.
