3Mutual dependency between lncRNA LETN and protein NPM1 in controlling the nucleolar structure and functions sustaining cell proliferation显示文摘Fundamental processes such as ribosomal RNA synthesis and chromatin remodeling take place in the nucleolus,which is hyperactive in fast-proliferating cells.The sophisticated regulatory mechanism underlying the dynamic nucleolar structure and functions is yet to be fully explored.The present study uncovers the mutual functional dependency between a previously uncharacterized human long non-coding RNA,which we renamed LETN,and a key nucleolar protein,NPM1.Specifically,being upregulated in multiple types of cancer,LETN resides in the nucleolus via direct binding with NPM1.LETN plays a critical role in facilitating the formation of NPM1 pentamers,which are essential building blocks of the nucleolar granular component and control the nucleolar functions.Repression of LETN or NPM1 led to similar and profound changes of the nucleolar morphology and arrest of the nucleolar functions,which led to proliferation inhibition of human cancer cells and neural progenitor cells.Interestingly,this inter-dependency between LETN and NPM1 is associated with the evolutionarily new variations of NPM1 and the coincidental emergence of LETN in higher primates.We propose that this human-specific protein–lncRNA axis renders an additional yet critical layer of regulation with high physiological relevance in both cancerous and normal developmental processes that require hyperactive nucleoli.Xianteng Wang Xiaolin Hu Wanlu Song Hui Xu Zhengtao Xiao Rongyao Huang Qingran Bai Fan Zhang Yongzhen Chen Yu Liu Jianhuo Fang Xin Li Qin Shen Haitao Zhao Xuerui Yang 2021Cell Research2021,31,6:3

83D-bioprinted cholangiocarcinoma-on-a-chip model for evaluating drug responses显示文摘Cholangiocarcinoma(CCA)is characterized by heterogeneous mutations and a refractory nature.Thus,the development of a model for effective drug screening is urgently needed.As the established therapeutic testing models for CCA are often ineffective,we fabricated an enabling three-dimensional(3D)-bioprinted CCA-on-a-chip model that to a good extent resembled the multicellular microenvironment and the anatomical microstructure of the hepato-vascular-biliary system to perform high-content antitumor drug screening.Specifically,cholangiocytes,hepatocytes,and vascular endotheliocytes were employed for 3D bioprinting of the models,allowing for a high degree of spatial and tube-like microstructural control.Interestingly,it was possible to observe CCA cells attached to the surfaces of the gelatin methacryloyl(GelMA)hydrogelembedded microchannels and overgrown in a thickening manner,generating bile duct stenosis,which was expected to be analogous to the in vivo configuration.Over 4000 differentially expressed genes were detected in the CCA cells in our 3D coculture model compared to the traditional two-dimensional(2D)monoculture.Further screening revealed that the CCA cells grown in the 3D traditional model were more sensitive to the antitumoral prodrug than those in the 2D monoculture due to drug biotransformation by the neighboring functional hepatocytes.This study provides proof-of-concept validation of our bioprinted CCA-on-a-chip as a promising drug screening model for CCA treatment and paves the way for potential personalized medicine strategies for CCA patients in the future.Qiong Liu Luis SMille Cesar Villalobos Ingrid Anaya Matthias Vostatek Sili Yi Wanlu Li Junlong Liao Huanghui Wu Yongteng Song Lize Xiong Yu Shrike Zhang 2023Bio-Design and Manufacturing2023,6,4:0

10Method to Appraise Dangerous Class of Building Masonry Component Based on DC-YOLO Model显示文摘This DC-YOLO Model was designed in order to improve the efficiency for appraising dangerous class of buildings and avoid manual intervention,thereby making the appraisal results more objective.It is an automated method designed based on deep learning and target detection algorithms to appraise the dangerous class of building masonry component.Specifically,it(1)adopted K-means clustering to obtain the quantity and size of the prior boxes;(2)expanded the grid size to improve identification to small targets;(3)introduced in deformable convolution to adapt to the irregular shape of the masonry component cracks.The experimental results show that,comparing with the conventional method,the DC-YOLO model has better recognition rates for various targets to different extents,and achieves good effects in precision,recall rate and F1 value,which indicates the good performance in classifying dangerous classes of building masonry component.Hongrui Zhang Wenxue Wei Xinguang Xiao Song Yang Wanlu Shao 2020Computers, Materials & Continua2020,,4:0
