1Oral uptake and persistence of the FnAb-8 protein characterized by in situ radio-labeling and PET/CT imaging显示文摘The absorption of peptides and proteins delivered orally is minimum because of the intestine epithelial barrier.There are few known active transport mechanisms for macromolecules including the neonatal Fc Receptor(FcRn)for the absorption and secretion of IgGs in infant and adult intestine.We had previously described the FnAb-8 protein that could bind to h FcRn tightly at pH 6.0 but barely at pH 7.4.In this study,we examined its uptake,biodistribution and pharmacokinetics after peroral administration in both wild-type and human FcRn transgenic(Tg)mice.FnAb-8 was modified to contain trans-cyclooctene(TCO)which could interact with18 F labeled tetrazine in situ via the bioorthogonal inverseelectron-demand Diels-Alder reaction.We showed that FnAb-8 had a tendency to distribute and persist in the Tg mice intestine for an extended duration of time.It could also be absorbed into the circulation and distributed systemically over a long period of time up to 172 h.The improvement in oral uptake and concentration in the intestine tissue may be valuable for designing oral delivery of biopharmaceuticals,especially for diseases involving the gastric intestinal tissue.Qian Wang Wangxi Hai Sanyuan Shi Jinliang Peng Yuhong Xu 2020Asian Journal of Pharmaceutical Sciences2020,15,6:3
2Effect of Different Irrigation Patterns on Transportation and Allocation of Carbohydrate During Grain Filling of Rice显示文摘Using intersubspecific hybrid rice Xieyou 9308 and Liangyou Peijiu as the tested materials, the effects of differentirrigation patterns on transportation and allocation of carbohydrate during grain filling stage was observed by the designwith three level of soil water content in irrigated field. The results showed that in the conventional flooding and thealternate dry-watering cultivations, the exported rate of stored carbohydrate from stem and photophate from the leaves were60 and 90 % respectively. The exported rate of carbohydrate was decreased significantly (P<0.01) in the non-floodingcultivation. There was no significant difference between the conventional flooding and dry-wet alternation treatments interm of the exported rate of carbohydrate. The filling grains were the major sinks for carbohydrate storage during grainfilling stage. Grains received nearly 50% of stored carbohydrate from leaf sheath and 80% of photophate from leaves. Atthe nonflooding condition the absorbing rates of grains were significantly decreased by 10 % from leaf sheath and 20 %from leaves photophate. 8Ionic strength directed self-assembled polyelectrolyte single-bilayer membrane for low-pressure nanofiltration显示文摘Layer-by-layer assembly is a versatile technique for fabricating nanofiltration membranes,where multiple layers of polyelectrolytes are usually required to achieve reasonable separation performance.In this work,an ionic strength directed self-assembly procedure is described for the preparation of nanofiltration membranes consisting of only a single bilayer of poly(diallyldimethylammoniumchloride)and poly(sodium-4-styrenesulfoate).The influence of background ionic strength as well as membrane substrate properties on the formation of singlebilayer membranes are systematically evaluated.Such a simplified polyelectrolyte deposition procedure results in membranes having outstanding separation performance with permeating flux of 14.21.5 L∙m^(–2)∙h^(-1)∙bar^(–1) and Na2SO4 rejection of 97.1%0.8%under a low applied pressure of 1 bar.These results surpass the ones for conventional multilayered polyelectrolyte membranes.This work encompasses an investigation of ionic strength induced coiling of the polyelectrolyte chains and emphasizes the interplay between-polyelectrolyte chain configuration and substrate pore profile.It thus introduces a new concept on the control of membrane fabrication process toward high performance nanofiltration.Feng Zhang Lu Tan Li Gong Shuqi Liu Wangxi Fang Zhenggong Wang Shoujian Gao Jian Jin 2022Frontiers of Chemical Science and Engineering2022,16,5:1
