1Chemopreventive effect of apple and berry fruits against colon cancer显示文摘Colon cancer arises due to the conversion of precancerous polyps(benign)found in the inner lining of the colon.Prevention is better than cure,and this is very true with respect to colon cancer.Various epidemiologic studies have linked colorectal cancer with food intake.Apple and berry juices are widely consumed among various ethnicities because of their nutritious values.In this review article,chemopreventive effects of these fruit juices against colon cancer are discussed.Studies dealing with bioavailability,in vitro and in vivo effects of apple and berry juices are emphasized in this article.A thorough literature survey indicated that various phenolic phytochemicals present in these fruit juices have the innate potential to inhibit colon cancer cell lines.This review proposes the need for more preclinical evidence for the effects of fruit juices against different colon cancer cells,and also strives to facilitate clinical studies using these juices in humans in large trials.The conclusion of the review is that these apple and berry juices will be possible candidates in the campaign against colon cancer.Saravana Kumar Jaganathan Muthu Vignesh Vellayappan Gayathri Narasimhan Eko Supriyanto Dyah Ekashanti Octorina Dewi Aqilah Leela T Narayanan Arunpandian Balaji Aruna Priyadarshini Subramanian Mustafa Yusof 2014World Journal of Gastroenterology2014,20,45:3
2Thymoquinone as a potential therapeutic for Alzheimer’s disease in transgenic Drosophila melanogaster model显示文摘Alzheimer’s disease(AD)is one of the most common forms of dementia.Cognitive dysfunction and memory loss are the two main clinical symptoms of AD.Drosophila melanogaster models of AD,which are based on overexpression of human amyloidβ(Aβ)or human tau(hTau)protein,have been used to study the mechanism underlying AD and to screen potential therapeutic compounds.Drugs that are currently available for AD provide only symptomatic relief.Huge unmet medical needs exists to slow,stop,or reverse the progression of AD.Thymoquinone(TQ)is an active ingredient isolated from Nigella sativa(NS)and possesses various pharmacological activities,and it is also a potential neuropharmacological agent.The current study was performed to investigate the effect of dietary administration of TQ at concentrations of 12.5μM and 25μM for 15 and 30 days on biochemical and behavioral parameters,gene,and protein expression of hTau,using Drosophila models of AD.Transgenic Drosophila models exhibiting pan-neuronal and eye-specific expression of hTau were generated using the GAL4/UAS system.Treatment with TQ at both concentrations resulted in a significant increase in behavioral activity,a significant reduction in the amount of reactive oxygen species(ROS),and restoration of depleted superoxide dismutase(SOD)and acetylcholine esterase(AChE)activities.A significant decrease in gene and protein expression of hTau was also observed at the molecular level for both concentrations of TQ.Therefore,TQ has the potential to be investigated as a potential therapeutic phytochemical for the treatment of AD in future studies.NARAYANAN NAMPOOTHIRI V.P. 3Molecular therapeutics in pancreas cancer显示文摘The emergence of the 'precision-medicine' paradigm in oncology has ushered in tremendous improvements in patient outcomes in a wide variety of malignancies. However, pancreas ductal adenocarcinoma(PDAC) has remained an obstinate challenge to the oncology community and continues to be associated with a dismal prognosis with 5-year survival rates consistently less than 5%. Cytotoxic chemotherapy with gemcitabine-based regimens has been the cornerstone of treatment in PDAC especially because most patients present with inoperable disease. But in recent years remarkable basic science research has improved our understanding of the molecular and genetic basis of PDAC. Whole genomic analysis has exemplified the genetic heterogeneity of pancreas cancer and has led to ingenious efforts to target oncogenes and their downstream signaling cascades. Novel stromal depletion strategies have been devised based on our enhanced recognition of the complex architecture of the tumor stroma and the various mechanisms in the tumor microenvironment that sustain tumorigenesis. Immunotherapy using vaccines and immune checkpoint inhibitors has also risen to the forefront of therapeutic strategies against PDAC. Furthermore, adoptive T cell transfer and strategies to target epigenetic regulators are being explored with enthusiasm.
