| 7 | Multi-spectroscopic investigation of the binding interaction of fosfomycin with bovine serum albumin显示文摘The interaction between fosfomycin(FOS) and bovine serum albumin(BSA) has been investigated effectively by multi-spectroscopic techniques under physiological p H 7.4. FOS quenched the intrinsic fl uorescence of BSA via static quenching. The number of binding sites n and observed binding constant K A were measured by the fl uorescence quenching method. The thermodynamic parameters ΔG0, ΔH0andΔS0were calculated at different temperatures according to the van't Hoff equation. The site of binding of FOS in the protein was proposed to be Sudlow's site I based on displacement experiments using site markers viz. warfarin, ibuprofen and digitoxin. The distance r between the donor(BSA) and acceptor(FOS) molecules was obtained according to the F?rster theory. The effect of FOS on the conformation of BSA was analyzed using synchronous fl uorescence spectra(SFS), circular dichroism(CD) and 3D fl uorescence spectra. It is a widely employed approach to retain the dosage form in the stomach for an extended period of time and release the drug slowly that can address many challenges associated with conventional oral delivery, including poor bioavailability.Different innovative approaches like magnetic field assisted gastro-retention, plug type swelling system, muco-adhesion technique, floating system with or without effervescence are being applied to fabricate GRDDS. Apart from in vitro characterization, successful GRDDS development demands well designed in vivo study to establish enhanced gastro-retention and prolonged drug release. Gama scintigraphy and MRI are popular techniques to evaluate in vivo gastric residence time. However, checking of their overall in-vivo efficacy still remains a major challenge for this kind of dosage form, especially in small animals like mice or rat.Reported in vivo studies with beagle dogs, rabbits, and human subjects are only a handful in spite of a large number of encouraging in vitro results. In spite of the many advantages,high subject variations in gastrointestinal physiological condition, effect of food, and variable rate of gastric emptying time are the challenges that limit the number of available GRDDS in the market. | 10 | Role of T cell death in maintaining immune tolerance during persistent viral hepatitis显示文摘Virus-specific T cells play an important role in the resolution of hepatic infection. However, during chronic hepatitis infection these cells lack their effector functions and fail to control the virus. Hepatitis B virus and hepatitis C virus have developed several mechanisms to generate immune tolerance. One of these strategies is the depletion of virus-specific T cells by apoptosis. The immunotolerogenic liver has unique property to retain and activate na ve T cell to avoid the over reactivation of immune response against antigens which is exploited by hepatotropic viruses to persist. The deletion of the virus-specific T cells occurs by intrinsic (passive) apoptotic mechanism. The pro-apoptotic molecule Bcl-2 interacting mediator (Bim) has attracted increasing attention as a pivotal involvement in apoptosis, as a regulator of tissue homeostasis and an enhancer for the viral persistence. | 11 | Effect of tool rotational speed on the particle distribution in friction stir welding of AA6092/17.5 SiCp-T6 composite plates and its consequences on the mechanical property of the joint显示文摘This study investigates the effect of tool rotational speed(TRS)on particle distribution in nugget zone(NZ)through quantitative approach and its consequences on the mechanical property of friction stir welded joints of AA6092/17.5 SiCp-T6 composite.6 mm thick plates are welded at a constant tool tilt angle of 2°and tool traverse speed of 1 mm/s by varying the TRS at 1000 rpm,1500 rpm and 2000 rpm with a taper pin profiled tool.Microstructure analysis shows large quantity of uniformly shaped smaller size SiC particle with lower average particle area which are homogeneously distributed in the NZ.The fragmentation of bigger size particles has been observed because of abrading action of the hard tool and resulting shearing effect and severe stress generation due to the rotation of tool.The particles occupy maximum area in the matrix compared to that of the base material(BM)due to the redistribution of broken particles as an effect of TRS.The migration of particles towards the TMAZ-NZ transition zone has been also encountered at higher TRS(2000 rpm).The microhardness analysis depicts variation in average hardness from top to bottom of the NZ,minimum for 1500 rpm and maximum for 2000 rpm.The impact strength at 1000 rpm and 1500 rpm remains close to that of BM(21.6 J)while 2000 rpm shows the accountable reduction.The maximum joint efficiency has been achieved at 1500 rpm(84%)and minimum at 1000 rpm(68%)under tensile loading.Fractographic analysis shows mixed mode of failure for BM,1000 rpm and 1500 rpm,whereas 2000 rpm shows the brittle mode of failure. | Uttam Acharya Barnik Saha Roy Subhash Chandra Saha | 2020 | Defence Technology(防务技术)2020,16,2: | 2 |
