| 1 | Antisense MMP-9 RNA inhibits malignant glioma cell growth in vitro and in vivo显示文摘The matrix-degrading metalloproteinases (MMPs), particularly MMP-9, play important roles in the pathogenesis and development of malignant gliomas. In the present study, the oncogenic role of MMP-9 in malignant glioma cells was investigated via antisense RNA blockade in vitro and in vivo. TJ905 malignant glioma cells were transfected with pcDNA3.0 vector expressing antisense MMP-9 RNA (pcDNA-AS-MMP9), which significantly decreased MMP-9 expression, and cell proliferation was assessed. For in vivo studies, U251 cells, a human malignant glioma cell line, were implanted subcutaneously into 4-to 6-week-old BALB/c nude mice. The mice bearing well-established U251 gliomas were treated with intratumoral pcDNA-AS-MMP9-Lipofectamine complex (AS-MMP-9-treated group), subcutaneous injection of endostatin (endostatin-treated group), or both (combined therapy group). Mice treated with pcDNA (empty vector)-Lipofectamine served as the control group. Four or eight weeks later, the volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity were assayed. We demonstrate that pcDNA-AS-MMP9 significantly decreased MMP-9 expression and inhibited glioma cell proliferation. Volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity in the antisense-MMP-9-treated and therapeutic alliance groups were significantly lower than those in the control group. The results suggest that MMP-9 not only promotes malignant glioma cell invasiveness, but also affects tumor cell proliferation. Blocking the expression of MMP-9 with antisense RNA substantially suppresses the malignant phenotype of glioma cells, and thus can be used as an effective therapeutic strategy for malignant gliomas. | Cuiyun Sun Qian Wang Hongxu Zhou Shizhu Yu Alain R. | 3 | Host Interferon-Stimulated Gene 20 Inhibits Pseudorabies Virus Proliferation显示文摘Host interferon-stimulated gene 20(ISG20)exerts antiviral effects on viruses by degrading viral RNA or by enhancing IFN signaling.Here,we examined the role of ISG20 during pseudorabies virus(PRV)proliferation.We found that ISG20 modulates PRV replication by enhancing IFN signaling.Further,ISG20 expression was upregulated following PRV infection and poly(I:C)treatment.Ectopic expression of ISG20 inhibited PRV proliferation in PK15 cells,whereas knockdown of ISG20 promoted PRV proliferation.In addition,ISG20 expression upregulated IFN-βexpression and enhanced IFN downstream signaling during PRV infection.Notably,PRV UL24 suppressed the transcription of ISG20,thus antagonizing its antiviral effect.Further domain mapping analysis showed that the N terminus(amino acids 1-90)of UL24 was responsible for the inhibition of ISG20 transcription.Collectively,these findings characterize the role of ISG20 in suppressing PRV replication and increase the understanding of host-PRV interplay. | Xiaoyong Chen Dage Sun Sujie Dong Huanjie Zhai Ning Kong Hao Zheng Wu Tong Guoxin Li Tongling Shan Guangzhi Tong | 2021 | Virologica Sinica2021,36,5: | 4 |
| 4 | A Classification–Detection Approach of COVID-19 Based on Chest X-ray and CT by Using Keras Pre-Trained Deep Learning Models显示文摘The Coronavirus Disease 2019(COVID-19)is wreaking havoc around the world,bring out that the enormous pressure on national health and medical staff systems.One of the most effective and critical steps in the fight against COVID-19,is to examine the patient’s lungs based on the Chest X-ray and CT generated by radiation imaging.In this paper,five keras-related deep learning models:ResNet50,InceptionResNetV2,Xception,transfer learning and pre-trained VGGNet16 is applied to formulate an classification-detection approaches of COVID-19.Two benchmark methods SVM(Support Vector Machine),CNN(Conventional Neural Networks)are provided to compare with the classification-detection approaches based on the performance indicators,i.e.,precision,recall,F1 scores,confusion matrix,classification accuracy and three types of AUC(Area Under Curve).The highest classification accuracy derived by classification-detection based on 5857 Chest X-rays and 767 Chest CTs are respectively 84%and 75%,which shows that the keras-related deep learning approaches facilitate accurate and effective COVID-19-assisted detection. | Xing Deng Haijian Shao Liang Shi Xia Wang Tongling Xie | 2020 | Computer Modeling in Engineering & Sciences2020,,11: | 4 |
| 5 | Reinforcement Learning Based Data Fusion Method for Multi-Sensors显示文摘In order to improve detection system robustness and reliability, multi-sensors fusion is used in modern air combat. In this paper, a data fusion method based on reinforcement learning is developed for multi-sensors. Initially, the cubic B-spline interpolation is used to solve time alignment problems of multisource data. Then, the reinforcement learning based data fusion(RLBDF) method is proposed to obtain the fusion results. With the case that the priori knowledge of target is obtained, the fusion accuracy reinforcement is realized by the error between fused value and actual value. Furthermore, the Fisher information is instead used as the reward if the priori knowledge is unable to be obtained. | 7 | Comparison of Viromes in Ticks from Different Domestic Animals in China显示文摘Ticks are involved in the transmission of various arboviruses and some tick-borne viruses pose significant threats to the health of humans or livestock.This study aimed to investigate the geographical distribution of tick species and tickassociated viruses in central and eastern China.Total 573 ticks from domestic animals including dogs,sheep and cattle were collected in 2017.Two genera of ticks were identified including Rhipicephalus and Haemaphysalis.Sequencing was performed on Miseq Illumina platform to characterize the tick viromes from the four different sampling locations.Following trimming,13,640 reads were obtained and annotated to 19 virus families.From these sequences,above 37.74%of the viral reads were related to the RNA viruses.Virome comparison study revealed that the tick viral diversity was considerably different in the two identified tick genera.The viral diversity of R.microplus was significantly different from that of other Rhipicephalus species.On the other hand,substantial overlap in viral species was observed between the same genera.In addition,we found no evidence that the natural host played a major role in shaping virus diversity based on the comparison of their viromes.Rather,the geographic location seems to significantly influence the viral families.Phylogenetic study indicated that the novel negative-sense RNA viruses identified in this study was closely related to Bole tick virus 1 and 3 viruses.In conclusion,the present study provides a baseline for comparing viruses detected in ticks,according to species,natural hosts,and geographic locations. | Tingting Zhao Haiyan Gong Xiaojuan Shen Wen Zhang Tongling Shan Xiangqian Yu Seong Jin Wang Li Cui | 2020 | Virologica Sinica2020,35,4: | 2 |

| 20 | Fusing Carbocycles of Inequivalent Ring Size to a Bis(imino)pyridine-Iron Ethylene Polymerization Catalyst: Distinctive Effects on Activity, PE Molecular Weight, and Dispersity显示文摘The 4,6-bis(arylimino)-1,2,3,7,8,9,10-heptahydrocyclohepta[b]quinoline-iron(II)chlorides(aryl=2,6-Me_(2)C_(6)H_(3) Fe1;2,6-Et_(2)C_(6)H_(3) Fe_(2);2,6-i-Pr_(2)C_(6)H_(3) Fe_(3);2,4,6-Me3C_(6)H_(2) Fe4;and 2,6-Et_(2)-4-Me_(2)C_(6)H_(2) Fe_(5))have been prepared in good yield by a straightforward one-pot reaction of 2,3,7,8,9,10-hexahydro-1H-cyclohepta[b]quinoline-4,6-dione,FeCl_(2)·4H_(2)O,and the appropriate aniline in acetic acid.All ferrous complexes have been characterized by elemental analysis and FT-IR spectroscopy.In addition,the structure of Fe_(3) has been determined by single crystal X-ray diffraction,which showed the iron center to adopt a distorted square pyramidal geometry with the saturated sections of the fused six-and sevenmembered carbocycles to be cis-configured.In combination with either MAO or MMAO,Fe1–Fe5 exhibited exceptionally high activities for ethylene polymerization(up to 15:86×10^(6) g(PE)mol^(−1)(Fe)h^(−1) at 40℃(MMAO)and 9:60×10^(6) g(PE)mol^(−1)(Fe)h^(−1) at 60℃(MAO))and produced highly linear polyethylene(HLPE,Tm≥128℃)with a wide range in molecular weights;in general,the MMAO-promoted polymerizations were more active.Irrespective of the cocatalyst employed,the 2,6-Me_(2)-substituted Fe1 and Fe4 proved the most active while the more sterically hindered 2,6-i-Pr_(2) Fe_(3) the least but afforded the highest molecular weight polyethylene(Mw:65.6–72.6 kg mol^(-1)).Multinuclear NMR spectroscopic analysis of the polymer formed using Fe4/MMAO at 40℃ showed a preference for fully saturated chain ends with a broad bimodal distribution a feature of the GPC trace(Mw/Mn=13:4).By contrast,using Fe4/MAO at 60℃ a vinyl-terminated polymer of lower molecular weight(Mw=14:2 kg mol^(−1))was identified that exhibited a unimodal distribution(Mw/Mn=3:8).Moreover,the amount of aluminoxane cocatalyst employed,temperature,and run time were also found to be influential on the modality of the polymer. | Zheng Wang Gregory A.Solan Yanping Ma Qingbin Liu Tongling Liang Wen-Hua Sun | 2019 | Research2019,,1: | 1 |
