| 1 | Anti-anxiety effect of Valeriana jatamansi Jones extract via regulation of the hypothalamus-pituitary-adrenal axis显示文摘BACKGROUND:Hypothalamus-pituitary-adrenal (HPA) axis dysfunction has been closely linked to anxiety.Previous studies have shown that Valeriana jatamansi Jones extract exhibits clear anxio-lytic effects, but it is unclear about the mechanism underlying regulation of the HPA axis dysfunction in these anxiolytic effects.OBJECTIVE:To observe the effects of Valeriana jatamansi Jones (Zhizhu Xiang) extract on HPA axis function in a rat model of anxiety, and to compare these effects with positive control estazolam.DESIGN, TIME AND SETTING:Randomized, controlled, animal experiment was performed at Chengdu University of Traditional Chinese Medicine, China, between February and September in 2006.MATERIALS:Estazolam was purchased from Shanghai Jiufu Pharmaceutical, China; Valeriana jatamansi Jones was purchased from the Lotus Pond Market for Chinese Herbal Medicine in Chengdu.It consisted of iridoids and flavonoid components.METHODS:A total of 72 Sprague Dawley rats, aged 2 months, were randomly assigned to 6 groups:low-, medium-, and high-dose Valeriana jatamansi Jones groups intragastrically injected with 0.3, 0.6, and 0.9 g/kg per day Valeriana jatamansi Jones extract, respectively; estazolam group intragastrically injected with 1.5 mg/kg per day estazolam; model and normal groups administered 5 mL physiological saline.Anxiety was established in all groups, except the normal group, through the use of elevated plus-maze test at 7 days following drug administration.MAIN OUTCOME MEASURES:Blood β-endorphin and corticosterone levels were determined using enzyme-linked immunosorbent assay following treatment with Valeriana jatamansi Jones extract.Expressions of HPA axis-related genes were measured by cDNA microarray.RESULTS:Blood β-endorphin and corticosterone levels were significantly greater in the model group than in the normal group.Compared with the model group, levels decreased with Valeriana jatamansi Jones extract or estazolam treatment, particularly in the low-dose Valeriana jatamansi Jones group (P<0.01).cDNA microarray results demonstrated that corticotropin-releasing hormone and Orexin, which are associated with HPA axis function, were differentially expressed; expression increased in the model group, but decreased in rats treated with Valeriana jatamansi Jones extract.CONCLUSION:Valeriana jatamansi Jones extract plays a role in regulating HPA axis function in a rat model of anxiety, and this effect was superior to estazolam. | Zhiyong Yan Tian'e Zhang Ting Xiao Lingzhen Pan Jinzhi Qin Zhanping Zhang Changying Zuo | 2010 | Neural Regeneration Research2010,5,14: | 9 |
| 2 | Plant growth,antioxidative enzyme,and cadmium tolerance responses to cadmium stress in Canna orchioides显示文摘The cadmium tolerance capacity of Canna orchioides was evaluated using glasshouse experiment to lay a theoretical foundation to broad applications of this species in cadmium(Cd)polluted areas.The plant growth,Cd absorption,and antioxidative enzyme activities of canna plants treated with different Cd concentrations(0,100,and 200mg·kg^(-1)per dry soil)for 30 days were investigated.The results indicated that the growth of roots,leaves and stems was restricted following Cd addition.The bio-concentration factor was the highest in the roots,followed by the leaves,rhizomes,and stems.Compared with the control,the translocation factor decreased sharply after 100mg·kg^(-1)or 200mg·kg^(-1)Cd treatment,whereas the tolerance index decreased significantly under 200mg·kg^(-1)Cd exposure.The Cd accumulation rate and total Cd accumulation significantly increased under 200mg·kg^(-1)Cd treatment than the control.Themalondialdehyde content remained unchanged at different Cd concentrations,while the relative electrical conductivity of the root increased significantly following 200mg·kg^(-1)Cd treatment.Cd stress increased significantly the activity of superoxide dismutase(SOD),peroxidase(POD),and ascorbate peroxidase(APX)in the leaves,whereas only SOD was raised in the roots.The catalase(CAT)and POD activity in the roots did not differ significantly at different Cd concentrations.Conversely,APX in the roots decreased significantly under Cd stress,which suggests that the protective mechanisms in the leaves differ from those in the roots.These results indicated that C.orchioides has a high tolerance for Cd.Furthermore,the reactive oxygen species caused by Cd stress could be effectively scavenged by the antioxidative system.The present study proved that C.orchioides constitutes a promising ornamental plant for use in the restoration of areas contaminated with Cd. | Wen’e Zhang Xuejun Pan Qian Zhao Ting Zhao | 2021 | Horticultural Plant Journal2021,7,3: | 3 |

| 4 | Rapid phenotypic evolution with shallow genomic differentiation during early stages of high elevation adaptation in Eurasian Tree Sparrows显示文摘Known as the‘third polar region’,the Qinghai-Tibet Plateau represents one of the harshest highland environments in the world and yet a number of organisms thrive there.Previous studies of birds,animals and humans have focused on well-differentiated populations in later stages of phenotypic divergence.The adaptive processes during the initial phase of highland adaptation remain poorly understood.We studied a human commensal,the Eurasian Tree Sparrow,which has followed human agriculture to the Qinghai-Tibet Plateau.Despite strong phenotypic differentiation at multiple levels,in particular in muscle-related phenotypes,highland and lowland populations show shallow genomic divergence and the colonization event occurred within the past few thousand years.In a one-month acclimation experiment investigating phenotypic plasticity,we exposed adult lowland tree sparrows to a hypoxic environment and did not observe muscle changes.Through population genetic analyses,we identified a signature of polygenic adaptation,whereby shifts in allele frequencies are spread across multiple loci,many of which are associated with muscle-related processes.Our results reveal a case of positive selection in which polygenic adaptation appears to drive rapid phenotypic evolution,shedding light on early stages of adaptive evolution to a novel environment. | Yanhua Qu Chunhai Chen Ying Xiong Huishang She Yong E Zhang Yalin Cheng Shane DuBay Dongming Li Per G P Ericson Yan Hao Hongyuan Wang Hongfeng Zhao Gang Song Hailin Zhang Ting Yang Chi Zhang Liping Liang Tianyu Wu Jinyang Zhao Qiang Gao Weiwei Zhai Fumin Lei | 2020 | National Science Review2020,7,1: | 2 |
