1Interaction of hepatitis C virus envelope glycoprotein E2 with the large extracellular loop of tupaia CD81显示文摘AIM: To further analyze the interaction of tupaia CD81 with hepatitis C virus (HCV) envelope protein E2. METHODS: A tupaia CD81 large extracellular loop (CD81 LEL), which binds to HCV E2 protein, was cloned and expressed as a GST-fusion protein, and interaction of HCV E2 protein with a tupaia CD81 LEL was evaluated by enzyme-linked immunosorbent assay (EIA). RESULTS: Although tupaia and human CD81 LEL differed in 6 amino acid changes, tupaia CD81 LEL was strongly recognized by anti-CD81 antibodies against human CD81 LEL conformation-dependent epitopes. Investigating LEL CD81-E2 interactions by EIA, we demonstrated that binding of tupaia CD81 LEL GST fusion protein to recombinant HCV E2 protein was markedly reduced compared to binding of human CD81 LEL GST fusion protein to recombinant HCV E2 protein. CONCLUSION: These data suggest that the structural differences in-between the tupaia and human CD81 may alter the interaction of the large extracellular loop with HCV envelope glycoprotein E2. 2Hepatitis C virus infection and apoptosis显示文摘Apoptosis is central for the control and elimination of viral infections. In chronic hepatitis C virus (HCV) infection, enhanced hepatocyte apoptosis and upregulation of the death inducing ligands CD95/Fas, TRAIL and TNFα occur. Nevertheless, HCV infection persists in the majority of patients. The impact of apoptosis in chronic HCV infection is not well understood. It may be harmful by triggering liver fibrosis, or essential in interferon (IFN) induced HCV elimination. For virtually all HCV proteins, pro- and anti-apoptotic effects have been described, especially for the core and NS5A protein. To date, it is not known which HCV protein affects apoptosis in vivo and whether the infectious virions act pro- or anti- apoptotic. With the availability of an infectious tissue culture system, we now can address pathophysiologically relevant issues. 3建立广州生物库进行慢性重大疾病研究概述显示文摘目的报告《建立广州生物库进行慢性重大疾病研究》过去十年的研究进展和今后十年的研究展望。方法选择≥50岁的30 518名有广州市户籍的居民,收集其个人背景资料、职业接触、居住和生活环境与习惯、营养摄入及个人与家族史等详细资料,并作体格检查,血生化检验、尿柯铁宁及心电图、X线胸片与肺功能检测等,同时储存其白细胞与血浆在-196℃液态氮库内使其永生化;定期重复体检随访及电话随访并建立亚队列进行基因、生活方式与职业环境等因素对慢性重大疾病影响的研究。计数资料采用χ2检验,计量资料符合正态分布的采用t检验,不符合正态分布的采用秩和检验,以双侧P<0.05为差异有统计学意义。结果 (1)建立液态氮生物库、条码识别技术与Fico11-Pague分离细胞室及分子流行病学研究室的管理系统;(2)分三个阶段收集30 518名研究对象建立生物库中老年队列;(3)基线资料分析表明,'三高'凸显,男女血压高值分别为24.0%和17.8%,高血压患病率分别为45.3%和41.3%,χ2值为39.89,P<0.001;血糖高值和糖尿病患者男性分别为11.1%和12.0%,略高于女性的9.9%和12.6%,χ2值分别为9.00和2.04,P=0.191和0.293,差异无统计学意义;而高脂血症更为明显,检出率则女性明显高于男性;(4)中央型肥胖、超重和全身性肥胖以及代谢综合征也呈现女性高于男性;短期记忆受损则男性为13.9%,稍高于女性的12.7%,χ2值为8.05,P=0.005;(5)发现不良的生活方式如吸烟、二手烟、饮酒、缺乏体力活动、不良睡眠状态及打鼾对健康的影响,经历不同社会经济环境(生命历程)成长发育对健康的影响,研究已发表论文80多篇;(6)第一次重复体检和电话随访已于2012年12月结束,除606名研究对象因户口迁出或身份证有误无法确定外,成功随访率达98.0%,已知1 801例死亡,随访资料仍在处理和分析中。结论既往十年《广州生物库中老年队列研究》项目已建立标准化的研究平台及完成第一次重复体检与电话随访,未来十年将继续重复体检为主的纵向随访及建立亚队列进行基因、职业与环境因素及生活方式对慢性重大疾病影响的研究,不断向深度和广度发展。江朝强 林大庆 郑家强 刘斌 张维森 靳雅丽 徐琳 Kin Bong Hubert Lam Graham Neil Thomas Peymane Adap 朱彤 梁卓伟 2013中国预防医学杂志2013,14,11:6

8Pseudomonas exotoxin antisense RNA selectively kills hepatitis B virus infected cells显示文摘AIM: To present an approach for selectively killing retrovirus-infected cells that combines the toxicity of Pseudomonas exotoxin (PE) and the presence of reverse transcriptase (RT) in infected cells. METHODS: PE antisense toxin RNA has palindromic stem loops at its 5' and 3' ends enabling self-primed generation of cDNA in the presence of RT. The RT activity expressed in retrovirus-infected cells converts 'antisense-toxin-RNA' into a lethal toxin gene exclusively in these cells. RESULTS: Using cotransfection studies with PE-expressing RNAs and β-gal expressing reporter plasmids, we show that, in HepG2 and HepG2.2.15 hepatoma cells as well as in duck hepatitis B virus (DHBV) infected cells, HBV or DHBV-polymerase reverse transcribe a lethal cDNA copy of an antisense toxin RNA, which is composed of sequences complementary to a PE gene and eukaryotic transcription and translation signals. 12Virus-host Interactions during Hepatitis C Virus Entry-Implications for Pathogenesis and Novel Treatment Approaches显示文摘Hepatitis C virus (HCV) is a member of the Flaviviridae family and causes acute and chronic hepatitis. Chronic HCV infection may result in severe liver damage including liver cirrhosis and hepatocellular carcinoma. The liver is the primary target organ of HCV, and the hepatocyte is its primary target cell. Attachment of the virus to the cell surface followed by viral entry is the first step in a cascade of interactions between the virus and the target cell that is required for successful entry into the cell and initiation of infection. This step is an important determinant of tissue tropism and pathogenesis; it thus represents a major target for antiviral host cell responses, such as antibody-mediated virus neutralization. Following the development of novel cell culture models for HCV infection our understanding of the HCV entry process and mechanisms of virus neutralization has been markedly advanced. In this review we summarize recent developments in the molecular biology of viral entry and its impact on pathogenesis of HCV infection, development of novel preventive and therapeutic antiviral strategies.Joachim Lupberger Mirjam B.
