| 1 | Functional analysis of the larval serum protein gene promoter from silkworm,Bombyx mori.显示文摘The regulation region of larval serum protein gene, Bombyx mori. (BmLSP), consisting of the first intron, the first exon, the central promoter region and 5′-upstream region, is cloned from genomic DNA from the silkworm va-riety of Suju譓inghu. Using PCR and restriction endonu-clease methods, a series of luciferase reporter plasmids, driven by different length of BmLSP promoters, are con-structed. Via the transient expression system in BmN cells, the effects of the regulation elements and foreign insect hor-mones on the BmLSP promoter activity are investigated. The results demonstrate that the promoter activity of BmLSP is 5.8- or 4.4-fold higher than that of BmLSPs whose first in-tron or the element in 5′-upstream region harboring the homologous sequence with the first intron of light-chain fib-roin gene (EHIF) is deleted, respectively, suggesting that both the first intron and EHIF contain the main positive cis-acting elements. However, the inactive mariner transposable ele-ment (MTE) in 5′-upstream region presents a negative effect. Furthermore, the effects of juvenile hormone analogue (JHA) on the BmLSP promoter activity show a typical dose-dependent manner, that is, low concentration treat-ments increase the BmLSP promoter activity and high con-centration treatments decrease it. | 2 | Interface and properties of epoxy resin modified by elastomeric nano-particles显示文摘Study on a new composite of epoxy resin/elastomeric nano-particles (ENP) is re- ported in this paper, which shows that, in comparison with pure epoxy resin and epoxy toughened with CTBN, the composites of epoxy resin/carboxylic nitrile-butadiene ENP and epoxy resin/styrene butadiene vinyl-pyridine ENP possess both higher toughness and heat resistance. Both ENPs used in the study have an average size of less than 100 nm. | 3 | Interaction between en- dogenous nitric oxide and carbon monoxide in the pathogenesis of hypoxic pulmonary hypertension显示文摘The aim of the study was to investigate the in-teraction between nitric oxygenase (NOS)/ nitric oxide (NO) and heme oxygenase (HO)/ carbon monoxide (CO) system in the pathogenesis of hypoxic pulmonary hypertension. On a rat model of hypoxic pulmonary hypertension, the pulmo-nary artery pressure was measured, and NO formation and expression of NOS in pulmonary tissues were examined after treatment with ZnPP-IX, an HO-1 inhibitor. The pulmonary artery pressure, CO formation and expression of HO-1 in pulmonary tissues were examined after treatment with L-NAME, a NOS inhibitor. We found that pulmonary hy-pertension developed after 2-week hypoxia, while the con-centration of NO in the pulmonary tissue homogenates and the expression of NOS in intrapulmonary artery endothelial cells decreased markedly. ZnPP-IX worsened pulmonary hypertension of hypoxic rats. However, it increased endoge-nous production of NO and the expression of NOS obviously. The concentration of CO in the pulmonary tissue homoge-nates and the expression of HO-1 in intrapulmonary artery smooth muscle cells increased markedly with hypoxic pul-monary hypertension. L-NAME worsened pulmonary hy-pertension of hypoxic rats, but inhibited CO formation and HO-1 expression (P < 0.01). | 4 | Physiological responses of osteoblasts to cyclic stretching and the change of intracellular calcium concentration显示文摘The development of bone tissues is regulated by mechanical stimulation. Cyclic stretching was applied to the osteoblasts that were delivered from rat calvarie. The results showed that stretching at 500 me increased the cell prolifera-tion while loading at 1000 me and 1500 me inhabited cell growth. Loading also increased the adhesive force between cells and substrate as well as spreading areas of osteobalsts. Furthermore, the fluorescence probe Fluo-3/AM was used to investigate the effect of stretching stimulation on the intra-cellular calcium concentration of osteoblasts. The intracellu-lar calcium concentration of osteoblasts that were stretched at 500 me for 5 min was 92.9% higher than the control. After being treated with the panax ontoginseng saponins, the stretched osteoblasts still expressed 28.6% higher intracellu-lar calcium concentration than that of the control, which proved that both the influx of extracellular calcium and the release of intracellular calcium store were involved in the increase of intracellular calcium concentration when os-teoblasts responded to the cyclic stretching. | 5 | The complete mitochondrial genomes sequences of Asio flammeus and Asio otus and comparative analysis显示文摘The complete mitochondrial genomes of Asio flammeus and Asio otus were se- quenced and found to span 18858 bp and 18493 bp, respectively. It is surprising to find the for- mer to be the largest among all avian mitochondrial genomes sequenced so far. The two ge- nomes have very similar gene order with that of Gallus gallus, neither contains the pseudo con- trol region, but both have a single extra base, namely Cytidine, at position 174 in ND3 gene. The control regions of Asio flammeus and Asio otus’ mitochondrial genomes span 3288 bp and 2926 bp respectively, which are the longest among vertebrates except for Myxine glutinosa and con- tribute to the large size of two genomes. The 3′end of the control region of Asio flammeus and Asio otus contains many tandemly repeated sequences, which are highly similar to a putative control element, i.e. Mt5, and may form stable stem-loop secondary structures. Such repeated sequences probably play an important role in regulating transcription and replication of mito- chondrial genome. The approach relies on ri-bozyme substrates modified at the two arms, with a fluores-cent moiety attached to the end of one arm and a non-fluorescent quenching moiety attached to the end of the other arm. MBR is employed to directly convert the cleavage information into fluorescence signal in real time. Compared with traditional approach, this method provides a no-radiolabeling, sensitive and effective way to research on the ribozyme activity, enzymatic dynamic process and ri-bozyme function during gene therapy. | 11 | Studies on the Coordination Polymer of Copper El ectropol yurushiol显示文摘The coordination polymer of copper electropolyurushiol(EPU-Cu2+) was obtained by the reaction of copper chloride with electropolyurushiol (EPU) in an isopropyl alcohol solution. The properties of EPU-Cu2+ were studied by means of ESR, FTIR, XPS, DMTA and TG-DTA.The results of AES show that the content of Cu2+ was 8.63%. The electric resistances of EPUCu2+ and EPU were determined to be 9.37×1010Ω and 7.90×1010Ω respectively. It was also ascertained that in EPU-Cu2+ each Cu2+ is coordinated with two units hydroxyl in EPU, making the EPU-Cu2+ cross-link further. | 12 | Imaging theory of nonlinear second harmonic and third harmonic generations in confocal microscopy显示文摘The imaging theory of nonlinear second harmonic generation (SHG) and third harmonic generation (THG) in confocal microscopy is presented in this paper. The nonlinear effect of SHG and THG on the imaging properties of confocal microscopy has been analyzed in detail by the imaging theory. It is proved that the imaging process of SHG and THG in confocal microscopy, which is different from conventional coherent imaging or incoherent imaging, can be divided into two different processes of coherent imaging. The three-dimensional point spread functions (3D-PSF) of SHG and THG confocal microscopy are derived based on the nonlinear principles of SHG and THG. The imaging properties of SHG and THG confocal microscopy are discussed in detail according to its 3D-PSF. There are two very strong peaks of UV-Vis spectrum in the range of 400-600 nm, which are similar to those of Mn catalase and Mn ribonuleotide reductase extracted from organisms. C2H2, C2H4, and C2H6 were irradiated by 800 nm, 100 fs laser pulses with the intensity range of 1013—1014 W/cm2. A TOF-mass spectrometer was coupled to the laser system and used to experimentally in-vestigate the field ionization of these molecules. The experi-mental ionization threshold intensities are obtained. Results show that conformational transitions from globular micelles→cylindrical micelles→flat films are observed in the process of the adsorption of CTAB to mica at twice the bulk critical concentration. However, in 0.5 cmc CTAB solution, the adsorbed CTAB molecules to mica form the layered film structures, and apparent conformational transitions do not appear.
