1Age-related rhesus macaque models of COVID-19显示文摘Background:Since December 2019,an outbreak of the Corona Virus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)in Wuhan,China,has become a public health emergency of international concern.The high fatality of aged cases caused by SARS-CoV-2 was a need to explore the possible age-related phenomena with non-human primate models.Methods:Three 3-5 years old and two 15 years old rhesus macaques were intratracheally infected with SARS-CoV-2,and then analyzed by clinical signs,viral replication,chest X-ray,histopathological changes and immune response.Results:Viral replication of nasopharyngeal swabs,anal swabs and lung in old monkeys was more active than that in young monkeys for 14 days after SARS-CoV-2 challenge.Monkeys developed typical interstitial pneumonia characterized by thickened alveolar septum accompanied with inflammation and edema,notably,old monkeys exhibited diffuse severe interstitial pneumonia.Viral antigens were detected mainly in alveolar epithelial cells and macrophages.Conclusion:SARS-CoV-2 caused more severe interstitial pneumonia in old monkeys than that in young monkeys.Rhesus macaque models infected with SARS-CoV-2 provided insight into the pathogenic mechanism and facilitated the development of vaccines and therapeutics against SARS-CoV-2 infection.Pin Yu Feifei Qi Yanfeng Xu Fengdi Li Peipei Liu Jiayi Liu Linlin Bao Wei Deng Hong Gao Zhiguang Xiang Chong Xiao Qi Lv Shuran Gong Jiangning Liu Zhiqi Song Yajin Qu Jing Xue Qiang Wei Mingya Liu Guanpeng Wang Shunyi Wang Haisheng Yu Xing Liu Baoying Huang Wenling Wang Li Zhao Huijuan Wang Fei Ye Weimin Zhou Wei Zhen Jun Han Guizhen Wu Qi Jin Jianwei Wang Wenjie Tan Chuan Qin 2020Animal Models and Experimental Medicine2020,3,1:22

3Identification and fine mapping of quantitative trait loci for seed vigor in germination and seedling establishment in rice显示文摘Seed vigor is an index of seed quality that is used to describe the rapid and uniform germination and the establishment of strong seedlings in any environmental conditions.Strong seed vigor in low-temperature germination conditions is particularly important in direct-sowing rice production systems. However, seed vigor has not been selected as an important breeding trait in traditional breeding programs due to its quantitative inherence. In this study, we identified and mapped eight quantitative trait loci(QTLs) for seed vigor by using a recombinant inbred population from a cross between rice(Oryza sativa L. ssp. indica) cultivars ZS97 and MH63.Conditional QTL analysis identified qSV-1, qSV-5b, qSV-6a, qSV-6b, and qSV-11 influenced seedling establishment and that qSV-5a, qSV-5c, and qSV-8 influenced only germination. Of these,qSV-1, qSV-5b, qSV-6a, qSV-6b, and qSV-8 were low-temperature-specific QTLs. Two major-effective QTLs, qSV-1, and qSV-5c were narrowed down to 1.13-Mbp and 400-kbp genomic regions, respectively. 4Facile metagrating holograms with broadband and extreme angle tolerance显示文摘The emerging meta-holograms rely on arrays of intractable meta-atoms with various geometries and sizes for customized phase profiles that can precisely modulate the phase of a wavefront at an optimal incident angle for given wavelengths.The stringent and band-limited angle tolerance remains a fundamental obstacle for their practical application,in addition to high fabrication precision demands.Utilizing a different design principle,we determined that facile metagrating holograms based on extraordinary optical diffraction can allow the molding of arbitrary wavefronts with extreme angle tolerances(near-grazing incidence)in the visible–near-infrared regime.By modulating the displacements between uniformly sized meta-atoms rather than the geometrical parameters,the metagratings produce a robust detour phase profile that is irrespective of the wavelength or incident angle.The demonstration of high-fidelity meta-holograms and in-site polarization multiplexing significantly simplifies the metasurface design and lowers the fabrication demand,thereby opening new routes for flat optics with high performances and improved practicality.Zi-Lan Deng Junhong Deng Xin Zhuang Shuai Wang Tan Shi Guo Ping Wang Yao Wang Jian Xu Yaoyu Cao Xiaolei Wang Xing Cheng Guixin Li Xiangping Li 2018Light(Science & Applications)2018,7,1:17
5Surgical treatment for both-column acetabular fractures using pre-operative virtual simulation and three-dimensional printing techniques显示文摘Background:Surgical treatment of both-column acetabular fractures is challenging because of the complex acetabular fracture patterns and the curved surface of the acetabulum.Seldom study has compared the application of three-dimensional(3D)printing technology and traditional methods of contouring plates intra-operatively for the surgical treatment of both-column acetabular fractures.We presented the use of both 3D printing technology and a virtual simulation in pre-operative planning for both-column acetabular fractures.We hypothesized that 3D printing technology will assist orthopedic surgeons in shortening the surgical time and improving the clinical outcomes.Methods:Forty patients with both-column acetabular fractures were recruited in the randomized prospective case-control study from September 2013 to September 2017 for this prospective study(No.ChiCTR1900028230).We allocated the patients to two groups using block randomization(3D printing group,n=20;conventional method group,n=20).For the 3D printing group,1:1 scaled pelvic models were created using 3D printing,and the plates were pre-contoured according to the pelvic models.The plates for the conventional method group were contoured during the operation without 3D printed pelvic models.The operation time,instrumentation time,time of intra-operative fluoroscopy,blood loss,number of times the approach was performed,blood transfusion,post-operative fracture reduction quality,hip joint function,and complications were recorded and compared between the two groups.Results:The operation and instrumentation times in the 3D printing group were significantly shorter(130.8±29.2 min,t=-7.5,P<0.001 and 32.1±9.5 min,t=-6.5,P<0.001,respectively)than those in the conventional method group.The amount of blood loss and blood transfusion in the 3D printing group were significantly lower(500[400,800]mL,Mann-Whitney U=74.5,P<0.001 and 0[0,400]mL,Mann-Whitney U=59.5,P<0.001,respectively)than those in the conventional method group.The number of the approach performed in the 3D printing group was significantly smaller than that in the conventional method group(pararectus+Kocher-Langenbeck[K-L]approach rate:35%vs.85%;x2=10.4,P<0.05).The time of intra-operative fluoroscopy in the 3D printing group was significantly shorter than that in the conventional method group(4.2±1.8 vs.7.7±2.6 s;t=-5.0,P<0.001).The post-operative fracture reduction quality in the 3D printing group was significantly better than that in the conventional method group(good reduction rate:80%vs.30%;x2=10.1,P<0.05).The hip joint function(based on the Harris score 1 year after the operation)in the 3D printing group was significantly better than that in the conventional method group(excellent/good rate:75%vs.30%;χ^2=8.1,P<0.05).The complication was similar in both groups(5.0%vs.25%;χ^2=3.1,P=0.182).Conclusions:The use of a pre-operative virtual simulation and 3D printing technology is a more effective method for treating bothcolumn acetabular fractures.This method can shorten the operation and instrumentation times,reduce blood loss,blood transfusion and the time of intra-operative fluoroscopy,and improve the post-operative fracture reduction quality.Ji-Hui Huang Hui Liao Xin-Yu Tan Wei-Rong Xing Qi Zhou Yu-Shi Zheng Hong-Yu Cao Can-Jun Zeng 2020Chinese Medical Journal2020,,4:15
6Topographic differentiations of biological soil crusts and hydraulic properties in fixed sand dunes, Tengger Desert显示文摘Biological soil crusts(BSCs) play an important role in surface soil hydrology. Soils dominated with moss BSCs may have higher infiltration rates than those dominated with cyanobacteria or algal BSCs. However, it is unnown whether improved infiltration in moss BSCs is accompanied by an increase in soil hydraulic conductivity or water retention capacity. We investigated this question in the Tengger Desert, where a 43-year-old revegetation program has promoted the formation of two distinct types of BSCs along topographic positions, i.e. the moss-dominated BSCs on the interdune land and windward slopes of the fixed sand dunes, and the algal-dominated BSCs on the crest and leeward slopes. Soil water retention capacity and hydraulic conductivity were measured using an indoor evaporation method and a field infiltration method. And the results were fitted to the van Genuchten–Mualem model. Unsaturated hydraulic conductivities under greater water pressure(<–0.01 MPa) and water retention capacities in the entire pressure head range were higher for both crust types than for bare sand. However, saturated and unsaturated hydraulic conductivities in the near-saturation range(>–0.01 MPa) showed decreasing trends from bare sand to moss crusts and to algal crusts. 7Management of granulomatous lobular mastitis: an international multidisciplinary consensus(2021 edition)显示文摘Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in the management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. This consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using PubMed from January 1, 1971 to July 31, 2020. Sixty-six international experienced multidisciplinary experts from 11 countries or regions were invited to review the evidence.Levels of evidence were determined using the American College of Physicians grading system, and recommendations were discussed until consensus. Experts discussed and concluded 30 recommendations on historical definitions,etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted definition. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combination of history, clinical manifestations, imaging examinations, laboratory examinations and pathology.The approach to treatment of GLM should be applied according to the different clinical stage of GLM. 8First observation of tropospheric nitrogen dioxide from the Environmental Trace Gases Monitoring Instrument onboard the GaoFen-5 satellite显示文摘The Environmental Trace Gases Monitoring Instrument(EMI)is the first Chinese satellite-borne UV–Vis spectrometer aiming to measure the distribution of atmospheric trace gases on a global scale.The EMI instrument onboard the GaoFen-5 satellite was launched on 9 May 2018.In this paper,we present the tropospheric nitrogen dioxide(NO2)vertical column density(VCD)retrieval algorithm dedicated to EMI measurement.We report the first successful retrieval of tropospheric NO_(2) VCD from the EMI instrument.Our retrieval improved the original EMI NO_(2) prototype algorithm by modifying the settings of the spectral fit and air mass factor calculations to account for the on-orbit instrumental performance changes.The retrieved EMI NO_(2) VCDs generally show good spatiotemporal agreement with the satellite-borne Ozone Monitoring Instrument and TROPOspheric Monitoring Instrument(correlation coefficient R of ~0.9,bias<50%).A comparison with ground-based MAX-DOAS(Multi-Axis Differential Optical Absorption Spectroscopy)observations also shows good correlation with an R of 0.82.The results indicate that the EMI NO_(2) retrieval algorithm derives reliable and precise results,and this algorithm can feasibly produce stable operational products that can contribute to global air pollution monitoring.Chengxin Zhang Cheng Liu Ka Lok Chan Qihou Hu Haoran Liu Bo Li Chengzhi Xing Wei Tan Haijin Zhou Fuqi Si Jianguo Liu 2020Light(Science & Applications)2020,9,1:11
9Artificial intelligence in drug design显示文摘Thanks to the fast improvement of the computing power and the rapid development of the computational chemistry and biology,the computer-aided drug design techniques have been successfully applied in almost every stage of the drug discovery and development pipeline to speed up the process of research and reduce the cost and risk related to preclinical and clinical trials.Owing to the development of machine learning theory and the accumulation of pharmacological data, the artificial intelligence(AI) technology, as a powerful data mining tool, has cut a figure in various fields of the drug design, such as virtual screening,activity scoring, quantitative structure-activity relationship(QSAR) analysis, de novo drug design, and in silico evaluation of absorption, distribution, metabolism, excretion and toxicity(ADME/T) properties. Although it is still challenging to provide a physical explanation of the AI-based models, it indeed has been acting as a great power to help manipulating the drug discovery through the versatile frameworks. 10Polymorphisms of MTHFD,plasma homocysteine levels, and risk of gastric cancer in ahigh-risk Chinese population显示文摘Purpose:Accumulative evidence suggests that folate has a protective effect on gastric cancer. The methylenetetrahyd-rofolate dlehydrogenase(MTHFD) plays an important role in folate and homocysteine metabolisms, and polymorphisms of MTHFD may result in disturbance of the folate-mediated homocysteine pathway. The aim of this study is to test the hypothesis that genetic variants of MTHFD and plasma homocysteine levels are associated with risk of gastric cancer and modulated by genotypes of methylenetetrahydrofolate reductase(MTHFR). Experimental Design: We genotyped G1958A and T401C in MTHFD and C677T in MTHFR and detected total plasma homocysteine(tHcy) levels in a case-control study of 589 gastric cancer cases and 635 cancer-free controls in a high-risk Chinese population. Results:The variant genotypes of MTHFD 1958AA and 401CC were associated with a significantly increased risk of gastric canceradjusted odds ratio(OR), 2.05; 95% confidence interval(95% CI),1.34-3.13 for 1958AA; adjusted OR,1.43; 95% CI,1.14-1.80 for 401CC compared with 1958GG/GA and 401TT/TC genotypes, respectively. Both of the effects were more evident in the subjects carrying MTHFR 677CT/TT genotypes. The average tHcy level was significantly higher in gastric cancer cases than in controls(P < 0.01), and the upper quartile of tHcy (> 13.6 mu mol/L) was associated with an 82% significantly increased risk of gastric cancer, compared with the lowest quartile of tHcy(<= 8.0 pmol/L;adjusted OR,1.82; 95% CI,1.20-2.75). Conclusions:The strong associations between MTHFD variants and the plasma tHcy levels and gastric cancer risk suggest, for the first time, a possible gene-environment interaction between genetic variants of folate-metabolizing genes and high tHcy levels in gastric carcinogenesis.Wang, L. 12TP53-induced glycolysis and apoptosis regulator alleviates hypoxia/ischemia-induced microglial pyroptosis and ischemic brain damage显示文摘Our previous studies have demonstrated that TP53-induced glycolysis and apoptosis regulator(TIGAR)can protect neurons after cerebral ischemia/reperfusion.However,the role of TIGAR in neonatal hypoxic-ischemic brain damage(HIBD)remains unknown.In the present study,7-day-old Sprague-Dawley rat models of HIBD were established by permanent occlusion of the left common carotid artery followed by 2-hour hypoxia.At 6 days before induction of HIBD,a lentiviral vector containing short hairpin RNA of either TIGAR or gasdermin D(LV-sh_TIGAR or LV-sh_GSDMD)was injected into the left lateral ventricle and striatum.Highly aggressively proliferating immortalized(HAPI)microglial cell models of in vitro HIBD were established by 2-hour oxygen/glucose deprivation followed by 24-hour reoxygenation.Three days before in vitro HIBD induction,HAPI microglial cells were transfected with LV-sh_TIGAR or LV-sh_GSDMD.Our results showed that TIGAR expression was increased in the neonatal rat cortex after HIBD and in HAPI microglial cells after oxygen/glucose deprivation/reoxygenation.Lentivirusmediated TIGAR knockdown in rats markedly worsened pyroptosis and brain damage after hypoxia/ischemia in vivo and in vitro.Application of exogenous nicotinamide adenine dinucleotide phosphate(NADPH)increased the NADPH level and the glutathione/oxidized glutathione ratio and decreased reactive oxygen species levels in HAPI microglial cells after oxygen/glucose deprivation/reoxygenation.Additionally,exogenous NADPH blocked the effects of TIGAR knockdown in neonatal HIBD in vivo and in vitro.These findings show that TIGAR can inhibit microglial pyroptosis and play a protective role in neonatal HIBD.The study was approved by the Animal Ethics Committee of Soochow University of China(approval No.2017LW003)in 2017.Lan-Lan Tan Xiao-Lu Jiang Li-Xiao Xu Gen Li Chen-Xi Feng Xin Ding Bin Sun Zheng-Hong Qin Zu-Bin Zhang Xing Feng Mei Li 2021Neural Regeneration Research2021,16,6:9
13A telomere-to-telomere gap-free reference genome of watermelon and its mutation library provide important resources for gene discovery and breeding显示文摘Watermelon,Citrullus lanatus,is the world's third largest fruit crop.Reference genomes with gaps and a narrow genetic base hinder functional genomics and genetic improvement of watermelon.Here,we report the assembly of a telomere-to-telomere gap-free genome of the elite watermelon inbred line G42 by incorporating high-coverage and accurate long-read sequencing data with multiple assembly strategies.All 11 chromosomes have been assembled into single-contig pseudomolecules without gaps,representing the highest completeness and assembly quality to date.The G42 reference genome is 369321829 bp in length and contains 24205 predicted protein-coding genes,with all 22 telomeres and 11 centromeres characterized.Furthermore,we established a pollen-EMS mutagenesis protocol and obtained over 200000M1 seeds from G42.In a sampling pool,48 monogenic phenotypic mutations,selected from 223M1and 78 M2 mutants with morphological changes,were confirmed.The average mutation density was 1 SNP/1.69Mband1 indel/4.55 Mb per M1 plant and 1SNP/1.08Mb and 1 indel/6.25 Mb per M2 plant.Taking advantage of the gap-free G42 genome,8039 mutations from 32 plants sampled from M1 and M2 families were identified with 100%accuracy,whereas only 25% of the randomly selected mutations identified using the 97103v2 reference genome could be confirmed.Using this library and the gap-free genome,two genes responsible for elongated fruit shape and male sterility(CiMs1)were identified,both caused by a single basechange from G to A.The validated gap-free genome and its EMS mutation library provide invaluable resources for functional genomics and genetic improvement of watermelon.Yun Deng Shoucheng Liu Yilin Zhang Jingsheng Tan Xiaopeng Li Xiao Chu Binghua Xu Yao Tian Yudong Sun Bosheng Li YunbiXu Xing Wang Deng Hang He Xingping Zhang 2022Molecular Plant2022,15,8:9
14A New Titanosaurian Sauropod from Late Cretaceous of Nei Mongol, China显示文摘A specimen collected from the Upper Cretaceous Erlian Formation of Nei Mongol (Inner Mongolia), China, represents a new genus and species of titanosaurian sauropod. The new taxon is named and described on the basis of the holotype and the only known specimen, which comprises several dorsal, sacral, and caudal vertebrae, several dorsal ribs, one anterior chevron, and much of the pelvis. Diagnostic features of the new species include a thick ridge extending down from the postzygapophysis on the lateral surface of the neural arch of the posterior dorsal vertebrae, a transversely oriented accessory lamina present between the anterior centroparapophyseal limina and the lateral centraprezygapophyseal lamina of the posterior dorsal vertebrae, long, anteroventrally directed caudal rib that bears two distinctive fossae on its posterior margin on the anterior caudal vertebrae and a prominent vertical ridge above the pubic peduncle on the medial surface of the ilium, among others. The ilium is pneumatic, a feature not common among non-avian dinosaurs. The new taxon has an unusual combination of primitive and derived character states. Preliminary character analysis shows a complex character distribution within the Titanosauriformes. 15Evaluation of the mold-filling ability of alloy melt in squeeze casting显示文摘The mold-filling ability of alloy melt in squeeze casting process was evaluated by means of the maximum length of Archimedes spiral line. A theoretical evaluating model to predict the maximum filling length was built based on the flowing theory of the incompressible viscous fluid. It was proved by experiments and calculations that the mold-filling pressure and velocity are prominent influencing factors on the mold-filling ability of alloy melt. The mold-filling ability increases with the increase of the mold-filling pressure and the decrease of the proper mold-filling velocity. Moreover, the pouring temperature relatively has less effect on the mold-filling ability under the experimental conditions. The maximum deviation of theoretical calculating values with experimental results is less than 15%. In this review,we update the studies on the BiCuSeO thin films first.Then, we focus on the most recent progress of multiple approaches that enhance the thermoelectric performance including advanced synthesized technologies,notable mechanisms for higher power factor(optimizing carrier concentration, carrier mobility, Seebeck coefficient)and doping effects predicted by calculation. 17Application of Avaatech X-ray fluorescence core-scanning in Sr/Ca analysis of speleothems显示文摘Application of X-ray fluorescence core-scanning(XRF-CS) on both marine and lake sediments has achieved remarkable results. However, its application has not been widely extended to the research on speleothems. In this study, we measure the Sr abundance and the Sr/Ca ratios of three stalagmites(two aragonite stalagmites, one calcite stalagmite) using the state-of-the-art fourth-generation Avaatech high-resolution XRF core scanner. Through comparisons among different scan paths and among different scan resolutions, as well as comparisons with inductively coupled plasma optical emission spectrometer(ICP-OES), Itrax XRF, and Artax XRF results, we confirm that the Avaatech XRF core scanner could precisely, quickly, and nondestructively analyze the high-resolution Sr abundance of speleothems. 18Development of composite PLGA microspheres containing exenatide-encapsulated lecithin nanoparticles for sustained drug release显示文摘This study aimed to prepare poly(D, L-lactic-co-glycolic acid) microspheres(PLGA-Ms)by a modified solid-in-oil-in-water(S/O/W) multi-emulsion technique in order to achieve sustained release with reduced initial burst and maintain efficient drug concentration for a prolonged period of time. Composite PLGA microspheres containing exenatideencapsulated lecithin nanoparticles(Ex-NPs-PLGA-Ms) were obtained by initial fabrication of exenatide-loaded lecithin nanoparticles(Ex-NPs) via the alcohol injection method,followed by encapsulation of Ex-NPs into PLGA microspheres. Compared to Ms prepared by the conventional water-in-oil-in-water(W/O/W) technique(Ex-PLGA-Ms), Ex-NPs-PLGAMs showed a more uniform particle size distribution, reduced initial burst release, and sustained release for over 60 d in vitro. Cytotoxicity studies showed that Ms prepared by both techniques had superior biocompatibility without causing any detectable cytotoxicity.In pharmacokinetic studies, the effective drug concentration was maintained for over 30 d following a single subcutaneous injection of two types of Ms formulation in rats, potentially prolonging the therapeutic action of Ex. 19Residues of Organochlorine Pesticides(OCPs) in Agricultural Soils of Zhangzhou City,China显示文摘A soil survey was conducted in Zhangzhou City,an important agricultural region in south of the Fujian Province,China.93 surface soil samples were collected in the paddy fields,vegetable lands,orchards and tea plantations from Zhangzhou City.An additional soil profile was sampled in a paddy field as previous research had indicated high concentrations of organochlorine pesticides(OCPs) in the paddy fields.Dichlorodiphenyltrichloroethanes(DDTs) ranged from 0.64-78.07 ng g 1 dry weight and hexachlorocyclohexanes(HCHs) ranged from 0.72-30.16 ng g 1 dry weight in the surface soil of the whole study region.Ratios of α-HCH/γ-HCH < 4 and o,p-DDT/p,p-DDT > 1 in all soil samples suggested that lindane and dicofol were widely applied in this region in the past.Concentrations of HCHs and DDTs in soils from the four land use types followed the orders:paddy fields > vegetable lands > tea plantations > orchards and tea plantations > orchards > paddy fields > vegetable lands,respectively.Analyses of the data showed no correlation(r < 0.1) between elevation and OCPs contents in paddy fields,orchards and vegetable lands,indicated no significantly different features in distribution of HCHs and DDTs in the soils from low lying plains and mountains and the unsystematic usage of OCPs,and highlighted the fragmented nature of agricultural production in Zhangzhou,as well as the reemission of OCPs from the soils,where high OCPs concentrations were found,in Longhai of Zhangzhou.In addition,no obvious relationship between the OCPs and total organic carbon(TOC)(r < 0.3) was observed in the soil profile.The mean contribution of dicofol in total DDTs was 66% in the whole Zhangzhou region.The approximate burdens of HCHs and DDTs in the surface layer of 0-20 cm were 0.44 and 1.55 t,respectively.The storage of both HCHs and DDTs in soil surface layer(0-20 cm) accounts for 40% burden of the soil layer of 0-50 cm(1.10 t HCHs and 3.87 t DDTs),in which the highest concentrations of OCPs were observed in soil profile.YANG Dan QI Shi-Hua ZHANG Jia-Quan TAN Ling-Zhi ZHANG Jun-Peng ZHANG Yuan XU Feng XING Xin-Li HU Ying CHEN Wei YANG Jun-Hua XU Mei-Hui 2012Pedosphere2012,22,2:7
20High manure load reduces bacterial diversity and network complexity in a paddy soil under crop rotations显示文摘Long-term application of chemical fertilizers causes soil degradation and nitrogen(N)loss,but these effects could be alleviated by organic fertilizers.In addition,crop rotation is a feasible practice to increase soil fertility,soil quality and crop yields comparing with monocultural cropping patterns.However,questions remain concerning how the soil microbiome responds to different manure application rates under crop rotations.Here,we collected soil samples from a rice-rape system to investigate the response of the soil microbiome to nine years of pig manure application at different rates(CK:0 kg ha^(-1),M1:1930 kg ha^(-1),M2:3860 kg ha^(-1) and M3:5790 kg ha^(-1)).Our results revealed that the bacterialα-diversity(Chao1 and Shannon index)in the rape season increased first and then decreased with increasing manure application rates,and a high manure load tended to decrease the bacterialα-diversity in the rice season.Long-term manure application enriched some copiotrophic bacteria,such as Proteobacteria and Actinobacteria,while it decreased the relative abundance of Nitrospirae.Redundancy analysis(RDA)and the Mantel test indicated that soil pH,TC,TN,AP,C/P and N/P ratios were the main factors influencing bacterial communities.Moreover,network analysis showed that a low manure application rate shaped a complexly connected and stable bacterial community,while higher manure application rate decreased the stability of the bacterial network.These findings improve our understanding of bacterial responses to longterm manure application under crop rotations and their relationships with soil factors,especially in the context of increasing fertilizer inputs.Haiyang Liu Xing Huang Wenfeng Tan Hongjie Di Jianming Xu Yong Li 2020Soil Ecology Letters2020,2,2:6
