1Down.regulation of E.cadherin enhances prostate cancer chemoresistance via Notch signaling显示文摘Background:The chemoresistance of prostate cancer(PCa)is invariably associated with the aggressiveness and metastasis of this disease.New emerging evidence indicates that the epithelial-to-mesenchymal transition(EMT)may play pivotal roles in the development of chemoresistance and metastasis.As a hallmark of EMT,E-cadherin is suggested to be a key marker in the development of chemoresistance.However,the molecular mechanisms underlying PCa chemoresistance remain unclear.The current study aimed to explore the association between EMT and chemoresistance in PCa as well as whether changing the expression of E-cadherin would affect PCa chemoresistance.Methods:Parental PC3 and DU145 cells and their chemoresistant PC3-Tx R and DU145-Tx R cells were analyzed.PC3-Tx R and DU145-Tx R cells were transfected with E-cadherin-expressing lentivirus to overexpress E-cadherin;PC3 and DU145 cells were transfected with small interfering RNA to silence E-cadherin.Changes of EMT phenotype-related markers and signaling pathways were assessed by Western blotting and quantitative real-time polymerase chain reaction.Tumor cell migration,invasion,and colony formation were then evaluated by wound healing,transwell,and colony formation assays,respectively.The drug sensitivity was evaluated using MTS assay.Results:Chemoresistant PC3-Tx R and DU145-Tx R cells exhibited an invasive and metastatic phenotype that associated with EMT,including the down-regulation of E-cadherin and up-regulation of Vimentin,Snail,and N-cadherin,comparing with that of parental PC3 and DU145 cells.When E-cadherin was overexpressed in PC3-Tx R and DU145-Tx R cells,the expression of Vimentin and Claudin-1 was down-regulated,and tumor cell migration and invasion were inhibited.In particular,the sensitivity to paclitaxel was reactivated in E-cadherin-overexpressing PC3-Tx R and DU145-Tx R cells.When E-cadherin expression was silenced in parental PC3 and DU145 cells,the expression of Vimentin and Snail was up-regulated,and,particularly,the sensitivity to paclitaxel was decreased.Interestingly,Notch-1 expression was up-regulated in PC3-Tx R and DU145-Tx R cells,whereas the E-cadherin expression was down-regulated in these cells comparing with their parental cells.The use ofγ-secretase inhibitor,a Notch signaling pathway inhibitor,significantly increased the sensitivity of chemoresistant cells to paclitaxel.Conclusion:The down-regulation of E-cadherin enhances PCa chemoresistance via Notch signaling,and inhibiting the Notch signaling pathway may reverse PCa chemoresistance.Wenchu Wang Lihui Wang Atsushi Mizokami Junlin Shi Chunlin Zou Jinlu Dai Evan T. 2Microbial Activity Indices: Sensitive Soil Quality Indicators for Trace Metal Stress显示文摘Physicochemical properties, total and DTPA (diethylenetriaminepentaacetic acid)-extractable Cu, Zn, Pb and Cd contents, microbial biomass carbon (C) content and the organic C mineralization rate of the soils in a long-term trace metal-contaminated paddy region of Guangdong, China were determined to assess the sensitivity of microbial indices to moderately metal-contaminated paddy soils. The mean contents of total Cu, Zn, Pb and Cd were 251, 250, 171, and 2.4 mg kg-1 respectively. DTPA-extractablc metals were correlated positively and significantly with total metals, CEC, and organic C (except for DTPA-extractable Cd), while they were negatively and highly significantly correlated with pH, total Fe and Mn. Metal stress resulted in relatively low ratios of microbial biomass C to organic C and in remarkable inhibition of the microbial metabolic quotient and C mineralization rate, which eventually led to increases in soil organic C and C/N. Moreover, microbial respiratory activity showed a stronger correlation to DTPA-extractable metals than to total metal content. Likewise, in the acid paddy soils some 'linked' microbial activity indices, such as metabolic quotient and ratios of basal respiration to organic C, especially during initial incuba.tion, were found to be more sensitive indicators of soil trace metal contamination than microbial biomass C or basal respiration alone.LI Yong-Tao T. 4Immune mediators in the tumor microenvironment of prostate cancer显示文摘Prostate cancer tissue is composed of both cancer cells and host cells.The milieu of host components that compose the tumor is termed the tumor microenvironment(TME).Host cells can be those derived from the tissue in which the tumor originates(e.g.,fibroblasts and endothelial cells)or those recruited,through chemotactic or other factors,to the tumor(e.g.,circulating immune cells).Some immune cells are key players in the TME and represent a large proportion of non-tumor cells found within the tumor.Immune cells can have both anti-tumor and pro-tumor activity.In addition,crosstalk between prostate cancer cells and immune cells affects immune cell functions.In this review,we focus on immune cells and cytokines that contribute to tumor progression.We discuss T-regulatory and T helper17 cells and macrophages as key modulators in prostate cancer progression.In addition,we discuss the roles of interleukin-6 and receptor activator of nuclear factor kappa-B ligand in modulating prostate cancer progression.This review highlights the concept that immune cells and cytokines offer a potentially promising target for prostate cancer therapy.Jinlu Dai Yi Lu Hernan Roca Jill M. 7Selective Smoothed Finite Element Method显示文摘The paper examines three selective schemes for the smoothed finite element method (SFEM) which was formulated by incorporating a cell-wise strain smoothing operation into the standard compatible finite element method (FEM). These selective SFEM schemes were formulated based on three selective integration FEM schemes with similar properties found between the number of smoothing cells in the SFEM and the number of Gaussian integration points in the FEM. Both scheme 1 and scheme 2 are free of nearly incompressible locking, but scheme 2 is more general and gives better results than scheme 1. In addition, scheme 2 can be applied to anisotropic and nonlinear situations, while scheme 1 can only be applied to isotropic and linear situations. Scheme 3 is free of shear locking. This scheme can be applied to plate and shell problems. Results of the numerical study show that the selective SFEM schemes give more accurate results than the FEM schemes.T. T. Nauven G. R. Liu K. 8Phosphorylated mesoporous carbon as effective catalyst for the selective fructose dehydration to HMF显示文摘Phosphorylated mesoporous carbons (PMCs) were investigated as catalysts in the dehydration of fructose to hydroxymethylfurfural (HMF). The acidic PMCs show better selectivity to HMF compared to sulfonated carbon catalyst (SC) despite lower activity. The concentration of P-O groups on the PMC was correlated with the activity/selectivity of the catalysts; the higher the P-O concentration, the higher the activity. However, the higher the P-O content, the lower the selectivity to HMF. Indeed, a lower concentration of the P-O groups minimized the degradation of HMF to levulinic acid and the formation of by-products, such as humines. Stability tests showed that these systems deactivate due to the formation of humines and water insoluble by-products derived from the dehydration of fructose which blocked the catalytically active sites.A.
