1Antioxidative phytoceuticals to ameliorate pancreatitis in animal models: An answer from nature显示文摘Despite enthusiastic efforts directed at elucidating critical underlying mechanisms towards the identification of novel therapeutic targets for severe acute pancreatitis(SAP), the disease remains without a specific therapy to be executed within the first hours to days after onset of symptoms. Although earlier management for SAP should aim to either treat organ failure or reduce infectious complications, the current standard of care for the general management of AP in the first hours to days after onset of symptoms include intravenous fluid replacement, nutritional changes, and the use of analgesics with a close monitoring of vital signs. Furthermore, repeated evaluation of severity is very important, as the condition is particularly unstable in the early stages. In cases where biliary pancreatitis is accompanied by acute cholangitis or in cases where biliary stasis is suspected,an early endoscopic retrograde cholangiopancreatography is recommended.However,practice guidelines regarding the treatment of pancreatitis are suboptimal.In chronic pancreatitis,conservative management strategies include lifestyle modifications and dietary changes followed by analgesics and pancreatic enzyme supplementation.Recently,attention has been focused on phytoceuticals or antioxidants as agents that could surpass the limitations associated with currently available therapies.Because oxidative stress has been shown to play an important role in the pathogenesis of pancreatitis,antioxidants alone or combined with conventional therapy may improve oxidativestress-induced organ damage.Interest in phytoceuticals stems from their potential use as simple,accurate tools for pancreatitis prognostication that could replace older and more tedious methods.Therefore,the use of antioxidative nutrition or phytoceuticals may represent a new direction for clinical research in pancreatitis.In this review article,recent advances in the understanding of the pathogenesis of pancreatitis are discussed and the paradigm shift underway to develop phytoceuticals and antioxidants to treat it is introduced.Despite the promise of studies evaluating the effects of antioxidants/phytoceuticals in pancreatitis,translation to the clinic has thus far been disappointing.However,it is expected that continued research will provide solid evidence to justify the use of antioxidative phytoceuticals in the treatment of pancreatitis.Jong-Min Park Sooyeon Lee Mi Kyung Chung Sung-Hun Kwon Eun-Hee Kim Kwang Hyun Ko Chang Il Kwon Ki Baik Hahm 2014World Journal of Gastroenterology2014,20,44:5
2Inhibition of Cdk5 increases osteoblast differentiation and bone mass and improves fracture healing显示文摘Identification of regulators of osteoblastogenesis that can be pharmacologically targeted is a major goal in combating osteoporosis,a common disease of the elderly population. Here, unbiased kinome RNAi screening in primary murine osteoblasts identified cyclin-dependent kinase 5(Cdk5) as a suppressor of osteoblast differentiation in both murine and human preosteoblastic cells. Cdk5 knockdown by si RNA, genetic deletion using the Cre-lox P system, or inhibition with the small molecule roscovitine enhanced osteoblastogenesis in vitro. Roscovitine treatment significantly enhanced bone mass by increasing osteoblastogenesis and improved fracture healing in mice. Mechanistically, downregulation of Cdk5 expression increased Erk phosphorylation, resulting in enhanced osteoblast-specific gene expression. Notably, simultaneous Cdk5 and Erk depletion abrogated the osteoblastogenesis conferred by Cdk5 depletion alone, suggesting that Cdk5 regulates osteoblast differentiation through MAPK pathway modulation. 7Investigating seed mineral composition in Korean landrace maize(Zea mays L.) and its kernel texture specificity显示文摘Mineral malnutrition affects billions of people all over the world and biofortification of staple crops provides a potential way to alleviate dietary mineral deficiencies.For example,nutritional quality is an important breeding target for fresh waxy maize(Zea mays L.),which is widely consumed in Asian countries.Successful improvement of mineral composition will require comprehensive profiling of the mineral composition of maize varieties and an understanding of the capacity for maize grains to accumulate minerals.Here,using inductively coupled plasma absorption emission spectrometry,we quantified 12 minerals from the seeds of 47 maize varieties,including 25 Korean landraces.We also compared the mineral contents in varieties with different seed starch profiles:waxy maize(which contains 100%amylopectin),dent maize(roughly 75%amylopectin and 25%amylose),and flint maize(similar to dent maize).The amounts of potassium,phosphorus,and sulfur were correlated with seed texture,waxy maize having higher amounts of phosphorus and potassium than dent maize and lower amounts of sulfur than flint maize or dent maize.In addition,a positive relationship was detected between the amount of phosphorus and that of potassium,magnesium,and manganese.These results provide information on maize seed mineral composition and indicate that it could be affected by starch composition.Furthermore,the landraces that exhibit high mineral contents could be used as germplasm materials for breeding programs aimed at producing biofortified maize cultivars.Sooyeon Lim Gibum Yi 2019Journal of Integrative Agriculture2019,18,9:1

14Reactive oxygen species-induced activation of Yes-associated protein-1 through the c-Myc pathway is a therapeutic target in hepatocellular carcinoma显示文摘BACKGROUND The Hippo signaling pathway regulates organ size by regulating cell proliferation and apoptosis with terminal effectors including Yes-associated protein-1(YAP-1).Dysregulation in Hippo pathway has been proposed as one of the therapeutic targets in hepatocarcinogenesis.The levels of reactive oxygen species(ROS)increase during the progression from early to advanced hepatocellular carcinoma(HCC).AIM To study the activation of YAP-1 by ROS-induced damage in HCC and the involved signaling pathway.METHODS The expression of YAP-1 in HCC cells(Huh-7,HepG2,and SNU-761)was quantified using real-time polymerase chain reaction and immunoblotting.Human HCC cells were treated with H2O2,which is a major component of ROS in living organisms,and with either YAP-1 small interfering RNA(siRNA)or control siRNA.To investigate the role of YAP-1 in HCC cells under oxidative stress,MTS assays were performed.Immunoblotting was performed to evaluate the signaling pathway responsible for the activation of YAP-1.Eighty-eight surgically resected frozen HCC tissue samples and 88 nontumor liver tissue samples were used for gene expression analyses.RESULTS H2O2 treatment increased the mRNA and protein expression of YAP-1 in HCC cells(Huh-7,HepG2,and SNU-761).Suppression of YAP-1 using siRNA transfection resulted in a significant decrease in tumor proliferation during H2O2 treatment both in vitro and in vivo(both P<0.05).The oncogenic action of YAP-1 occurred via the activation of the c-Myc pathway,leading to the upregulation of components of the unfolded protein response(UPR),including 78-kDa glucoseregulated protein and activating transcription factor-6(ATF-6).The YAP-1 mRNA levels in human HCC tissues were upregulated by 2.6-fold compared with those in nontumor tissues(P<0.05)and were positively correlated with the ATF-6 Levels(Pearson’s coefficient=0.299;P<0.05).CONCLUSION This study shows a novel connection between YAP-1 and the UPR through the c-Myc pathway during oxidative stress in HCC.The ROS-induced activation of YAP-1 via the c-Myc pathway,which leads to the activation of the UPR pathway,might be a therapeutic target in HCC.Yuri Cho Min Ji Park Koeun Kim Sun Woong Kim Wonjin Kim Sooyeon Oh Joo Ho Lee 2020World Journal of Gastroenterology2020,26,42:1

19Loss of Notch signaling in skeletal stem cells enhances bone formation with aging显示文摘Skeletal stem and progenitor cells(SSPCs) perform bone maintenance and repair. With age, they produce fewer osteoblasts and more adipocytes leading to a loss of skeletal integrity. The molecular mechanisms that underlie this detrimental transformation are largely unknown. Single-cell RNA sequencing revealed that Notch signaling becomes elevated in SSPCs during aging. To examine the role of increased Notch activity, we deleted Nicastrin, an essential Notch pathway component, in SSPCs in vivo. Middle-aged conditional knockout mice displayed elevated SSPC osteo-lineage gene expression, increased trabecular bone mass, reduced bone marrow adiposity, and enhanced bone repair. Thus, Notch regulates SSPC cell fate decisions, and moderating Notch signaling ameliorates the skeletal aging phenotype, increasing bone mass even beyond that of young mice. 20Reduction-induced metal/oxide interfacial sites for selective CO_(2) hydrogenation显示文摘The interfacial structures of bimetallic-derived catalysts play an important role in promoting the activation of reactants such as CO_(2).In particular,both the physical property(e.g.,local bonding environment)and the electronic property(e.g.,oxidation state)can evolve from their native states under different environments,such as upon reduction and during the catalytic reaction.Hence,taking the CO_(2) hydrogenation reaction over Rh-based catalysts as a case study,the present work compares the interfacial structures in tuning the selectivity toward CH_(4) or CO.The combination of ex situ and in situ characterization reveals two representative interfacial structures:the Rh/CeO_(x) interface formed over Rh/CeO_(2) is active and selective to produce CH_(4)(~95%)by following a formate-mediated pathway;in comparison,the InO_(x)/Rh interface derived after reduction is active for CO_(2) activation and enables a redox mechanism for the exclusive formation of CO(~100%).This work provides insights into the environment-induced structural evolution at the metal−oxide interfaces,as well as the role of distinct interfacial active sites in tuning the selectivity of CO_(2) hydrogenation.Zhenhua Xie Sooyeon Hwang Jingguang G.Chen 2023SmartMat2023,4,4:0
